この問題を解決するため、亜塩素酸塩にクエン酸などの有機酸からなるpH調整剤を加えたものを溶存二酸化塩素液剤に混ぜて3成分とし、二酸化塩素濃度を長期間、略一定に維持させるといった提案がなされた(特許文献2参照)。これにより、二酸化塩素の安定性が飛躍的に向上し、二酸化塩素濃度を長期間一定に保持できるようになった。
本発明者は、溶存二酸化塩素液剤の長期保存技術に関し、さらに効果的な技術を見出すべく研究開発を重ねて鋭意検討した結果、必須成分であると考えられていたpH調整剤を除く2成分(溶存二酸化塩素と亜塩素酸塩)からなる液剤組成物をガラス製容器あるいは琺瑯容器に密封することにより、長期間にわたって溶存二酸化塩素濃度を略一定に保持できることを見出し、本発明に至った。
また、本発明の二酸化塩素液剤組成物の特徴構成は、溶質が溶存二酸化塩素および亜塩素酸塩の2成分からなることを特徴とする。
さらに、本発明の二酸化塩素液剤組成物の特徴構成は、溶質が溶存二酸化塩素および亜塩素酸塩からなり、pH調整剤を実質的に含まないことを特徴とする。
上記二酸化塩素液剤組成物は、真菌性疾患、ウイルス性疾患および細菌性疾患の治療用に供するのが好ましい。また、溶存二酸化塩素の濃度は0.1~500ppmが好ましく、亜塩素酸塩が亜塩素酸アルカリ金属塩または亜塩素酸アルカリ土類金属塩であり、さらに亜塩素酸アルカリ金属塩が亜塩素酸ナトリウムまたは亜塩素酸カリウムであって、その濃度が500~50000ppmであるのが好ましい。
本発明の二酸化塩素液剤組成物は、溶質として二酸化塩素および亜塩素酸塩のみを備え、必須成分であると思われていたpH調整剤を含まなくても、該二酸化塩素液剤組成物における溶存二酸化塩素の濃度を長期間ほぼ一定に維持できる。pH調整剤を配合しなくても優れた保存安定性が得られると、当該二酸化塩素液剤組成物を臨床的に適用する場合(特に人体に適用する場合)において好都合である。
本発明の二酸化塩素液剤は、例えば次のように製造される。
即ち、二酸化塩素ガスを水中にバブリングして溶解することにより100~2900ppmの溶存二酸化塩素(二酸化塩素水溶液)(a)を調製する。一方、亜塩素酸塩を水に溶解して2000~180000ppmの亜塩素酸塩水溶液(b)を調製する。そして、溶存二酸化塩素水溶液(a)の5.0~990ml、好ましくは50~800ml、および、亜塩素酸塩水溶液(b)の5.0~990ml、好ましくは50~800mlを混合し、室温で十分に攪拌して、本発明の二酸化塩素液剤組成物とする。
亜塩素酸塩は、例えば、亜塩素酸アルカリ金属塩や亜塩素酸アルカリ土類金属塩が挙げられる。亜塩素酸アルカリ金属塩としては、例えば亜塩素酸ナトリウム、亜塩素酸カリウム、亜塩素酸リチウムが挙げられ、亜塩素酸アルカリ土類金属塩としては、亜塩素酸カルシウム、亜塩素酸マグネシウム、亜塩素酸バリウムが挙げられる。なかでも、入手が容易という理由のみならず、二酸化塩素ガス発生の持続性の点から、亜塩素酸ナトリウム、亜塩素酸カリウムが好ましく、亜塩素酸ナトリウムがさらに好ましい。
液剤組成物を保存する容器としては、容器本体のほとんど(表面積の90%以上、好ましくは95%以上の部分)がガラス製であり、充填物(液剤組成物)をしっかり密封できるものであればどのようなものでもよく、例えばアンプル、バイアル、スクリューキャップバイアル、ガラス製密封保存瓶などが使用可能である。
容器本体は透明のものであっても、遮光性能を持たせた褐色のものであってもよいが、液剤組成物の保存安定性に優れているという点で褐色の遮光性ガラスからなる方が好ましい。市販された公知のアンプル充填溶閉機(アンプルシーラー)やバイアル用充填・打栓機を用いて、液剤組成物の充填および溶閉を行い、必要に応じて窒素やアルゴン置換などの作業を行えばよい。
また、頻回に開栓して液剤を使用するような容器の場合においても、栓の部分の気密性は高いものが望ましく、非開栓時(非使用時)に二酸化塩素ガスの空気中への散逸を少なくするような構造が望ましい。
