Herstellung eines Mittels gegen Hepatitis B-, Hl-, Paramyxo- und Orthomyxo-Viren
Beschreibung
Die Erfindung betrifft die Verwendung einer Substanz der allgemeinen Formel I zur Herstellung eines Mittels zur Behandlung von mit Hepatitis-B- Viren (HBV), HI-Viren, Viren der Paramyxogruppe oder/und der Orthomy- xogruppe assoziierten Erkrankungen, ein Verfahren zum Vermindern oder Inhibieren des Wachstums oder Vermehrung derartiger Viren, und ein Verfahren zum Nachweis derartiger Viren.
Hepatitis-B-Virus-Infektion führt zu akuter und in bestimmten Fällen chronischer Erkrankung der Leber. Weltweit sind nach Schätzungen der WHO etwa 350 Millionen Menschen persistent mit HBV infiziert. Chronisch Infizierte tragen ein hohes Risiko, an chronischer aktiver Hepatitis, Leberzirrhose oder hepatozellulärem Carcinom zu erkranken. Obwohl hochwirksame HBV Vakzine seit 1 982 verfügbar sind, können bereits infizierte Patienten damit nicht therapiert werden. Die verfügbare antivirale Therapie mit Interferon alpha ist mit Nebenwirkungen verbunden und nur bei einem Teil der Patienten einsetzbar und wirksam. Neuere Behandlungsmöglichkeiten mit Nucleosidanalogen wie 3'-Thiacytidin erscheinen erfolgversprechend und verträglich, aber die Langzeitwirksamkeit ist bisher unbekannt, vor allem in Anbetracht bereits beobachteter Resistenzbildung . Die Bereitstellung von Mitteln mit geringen Nebenwirkungen, die nicht zur Resistenzbildung führen, ist daher wünschenswert. Die zunehmende Bedeutung von Kombinationstherapien erfordert die Entwicklung zusätzlicher Mittel, die mit anderen Stadien des Infektionszyklus oder des Krankheitsver- laufs interferieren.
Eine HIV-Infektion führt in den meisten Fällen nach unterschiedlich langen Latenzphasen zur Ausprägung von AIDS (acquired immune deficiency syndrome), dessen Krankheitsbild und -verlauf neben den Auswirkungen der Primärinfektion durch den HI-Virus in erheblichem Maß von Sekundärinfektionen bestimmt wird. Die Kombinationstherapie mit mehreren Wirkstoffen (Nucleosidanaloge, Proteaseinhibitoren) spielt auch in der Therapie HlV-infizierter Patienten die entscheidende Rolle, allerdings ist damit nur eine Verzögerung und Erleichterung des Krankheitsverlaufs möglich, keine Heilung von AIDS. Ein Impfstoff steht derzeit nicht zur Verfügung.
Viren der Familie Orthomxyviridae umfassen Influenzaviren der Gruppen A, B und C. Influenza A und B Viren verursachen in infizierten Patienten ein Spektrum an klinischen Symptomen, das von asymptomatisch bis zu viraler Pneumonie mit tödlichem Ausgang reicht. Diese Viren können, bedingt durch Rekombination genetischer Information zwischen verschiedenen Stämmen, weltweite Epidemien mit exzessiver Mortalitätsrate auslösen. Eine Impfung von Risikogruppen mit der jährlich von der WHO empfohlenen Zusammensetzung inaktivierter Vakzine wird routinemäßig durchgeführt. Amantadine und Rimantadine stehen zur Prophylaxe und Therapie als antivirales Mittel zur Verfügung, sind allerdings nur gegen Influenza A wirksam, nicht gegen Influenza B, das in einem Viertel der Fälle für Hospitalisierung verantwortlich ist, oder gegen resistente Stämme. Zu den Paramyxoviren gehören Mumps-, Masern-, Hundestaupe-, Rinderpest- und Respiratory Syncytial Virus.
Daher besteht seit langem das Bedürfnis für wirksame Mittel gegen Hepatitis-B-Viren, HI-Viren, Viren der Paramyxo- und Orthomyxogruppe, die geringe Nebenwirkungen aufweisen und kostengünstig in geeigneten Darreichungsformen bereitgestellt werden können.
Eine Aufgabe der vorliegenden Erfindung ist es deshalb, Mittel zur Behandlung von Hepatitis-B-Viren, HI-Viren, Viren der Paramyxo- oder/und
der Orthomyxogruppe bereitzustellen, die eine gute Wirksamkeit gegen diese Viren haben sowie geringe Nebenwirkungen aufweisen.
