一種酸性飲料,其係含有(A)吡咯并喹啉醌或其鹽、(B)抗壞血酸及(C)環糊精,並使全部成分溶解。

本發明為關於酸性飲料及析出抑制方法。
酸性飲料係pH為在酸性區域,且特徵在於恰當的酸味與清爽的風味之偏好性為較高的飲料。為了調整如此般的酸性飲料的pH或風味,並防止氧化,而廣泛地使用了抗壞血酸。
吡咯并喹啉醌係存在於細菌、真核生物的霉、酵母中的輔酶。吡咯并喹啉醌係被提案著作為新的維生素的可能性,因而作為在健康輔助食品、化粧品等中為有用的物質而備受關注。又,近年來對於吡咯并喹啉醌,已清楚得知具有細胞的增殖促進作用、抗白內障作用、肝臟疾患預防治療作用、創傷治癒作用、抗過敏作用、逆轉錄酶抑制作用及乙二醛酶I抑制作用(抗癌作用)等多種的生物活性(參考例如非專利文獻1)。迄今已有報告著,為了使吡咯并喹啉醌穩定化,而製成與環糊精的包合物(參考例如專利文獻1)。

[發明所欲解決之課題]
本發明人在對於含有吡咯并喹啉醌的酸性飲料進行研究時發現,與抗壞血酸共存下,吡咯并喹啉醌會產生析出之問題。
因此,本發明之目的在於提供一種在抗壞血酸存在下,吡咯并喹啉醌難以析出的良好的酸性飲料。

本發明之第一實施形態相關的酸性飲料係含有(A)吡咯并喹啉醌及/或其鹽、(B)抗壞血酸、(C)環糊精、任意的抗壞血酸以外的(D)酸化劑或其他的成分。在環糊精之存在下,以吡咯并喹啉醌作為代表的所有成分為溶解在酸性飲料中。
本實施形態的酸性飲料係含有吡咯并喹啉醌(以下亦記載為「PQQ」)及/或其鹽來作為成分(A)。

