本発明の口腔内崩壊錠において使用される活性成分としては、医薬活性成分として疾患の治療や予防に供され、経口投与可能なものであれば特に限定されない。例えば、滋養強壮保健薬;解熱鎮痛消炎薬;抗精神病薬;催眠鎮静薬;鎮痙薬;中枢神経作用薬;脳代謝改善薬;脳循環改善薬;抗てんかん薬;交感神経興奮剤;健胃消化剤;抗潰瘍剤;消化管運動機能改善剤;制酸剤;鎮咳去痰剤;腸運動抑制薬;鎮吐剤;呼吸促進剤;気管支拡張剤;アレルギー用薬;抗ヒスタミン剤;強心剤;不整脈用剤;利尿剤;ACE阻害剤;Ca拮抗剤;AII拮抗薬;血管収縮剤;冠血管拡張剤;血管拡張薬;末梢血管拡張薬;高脂血症用剤;利胆剤;セフェム系抗生物質;経口抗菌薬;化学治療剤;スルフォニル尿素薬;αグルコシダーゼ阻害薬;インスリン抵抗性改善薬;速効性インスリン分泌促進剤;DPPIV阻害薬;糖尿病合併症治療薬;骨粗しょう症剤;抗リウマチ剤;骨格筋弛緩剤;アルカロイド系麻薬;サルファ剤;痛風治療剤;血液凝固阻止剤;抗悪性腫瘍剤等が挙げられる。
本発明における必須成分の一つであるマンニトールとしては、特に限定は無いが、D-マンニトールが好ましい。「日本薬局方」または「医薬品添加物規格」に記載されたものを通常に用いることができる。その結晶形は特に限定されず、α型、β型、δ型のいずれを用いても良いし、噴霧乾燥して得られる無晶形のものであっても良い。一方、特開平11-92403号公報に記載されたような、球形で密度の高いマンニトールを使用しても良い。
本発明における必須成分の一つである結晶セルロースは、口腔内崩壊錠に配合する際のその集合体のかさ密度が重要である。即ち、その集合体のかさ密度が、通常0.18g/cm3以下のものを使用することを特徴とする。好ましくは0.17g/cm3以下のものを使用し、さらに好ましくは0.15g/cm3以下のものがよい。
本発明においては、低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも2種の特定成分を含有することを特徴とする。上記特定の結晶セルロースとの組み合わせにおいて、所望の効果が得られるのは、この3種の特定成分のうち、2種以上を配合する場合である。言い換えれば、特定成分を配合しない、または1種のみを配合する場合には所望の効果が得られず(比較例1~5および比較例6~10参照)、2種または3種配合することによってはじめて本発明の目的を達成できることを見出したものである。
本発明で使用される低置換度ヒドロキシプロピルセルロースは、ヒドロキシプロポキシ基の置換割合に特に限定はなく、局方に適合するものであればよい。通常は7.0~12.9%である。その平均粒子径としては特に限定はないが、用いる低置換度ヒドロキシプロピルセルロースの平均粒子径が大きいと、口腔内で崩壊した後にザラツキを感じるため、服用感の観点からは、原料とする低置換度ヒドロキシプロピルセルロースの平均粒子径は、好ましくは10~200μm、より好ましくは10~150μm、さらに好ましくは10~100μmである。所望の粒子径とするため、必要に応じて適宜粉砕してもよい。粉砕方法としては、例えば気流粉砕機やハンマー式粉砕機による方法が挙げられる。低置換度ヒドロキシプロピルセルロースの配合量は、口腔内崩壊錠100重量%あたり通常1~20重量%であるが、好ましくは1~16重量%、より好ましくは1~10重量%、さらに好ましくは1~6重量%である。
本発明で使用されるコーンスターチは、その平均粒子径としては特に限定はないが、用いるコーンスターチの平均粒子径が大きいと、口腔内で崩壊した後にザラツキを感じるため、服用感の観点からは、原料とするコーンスターチの平均粒子径は、好ましくは10~200μm、より好ましくは10~100μm、さらに好ましくは10~50μmである。所望の粒子径とするため、必要に応じて適宜粉砕してもよい。粉砕方法としては、例えば気流粉砕機やハンマー式粉砕機による方法が挙げられる。コーンスターチの配合割合は、口腔内崩壊錠100重量%あたり通常1~20重量%、好ましくは、5~20重量%である。