Gelöst wird diese Aufgabe erfindungsgemäß durch die Verwendung einer Substanz der allgemeinen Formel I
worin (A) RT und R2 jeweils Wasserstoff sind, R3 ein Halogen, -CF3, Alkoxy mit
1 oder 2 Kohlenstoffatomen oder ein halogensubstituiertes Alkoxy mit 1 oder 2 Kohlenstoffatomen ist, und R4 ein Ci- -Alkyl oder C3-
C4-Cycloalkyl, insbesondere CH3 ist,
(B) RT Wasserstoff ist, R2 und R3, die gleich oder unterschiedlich sind, ein
Halogen oder -CF3 sind, und R4 ein C C4-Alkyl oder C3-C4-Cycloalkyl, insbesondere CH3 ist,
(C) R-, Wasserstoff ist, R2 Alkyl mit 1 oder 2 Kohlenstoffatomen ist, R3 ein Halogen, -CF3, Alkoxy mit 1 oder 2 Kohlenstoffatomen oder ein halogensubstituiertes Alkoxy mit 1 oder 2 Kohlenstoffatomen ist und
R4 ein C C4-Alkyl oder C3-C4-Cycloalkyl, insbesondere CH3 ist
(D) RT Wasserstoff ist, R2 und R3 zusammen 3',4'-Methylendioxy und R4 ein CrC4-Alkyl oder C3-C4-Cycloalkyl, insbesondere CH3 ist,
oder eines Derivats davon mit offenkettigen Isoxazolring oder eines physiologisch verträglichen Salzes der genannten Substanzen, beispielsweise eines Alkalimetall-, eines Ammonium- oder substituierten Ammoniumsalzes, insbesondere des Natriumsalzes oder des Salzes einer basischen Aminosäure wie Lysin, zur Herstellung eines Mittels zur Prävention oder
Behandlung von Infektionen mit Hepatitis-Viren, HI-Viren, Viren der Paramyxogruppe oder/und der Orthomyxogruppe.
Die Verbindungen der Formel I lassen sich unter Leflunomide einordnen. Leflunomid bezeichneteine Substanzgruppevon N-(4-Trifluormethylphenyl)- 5-methylisoxazol-4-carboxamiden, die ursprünglich als Herbizide in der Landwirtschaft zum Einsatz kamen. Für sie wurde mittlerweile auch eine entzündungshemmende und immunsuppressive Wirkung festgestellt, sodass ihr Einsatz, insbesondere bei Transplantationspatienten, zur Unterdrückung einer Transplantatabstoßung möglich erscheint. Eine antivirale Aktivität für Leflunomide oder seine Derivate wurde bisher jedoch nicht berichtet.
Es war überraschend, dass die erfindungsgemäß verwendeten Substanzen neben einer Herbizidaktivität und einer entzündungshemmenden sowie einer immunsuppressiven Wirkung auch eine gute antivirale Wirksamkeit insbesondere gegen Hepatitis-B-Viren, HI-Viren, Viren der Paramyxogruppe und der Orthomyxogruppe zeigen.
Die erfindungsgemäß erhaltenen Mittel mit Substanzen gemäß Formel I oder offenkettigen Derivaten oder Salzen davon als Wirkstoffe zeichnen sich durch ihre hohe Wirksamkeit aus, sodass bereits geringe Mengen für eine erfolgreiche Behandlung ausreichend sind. Andererseits sind die Substanzen gut verträglich, sodass die in dem erfindungsgemäß erhaltenen Mittel verwendete Dosis nicht durch nachteilige Nebenwirkungen beschränkt wird. Darüber hinaus sind die erfindungsgemäß verwendeten Substanzen unproblematisch in ihrer Herstellung und somit in großen Mengen leicht und kostengünstig verfügbar.
In der Substanz gemäß Formel I liegt der Isoxazolring geschlossen oder offen vor. Nach Einnahme eines erfindungsgemäß erhaltenen Mittels, das die Substanz gemäß Formel I mit geschlossenem Isoxazolring beinhaltet, kann diese im Körper metabolisiert und der Isoxazolring geöffnet werden,
wodurch eine physiologisch wirksamere Form der Substanz (Grundstruktur: N-(4-Trifluoromethylphenyl)-2-cyano-3-hydroxycrotonamid (A771 726)) entsteht.