本實施形態中所謂PQQ係指式1所記載之化合物。
作為PQQ的鹽未特別限定,但可舉出金屬鹽,例如,單鈉鹽、二鈉鹽、三鈉鹽、單鉀鹽、二鉀鹽、三鉀鹽、單鋰鹽、二鋰鹽、三鋰鹽等。較佳為二鈉鹽。
本實施形態的酸性飲料中的成分(A)之含量,若成分(C)為α-環糊精之情形時,以5mg/L~100mg/L為較佳,以10mg/L~100mg/L為又較佳,以10mg/L~40mg/L為特佳。又,本實施形態的酸性飲料中的成分(A)之含量,若成分(C)為γ-環糊精之情形時,以5mg/L~550mg/L為較佳,以10mg/L~80mg/L為又較佳,以10mg/L~40mg/L為特佳。
成分(A)之含量係可藉由一般已知的吡咯并喹啉醌的分析法之中適合於測定樣品的狀況的分析法來進行測定。具體而言能夠利用後述揭示的實施例所記載之液相層析法來將成分(A)進行定量分析。尚,包含吡咯并喹啉醌的樣品係可因應所需施予適宜處理。例如,可於測定之際為了適合於裝置的檢測區域而將樣品進行凍結乾燥,或為了適合於裝置的分離能力而去除樣品中的夾雜物等。
成分(B)抗壞血酸係能夠被添加至酸性飲料中來作為酸化劑或抗氧化劑等。抗壞血酸係可調配與成分(A)吡咯并喹啉醌為相同、或者較其為多。具體而言,成分(A)與成分(B)之質量比[(B)/(A)]係以1~1000為較佳,以2~350為又較佳,以10~100為特佳。相對於吡咯并喹啉醌,若抗壞血酸未滿1之質量比之情形時,不會產生析出的問題。另一方面,相對於吡咯并喹啉醌,若抗壞血酸超過1000之質量比之情形時,則風味的平衡將會變差。
所使用的環糊精為α、γ-環糊精中任一種而無限制。就析出抑制效果之觀點而言,以γ-環糊精為較佳。酸性飲料中的γ-環糊精之含量係以0.3~24重量%為較佳,以0.4~13重量%為又較佳,以1~10重量%為特佳。
酸性飲料中的α-環糊精之含量係以3~13重量%為較佳,以5~13重量%為又較佳,以6~10重量%為特佳。
即使環糊精的濃度為上述之範圍外之情形時,相較於無添加環糊精之情形,亦可使析出物的產生減少,但若環糊精的濃度低於上述下限之情形時,將會有發現吡咯并喹啉醌的析出之情況。若環糊精的濃度高於各上限之情形時,將會有環糊精本身析出之情況。
作為(D)酸化劑(acidifier)未特別限定,可舉例如檸檬酸、乳酸、蘋果酸、磷酸、琥珀酸等。本實施形態的酸性飲料中,酸化劑係對於展現出清涼感為有效。酸化劑之含量係可從味覺之方面來做適時設定,例如以0.001~5重量%為較佳,以0.01~3重量%為又較佳,以0.1~3重量%為特佳。
本實施形態的酸性飲料中,pH係以2~5.4為較佳,以2~4為又較佳,以2~3為特佳。為了展現出清涼感而需要具有酸性。能夠藉由適量調配酸化劑來控制pH。
本實施形態的酸性飲料係以含有甜味劑為較佳。作為甜味劑,可舉例如糖質系甜味劑、高甜度甜味劑等。於此,本說明書中所謂「高甜度甜味劑」係指與蔗糖相比較下具有十倍~千倍的甜味,利用微量的添加而可對飲食品賦予甜味的人工或天然的甜味劑之涵義。
作為糖質系甜味劑,可舉例如果糖、葡萄糖、塔格糖、阿拉伯糖、D-阿洛酮糖、D-阿洛糖等的單糖;乳糖、海藻糖、麥芽糖、蔗糖、纖維二糖等的二糖;赤蘚醇、木糖醇、麥芽糖醇、山梨糖醇、甘露糖醇、麥芽糖醇、還原巴拉金糖、乳糖醇、還原澱粉糖化物等的糖醇。又,作為高甜度甜味劑,可舉例如蔗糖素、艾司沙芬鉀、阿斯巴特糖、甜菊(甜菊雙糖苷(rebaudioside)、甜菊糖)、索馬甜(thaumatin)、糖精 (saccharin)、糖精 (saccharin)鈉、甘草萃取物、羅漢果萃取物、紐甜(neotame)、馬檳榔甜蛋白(mabinlin)、巴西甜蛋白(brazzein)、莫内林甜蛋白(monellin)、甘草甜素、阿力甜(alitame)、甜蜜素、吡咯并喹啉醌、新橙皮苷等。甜味劑係可使用1種或2種以上。作為高甜度甜味劑係以選自蔗糖素、艾司沙芬鉀、阿斯巴特糖、甜菊及索馬甜(thaumatin)之1種或2種以上為較佳。
甜味劑之含量係可因應最終製品中所需要的甜度予以適宜調整。例如甜味劑係蔗糖換算下以1~20重量%為較佳,以1~15重量%為又較佳,以1~10重量%為特佳。
本實施形態的酸性飲料為了對呈味賦予刺激感而可含有碳酸。
碳酸之含量係以0.01~1重量%為較佳,以0.1~0.5重量%為又較佳,以0.3~0.4重量%為特佳。