また、金属、陶磁器、ガラスなどの素地に釉薬を塗って焼き、ガラス質に変えて前記素地の表面にガラス層を設けてなる琺瑯(ほうろう)素材の容器を用いることも好適である。具体的には、鉄やアルミニウムなどの金属材料や陶磁器の表面にシリカ(二酸化ケイ素)を主成分とするガラス質の釉薬を高温で焼き付けたものが挙げられる。
尚、本発明の液剤組成物は、pH調整剤を積極的に排除したものであり、実質的にpH調整剤は含有されない。ここでいうpH調整剤としては、例えば有機酸またはその塩、無機酸またはその塩が挙げられる。有機酸またはその塩としては、蟻酸、酢酸、プロピオン酸、酪酸、乳酸、ピルビン酸、クエン酸、リンゴ酸、酒石酸、グルコン酸、グリコール酸、フマル酸、マロン酸、マレイン酸、シュウ酸、コハク酸、アクリル酸、クロトン酸、シュウ酸、グルタル酸、及びこれらの塩が挙げられる。また、無機酸としては、リン酸、ホウ酸、メタリン酸、ピロリン酸、スルファミン酸などが挙げられる。無機酸の塩としては、例えば、リン酸二水素ナトリウム、リン酸二水素ナトリウムとリン酸一水素ナトリウムの混合物などが挙げられる。
本発明の二酸化塩素液剤組成物を、例えば飲料水、食品の処理水、プール用水等に添加してそれらを殺菌消臭処理することができる。具体的には、上述したように調製した二酸化塩素液剤を10倍程度に水で薄めた水溶液に野菜、食器、布巾等を浸漬して殺菌消毒処理することができ、5倍程度に薄めてホテル、レストラン、外食産業、学校、家庭等のキッチンの設備、家庭の室内、便器、あるいは、自動車内にスプレーすることによりこれらを殺菌消臭処理することができる。
(1)二酸化塩素液剤組成物をしみ込ませた不織布や編織物を合成樹脂あるいは金属箔を用いて包装し、当該包装体を携帯して用事開封しておしぼり(手拭き簡易タオル)にしたり、レストランで客に提供する。
(2)二酸化塩素が保有する抗ウイルス作用、抗菌作用および抗真菌作用を利用して、臨床的に感染症治療に応用することができる。治療に際しては、例えば皮膚や皮膚粘膜疾患の場合は、0.1~500ppm好ましくは10~250ppmの二酸化塩素、500~50000ppm好ましくは700~20000ppmの亜塩素酸ナトリウムを含む本発明の液剤組成物をガーゼや綿花あるいは不織布にしみ込ませ、これを1日1~3回、1回につき少なくとも5分間、好ましくは10分間以上、患部に当てがったり患部に挿入する。点眼や点鼻する場合は、1日1~6回程度、1回につき濃度0.1~500ppm、好ましくは10~250ppmの二酸化塩素を含む本発明の液剤組成物を使用すればよい。
(3)従来公知の装置を使用して、本発明の二酸化塩素液剤組成物を室内に空中散布することにより、インフルエンザウイルスなど、後述するウイルス性疾患、細菌性疾患ならびに真菌性疾患を予防する。
(4)二酸化塩素液剤組成物をマスクや眼帯に直接しみ込ませ、あるいは二酸化塩素液剤組成物をしみ込ませたガーゼや不織布をマスクや眼帯に装着し、これにより後述するウイルス性疾患、細菌性疾患ならびに真菌性疾患を予防する。
ウイルス性疾患の原因病原体としては、例えばインフルエンザウイルス(A型、B型、C型)、鳥インフルエンザウイルス、ノロウイルス(ネコカリシウイルス)、ヒトパピローマウイルス(子宮頸部炎、子宮頸部悪性腺腫、尖圭コンジローマの病原体であるヒト乳頭種ウイルス、HPV)、コクサッキーウイルス(手足口病、無菌性髄膜炎、夏かぜ、熱性疾患、麻痺や気道性疾患の病原体)、エイズウイルス(HIV)、B型肝炎ウイルス、イヌパルボウイルス、ロタウイルス、HHV-1(単純ヘルペスウイルス1型(HSV-1))、HHV-2(単純ヘルペスウイルス2型(HSV-2))、HHV-3(水痘・帯状疱疹ウイルス(VZV))、HHV-5(サイトメガロウイルス(CMV))、眼科領域のウイルス(咽頭結膜熱(プール熱)や流行性角結膜炎(はやり目):アデノウイルス、急性出血性結膜炎:エンテロウイルス)など、幅広く挙げることができる。