Das erfindungsgemäß erhaltene Mittel kann weiterhin übliche Träger-, Hilfsoder/und Zusatzstoffe enthalten, die dem Fachmann bekannt sind und deshalb nicht explizit genannt werden müssen. Besonders bevorzugt sind Träger-, Hilfs- oder/und Zusatzstoffe, die die Lagerfähigkeit, die Resorption, Kinetik oder/und die Verträglichkeit des erfindungsgemäßen Mittels erhöhen.
Vorzugsweise enthält das Mittel weiterhin mindestens ein Pyrimidin, vorzugsweise Uridin, Cytidin oder/und Thymidin. Durch die Verwendung von Pyrimidinen in dem erfindungsgemäßen Mittel wird die Verträglichkeit für die verwendete Substanz weiter erhöht und mögliche Nebenwirkungen können vermindert werden. Darüber hinaus wird es dadurch möglich, die Dosis der erfindungsgemäß verwendeten Substanz zu erhöhen. Somit kann auch eine schwerwiegende Virusinfektion wirksam bekämpft werden. Der Gehalt des Pyrimidins kann bis zu 90 Gew.-% betragen, bevorzugt 5 bis 70 Gew.-%, mehr bevorzugt 1 0 bis 60 Gew.-% und am meisten bevorzugt 30 bis 40 Gew.-% .
In bevorzugten Ausführungsformen wird die Substanz zur Herstellung von Mitteln zur Bekämpfung, d.h. Prävention oder Behandlung von Infektionen mit Viren aus der Gruppe Hepatitis B-Virus, HIV-1 oder HIV-2-Virus, aviäres Paramyxovirus, Parainfluenzavirus, bevorzugt vom Typ 1 , Mumps-Virus, Masern-Virus, Hundestaupe-Virus, Rinderpest-Virus, Pneumovirus und Influenzavirus, bevorzugt Influenza-A-Virus, verwendet.
Als Substanz für das Mittel werden beispielsweise (A) N-(4-Trifluor- methylphenyl)-5-methylisoxazol-4-carboxamid, (B) die Form mit offenem
Isoxazolring von (A) N-(4-Trifluormethylphenyl)-2-cyano-3-hydroxycroton-
amid, (C) ein Natriumsalz oder Lysinsalz von N-(4-Trifluormethylphenyl)-2- cyano-3-hydroxy-crotonamid oder/und ein Gemisch davon verwendet.
Der Gehalt der Substanz gemäß Formel I in dem erfindungsgemäßen Mittel kann je nach dem zu behandelnden Virustyp und gegebenenfalls der Schwere der Virusinfektion ausgewählt und angepaßt werden.
Das erfindungsgemäß hergestellte Mittel wird im allgemeinen in solcher Menge verabreicht, die 0,01 bis 1000 mg/Tag, bevorzugt 0, 1 bis 1 00 mg/Tag an Substanz entspricht, abhängig von der Proteinbindung der jeweiligen Substanz.
In einer weiteren Ausführungsform kann das Mittel weitere antiviral wirksame Substanzen enthalten. Vorteilhaft ist die Auswahl von Sub- stanzen, die die gleiche Virustypspezifität wie die erfindungsgemäß verwendete Substanz haben. Dies hat den Vorteil, dass die Wirksamkeit gegenüber einem bestimmten Virustyp, gegebenenfalls durch synergistische Effekte der Substanzen erhöht werden kann. Gegebenenfalls zeigt diese Substanz eine andere Virustyp-Spezifität als die erfindungsgemäße Substanz, so daß andererseits neben dem Primärvirus gleichzeitig auch Sekundärvirus-Infektionen bekämpft werden können.
Das erfindungsgemäß hergestellte Mittel kann in jeder geeigneten Darreichungsform zubereitet werden. Vorzugsweise wird das Mittel in Form einer Tablette, einer Retardtablette, eines Dragees, einer Kapsel, eines Granulats, einer Ampulle, einer Infusionslösung, einer Injektionslösung oder eines Konzentrats zur Herstellung einer Infusionslösung oder einer Injektionslösung zubereitet.
In einer weiteren Ausführungsform betrifft die Erfindung ein Verfahren zum Vermindern oder Inhibieren des Wachstums von Hepatitis-B-Viren, HI-Viren, Viren der Paramyxogruppe oder/und der Orthomyxogruppe in mit diesen
Viren infizierten Zellen, umfassend Inkonktaktbringen der Zellen mit einer ausreichend wirksamen Menge einer erfindungsgemäßen Substanz, um die Virusvermehrung zu verlangsamen oder zu inhibieren.
