生産されるβ-カロテン、β-クリプトキサンチン、エキネノン、カンタキサンチン、3-ヒドロキシエキネノン、3’-ヒドロキシエキネノン、ゼアキサンチン、フェニコキサンチン、アドニキサンチンおよびアスタキサンチンを含むカロテノイド類において、カンタキサンチンが前記カロテノイド類の合計量の90重量%以上しめ、カンタキサンチンを選択的に生産するカロテノイド生産性パラコッカス属細菌、およびそれを用いて培養し、培養後の菌体または培養液からカロテノイド類を回収するカンタキサンチンの製造方法を用いる。

[0001] 本発明は新規微生物およびそれを用いたカロテノイド類、特にカンタキサンチンの 製造方法に関する。
背景技術
[0002] ァスタキサンチン、ゼアキサンチン、フエ-コキサンチン、カンタキサンチン、アド- キサンチン、 /3一力口テン等のカロテノイドは天然物由来の色素として食品、医薬品、 医薬部外品、化粧品等に添加され利用されている。あるいは、抗酸化機能が注目さ れ抗酸化物質としても広く利用されている。これらのカロテノイド類は、植物、動物、 微生物に広く分布し、天然において数百種類同定されてきた (例えば、非特許文献 1 参照)。
[0003] 機能性カロテノイドを合成する微生物として、横山らによってァグロバタテリゥム属( 後に、パラコッカス属に再分類された)に属する細菌が報告された (例えば、非特許 文献 2参照)。同細菌は機能性カロテノイドであるァスタキサンチンを高含量に合成す ることが特徴である。また、同細菌においてはァスタキサンチンの生合成経路の解明 が行われ、遺伝子レベルまでの詳細な機構が報告された (例えば、非特許文献 3参 照)。報告された生合成経路によると、カロテノイドは基本代謝物であるメバロン酸を 出発物質としてイソプレノイド生合成系の代謝合成経路を通り、生じた炭素数 15のフ アルネシルピロリン酸 (FPP)、さらに、生じたフアルネシルピロリン酸に炭素数 5のイソ ペンテニル 2リン酸 (IPP)と縮合することにより、炭素数 20のゲラ -ルゲラニル 2リン酸 (GGPP)が合成される。次いで、 2分子のゲラ -ルゲラニル 2リン酸が縮合して、フイト ェン(phytoene)が合成される。フイトェンは一連の不飽和反応により、リコペン(lyc opene)に変換され、さらに、このリコペンは環化反応により j8—力口テン(carotene) に合成される。次いで、 j8—力口テンがケト化反応により酸ィ匕されるとカンタキサンチ ン(canthaxanthin)となり、 13—カロテンが水酸化されるとゼアキサンチン(zeaxant hin)となる。さらに、ケト化、水酸化による酸化反応が進み、ァスタキサンチン (astax
anthin)が合成される。以上のカロテノイド生合成経路を図 1に図示する。
[0004] 一方で一部の微生物においては前述の j8—力口テンをケト化する酵素をコードす る遺伝子を有せず、ゼアキサンチンのみを選択的に合成することが報告されている( 例えば、非特許文献 4参照)。カンタキサンチンについても同様に考えると、 j8—力口 テン水酸ィ匕酵素が欠損等している微生物が存在すればカンタキサンチンを選択的に 合成することが可能であるが、カンタキサンチンを選択的に合成する新規微生物分 離は報告されていない。
[0005] カンタキサンチンは機能性カロテノイドとして養殖魚'鶏卵等の色揚剤として有用で あるが、カロテノイド抽出物から精製し利用されているに過ぎない。カロテノイド抽出 物には、カンタキサンチンと化学的物性が類似したカロテノイド類が多く含まれ、さら に、抽出'精製を困難にしている(例えば、特許文献 1および 2参照)。そこで、カンタ キサンチンを高含量に蓄積する微生物等の含有物が望まれていた。

[0006] 本発明は、カロテノイド類であるカンタキサンチンを簡便に抽出、精製することがで きるカンタキサンチン選択合成細菌を提供することを目的とする。また、本発明は培 養法によるカンタキサンチンの製造方法を提供することも目的とする。
課題を解決するための手段