本発明で使用されるカルメロースは特に限定はないが、通常、日本薬局方に適合しているものを用いる。その平均粒子径としては特に限定はないが、用いるカルメロースの平均粒子径が大きいと、口腔内で崩壊した後にザラツキを感じるため、服用感の観点からは、原料とするカルメロースの平均粒子径は、好ましくは10~200μm、より好ましくは10~150μm、さらに好ましくは10~100μmである。所望の粒子径とするため、必要に応じて適宜粉砕してもよい。粉砕方法としては、例えば気流粉砕機やハンマー式粉砕機による方法が挙げられる。カルメロースの配合量は、口腔内崩壊錠100重量%あたり通常1~20重量%であるが、好ましくは1~10重量%、より好ましくは1~5重量%、もっとも好ましくは1~3重量%である。
本発明においては、さらに混合性または含量均一性を向上させるため、造粒する際の結合剤として、水溶性高分子を加えてもよい。本発明で使用される水溶性高分子は、例えば、メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ヒプロメロースおよびポリビニルピロリドン等が挙げられる。好ましくは、メチルセルロースおよびヒドロキシプロピルセルロースが挙げられる。水溶性高分子はその結合力から崩壊性に悪影響を与えるため、通常の場合は添加されないか、添加するとしても配合割合はごく少量添加できるのみである。しかし、本発明の場合、水溶性高分子を口腔内崩壊錠100重量%あたり0.5~10重量%添加することが可能である。崩壊性の観点から、好ましくは0.5~8重量%、より好ましくは0.5~5重量%である。
本発明の口腔内崩壊錠においては、上記の成分以外にも、通常、その他の製剤化成分を加えて製剤化される。本発明で用いられることもある「その他の製剤化成分」としては、配合しても薬剤の初期および加湿後の硬度および崩壊時間に影響が無いか、または極めて少なく、製剤化するのに支障の無い成分であればいずれでもよい。例えば、賦形剤、崩壊剤、結合剤、甘味剤、矯味剤・矯臭剤、安定化剤、界面活性剤、流動化剤、帯電防止剤、コーティング剤、滑沢剤、着色剤、香料等がその例として挙げられる。「その他の製剤化成分」の配合量は、口腔内崩壊錠100重量%あたり0.01~25重量%であり、錠剤中に、これら製剤化成分を含むときは、その配合量に応じて上記の成分の量が減じられる配合比でもって構成される。
賦形剤としては、例えば、キシリトール、ソルビトール、エリスリトール、トレハロース、ブドウ糖、白糖、乳糖水和物、硫酸カルシウム、炭酸カルシウム等が挙げられる。
崩壊剤としては、例えば、クロスカルメロースナトリウム;カルボキシメチルスターチナトリウム;部分アルファー化デンプン、バレイショデンプン、コメデンプンなどのデンプン類;カルメロースナトリウム、カルメロースカルシウム、クロスポビドン等が挙げられる。崩壊剤の配合量は、錠剤の硬度維持に支障のない量であればよい。
結合剤としては、例えば、アラビアゴム、アラビアゴム末、部分アルファー化デンプン、ゼラチン、カンテン、デキストリン、プルラン、ポビドン、エチルセルロース、カルボキシメチルエチルセルロース、カルメロースナトリウム、ヒドロキシエチルセルロース、ヒドロキシエチルメチルセルロース等が挙げられる。結合剤の配合量は、錠剤の硬度を維持し、且つ口腔内での崩壊に支障のない量であればよい。
甘味剤としては、例えば、アスパルテーム、ネオテーム、アセスルファムカリウム、果糖、還元麦芽糖水アメ、グリチルリチン酸二カリウム、サッカリン、サッカリンナトリウム、スクラロース、ステビア、ソーマチン等が挙げられる。
矯味剤・矯臭剤としては、例えば、アスパラギン酸ナトリウム、アラニン、アルギニン、グリシン、グルタミン、グルタミン酸、グルタミン酸塩酸塩、グルタミン酸ナトリウム等の各種アミノ酸またはその塩、アジピン酸、アスコルビン酸、クエン酸、コハク酸、酒石酸、リンゴ酸等の有機酸、カンゾウ、クエン酸トリエチル、タウリン、タンニン酸等が挙げられる。