In einer bevorzugten Ausführungsform dieses Verfahrens wird das Vermindern bzw. Inhibieren des Wachstums oder der Vermehrung der Viren durch eine Verlangsamung oder Inhibierung der Virusassemblierung erreicht.
Die wirksame Menge einer erfindungsgemäßen Substanz wird dabei an den jeweiligen Virustyp und die infizierten Zellen in geeigneter Weise angepasst. Im Allgemeinen liegt die wirksame Menge der Substanz in einer Konzentration von 1 bis 1 000 yM.
In noch einer weiteren Ausführungsform betrifft die Erfindung ein Verfahren zum Nachweis eines Hepatitis-B-Virus, eines Hl-Virus oder eines Virus der Paramyxogruppe oder der Orthomyxogruppe in Zellen, das dadurch gekennzeichnet ist, dass 1 . zu Patientenzellen in Kultur eine Substanz gemäß Formel I zu dem Kulturmedium zugegeben wird und 2. cytopathische oder zeilverändernde Effekte im Vergleich mit einer Kontrollkultur ohne Zugabe dieser Substanz gemessen werden. Dieses Verfahren hat den Vorteil, dass durch einfache visuelle Kontrolle ein Nachweis der oben genannten Viren möglich ist.
In einer weiteren Ausführungsform betrifft die Erfindung ein Verfahren zur prophylaktischen oder therapeutischen Behandlung von Infektionen durch Hepatitis-B-Viren, HI-Viren, Viren der Paramyxogruppe oder/und der Orthomyxogruppe durch Verabreichung eines Mittels an einen Patienten, das eine Substanz der allgemeinen Formel I enthält. Die Menge der Substanz wird an den jeweils vorliegenden Virus-Typ und die Schwere der Virusinfek- tion angepasst. Zweckmäßig wird dem Patienten eine Tagesdosis von 0,01 bis 1 000 mg, bevorzugt 0, 1 bis 1 00 mg der Substanz verabreicht. Die Verabreichung des erfindungsgemäß erhaltenen Mittels kann auf jede geeignete
Weise erfolgen, wobei bevorzugt eine systemische Applikation durch z.B. orale, intravenöse, intramuskuläre oder subkutane Applikation erfolgt. Das Mittel mit der Substanz kann dem Patienten auch in Form eines Aerosols verabreicht werden. Eine andere mögliche Art der Applikation ist topisch, z.B. in Form von Tropfen, Cremes, Pflaster oder Suppositorien.
Als Substanz werden (A) N-(4-Trifluormethylphenyl)-5-methylisoxazol-4- carboxamid, (B) die Form mit offenem Oxazolring von (A) N-(4-Trifluorme- thylphenyl)-2-cyano-3-hydroxycrotonamid, (C) ein Natrium- oder Lysinsalz von N-(4-Trifluormethylphenyl)-2-cyano-3-hydroxy-crotonamid oder Gemische davon bevorzugt.
Um die Wirksamkeit oder/und die Verträglichkeit der Substanz gemäß Formel I weiter zu erhöhen, kann das erfindungsgemäße Mittel weiterhin mindestens ein Pyrimidin, vorzugsweise Uridin, Cytidin oder/und Thymidin enthalten.
Die Substanz gemäß Formel I bzw. das Derivat mit geöffnetem Isoxazolring oder das Salz davon kann zusammen mit weiteren antiviral wirksamen Mitteln verabreicht werden, wobei dies gleichzeitig oder nacheinander erfolgen kann. Beispiele für weitere antiviral wirksame Substanzen beinhalten Aciclovir, Ganciclovir, Vidaravidin, Foscarnet, Cidofovir, Amantadin, Ribavirin, Trifluorthymidin, Interferon-σ, Cidovudin, Didanosin oder Calcitabin.
Die Erfindung wird durch die folgenden Figuren und Beispiele weiter erläutert.

