Claims (47)
1. Аматоксин формулы I
где R1 выбран из С=O, C=S, C=NR6 и CR7R8;
R2 выбран из S=O, SO2 и S;
R3 выбран из NHR5 и OR5;
R4 выбран из Н, OR5 и OC1-6-алкила;
R6 выбран из С1-6-алкилен-R5, циклоалкилен-R5, гетероциклоалкилен-R5, арилен-R5 и гетероарилен-R5;
R7 и R8 независимо выбраны из Н, С1-6-алкилен-R5, циклоалкилен-R5, гетероциклоалкилен-R5, арилен-R5 и гетероарилен-R5;
где:
(i) каждый R5 представляет собой Н;
(ii) один из R5 представляет собой -Ln-X, где L является линкером, n выбран из 0 и 1, и X представляет собой химический фрагмент, который может быть связан с нацеливающим фрагментом и где оставшиеся R5 являются Н; или
(iii) один из R5 представляет собой -Ln-X*-Y, где L является
линкером, n выбран из 0 и 1, Y представляет собой нацеливающий фрагмент, и X* является химическим фрагментом, образующимся в результате связывания X с функциональной группой Y, и где оставшиеся R5 являются Н.
2. Аматоксин по п. 1 (iii), где указанной функциональной группой Y является аминогруппа.
3. Аматоксин по п. 2, где X* является фрагментом мочевины.
4. Аматоксин по любому из пп. 1 (iii), 2 и 3, где указанный остаток R5, представляя собой -Ln-X*-Y, является:
(v) присутствующим в R1;
(vi) присутствующим в R3;
(vii) присутствующим в R4; или
(viii) присутствующим в R5.
5. Аматоксин по любому из пп. 1, 2 или 3, где аматоксин выбран из α-аманитина, β-аманитина, аманина, аманинамида или из их солей или аналогов.
6. Аматоксин по любому из пп. 1, 2 или 3, где n равно 1, и где линкер имеет длину до 12 атомов, предпочтительно от 2 до 10, более предпочтительно от 4 до 9 и наиболее предпочтительно от 6 до 8 атомов.
7. Аматоксин по любому из пп. 1, 2 или 3, где линкером L является группа алкилен, гетероалкилен, алкенилен, гетероалкенилен, алкинилен, гетероалкинилен, циклоалкилен, гетероциклоалкилен, арилен, гетероарилен, аралкилен или гетероаралкилен, состоящая из 1-4 гетероатомов, выбираемых из N, О и S, где указанный линкер является необязательно замещенным.
8. Аматоксин по любому из пп. 1, 2 или 3, где линкер L включает фрагмент, выбираемый из: дисульфида, простого эфира, тиоэфира, амина, сложного эфира, карбоксамида, уретана и фрагмента мочевины.
9. Аматоксин по любому из пп. 1, 2 или 3, где мишень-связывающий фрагмент специфически связывается с эпитопом, который присутствует на опухолевой клетке, предпочтительно где мишень-связывающий фрагмент специфически связывается с эпитопом рецептора 2 человеческого эпидермального фактора роста (HER2).
10. Аматоксин по любому из пп. 1, 2 или 3, где мишень-связывающий фрагмент выбран из группы, состоящей из:
(i) антитела или его антиген-связывающего фрагмента;
(ii) антитело-подобного белка; и
(iii) аптамера нуклеиновой кислоты.
11. Аматоксин по п. 10, где антитело или его антиген-связывающий фрагмент выбран из диатела, тетратела, нанотела, химерного антитела, деиммунизированного антитела, гуманизированного антитела или человеческого антитела.
12. Аматоксин по п. 11, где антиген-связывающий фрагмент выбран из группы, состоящей из Fab, F(ab')2, Fd, Fv, одноцепочечного Fv и дисульфидно-связанного Fv (dsFv).
13. Аматоксин по любому из пп. 1, 2, 3, 11 или 12 для применения в качестве лекарственного препарата.
14. Аматоксин по любому из пп. 1, 2, 3, 11 или 12 для применения в лечении рака у пациента, предпочтительно когда рак выбран из группы, состоящей из рака молочной железы, рака поджелудочной железы, холангиокарциномы, колоректального рака, рака легкого, рака предстательной железы, рака яичника, рака желудка, рака почки, злокачественной меланомы, лейкоза и злокачественной лимфомы.
15. Фармацевтическая композиция, включающая аматоксин по любому из пп. 1 (iii)-12, и дополнительно включающая один или более фармацевтически приемлемых разбавителей, носителей, вспомогательных веществ, наполнителей, вяжущих веществ, смазывающих веществ, глидантов, дезинтегрирующих веществ, адсорбентов и/или консервантов.
16. Аматоксин по п. 1 (ii), где X представляет собой производное карбамовой кислоты -NH-C(O)-Z, где Z представляет собой уходящую группу, которая может быть заменена нуклеофильной группой мишень-связывающего фрагмента, предпочтительно первичным амином мишень-связывающего фрагмента.
17. Аматоксин по п. 16, где Z выбран из: -tбутилокси, -сукцинимидилокси, -1-О-сукцинимидилокси-3-сульфоната (-сульфо-NHS), -О-(4-нитрофенилокси), -О-(3-нитрофенилокси), -О-(2,4-динитрофенилокси), -О-(2,4-дихлор-6-нитрофенилокси), -
пентафторфенилокси, -пентахлорфенилокси, -О-(2,4,5-трихлорфенилокси), -О-(3,4-дигидро-3-гидрокси-4-оксо-1,2,3-бензотриазин-3-ил), -О-(эндо-1-гидрокси-5-норборнен-2,3-дикарбоксимид-1-ил), -1-фталимидоилокси, -1-бензотриазолилокси, -1-(7-азабензотриазолил)окси и -N-имидазолила.
18. Способ синтеза аматоксина по п. 16 или 17, включающий стадию реакции аматоксина формулы II
где R2 выбран из S=O, SO2 и S;
R3 выбран из NHR5 и OR5;
R4 выбран из Н, OR5 и OC1-6-алкила;
R6 выбран из С1-6-алкилен-R5, циклоалкилен-R5, гетероциклоалкилен-R5, арилен-R5 и гетероарилен-R5;
R7 и R8 независимо выбраны из Н, С1-6-алкилен-R5, циклоалкилен-R5, гетероциклоалкилен-R5, арилен-R5 и гетероарилен-R5;
где один из R5 представляет собой -Ln-X, где L является линкером, n выбран из 0 и 1, и X является химическим фрагментом, который может быть связан с нацеливающим фрагментом, и где
оставшиеся R5 являются Н;
с (i) N,N'-дисукцинимидилкарбонатом (DSC), (ii) тиокарбонилирующим реагентом, предпочтительно тиофосгеном, 1,1'-тиокарбонилдиимидазолом или 1,1'-тиокарбонилди-2(1Н)-пиридоном; (iii) иминокарбонилирующим реагентом, предпочтительно дихлоридом изоцианида или фенилизотиоцианатом; или (iv) альдегидом, кетоном или ациклическим ацеталем.