本實施形態的酸性飲料,在所希望的效果不受阻礙的範圍內,亦可包含一般作為飲料所調配的添加劑。作為如此般的添加劑未特別限定,可舉例如著色料、抗氧化劑、香料、鹽類、穩定劑等。作為著色料未特別限定,可舉例如焦糖色素、梔子色素、果汁色素、蔬菜色素、合成色素等。作為抗氧化劑未特別限定,可舉例如維生素C、維生素E、多元酚等。作為鹽類未特別限定,可舉例如氯化鈉、氯化鉀等。又,作為穩定劑未特別限定,可舉例如果膠、水溶性大豆多糖類等。
又,本實施形態的酸性飲料係因應最終製品的種類,亦可包含其他的添加劑。作為其他的添加劑未特別限定,可舉例如桉葉醇(eudesmol)、菊粉、原花青素C1、諾卡酮(nootkatone)、甲氧基黃酮、羥基酪醇、茶胺酸、l-薄荷醇、香草醛、原花青素(proanthocyanidin)、皂素、蘆丁(rutin)、辣椒素(capsaicin)、果聚糖(fructan)、木麻黃素、蝦青素、胎盤、花青素、葡糖甘露聚糖、阿拉伯木聚糖、丙烯醯胺、鞘磷脂、瓜胺酸(citrulline)、糠基硫醇、異麥芽酮糖、膠原蛋白肽、酪蛋白肽、胺基酸(脯胺酸、甘胺酸等)、L-阿拉伯糖、2,5-哌嗪二酮、3,6-雙(苯基甲基)-,(3S,6S)、6-O-PUFA抗壞血酸酯、α-葡聚醣、β-葡聚醣、綠原酸、丙酮酸、二咖啡奎寧酸、脫氫抗壞血酸、α酸或異α酸、β酸、乳清酸、硫辛酸、寡糖(麥芽寡糖等)、異槲皮素或其糖加成物、橙皮苷及橙皮苷糖加成物、辛烯基琥珀酸澱粉、輔酶Q10、聚合或非聚合兒茶素類、葉綠素類、茶黃素類、麥芽萃取物、植物萃取物、香氛萃取物、咖啡因、薑黃、果實味道、果汁、葉肉等。
進而,本實施形態的酸性飲料係可包含除了吡咯并喹啉醌以外的機能性成分。作為機能性成分未特別限定,可舉例如BCAA(分支鏈胺基酸)、胺基酸(組胺酸、L-絲胺酸、白胺酸40%調配必須胺基酸等)、β‐隱黃素、DHA(二十二碳六烯酸)、EPA(二十碳五烯酸)、GABA(γ-胺基丁酸)、乙酸、HMB(雙-3-羥基-3-甲基丁酸一水合物)、N-乙醯基葡糖胺、蝦青素、異黃酮(來自葛花的異黃酮等)、銀杏葉烯帖內酯、銀杏葉黃酮醇配糖體、表沒食子兒茶素沒食子酸酯、甲基化兒茶素、鳥胺酸、可可黃烷醇、乳酸菌(加氏乳桿菌(Lactobacillus gasseri)SP菌株等)、雙歧桿菌、來自寒天的半乳聚糖(galactan)、胍爾豆膠(guar gum)分解物、胺基葡萄糖、葡糖苷酰鞘氨醇、來自車前子種皮的食物纖維、來自五層龍屬(salacia)的桫拉烯醇(salacinol)、銀椴苷(tiliroside)、玻尿酸Na、胡椒鹼、單葡糖基橙皮苷、乳三肽、咪唑二肽化合物、番茄紅素、蘋果多元酚等。
本實施形態的酸性飲料,可藉由例如使上述之(A)~(C)成分溶解至水中而來製造。
本實施形態的酸性飲料係可在調製後所希望的時機下進行加熱殺菌。作為加熱殺菌方法,只要符合應該適用的法規(在日本為食品衛生法)所規定的條件下即可未特別限定。可舉例如殺菌釜滅菌法、高溫短時間殺菌法(HTST法)、超高溫殺菌法(UHT法)、填充後殺菌法(巴斯德消毒法)等。
加熱殺菌法係可適宜選擇。例如金屬罐、瓶般地將飲料填充至容器中後,可連同容器進行加熱殺菌(例如以60~140℃、1~60分)之情形時,可採用殺菌釜滅菌或填充後殺菌法(巴斯德消毒法)。若為填充後殺菌法(巴斯德消毒法)之情形時,可例如以65℃下進行1~60分鐘,較佳以65℃下進行5~30分鐘,更佳以65℃下進行10~20分鐘來做加熱殺菌。
又,對於如PET塑料瓶般地無法進行殺菌釜滅菌者,可採用與上述相同的殺菌條件(例如以65~140℃下進行0.1秒~30分鐘、較佳以70~125℃下進行1秒~25分鐘、更佳以75~120℃下進行10秒~20分鐘)預先對飲料進行加熱殺菌、填充至在無菌環境下經殺菌處理的容器之無菌填充、或高溫填充(hot filling)等。
第二實施形態中,本發明係提供一種吡咯并喹啉醌的析出抑制方法,其係包含在抗壞血酸之存在下,使吡咯并喹啉醌或其鹽與環糊精接觸之步驟。
關於析出抑制方法中使用的各成分係如上述般,各成分量係可因應所希望的效果來予以適宜決定。析出抑制方法係可進而包含在酸性飲料的製造中所使用之步驟,例如,上述之填充或殺菌等的步驟。