Figur 1 die Verminderung des cytopathischen Effekts (CPE) von HIV-1 bei C81 66-Zellen durch die Substanzen N-(4-Trifluormethyl-
phenyl)-5-methylisoxazol-4-carboxamid (A), der metabolisier- ten, aktiven offenen Ringform von (A) N-(4-Trifluormethylphe- nyl)-2-cyano-3-hydroxycrotonamid (B) und dessen Natriumsalz N-(4-Trifluormethylphenyl)-2-cyano-3-hydroxycrotonamid (C) in verschiedenen Verdünnungen,
Figur 2 den Einfluss der Substanzen A, B und C in verschiedenen Verdünnungen auf die Ausbildung des CPE (Syncytien-Bildung),
Figur 3, Figur 4 und Figur 5 die Auswirkung von N-(4-Trifluormethyl phenyl)-5-methylisoxazol~4-carboxamid in verschiedenen Verdünnungen zu unterschiedlichen Zeitpunkten (Tag 2-5, 5-9 bzw. 9-1 2) auf die Enten-Hepatitis-B-Virus-Produktion in primären Entenhepatozyten.
Figur 6 den Einfluss einer Verabreichung der Substanz A vor Infektion auf die Enten-Hepatitis-B-Virus-Produktion in primären Entenhepatozyten.
Beispiel 1
Testprinzip: Durchführung von Infektionsreihen von C81 66-Zellen mit HIV- 1 und anschließende Auswertung des cytopathischen Effekts (CPE = Riesenzellbildung durch virusbedingte Zellverschmelzungen (Syncytien)), wobei jeweils eine Virus-Verdünnungs- reihe ohne Substanz als Kontrolle verwendet wurde. Bei diesen
Experimenten wurden die Infektionen mit HIV jeweils ohne Substanz in einem Falcon-Röhrchen durchgeführt, nach 1 Stunde Adsorption wurde abzentrifugiert und gewaschen, um die Zellen dann auf die Platte auszubringen. Das Waschen verringert die Zahl der ungebundenen Viren im Überstand auf ca. 100/ml. Auf der Platte sind weitere Indikatorzellen und Medium mit entsprechender Substanz vorgelegt.
Zelllinie: C81 66, humane T-Lymphozyten, Indikatorzelllinie für HIV- 1
Virus: HIV-1 stammte aus dem Kulturüberstand von infizierten H9
Zellen (Isolat aus einem männlichen homosexuellen Münchner; zur Verfügung gestellt von Herrn Prof.L. Gürtler, München) .
Virustiter: ca. 3 x 1 03 - 1 ,6 x 1 04/ml (festgestellt über CPE nach 3-4 Tagen)
Der cytopathische Effekt wurde jeweils nach 3-4 Tagen ausgewertet.
Testsubstanzen:
A = N-(4-Trifluormethylphenyl)-5-methylisoxazol-4-carboxamid
B = metabolisierte, aktive offene Ringform von (A) : N-(4-Trifluor- methylphenyl)-2-cyano-3-hydroxycrotonamid
C = das Natriumsalz von B: N-(4-Trifluormethylphenyl)-2-cyano-3- hydroxy-crotonamid
Der Test wurde für Substanz A auf 24-Well-Platten durchgeführt, alle weiteren Tests wurden auf 48-Well-Platten durchgeführt. Die Zahl der eingesetzten C81 66 Zellen war ca. 1 03-1 04/Well. Die erste Spalte der Platte blieb virusfrei; die weiteren Spalten enthalten von links nach rechts HIV-1 Überstand in zunehmender Verdünnung ( 1 :5, 1 :25, 1 : 1 25, 1 :625, 1 :31 25, 1 : 1 5625; 1 :781 25) . Die erste Zeile der Platte war in der Regel der Kontrollverdünnungsreihe vorbehalten, in den weiteren Reihen wurden die entsprechenden Substanzen ins Medium gegeben (EK im Medium: 1 00 μM, 50 μM und 20 μM). Nach drei bis vier Tagen, wenn der cytopathische Effekt in der Kontrollreihe voll ausgebildet war, wurden die Syncytien in jedem Well ausgewertet.
Ergebnisse:
Substanz A: Rückgang des CPE um den Faktor 5 bis 25; es ist keine deutliche Konzentrationsabhängigkeit zu erkennen. Substanz B: Rückgang des CPE um den Faktor 25 bis 1 25; es ist nur eine schwache Konzentrationsabhängigkeit zu erkennen.
Substanz C: Rückgang des CPE um den Faktor 5 bis 1 25; es ist nur eine schwache Konzentrationsabhängigkeit zu erkennen. Diese Ergebnisse sind in Figur 1 dargestellt.