安定化剤としては、エデト酸ナトリウム、トコフェロール等が挙げられる。
界面活性剤としては、ラウリル硫酸ナトリウム、ポリソルベート、硬化油等が挙げられる。
流動化剤としては、例えば、含水二酸化ケイ素、軽質無水ケイ酸、重質無水ケイ酸、酸化チタン、メタケイ酸アルミン酸マグネシウム、ケイ酸カルシウム等が挙げられる。
帯電防止剤としては、例えば、含水二酸化ケイ素、軽質無水ケイ酸、タルク等が挙げられる。
コーティング剤としては、例えば、アクリル酸エチル・メタクリル酸メチルコポリマー分散液、アミノアルキルメタクリレートコポリマー、アラビアゴム末、エチルセルロース、オパドライ、カルナウバロウ、カルボキシビニルポリマー、カルボキシメチルエチルセルロース、カルメロースナトリウム、乾燥メタクリル酸コポリマー、ステアリルアルコール、セタノール、セラック、ゼラチン、ヒドロキシプロピルメチルセルロースアセテートサクシネート、プルラン、ポビドン、ヒドロキシエチルセルロース、ヒドロキシエチルメチルセルロース、ポリビニルアルコールコポリマー、ジメチルアミノエチルメタアクリレート・メチルメタアクリレートコポリマー、ヒドロキシプロピルメチルセルロースフタレート、フマル酸・ステアリン酸・ポリビニルアセタールジエチルアミノアセテート・ヒドロキシプロピルメチルセルロース混合物、ポリビニルアセタールジエチルアミノアセテート、メタクリル酸コポリマー、2-メチル-5-ビニルピリジンメチルアクリレート・メタクリル酸コポリマー等が挙げられる。
滑沢剤としては、例えば、ステアリン酸、ステアリン酸金属塩、フマル酸ステアリルナトリウム、ショ糖脂肪酸エステル、タルク、硬化油、マクロゴール等が挙げられる。ステアリン酸金属塩としては、ステアリン酸マグネシウム、ステアリン酸カルシウム、ステアリン酸アルミニウム等が挙げられるが、滑沢剤の中でステアリン酸またはステアリン酸金属塩、特に、ステアリン酸マグネシウムが好ましい。製剤化する前の滑沢剤の平均粒子径は0.5~50μmであり、好ましくは1~30μmである。滑沢剤の配合割合は、通常口腔内崩壊錠100重量%あたり0.01~3.0重量%であり、好ましくは0.1~2.0重量%である。さらに好ましくは0.3~1.2重量%である。
着色剤としては、例えば、食用赤色3号、食用黄色5号、食用青色1号などの食用色素、黄色三二酸化鉄、三二酸化鉄、褐色酸化鉄、黒酸化鉄、銅クロロフィル、銅クロロフィルナトリウム、リボフラビン、抹茶末等が挙げられる。
香料としては、例えば、オレンジエッセンス、オレンジ油、カラメル、カンフル、ケイヒ油、ペパーミントおよびスペアミント等のミント系油、ストロベリーエッセンス、チョコレートエッセンス、チェリーフレーバー、トウヒ油、パインオイル、ハッカ油、バニラフレーバー、ビターエッセンス、フルーツフレーバー、ヨーグルトフレーバー、ペパーミントおよびスペアミント等のミント系パウダーおよびミント系フレーバー、ミックスフレーバー、メントール、レモンパウダー、レモン油、ローズ油等が挙げられる。
本発明の口腔内崩壊錠は、製剤分野において慣用の方法を用いて製造することができる。上記(1)~(4)の各成分(および必要に応じて(5)の成分)、並びに必要に応じて上記のその他の製剤化成分を含む組成物を公知手段で製剤化することによって得ることができる。例えば、上記のような諸成分を均一に混合した後、その混合物を通常の打錠機等を用いた打錠等の圧縮成形を行うことによって錠剤とすることができる。また、上記成分を混合し(必要に応じて造粒を行って顆粒とし)、これに滑沢剤を加えて圧縮成形することによって錠剤とすることができる。また、活性成分(1)をあらかじめ、(2)~(4)の各成分の一部、およびその他製剤化成分とともに造粒やコーティング等公知手段で前処理してから、残りの製剤化成分とともに均一に混合した後、その混合物を公知手段で錠剤とすることもできる。