本實施形態中使用的吡咯并喹啉醌二鈉為使用三菱瓦斯化學股份有限公司製BioPQQ(註冊商標)。α-環糊精及γ-環糊精為使用和光純藥製造者。
[實施例1]
製作每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、α-環糊精4.0%重量、吡咯并喹啉醌二鈉0.008重量%(80mg/L)的水溶液。抗壞血酸/PQQ二鈉之質量比為37.5。將此者作為含有吡咯并喹啉醌的酸性飲料。pH為2.3。將含有吡咯并喹啉醌的酸性飲料以4℃下保存1天,評估吡咯并喹啉醌的析出之有無。在調配α-環糊精的含有吡咯并喹啉醌的酸性飲料中,未確認到吡咯并喹啉醌的析出。
製作每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、α-環糊精4.0%重量、吡咯并喹啉醌二鈉0.008重量%(80mg/L)的水溶液。抗壞血酸/PQQ二鈉之質量比為37.5。將此者作為含有吡咯并喹啉醌的酸性飲料。pH為2.3。將含有吡咯并喹啉醌的酸性飲料以4℃下保存1天,評估吡咯并喹啉醌的析出之有無。在調配α-環糊精的含有吡咯并喹啉醌的酸性飲料中,未確認到吡咯并喹啉醌的析出。
[實施例2]
製作每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、γ-環糊精4.0%重量、吡咯并喹啉醌二鈉0.008重量%(80mg/L)的含有吡咯并喹啉醌的酸性飲料。抗壞血酸/PQQ二鈉之質量比為37.5。pH為2.3。將此者以4℃下保存1天,評估吡咯并喹啉醌的析出之有無。即使在調配γ-環糊精的酸性飲料中,亦未確認到吡咯并喹啉醌的析出。

除了未添加環糊精以外,與上述之實施例1或2相同地添加,以成為每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、吡咯并喹啉醌二鈉0.008重量%(80mg/L),來調製含有吡咯并喹啉醌的酸性飲料。抗壞血酸/PQQ二鈉之質量比為37.5。pH為2.3。若將含有吡咯并喹啉醌的酸性飲料以4℃下保存1天時,會析出暗紅色的沉澱。
除了未添加環糊精以外,與上述之實施例1或2相同地添加,以成為每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、吡咯并喹啉醌二鈉0.008重量%(80mg/L),來調製含有吡咯并喹啉醌的酸性飲料。抗壞血酸/PQQ二鈉之質量比為37.5。pH為2.3。若將含有吡咯并喹啉醌的酸性飲料以4℃下保存1天時,會析出暗紅色的沉澱。
由以上之結果可得知,環糊精的添加係具有抑制在酸性溶液中的吡咯并喹啉醌的析出之效果。
[比較例2~4]
除了未添加環糊精以外,與上述之實施例1或2相同地調製每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、吡咯并喹啉醌二鈉0.008重量%(80mg/L)的酸性飲料。抗壞血酸/PQQ二鈉之質量比為37.5。
以成為5.0重量%之方式來添加葡萄糖、果糖、山梨糖醇,將該酸性飲料以4℃下保存1天並確認析出之有無。將結果記載於表1中。
除了未添加環糊精以外,與上述之實施例1或2相同地調製每1L中含有抗壞血酸0.3重量%、檸檬酸0.6重量%、吡咯并喹啉醌二鈉0.008重量%(80mg/L)的酸性飲料。抗壞血酸/PQQ二鈉之質量比為37.5。
以成為5.0重量%之方式來添加葡萄糖、果糖、山梨糖醇,將該酸性飲料以4℃下保存1天並確認析出之有無。將結果記載於表1中。