Durch die erfindungsgemäße Verwendung der Substanzen A, B bzw. C konnte jeweils eine deutliche reproduzierbare Verringerung des cytophati- schen Effekts von HIV-1 Viren auf C81 66 Indikatorzellen festgestellt werden.
Beispiel 2
Verminderung des cytopathischen Effekts von HIV-1 (lllb) auf C8166-Zellen durch die Substanzen A, B und C
Testprinzip: Durchführung von Infektionsreihen von C81 66-Zellen mit HIV- 1 und anschließender Auswertung des cytopathischen Effekts. Eine Virus-Verdünnungsreihe wurde als Kontrollwert verwendet, wobei jeweils eine Virus-Verdünnungsreihe pro getesteter Substanz angesetzt wurde. Zelllinie: C81 66, humane T-Lymphozyten, Indikatorzelllinie für HIV-1
Virus: HIV-1 (HIV lllb) stammte aus dem Kulturüberstand von infizierten HUT78 Zellen;
Virustiter: ca. 5 x 1 02 - 1 x 1 03/ml
Testsubstanzen: A, B, C (50 mM), wurden in einer 1 :500 Verdünnung verwendet.
A = N-(4-Trifluormethylphenyl)-5-methylisoxazol-4-carboxamid B = metabolisierte, aktive offene Ringform von (A) N-(4-Trifluor- methylphenyl)-2-cyano-3-hydroxycrotonamid
C = das Natriumsa!zvon (B) N-(4-Trifluormethylphenyl)-2-cyano-3- hydroxy-crotonamid
Der Test wurde auf 24-Well-Platten durchgeführt. Die Zahl der eingesetzten C81 66-Zellen war ca. 1 03-1 04/Well. Die erste Spalte der Platte blieb virusfrei; die weiteren Spalten enthalten von links nach rechts HIV lllb Überstand in zunehmender Verdünnung (1 :5, 1 :25, 1 : 1 25, 1 :625; 1 :31 25) . Die erste Reihe der Platte war jeweils der Kontrollverdünnungsreihe vorbehalten, in den weiteren drei Reihen wurden die Substanzen A, B und C ins Medium gegeben. Nach drei Tagen, wenn der CPE in der Kontrollreihe voll ausgebildet war, wurden die Syncytien in jedem Well ausgezählt. Der Versuch wurde dreimal wiederholt. Figur 2 zeigt die gemittelten Werte.
Das Ergebnis zeigt deutlich, dass die Syncytienbildung von HIV-1 lllb bei C81 66-Zellen durch Anwendung der erfindungsgemäßen Substanzen um einen Faktor von ca. 1 00 unterdrückt wird. Aufgrund des geringen Virustiters sind die erhaltenen Werte jedoch an der Auflösungsgrenze des Tests.
Beispiel 3
Verringerung der Vermehrung von Ortho- und Paramyxoviren durch die Substanzen A, B und C
Testprinzip: Durchführung von Infektionen von Zellen in Gegenwart der einzelnen Substanzen und Quantifizierung der freigesetzten Viren durch HA-Test (Orthomyxoviren) bzw. TCID-Test
(Paramyxoviren) . Zusätzliche mikroskopische Beurteilung des durch die Substanzen allein oder durch Viren in Gegenwart der Substanzen ausgelösten cytopathischen Effekts (CPE) .
Viren: Influenza A Virus, 2560 HA/ml; Parainfluenza Typ 1 (Sendai) Virus, 6,4 x 1 05 TCID50/ml;
Zelllinien: MDCK-Zellen für Influenza A Viren; MCF-1 0A Zellen und HeLa- Zellen für Sendai-Viren;
Experiment: Zellen in 6-Well-Platten wurden mit 1 28 HA Influenza A Virus, 1 , 6 x 1 04 TCID50 Sendai-Virus für zwei Stunden infiziert; nicht aufgenommene Viren wurden durch Waschen entfernt und die Zellen in Serum-freien Medium in Gegenwart der Testsub- stanzen für 24 oder 48 Std. inkubiert; anschließend wurde die
Anzahl der in den Zellkulturüberstand freigesetzten Viren im HA- bzw. TCID50-Test bestimmt.