本発明は特に、活性成分として、不快な味を有する薬物である4-アミノ-5-クロロ-2-エトキシ-N-〔〔4-(4-フルオロベンジル)-2-モルホリニル〕メチル〕ベンズアミド(化合物A)またはその薬学的に許容される塩を用いる場合に適用されるのが好ましい。該化合物(またはその酸付加塩或いはこれらの水和物)は、選択的セロトニン4受容体アゴニストであり、良好な消化管運動促進作用を示す(米国特許第4,870,074号公報参照)。該化合物は、例えば、米国特許第4,870,074号公報に記載の方法またはこれに準じる方法によって製造することができる。この化合物Aは、ラセミ体であっても、一方の光学活性体であってもよい。また、該化合物はフリー体であってもよいし、その薬学的に許容される塩であってもよい。塩としては好ましくは酸付加塩がよい。たとえば有機酸の付加塩としては、ギ酸塩、酢酸塩、乳酸塩、アジピン酸塩、クエン酸塩、酒石酸塩、フマル酸塩、メタンスルホン酸塩、マレイン酸塩等が挙げられ、無機酸の付加塩としては、塩酸塩、硫酸塩、硝酸塩、リン酸塩等が例示できる。この中でも特にクエン酸塩が好ましい。さらに、該活性成分またはその薬学的に許容される塩は、溶媒和物であってもよく、水和物および非水和物であってもよい。好ましくはクエン酸塩の水和物であり、とりわけクエン酸塩・2水和物が好ましい。
化合物Aを用いた好ましい活性成分含有粒子は、(1a)化合物Aまたはその薬学的に許容される塩、(2a)マンニトール、並びに(5a)メチルセルロースおよびヒドロキシプロピルセルロースからなる群から選ばれる少なくとも1種の水溶性高分子を含む組成物を混合し、粒子化したものである。
上記活性成分含有粒子を本発明の口腔内崩壊錠に適用することによって、不快な味を遮蔽し、且つ優れた崩壊性および適度な硬度を有し、加湿条件下においても速やかな口腔内崩壊性を保持し、自動分包機で分包するために必要な一定以上の硬度を維持し得る口腔内崩壊錠を得ることができる。

得られる活性成分含有粒子、(2b)マンニトール、(3b)結晶セルロース、(4b)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも1種の特定成分、並びに(5b)メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ヒプロメロースおよびポリビニルピロリドンからなる群から選ばれる少なくとも1種の水溶性高分子を含む混合物を造粒し、
得られる造粒顆粒、(3c)結晶セルロース、(4c)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも1種の特定成分を含む混合物を成形することによって解決できることを見出したものである。ただし、該崩壊錠100重量%における配合割合が(1a)0.1~10重量%、(2a)および(2b)の合計が20~86重量%、(3b)および(3c)の合計が10~30重量%、(4b)および(4c)において、少なくとも2種の特定成分を含有し、その合計における各々の特定成分の配合割合が1~20重量%であり、且つ配合する特定成分の全体の合計が3~60重量%、並びに(5a)および(5b)の合計が0.5~10重量%である。
(1a)化合物Aまたはその薬学的に許容される塩、(2a)マンニトール、並びに(5a)メチルセルロースおよびヒドロキシプロピルセルロースからなる群から選ばれる少なくとも1種の水溶性高分子を混合し粒子化し、

かくして得られる本発明の口腔内崩壊錠は、製剤を服用するために水を摂取することなく、口腔内で速やかな崩壊性を示すものを意味する。具体的には、本発明の口腔内崩壊錠は、口腔内で主として唾液により、約40秒以内に崩壊する製剤を意味し、通常、40秒以内、好ましくは30秒以内で崩壊するものである。

本粒子は、表26に示す処方および表27に示す配合割合に従って製造した。すなわち、マンニトール(2a)、メチルセルロース(5a)を30メッシュ篩で篩過後、流動層造粒乾燥機(パウレック製、MP-01)にて、ヒドロキシプロピルセルロース(5a)が4重量%水溶液になるように調製した結合液に化合物A-b・2水和物(1a)を懸濁させた懸濁液を噴霧して造粒、乾燥した。できた顆粒を22メッシュ篩で篩過し、平均粒子径が約100μmの活性成分含有粒子を得た。