[0007] 本発明者らは、上記課題を達成すべく鋭意検討した結果、カロテノイド類を生産す るパラコッカス属細菌に変異を導入することにより、カンタキサンチンを選択的に合成 することを見出し本発明に至った。すなわち、本発明は、カロテノイド生産パラコッカ ス属細菌の育種により得られることを特徴とするカンタキサンチンを選択的に合成す るパラコッカス属細菌である。また、本発明は、そのような細菌を培養し、菌体または 培養液力 カンタキサンチンを回収することを特徴とするカンタキサンチンの製造方 法である。すなわち、本発明は以下に関するものである。
[0008] (1)カンタキサンチンを選択的に生産するカロテノイド生産性パラコッカス属細菌。
[0009] (2)生産される β一力口テン、 13 クリプトキサンチン、ェキネノン、カンタキサンチン 、 3—ヒドロキシェキネノン、 3,ーヒドロキシェキネノン、ゼアキサンチン、フエニコキサ ンチン、アドニキサンチンおよびァスタキサンチンを含むカロテノイド類において、力 ンタキサンチンが前記カロテノイド類の合計量の 90重量%以上しめる(1)の細菌。

発明を実施するための最良の形態
[0019] 以下、本発明を詳細に説明する。
[0020] 本発明に使用する微生物としては、パラコッカス(Paracoccus)属に属し、カンタキ サンチン合成能を有する微生物であれば野生株でも変異株でもよぐ通常の変異処 理により変異させた変異株でも、自然突然変異株でも、何れも用いることができる。本 発明に用いる野生株としては Paracoccus sp. N81106株(特許生物寄託センター 受託番号: FERM P— 14023)を挙げることができる。本発明においてはカロテノィ
ド類を合成する野生株あるいは変異株に変異をせしめてカンタキサンチン合成を選 択的に行う微生物がさらに好ましい。
[0021] 本発明においては、ァスタキサンチン合成能を有する微生物への変異導入により カンタキサンチン合成能を付与することができる。図 1に図示した通り、ァスタキサン チン合成能を有する微生物においては、 β一力口テンを水酸ィ匕およびケト化し最終 生産物であるァスタキサンチンを合成する。すなわち、カンタキサンチンを選択的に 合成するためには β一力口テン水酸ィ匕酵素をコードする遺伝子である crtZに変異を 導入し、水酸ィ匕酵素の活性を低下させればよい。あるいは j8—カロテン水酸ィ匕酵素 をコードする遺伝子である crtZに変異を導入し、該遺伝子が機能しないように破壊等 すればよい。
[0022] 本発明において変異を導入する細菌としては、カロテノイド類を高含量に細胞内に 蓄積しても、高 、増殖能を有するカロテノイド代謝アナログ耐性を有する細菌がさら に好ましい。カロテノイド代謝アナログとしては、当業者としては周知の a—ョノン (io none)または |8—ョノン (ionone)を例示することができる力 カロテノイド類の蓄積に よる細胞内調節機構の調節を解除する物質であればこれらの物質に限定されない。 本明細書において「耐性を有する」とは、親株が生育阻害を受ける濃度の薬剤存在 下でも生育しうることをいう。
[0023] 変異処理とは、変異原物質や紫外線で細胞を処理することによって変異を加速さ せる当業者には周知な方法である。変異原物質を例示すると N—メチル—N'—-ト ロー N— -トロソグァ-ジン、メタンスルホン酸ェチル等の化合物を挙げることができ る。
[0024] また、本発明においては、 β—カロテン水酸化酵素遺伝子を破壊、欠損してもよい 。破壊、欠損する方法としては、当業者としては周知の遺伝子工学的手法の一つで ある相同組換え法等により、ターゲットとなる遺伝子に適当な DNAを挿入する方法な どを挙げることができるがこれらに限定されない。
[0025] 本発明にお ヽては、変異処理後、変異株を分離し、所望の機能を評価する育種を 繰返すことが好ましい。育種方法の一例をあげると、予め培養して得たパラコッカス属 細菌 TSTT052株(特許生物寄託センター受託番号 FERM Ρ— 20690)の菌体を
これらの変異原物質の水溶液に懸濁して一定時間放置した後に遠心分離などの方 法で菌体を回収して変異原物質を除去する。その後平板培地上で培養し、優良菌 株のコロニーを選択する。コロニーの選択は色調の濃いコロニーを選択、分離後液 体培養を行ない、次いで、菌体力ゝらカロテノイドを抽出してカロテノイド生成量や組成 を HPLCなどで分析し、生産性の向上した菌株を絞り込むことにより優良な菌株を得 ることがでさる。