細菌性疾患の原因菌としては、例えば黄色ブドウ球菌、緑膿菌、連鎖球菌、淋菌、梅毒菌などが挙げられ、これら細菌によって引き起こされる次のような疾患の治療用として使用することができる。すなわち、尋常性ざ瘡(ニキビ)、伝染性膿痂疹(とびひ)、褥瘡(とこずれ)、麦粒腫(ものもらい)、眼瞼縁炎、歯槽膿漏などの歯周病、蜂窩織炎(ほうかしきえん)、毛包炎、ブドウ球菌性熱傷様皮膚症候群などの皮膚疾患が挙げられ、外傷、熱傷および手術創などの二次感染の予防にも使用することができる。
真菌性疾患の原因菌としては、例えば白癬菌、マラセチア菌、カンジダなどが挙げられ、これら真菌によって引き起こされる次のような疾患の治療用として使用することができる。すなわち、足部白癬、体部白癬、股部白癬、爪白癬、その他の白癬菌感染症、皮膚カンジダ症、外陰カンジダ症のようなカンジダ症、指間びらん症、乳児寄生菌性紅斑、爪囲炎、でん風、脂漏性皮膚炎などの皮膚疾患が挙げられる。
本発明の液剤組成物が使用できる身体の部位としては、例えば頭皮、顔、目(眼瞼、結膜または角膜)、耳(外耳、外耳道など)、鼻(鼻孔、鼻粘膜など)、口唇、口腔内、咽頭、喉頭、声門、気道粘膜、食道、胃、十二指腸、小腸、結腸、舌、歯肉(歯茎)、首、胴体部分や手足(手指・足指の間隙を含む)、陰茎、包皮陰部、外陰、膣、子宮頚部、子宮内膜、肛門、直腸、爪などが挙げられ、粘膜にも好適に使用可能である。また、人体のみならず、犬や猫などのペットや、家畜(ウシ、ブタ、ニワトリ、ヒツジなど)の皮膚や皮膚粘膜にも使用することができる。
次のようにして二酸化塩素液剤組成物を調製した。
即ち、9%HClおよび25%亜塩素酸ナトリウムにて二酸化塩素ガスを発生させ、この二酸化塩素ガスを精製水に通気して二酸化塩素ガスを溶存させ、1200ppmの溶存二酸化塩素水溶液を調製した。
この1200ppmの溶存二酸化塩素水溶液750gおよび25%亜塩素酸ナトリウム水溶液20gを精製水と混合して全量を5リットルとし、二酸化塩素濃度が150ppm、亜塩素酸ナトリウム濃度が1000ppmの二酸化塩素液剤組成物を調製した。
調製した二酸化塩素液剤組成物20mlを市販のガラス製の褐色(遮光性)アンプル(容量20ml)に封入し、従来公知の方法に従ってアンプル充填溶閉機(アンプルシーラー)で密封して液剤組成物封入体とした(n=4)。このアンプル(検体)を60℃下にて20日間の苛酷試験を実施し、5日後、10日後、20日後における二酸化塩素の残存率(%)を紫外線可視分光光度計を用いて測定した。結果を表1および図1に記載する。
実施例1で調製した二酸化塩素液剤組成物を褐色(遮光性)のガラス密封ボトルと褐色(遮光性)の合成樹脂製(PET)密封ボトルにそれぞれ封入した。なお、ガラス密封ボトルと合成樹脂製(PET)密封ボトルの容積や容器本体の厚みはどちらも同一とし、この容器に対して上記の二酸化塩素液剤組成物をそれぞれ同量ずつ充填し、のち密封して液剤組成物封入体とした。密封後、封入した液剤組成物(液剤組成物封入体)における溶存二酸化塩素濃度の経時的な変化を紫外線可視分光光度計を用いて測定した。結果を図2に示す。
実施例1で調製した二酸化塩素液剤組成物20mlを市販の褐色(遮光性)ガラス密封ボトルに封入し、従来公知の方法に従ってアンプル充填溶閉機(アンプルシーラー)で密封して液剤組成物封入体とした。このアンプルを3ヶ月間冷所保存したあと、患者と医師の協力の下で臨床試験に供した。なお、試験には、使用時にアンプルを開封して取り出した前記二酸化塩素液剤組成物を、筒状に丸めた20cm×20cmのガーゼにしみ込ませて試験に供した(二酸化塩素液剤組成物をしみ込ませた後はガーゼは絞らない)。以下詳述する。
52歳の女性が、外陰部が痒くなり、おりものが多くなってから2日経過したのち来院した。治療前の所見としては、外陰部が充血し、大陰唇・小陰唇ともに塊状の分泌液(おりもの)が付着していた。また、膣粘膜も充血しており、塊状の分泌液(おりもの)が見られた。主治医は真菌性膣炎(カンジダ膣炎)と診断し、治療すべく患者に、上記した筒状に丸めたガーゼ(二酸化塩素液剤組成物を含ませたガーゼ)を膣内に挿入し、15分後に取り出し、これを毎日続けるように指導した。