依據本發明,可提供一種即使是抗壞血酸之存在下,亦能抑制吡咯并喹啉醌的析出之穩定的飲料。
[實施例12]
在Wilkinson的碳酸塑料瓶中添加下述之成分來調製碳酸飲料。以每1L中成為砂糖0.5重量%、檸檬酸0.05重量%、抗壞血酸0.3重量%、γ-環糊精0.4重量%、吡咯并喹啉醌二鈉0.008重量%(80mg/L)、艾司沙芬鉀(acesulfame potassium)0.015重量%方式來進行添加。抗壞血酸/PQQ二鈉之質量比為37.5。pH為3。雖將該飲料以4℃下來做保存,但未確認到析出。為具有清涼感的甘甜飲料。
在Wilkinson的碳酸塑料瓶中添加下述之成分來調製碳酸飲料。以每1L中成為砂糖0.5重量%、檸檬酸0.05重量%、抗壞血酸0.3重量%、γ-環糊精0.4重量%、吡咯并喹啉醌二鈉0.008重量%(80mg/L)、艾司沙芬鉀(acesulfame potassium)0.015重量%方式來進行添加。抗壞血酸/PQQ二鈉之質量比為37.5。pH為3。雖將該飲料以4℃下來做保存,但未確認到析出。為具有清涼感的甘甜飲料。
[比較例6]
在Wilkinson的碳酸塑料瓶中添加下述之成分來調製碳酸飲料。以每1L中成為砂糖0.5重量%、檸檬酸0.05重量%、抗壞血酸0.3重量%、吡咯并喹啉醌二鈉0.006重量%(60mg/L)、艾司沙芬鉀0.015重量%之方式來進行添加。抗壞血酸/PQQ二鈉之質量比為50。pH為3。雖將該飲料以4℃下來做保存,但吡咯并喹啉醌為析出。為具有清涼感的甘甜飲料。
在Wilkinson的碳酸塑料瓶中添加下述之成分來調製碳酸飲料。以每1L中成為砂糖0.5重量%、檸檬酸0.05重量%、抗壞血酸0.3重量%、吡咯并喹啉醌二鈉0.006重量%(60mg/L)、艾司沙芬鉀0.015重量%之方式來進行添加。抗壞血酸/PQQ二鈉之質量比為50。pH為3。雖將該飲料以4℃下來做保存,但吡咯并喹啉醌為析出。為具有清涼感的甘甜飲料。
若依據本實施形態時,即使是碳酸飲料中亦可抑制析出。可得知特別是γ-環糊精的析出抑制能力為高,故即使是更高濃度亦未產生析出。又,可得知環糊精的添加係對於風味並無影響,故用來製作飲料為優異。
[實施例13~24]
如表5及6所表示般來添加抗壞血酸、檸檬酸、α-環糊精(α-CD)或者γ-環糊精(γ-CD)、吡咯并喹啉醌二鈉(PQQ二鈉),從而製作水溶液。將此者作為含有吡咯并喹啉醌的酸性飲料。各酸性飲料的pH為2。將此者以4℃下保存1天,並評估酸性飲料成分的析出。將評估結果表示於表5及6中。
如表5及6所表示般來添加抗壞血酸、檸檬酸、α-環糊精(α-CD)或者γ-環糊精(γ-CD)、吡咯并喹啉醌二鈉(PQQ二鈉),從而製作水溶液。將此者作為含有吡咯并喹啉醌的酸性飲料。各酸性飲料的pH為2。將此者以4℃下保存1天,並評估酸性飲料成分的析出。將評估結果表示於表5及6中。
若添加環糊精之情形時,即使是任何的濃度,相較於無添加之情形,亦可減少析出物的產生。特別是若α-環糊精的濃度為未滿14%之濃度時,則完全抑制了析出。若γ-環糊精的濃度為未滿25%時,則完全抑制了析出。

- 一種酸性飲料,其係含有 (A)吡咯并喹啉醌或其鹽、 (B)抗壞血酸及 (C)環糊精。
- 如請求項1之酸性飲料,其中,成分(C)係α-環糊精,其含量係3~13重量%。
- 如請求項1之酸性飲料,其中,成分(C)係γ-環糊精,其含量係0.3~24重量%。
- 如請求項1~3中任一項之酸性飲料,其中,成分(A)與成分(B)之質量比[(B)/(A)]係1~1000。
- 如請求項1之酸性飲料,其pH係2~5.4。
- 如請求項1之酸性飲料,其中,進而含有抗壞血酸以外的酸化劑。