[0026] 本発明ではパラコッカス(Paracoccus)属に属し、カンタキサンチン合成能を有す る微生物であれば野生株でも変異株でもよぐ例を挙げれば本願発明者らによって 榭立されたパラコッカス(Paracoccus) sp. TSA0538株がある(受託番号 FERM P— 20707)。

[0028] 本発明において、選択的に合成できるとは、カロテノイド類を合成する微生物にお いてカンタキサンチンが分離、精製することが容易となる程度にカンタキサンチンが 合成されればよぐ好適には、生産される β一力口テン、 j8—クリプトキサンチン、ェキ ネノン、カンタキサンチン、 3—ヒドロキシェキネノン、 3,ーヒドロキシェキネノン、ゼァ キサンチン、フエニコキサンチン、アドニキサンチンおよびァスタキサンチンを含む力 ロテノイド類にぉ 、て、カンタキサンチンがカロテノイド類の合計量の 90重量%以上 しめるものである。なお、カロテノイド類とは、 β一力口テン、 j8—クリプトキサンチン、 ェキネノン、カンタキサンチン、 3—ヒドロキシェキネノン、 3 '—ヒドロキシェキネノン、 ゼアキサンチン、フエニコキサンチン、アドニキサンチン、ァスタキサンチン等であり、 さらに、その微生物内に蓄積されるその他のカロテノイド類を含む。
[0029] 本発明において、優良菌株は適当な菌株を評価し決定することができる。固体培 地を利用した場合には、任意のコロニーをピックアップし、液体培養を行い、増殖能、 カロテノイド生産能を評価することにより評価可能である。
[0030] 本発明の方法で使用された微生物は従来方法として知られている栄養培地中で培 養することができる。なお、本発明に用いる培地としては、細菌が増殖しカロテノイド 類を生産し得るものであればいずれを使用してもよぐ炭素源には廃糖蜜、ダルコ一 ス、フルクトース、マルトース、ショ糖、デンプン、乳糖、グリセロール、酢酸など力 窒 素源にはコーンスティープリカ一、ペプトン、酵母エキス、肉エキス、大豆粕等の天然 成分や、酢酸アンモ-ゥム、塩化アンモ-ゥム、硫酸アンモ-ゥム等のアンモ-ゥム 塩等やグルタミン酸、ァスパラギン酸、グリシン等のアミノ酸類力 無機塩にはリン酸 1 ナトリウム、リン酸 2ナトリウム、リン酸 1カリウム、リン酸 2カリウム等のリン酸塩や塩ィ匕ナ トリウムなどが、金属イオンには塩ィ匕マグネシウム、硫酸マグネシウム、硫酸第 1鉄、硫 酸第 2鉄、塩化第 1鉄、塩化第 2鉄、クェン酸鉄、硫酸アンモニゥム鉄、塩化カルシゥ ム · 2水和物、硫酸カルシウム、硫酸亜鉛、塩化亜鉛、硫酸銅、塩化銅、硫酸マンガ ン、塩化マンガンなど力 ビタミン類として酵母エキスやピオチン、ニコチン酸、チアミ
ン、リボフラビン、イノシトール、ピリドキシン等が使用できる。
[0031] 培養工程で使用される菌株は、公知方法に従って、画線培養プレートから発酵容 器に移すことができる。好適な方法は、寒天平板培地、斜面寒天培地およびフラスコ 培養液を用いた方法である。
[0032] 本発明のカンタキサンチン等のカロテノイドを製造せしめる条件での新規微生物の 培養の条件については、いずれの一般方法を用いて実施してもよい。本方法の好適 な実施態様としては、培養は培養液中で行うことが好ましい。この液内培養に関して は通常の液内培養で用いられる条件を使用してもよい。好適には、培養温度を 10〜 35°Cに、培地の pHを 6〜9の範囲に設定し、 20〜200時間発酵させるのが好ましい 。培養温度については培養初期、中期、後期に区別してそれぞれの段階で温度を 変えてもよい。本発明の栄養培地を使用した好適な条件は、培養温度 20〜27°C、 p Hが約 7. 0、培養時間が 50〜150時間である。
[0033] 培地の pHは 6. 5〜8. 0に調整される力 好ましくは約 7. 0である。 pHの調整には 水酸化ナトリウム、水酸ィ匕カリウム若しくはアンモニアの水溶液を使用する力 これら の物に限定されない。
[0034] 本発明におけるカロテノイド類の分析方法は、菌体または培養液力 安定に効率 良く回収されれば特に限定はなぐ例えば抽出溶媒としてはメタノール、エタノール、 イソプロピルアルコール、アセトン、メチルェチルケトン、メチルイソブチルケトン、ジク ロロメタン、クロロフオルム、ジメチルフオルムアミド、ジメチルスルフォキシド等がよい。 抽出されたカロテノイドの定量は、各種カロテノイドが分離され定量性に優れる高速 液体クロマトグラフィーにより行なうことが好ましい。