その結果、3日目より外陰部の痒みがなくなり、14日目には外陰部の充血が消え始め、21日目には外陰部、膣粘膜とも、充血が完全に消え、塊状の分泌液もなくなった。
来院した45歳の女性(腰痛あり)を診察した結果、おりものが多く、子宮頸部のびらんが肥大し(子宮頸部びらんII度)、また充血や傷が認められた。主治医は慢性子宮頸管炎と診断し、治療すべく患者に、上記した筒状に丸めたガーゼ(二酸化塩素液剤組成物を含ませたガーゼ)を膣内に挿入し、15分後に取り出し、これを毎日続けるように指導した。
その結果、7日目より子宮口が癒合し、比較的軽症になっていることが確認された。7日目におけるヒトパピローマウイルス(HPV)測定を従来公知の方法により行った結果、256.28pg/mLであった。同様の治療を続け、治療開始から14日目には、おりものが顕著に減少し、腰痛も改善された。また、21日目には、おりものや腰痛がさらに改善し、子宮頸部の充血もびらんも軽くなっていることが確認できた。21日目におけるHPV測定値は183.56pg/mLまで下がっていた。
長年にわたり膿によるおりものが多い22歳の女性が来院し、診察した結果、主治医は子宮頸部びらん、及びHPV(ヒトパピローマウイルス)感染症と診断した。主治医は治療すべく患者に、上記した筒状に丸めたガーゼ(二酸化塩素液剤組成物を含ませたガーゼ)を膣内に挿入し、15分後に取り出し、これを毎日続けるように指導した。
その結果、治療前はHPV値が1176.42pg/mLであったのが、7日目には、357.21pg/mLと大幅に減少していた。また、おりものが顕著に減少していた。
おりものが多いことから来院した49歳の女性を診察した結果、おりものは膿性であり、おりもの中に細菌が多く存在していることがわかった。また外陰部や膣粘膜に充血(炎症)が見られ、主治医は細菌性膣炎(BV)であると診断した。主治医は治療すべく患者に、上記した筒状に丸めたガーゼ(二酸化塩素液剤組成物を含ませたガーゼ)を膣内に挿入し、15分後に取り出し、これを毎日続けるように指導した。
その結果、7日目には外陰部や膣粘膜の充血(炎症)が消えており、またおりものも減少していた。
Claims (13)
- 溶質が溶存二酸化塩素および亜塩素酸塩からなる二酸化塩素液剤組成物。
- 溶質が溶存二酸化塩素および亜塩素酸塩の2成分からなることを特徴とする二酸化塩素液剤組成物。
- 溶質が溶存二酸化塩素および亜塩素酸塩からなり、pH調整剤を実質的に含まないことを特徴とする二酸化塩素液剤組成物。
- 請求項1~3の何れか1項に記載の二酸化塩素液剤組成物が、ガラス製容器あるいは琺瑯容器に密封されてなることを特徴とする液剤組成物封入体。
- 請求項1~3の何れか1項に記載の二酸化塩素液剤組成物を、ガラス製容器あるいは琺瑯容器に密封することを特徴とする二酸化塩素液剤組成物の保存方法。
- 請求項1~3の何れか1項に記載の二酸化塩素液剤組成物を、ガラス製容器あるいは琺瑯容器に密封することを特徴とする二酸化塩素液剤組成物における溶存二酸化塩素の濃度維持方法。
- 真菌性疾患の治療用であることを特徴とする請求項1~3の何れか1項に記載の二酸化塩素液剤組成物。
- ウイルス性疾患の治療用であることを特徴とする請求項1~3の何れか1項に記載の二酸化塩素液剤組成物。
- 細菌性疾患の治療用であることを特徴とする請求項1~3の何れか1項に記載の二酸化塩素液剤組成物。
- 前記溶存二酸化塩素の濃度が0.1~500ppmである請求項1~3の何れか1項に記載の二酸化塩素液剤組成物。
- 前記亜塩素酸塩が、亜塩素酸アルカリ金属塩または亜塩素酸アルカリ土類金属塩である請求項1~3の何れか1項に記載の二酸化塩素液剤組成物。
- 前記亜塩素酸アルカリ金属塩が亜塩素酸ナトリウムまたは亜塩素酸カリウムである請求項11に記載の二酸化塩素液剤組成物。
- 前記亜塩素酸塩の濃度が500~50000ppmである請求項1~3の何れか1項に記載の二酸化塩素液剤組成物。