本錠剤は、表26に示す処方および表27に示す配合割合に従って製造した。すなわち、活性成分含有粒子、マンニトール(2b)、結晶セルロース(3b)、カルメロース(4.3b)、コーンスターチ(4.2b)、軽質無水ケイ酸、アスパルテームおよびステビアの中から処方に記載の成分を混合し、30メッシュ篩で篩過後、流動層造粒乾燥機(パウレック製、MP-01)でヒドロキシプロピルセルロース(5b)が4重量%水溶液になるように調製した結合液を噴霧して造粒、乾燥した。得られた顆粒を22メッシュ篩で篩過し、平均粒子径が約100μmの造粒顆粒を得た。該顆粒とステアリン酸マグネシウム以外の処方に記載の粉末成分を混合した。続いて、その混合物にステアリン酸マグネシウムを添加し、得られた混合物を打錠機(菊水製作所、VIRGO)で打錠し、1錠重量200mg、直径8mmの錠剤を得た。なお、打錠圧は、初期の絶対硬度が2.5~3.0N/mm2になるように調整しながら打錠した。そのときの打錠圧は、下記の試験例の表50に示した。
<活性成分を粒子化し活性成分含有粒子とした場合>
本粒子は、表28に示す処方および表29に示す配合割合に従って製造した。すなわち、マンニトール(2a)、メチルセルロース(5a)を30メッシュ篩で篩過後、流動層造粒乾燥機(パウレック製、MP-01)にて、ヒドロキシプロピルセルロース(5a)が4重量%水溶液になるように調製した結合液に化合物A-b・2水和物(1a)を懸濁させた懸濁液を噴霧して造粒、乾燥した。できた顆粒を22メッシュ篩で篩過し、平均粒子径が約100μmの活性成分含有粒子を得た。
本錠剤は、表28に示す処方および表29に示す配合割合に従って製造した。すなわち、活性成分含有粒子、マンニトール(2b)、結晶セルロース(3b)、カルメロース(4.3b)、コーンスターチ(4.2b)、各種低置換度ヒドロキシプロピルセルロース(4.1b)、軽質無水ケイ酸の中から処方に記載の成分を混合し、30メッシュ篩で篩過後、流動層造粒乾燥機(パウレック製、MP-01)でヒドロキシプロピルセルロース(5b)が4重量%水溶液になるように調製した結合液を噴霧して造粒、乾燥した。得られた顆粒を22メッシュ篩で篩過し、平均粒子径が約100μmの造粒顆粒を得た。該顆粒とステアリン酸マグネシウム以外の処方に記載の粉末成分を混合した。続いて、その混合物にステアリン酸マグネシウムを添加し、得られた混合物を打錠機(菊水製作所、VIRGO)で打錠し、1錠重量200mg、直径8mmの錠剤を得た。なお、打錠圧は、初期の絶対硬度が2.5~3.0N/mm2になるように調整しながら打錠した。そのときの打錠圧は、下記の試験例の表50に示した。

本顆粒は、表63に示す配合割合に従って製造した。すなわち、A-b・2水和物、乳糖造粒粉末、コーンスターチ、低置換度ヒドロキシプロピルセルロース31を流動層造粒乾燥機(パウレック製、MP-01)にて、ヒドロキシプロピルセルロースに22倍重量の精製水を添加して調製した結合液に、黄色三二酸化鉄を懸濁させた懸濁液を噴霧して造粒し、乾燥した。得られた顆粒を22メッシュ篩で篩過し、顆粒Bを得た。
本顆粒は、表64に示す配合割合に従って製造した。すなわち、マンニトールS、マンニトール1.059、低置換度ヒドロキシプロピルセルロース11を流動層造粒乾燥機(パウレック製、MP-01)に仕込み、精製水306.6gにD-マンニトール、無水クエン酸、黄色三二酸化鉄を懸濁させた懸濁液を噴霧しながら造粒、乾燥した。得られた顆粒を22メッシュ篩で篩過し、顆粒Cを得た。