[0035] 培養後に、菌体又は培養液力もカロテノイド類および Zまたはカンタキサンチンを 回収するには、たとえば、遠心分離操作等により培養液から菌体を分離し、適当な有 機溶媒によりそれぞれから抽出すればよい。有機溶媒はメタノール、エタノール、イソ プロピルアルコール、アセトン、メチルェチルケトン、メチルイソブチルケトン、ジクロロ メタン、クロロフオルム、ジメチルフオルムアミド、ジメチルスルフォキシド等が挙げられ る。好適にはアセトンを例示できる。さらには、液体クロマトグラフィー等を利用して高 純度に分離、精製することも可能である。液体クロマトグラフィーの分離原理としては
イオン交換、疎水性相互作用、分子ふるい等を挙げることができる。好ましくは、逆相 クロマトグラフィー、順相クロマトグラフィーである。または、超臨界流体抽出によって 細胞から抽出する。
[0036] あるいは、培養終了後、菌体を培地から、例えば、遠心分離操作、デカンテーショ ンまたはろ過等の方法を用いて分離してもよい。得られた菌体は、使用し易い粘度に まで水を加えてスラリーとする。カンタキサンチン等のカロテノイド類の分解を防ぐた めには、このスラリー中適当な添加剤をカ卩えてもよい。添加剤としてはァスコルビン酸 等の酸ィ匕防止剤を例示することができるが、これらには限定されない。その後、調製 スラリーを例えばガラスビーズ、ジノレコ-ァビーズを用いた破碎機または高圧ホモジ ナイザーを使用して均一化し、後に使用するため乾燥しておく。好ましい乾燥法はス プレー乾燥法である。
[0037] この菌体を、例えば、このまま養殖魚等の飼料中へ添加してもよい。または、前述し たように極性有機溶媒等により抽出して使用してもよい。カンタキサンチン等のカロテ ノイドを抽出した後に残った、ほとんど色素を含まない細胞体は、家禽を飼育するうえ で理想的な蛋白質およびビタミン類の供給源として使用することができる。
実施例
[0038] 以下、実施例を用いてさらに詳細に説明するが、本発明はこれらに限定されるもの ではない。
[0039] (実施例 1)
ノ ラコッカス属細菌への変異導入および変異株のプレート評価
パラコッカス属細菌 TSTT052株(受託番号 FERM P— 20690)を表 1にお!/、て 表す液体培地 5mlに植菌し、 25°C、 150rpmで 1日間振とう培養を行った。この培養 液のうち lmlを 1. 5mlのエツペンドルフチューブに移し、 15, 000回転、 10分間の遠 心分離により菌体を回収した。この菌体を pH7. 0の 0. 1Mリン酸カリウム緩衝液 (緩 衝液 A) lmlに懸濁し、次いで 3mgZmlの N-メチル N, 一-トロ一 N -トロソグァ ジ-ジン(以下 NTGと略記する)水溶液 10 1を加え、 30〜60分間静置した。その 後、遠心分離して上清を除去し、緩衝液 Aに再懸濁する操作を 2回繰返して NTGを 除去した。さらに、 0. 5ml緩衝液 Aに菌体を懸濁し、表 1に示す培地 3mlに植菌して
4〜5時間培養した。得られた培養液を、表 1に示す組成の培地にさらに寒天を添カロ し固化させたプレートに適当量スプレッドし、 25°Cで 5日間培養を行った。培養 3日目 から、コロニーを目視により確認することができ、コロニーの赤色の濃淡を区別するこ とも可能となった。
(実施例 2)
ノ ラコカツカス属細菌変異株の培養
実施例 1で得られた菌株をランダムに選択し、表 1にお ヽて示す培養液 5mlに植菌 し、 25°C、 1日間培養後した。次いで、表 1に示した組成の培地 60mlを 100ml容バ ッフル付三角フラスコに入れ滅菌し、先に培養した培養液 2mlを植菌した。 25°Cで 7 日間振とう(lOOrpm)培養後、培養液を適宜抜き取り、濁度 (OD660 nm)および カロテノイドの定量を行った。カロテノイドの定量は以下の様に行なった。まず培養液 lmlを 1. 5ml容エツペンドルフチューブに入れ、 15, 000回転、 5分間遠心分離して 菌体ペレットを得た。この菌体に 20 μ 1の純水に懸濁し、次いで 200 μ 1のジメチルフ オルムアミドおよび 500 1のアセトンを加え振とうしてカロテノイドを抽出した。