Ergebnisse: In mehreren unabhängigen Versuchsreihen konnte eine Reduktion der Replikation von Ortho- und Paramyxoviren in Abhängigkeit von der Konzentration der eingesetzten Substanzen (z.B. Substanz B) festgestellt werden:

Die Substanz B zeigte eine um den Faktor 2 bessere Hemmung der Virusreplikation im Vergleich zu den Substanzen A und C.
In Gegenwart der Substanz B verringerte sich der durch die Virusinfektion bedingte CPE auf 90 % - 75 % gegenüber der Positivkontrolle, insbesondere die Veränderung der Zellform in Richtung "spindelförmig" war reduziert.
Beispiel 4 Verringerung der Duck Hepatitis B Virus (DHBV) Sekretion aus primären
Entenhepatozyten durch N-(4-Trifluormethylphenyl)-5-methylisoxazol-4- carboxamid (Substanz A).
Methoden: Ein Entenküken wurde einen Tag nach dem Schlüpfen intravenös mit 200 μl Entenserum injiziert, das 1 010 DHBV DNA-Genomäquivalente/ml enthielt. Zwei Wochen nach Infektion wurden primäre Enten-Hepatozyten
(PDH) wie beschrieben präpariert und kultiviert (JV ( 1 986) 58, 1 7-251 ; JV
( 1 992) 66, 2829-2836) : Hepatozyten wurden durch Zwei-Schritt
Collagenase-Perfusion isoliert und in 6-Well-Platten mit ca. 1 06 Zellen/Well ausgesät. Die Zellen wurden bei 37 °C in 5 % CO2 in William's Medium E (Gibco BRL) gehalten, das mit Gentamycin (50 μg/ml), L-Glutamin (2,25 mM), Glucose (0,06 %), HEPES pH 7,4 (23 mM), Hydrocortison (4,8 μg/ml) , Inosin ( 1 μg/ml), Penicillin (50 lU/ml), Streptomycin (50 μg/ml) und Dimethylsulfoxid ( 1 ,7 %) supplementiert worden war. Ein Mediumwechsel erfolgte an den Tagen 2, 5, 9 und 1 2 nach Ausplattieren.
Die Substanz A wurde am Tag 5 bzw. am Tag 9 nach dem Plattieren zu- gegeben. Die Kulturüberstände wurden am Tag 5, 9 und 1 2 nach dem Plattieren gesammelt. Pro Well wurde ein Viertel des Kulturmediums (entspricht 2,5 x 105 Zellen) mit Hilfe von Dot-Blot-Hybridisierung relativ zu einem DNA-Standard analysiert. Ein linearisiertes Plasmid, das eine Kopie des DHBV-Genoms trägt, wurde mit "random priming" auf eine spezifische Aktivität von etwa 108 Ipm/μg markiert und als Sonde zur Hybridisierung eingesetzt. Die Quantifizierung der Signale erfolgte mit einem Molecular Dynamics Phosphoimager.
Erläuterung zu den Figuren: Figur 3 zeigt den DNA-Dot-blot der PDH-Überstände. PDH wurden mit Substanz A gelöst in DMSO (comp. A, comp. A rev) in den angezeigten Konzentrationen 5 Tage nach dem Plattieren behandelt. Als Kontrolle wurde DMSO alleine getestet. Am Tag 9 nach dem Plattieren wurde frisches Medium mit (comp. A) oder ohne Substanz A (comp. A rev) zu den Zellen gegeben. Der DNA-Standard, der zur Umrechnung der Daten in das Diagramm von Figur 4 diente, ist ebenfalls gezeigt. Die Zahlen in Figur 4 repräsentieren die Medianwerte aus 3 (A rev), 6 (unbehandelt d9, unbehan- delt d 1 2) oder 1 2 individuellen Wells (unbehandelt d5, A), von denen der unspezifische Hintergrund (7,7 x 1 06 DNA Äquivalente/ml) bereits abgezogen wurde. "d2-5" steht für Überstände, die am Tag 5 gesammelt wurden, "d5-9" am Tag 9 und "d9-1 2" am Tag 1 2.
Das Diagramm eines Dot-blots mit Überständen von PDH, die mit Substanz A in Konzentrationen von 5 bis 200 μM behandelt wurden, ist in Figur 5 dargestellt. Zahlen repräsentieren hier Medianwerte von zwei (mit A behandelt) oder drei (unbehandelt) individuellen Wells, der unspezifische Background entsprach 4,3 x 1 06 DNA Äquivalenten/ml .