本錠剤は、表65に示す配合割合に従って製造した。すなわち、顆粒B、顆粒C、アスパルテーム、ステアリン酸マグネシウムおよび結晶セルロースKG-1000、結晶セルロースKG-802、結晶セルロースPH-101の中から処方に記載した成分のうちステアリン酸マグネシウム以外の粉末成分を混合した。続いて、その混合物にステアリン酸マグネシウムを添加し、得られた混合物を打錠機(理研製、油圧式プレス機)で打錠し、1錠重量250mg、直径9mmの錠剤を得た。なお、打錠圧は、崩壊時間が25秒前後になるように調整しながら打錠した。そのときの打錠圧は、下記の試験例の表69に示した。
<本発明の処方とは異なり、特許文献5に開示された処方による場合>
本混合粉末は、表66に示す配合割合に従って製造した。すなわち、A-b・2水和物、乳糖造粒粉末、コーンスターチ、低置換度ヒドロキシプロピルセルロース31、ヒドロキシプロピルセルロース、黄色三二酸化鉄を混合し、22メッシュ篩で篩過し、混合粉末Dを得た。
本混合粉末は、表67に示す配合割合に従って製造した。すなわち、マンニトールS、マンニトール1.059、低置換度ヒドロキシプロピルセルロース11、D-マンニトール、無水クエン酸、黄色三二酸化鉄を混合し、22メッシュ篩で篩過し、混合粉末Eを得た。
本錠剤は、表68に示す配合割合に従って製造した。すなわち、混合粉末D、混合粉末E、アスパルテーム、ステアリン酸マグネシウムおよび結晶セルロースKG-1000、結晶セルロースKG-802、結晶セルロースPH-101の中から処方に記載した成分のうち、ステアリン酸マグネシウム以外の処方に記載の粉末成分を混合した。続いて、その混合物にステアリン酸マグネシウムを添加し、得られた混合物を打錠機(理研製、油圧式プレス機)で打錠し、1錠重量250mg、直径9mmの錠剤を得た。なお、打錠圧は、崩壊時間が25秒前後になるように調整しながら打錠した。そのときの打錠圧は、下記の試験例の表69に示した。
上記比較例66~71で製造された錠剤の初期および加湿後の崩壊時間および硬度を測定し、表69の結果を得た。

Claims (24)
- (1)活性成分、(2)マンニトール、(3)結晶セルロース並びに(4)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも2種の特定成分を含有する口腔内崩壊錠であって、該崩壊錠100重量%における各成分の配合割合が、(1)0.01~50重量%、(2)20~86重量%、(3)10~30重量%、および(4)各々の特定成分の配合割合が1~20重量%であり、且つ配合する特定成分の合計が3~60重量%であって、配合する結晶セルロース(3)の集合体のかさ密度が0.18g/cm3以下である、口腔内崩壊錠。
- 特定成分(4)が、カルメロースおよびコーンスターチを含む、請求項1に記載の口腔内崩壊錠。
- 該崩壊錠100重量%におけるマンニトール(2)の配合割合が、20~75重量%である、請求項1または2に記載の口腔内崩壊錠。
- 特定成分(4)において、該崩壊錠100重量%中、カルメロースを1~10重量%、コーンスターチを5~20重量%の配合割合で含む、請求項3に記載の口腔内崩壊錠。
- (4)の特定成分が、低置換度ヒドロキシプロピルセルロース、カルメロースおよびコーンスターチを含む、請求項1に記載の口腔内崩壊錠。
- 特定成分(4)において、該崩壊錠100重量%中、低置換度ヒドロキシプロピルセルロースを1~10重量%、カルメロースを1~10重量%およびコーンスターチを5~20重量%の配合割合で含む、請求項5に記載の口腔内崩壊錠。
- 活性成分(1)が、4-アミノ-5-クロロ-2-エトキシ-N-〔〔4-(4-フルオロベンジル)-2-モルホリニル〕メチル〕ベンズアミド(以下「化合物A」という)またはその薬学的に許容される塩である、請求項1~6のいずれか1項に記載の口腔内崩壊錠。
- さらに、(5)メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ヒプロメロースおよびポリビニルピロリドンからなる群から選ばれる少なくとも1種の水溶性高分子を含み、口腔内崩壊錠100重量%における該水溶性高分子(5)の配合割合の合計が0.5~10重量%である、請求項1~7のいずれか1項に記載の口腔内崩壊錠。