この抽 出液を 15, 000回転、 5分間遠心分離により残渣を除去後、 TSKgel— ODS80TM カラム (東ソ一社製)を用いた高速液体クロマトグラフィー(以下 HPLCと略記する)で 各種カロテノイドを定量した。なおカロテノイドの分離は A液として純水とメタノールの 5対 95の混合溶媒、 B液としてメタノールとテトラヒドロフランの 7対 3の混合溶媒を用 い、 lmlZminの流速で A液を 5分間カラムに通過させた後、同じ流速 A液力 B液 へ 5分間の直線濃度勾配を行ない、さらに B液を 5分間通過させることにより行なった 。カンタキサンチン濃度は 470nmの吸光度をモニターし、既知濃度のカンタキサン チン標準試薬で作成した検量線より濃度を算出した。評価した変異株のなかから、力 ンタキサンチン選択合成を確認することができた新規変異株として TSA0538株 (特 許生物寄託センター寄託した菌株であり、受託番号 FERM P 20707)を分離し た。カンタキサンチンであることは標準試薬の HPLC上の保持時間から確認した。図 2に TSA0538株の抽出カロテノイドの HPLCパターンと標準試薬のそれを図示した 。図 2の通り、 TSA0538株の抽出カロテノイドは標準試薬のカンタキサンチンのピー クと重なり、選択的にカンタキサンチンを合成するこが確認できた。
[0041] 同様にして培養した変異導入前の株である TSTT052株の培養 4日目のカロテノィ ド類の生成パターンを図 3に示した。図 2および図 3の通り、 TSA0538株は TSTTO 52株とは異なり、カンタキサンチンを選択的に合成し、カロテノイド類に占めるカンタ キサンチンの割合は 90%を超えた。選択合成割合は、 HPLC法 (例えば、非特許文 献 2参照)により、ァスタキサンチン、ゼアキサンチン、カンタキサンチン、 β一力ロテ ン、フエニコキサンチン、アドニキサンチン、ェキネノンの生産量を求め、それに対す るカンタキサンチンの生産量として算出した。結果を表 2に示す。
[0042] 図 4に TSA0538株のカンタキサンチン合成における時間変化を示したグラフを図 示した。図 4の通り、 TSA0538株は培養の経過と共に、カンタキサンチンを合成し、 培養 5曰目においてもカンタキサンチンを分解することなく細胞内に含有していた。 培養 4日目においては菌体あたりのカンタキサンチン含有量は約 1. 2重量%であつ た。重量%は乾燥菌体の重量を求めることにより算出した。

産業上の利用可能性
本発明の新規なカンタキサンチン選択合成細菌は、カロテノイド類であるカンタキサ ンチンを簡単に抽出、精製することができるので、食品または飼料として有用なカンタ キサンチンを工業規模で製造することができる。
Claims
[1] カンタキサンチンを選択的に生産するカロテノイド生産性パラコッカス属細菌。
[2] 生産される j8—力口テン、 13 クリプトキサンチン、ェキネノン、カンタキサンチン、 3 ーヒドロキシェキネノン、 3,ーヒドロキシェキネノン、ゼアキサンチン、フエニコキサン チン、アドニキサンチンおよびァスタキサンチンを含むカロテノイド類において、カンタ キサンチンが前記カロテノイド類の合計量の 90重量%以上しめることを特徴とする請 求項 1記載の細菌。
[3] ァスタキサンチン合成細菌の変異処理により β—カロテン水酸化酵素遺伝子に変 異が導入され、 β一力口テン水酸ィ匕酵素活性が低下することによりカンタキサンチン を選択的に合成する生産能を有する細菌。
[4] ァスタキサンチン合成細菌の変異処理により β—カロテン水酸化酵素遺伝子に変 異が導入され、 β一力口テン水酸ィ匕酵素遺伝子が欠損することによりカンタキサンチ ンを選択的に合成する生産能を有する細菌。
[5] 菌体にカンタキサンチンを 1重量% (乾重量換算)以上含むカロテノイド類合成細菌
[6] 細菌力 カロテノイド生産性パラコッカス属細菌 TSA0538株(受託番号 FERM Ρ
20707)である請求項 1記載のパラコッカス属細菌。
[7] 請求項 1ないし 6の何れか一項に記載の細菌を培養し、培養後の菌体または培養 液力 カロテノイド類を回収することを特徴とするカロテノイド類の製造方法。