Ergebnisse:
Die Virusabgabe der unbehandelten PDH war in den ersten Tagen nach Plattierung niedrig, stieg jedoch bei Tag 1 2 auf 3,8 x 1 08 DNA Äquivalente/ml an (Fig. 4 (-), vgl. d2-5, d5-9, d9- 1 2) . Im Gegensatz dazu verblieb die Produktion von Nachkommenviren in PDH, die mit 20 oder 50 μM Substanz A behandelt worden waren, selbst am Tag 1 2 auf dem niedrigen Anfangsniveau von 5 bzw. 7 x 1 07 DNA Äquivalenten/ml (Fig. 4 20 μM A, 50 μM A) . Vergleichbare Ergebnisse wurden in einem Konzen- trationsbereich von 5 μM bis 200 μM A (Fig. 5) erzielt. Das Entfernen von Substanz A am Tag 9 (Fig. 4 A rev) führte zu höherer Virusfreisetzung zwischen Tag 9 und 1 2 im Vergleich zu weiterhin behandelten Zellen (Fig. 4 A), was darauf hinweist, dass die Wirkung von Substanz A reversibel ist. Zusammenfassend zeigte sich, dass Substanz A den Anstieg an Virussekre- tion verhinderte, der bei nicht behandelten Zellen ungefähr 1 ,5 Wochen nach Plattierung beobachtet wird .
Beispiel 5
N-(-4-trifluormethylphenyl)-5-methylisoxazol-4-carboxamid (Substanz A) interferiert mit der Infektion von primären Entenhepatozyten durch Duck Hepatitis B Virus (DHBV)
Methoden:
Primäre Enten-Hepatozyten (PDH) eines zwei Wochen alten Entenkükens wurden wie beschrieben präpariert und kultiviert (siehe Beispiel 4) . Substanz
A wurde am Tag 6 nach dem Plattieren zugegeben. Am Tag 7 nach dem
Plattieren wurden die Zellen mit 1 x1 08 DHBV DNA Äquivalenten pro 1 x1 06
Zellen für 1 2 h infiziert, nach Entfernung der nicht-aufgenommenen Viren wurde erneut Substanz A zugegeben. Am Tag 9 nach Plattierung erfolgte ein Mediumwechsel mit erneuter Substanz-Zugabe, am Tag 1 2 nach Plattierung (entspricht Tag 6 nach Infektion) wurden die Zellkulturüber- stände gesammelt. Pro Well wurde ein Viertel des Kulturmediums (entspricht 2,5x 1 05 Zellen) mit Hilfe von Dot-Blot-Hybridisierung relativ zu einem DNA-Standard analysiert. Ein linearisiertes Plasmid, das eine Kopie des DHBV-Genoms trägt, wurde mit "random priming" auf eine spezifische Aktivität von etwa 1 08 Ipm/μg markiert und als Sonde zur Hybridisierung eingesetzt. Die Zellen wurden mit 1 00% Methanol fixiert und mit polyklonalen Kaninchen-Antiseren gegen DHBV core Protein oder DHBV preS Protein inkubiert. Das Binden des ersten Antikörpers an Antigen wurde mit einem Ziege-Anti-Kaninchen Fluorescein Thioisocyanat konjugierten Zweitantikörper (Dianova, Hamburg) detektiert.
Erläuterung zu der Figur: Figur 6 zeigt in Diagramm-Form die Ergebnisse des DNA Dotblots mit den PDH-Überständen 6 Tage nach Infektion. Die Zellen wurden mit 20 oder 50 μM Substanz A (A20, A50) oder DMSO (DMSO) einen Tag vor Infektion vorbehandelt. Die Balken repräsentieren Mittelwerte aus 6 individuellen Wells, der unspezifische Hintergrund entsprach 7 x 1 06 DNA Äquivalen- ten/ml.
Ergebnisse:
Die Vorbehandlung primärer Enten-Hepatozyten mit 20 μM und 50 μM Substanz A einen Tag vor Infektion führte zu einer deutlichen Reduktion der Virusausbeute in den Zellkulturüberständen, gemessen durch DNA-Dotblot Analyse (Figur 6 vgl. no comp und A20 bzw. A50) . Diese Verringerung der Virusvermehrung zeigte sich auch beim Anfärben der Zellen mit Capsid-
oder Envelope-Protein spezifischen Antikörpern. Die Replikation des Genoms und die Infektion weiterer Zellen sind in Anwesenheit von Substanz A deutlich reduziert.