得られる活性成分含有粒子、(2b)マンニトール、(3b)結晶セルロース、(4b)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも2種の特定成分、並びに必要に応じて(5b)メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ヒプロメロースおよびポリビニルピロリドンからなる群から選ばれる少なくとも1種の水溶性高分子を混合し、成形する口腔内崩壊錠の製造方法であって、該崩壊錠100重量%における配合割合が、(1a)0.01~50重量%、(2a)および(2b)の合計が20~86重量%、(3b)10~30重量%、(4b)の特定成分の各配合割合が1~20重量%であり、且つ配合する特定成分の配合割合の合計が3~60重量%で、(5a)および(5b)を配合する場合の配合割合の合計が0.5~10重量%であって、配合する(3b)の結晶セルロースの集合体のかさ密度が0.18g/cm3以下である、口腔内崩壊錠の製造方法。 - (1)活性成分、(2b)マンニトール、(3b)結晶セルロース並びに(4b)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも1種の特定成分、並びに必要に応じて(5b)メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ヒプロメロースおよびポリビニルピロリドンからなる群から選ばれる少なくとも1種の水溶性高分子をを含む混合物を造粒し、
得られる造粒顆粒、(3c)結晶セルロースおよび(4c)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも1種の特定成分含む混合物を成形してなる口腔内崩壊錠の製造方法であって、該崩壊錠100重量%における各成分の配合割合が、(1)0.01~50重量%、(2b)20~86重量%、(3b)および(3c)の合計が10~30重量%、および(4b)および(4c)において少なくとも2種の特定成分を含有し、各特定成分の配合割合が1~20重量%であり、且つ配合する特定成分の配合割合の合計が3~60重量%、並びに配合される場合の(5b)の配合割合は0.5~10重量%であって、配合する(3b)および(3c)の結晶セルロースの集合体のかさ密度が0.18g/cm3以下であることを特徴とする、口腔内崩壊錠の製造方法。 - (1a)化合物Aまたはその薬学的に許容される塩、(2a)マンニトール、並びに(5a)メチルセルロースおよびヒドロキシプロピルセルロースからなる群から選ばれる少なくとも1種の水溶性高分子を混合し粒子化し、
得られる活性成分含有粒子、(2b)マンニトール、(3b)結晶セルロース、(4b)低置換度ヒドロキシプロピルセルロース、コーンスターチおよびカルメロースからなる群から選ばれる少なくとも2種の特定成分、並びに必要に応じて(5b)メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ヒプロメロースおよびポリビニルピロリドンからなる群から選ばれる少なくとも1種の水溶性高分子を混合し、成形する口腔内崩壊錠の製造方法であって、該崩壊錠100重量%における配合割合が、(1a)0.1~10重量%、(2a)および(2b)の合計が20~86重量%、(3b)10~30重量%、(4b)の特定成分の各配合割合が1~20重量%であり、且つ配合する特定成分の配合割合の合計が3~60重量%で、(5a)および(5b)を配合する場合の配合割合の合計が0.5~10重量%であって、配合する(3b)の結晶セルロースの集合体のかさ密度が0.18g/cm3以下である、口腔内崩壊錠の製造方法。 - (1a)化合物Aまたはその薬学的に許容される塩、(2a)マンニトール、並びに(5a)メチルセルロースおよびヒドロキシプロピルセルロースからなる群から選ばれる少なくとも1種の水溶性高分子を混合し粒子化し、
