Description
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av L-pyroglutamyl-L-prolinamid og dets anvendelse ved behandling av tiltagende alderdom med henblikk på å for-bedre alderdomsmangler.
Ifølge oppfinnelsen kan den nye forbindelse L-pyroglutamyl-L-prolinamid fremstilles ved omsetning av L-pyroglutaminsyre med L-prolinamld i nærvær av en koblingsreaktant etter følgende reaksjonsligning:
De to anvendte utgangsmaterialer for fremstilling av L-pyroglutamyl-L-prolinamid er kjent og kommersielt tilgjenge-lig. Således markedsføres L-pyroglutaminsyre av Aldrich Chemical Co. Inc, mens L-prolinamid markedsføres av Cyclo Chemical. Den sistnevnte forbindelse kan også fremstilles ved ammonlyse av den tilsvarende ethylester (se R.W. Chambers and F.M. Carpenter, J.Am.chem. Soc. 77 (.1955), 1522 - 1526.
Konvensjonelle koplingsmidler, i særdeleshet dicyclohexylcarbodiimid og l-ethoxycarbonyl-2-ethoxy-l,2^ethoxy-l,2-dihydrokinolin kan anvendes som koblingsreaktanter for denne reaksjon. Med hensyn til de substanser som kan anvendes for dette formål kan nevnes de som ér oppført av Y.S. Klausner and M. Bodansky, publisert i Synthesis, 1972, s. 453 - 463.
Et organisk løsningsmiddel slik som tetrahydrofuran, dimethylformamid, dioxan, ethylacetat, diklormethan eller lig-nende kan anvendes som reaksjonsmedium for fremgangsmåten ifølge oppfinnelsen.
Det etterfølgende eksempel illustrerer oppfinnelsen.
Eksempel
283,3 g (1,2 mol) L-pyroglutaminsyre og 193,1 g
(1,7 mol) L-prolinamid ble først innført i en 10-liters tre-halset kolbe utstyrt med en rører og en dråpetrakt.
4 liter dimethylformamid ble deretter tilsatt og den resulterende løsning avkjølt til 0° C og under omrøring ble 488 g (2,37 mol) dicyclohexylcarbodiimid løst i 2 liter diklormethan tilsatt i løpet av 1 time. Under tilsetningen . ble temperaturen holdt mellom 0 og 5°.C. Omrøringen ble fort-satt i 4 8 timer mens blandingen gradvis fikk oppvarmes til omgivende temperatur. 3 liter vann ble deretter tilsatt for å utfelle det dicyclohexylurea som dannes fra dicyclohexyl-carbodiimidet. Bunnfallet ble filtrert fra og vasket med vann.' Filtratet ble fordampet til tørrhet og residuet tatt opp i 1 liter vann. Et bunnfall ble dannet og ble fjernet ved filtrering. Etter fordampning av vannet ble det erholdte oljeaktige residuum løst i 1 liter varm acetonitril og den gjennomsiktige løsning fikk krystallisere i et kjøleskap. Krystallene ..erholdt på denne måte ble filtrert fra, vasket med 100 ml acetonitril og tørket.
En første sats på 262,6 g L-pyroglutamyl-L-prolinamid ble således erholdt i ét utbytte på 63,5 % beregnet på pfolinamidutgangsmaterialet. En annen sats av produktet kan gjenvinnes fra modervæskene slik at utbyttet kan heves til ca-70 % av det teoretiske. L-pyroglutamyl-L-prolinamid-krystalliserer i form av et monohydrat, sm.p. 98° C, [a]D = -100°.

Kjernemagnetisk resonansspektrum nedtegnet med en
60 MHz Perkins-Elmer-apparatur og med D20/TMS:
Protonene av- amidgruppene av L-pyroglutaminsyren og av L-prolinamidet gjennomgår forandring med D^O og fremkommer derfor ved S . = 4,65 ppm.
Resonansene er blitt forklart ved sammenligning med resonansspekteret av pyroglutamyl og prolinamid-rester i molekylet av TRH (tyrodin-frigivende hormon) (se S. Fermand-. jian et al., FEBS Letters, 28 (1972),'156 og J.C. Boilot et al, CR. Acad. Sei. Paris, Series C 276 (1973), 217 - 220 .
Analyse av syrehydrolysatet av molekylet indikerer den korrekte støkiometri for de forskjellige bestanddeler: Glu/Pro/NH3:1/1/1.
Farmakologiske egenskaper
Det er kjent at som et resultat av aldring har gamle mennesker som tilsynelatende ikke lider av noen veldefinert sykdom en økende serie av mer eller mindre hurtige involusjons-prosesser som fremtrer som forstyrrelser i den generelle homeostasis.
Disse svekkelser er hyppige og fremtrer hovedsakelig ved nivået for tre endocrine akser: Insulinaksen, thyroid-aksen og gonadaksen.
Det er f.eks. vel kjent at insulinresponsen til for-døyelses- eller fremkalt hyperglycemi er forskjellige og be-tydelig redusert hos gamle mennesker i forhold til den respons som finnes i friske barn (prediabetes). Det samme gjelder jodbindingskapasiteten til thyroidkjertel (se Handbook of Endocrinology, R.D. Dillon, Lea and Febiger, Philadelphia., 1973) .
For tiden foreligger det ingen terapi som muliggjør
en effektiv bekjempelse av disse typer av svekkelser.
Det er nå funnet at den nye forbindelse ifølge oppfinnelsen, L-pyroglutamyl-L-prolinamid, er anvendbar for opp-retting av metabole eller endocrine forstyrrelser forbundet med alderdom. Denne forbindelse restituerer i særdeleshet organismens evne og dennes respons overfor visse stimuli,
f.eks. indusert hyperglycemi, mens den samtidig ikke stimule-rer frigivelse av insulin.
Farmakologiske tester
1. Indusert hyperglycemi eller glucose toleransetest

Glucosetoleransetesten består i oral administrering
til rotter av en dose på 1 g/kg glucose løst i 1 ml vann og måling i plasma av mengden frigitt insulin (IRI) etter 0, 15, 30, 60 og 120 minutter. Den normale respons på denne test er
en trekantet figur hvori den maksimale insulinfrigivelse er ved ca. 15 minutter.
Ti tolv-måneders gamle rotter behandles i 1 uke med fysiologisk saltvannløsning. På den siste dag gies de et overskudd av glucose i henhold til den ovenfor beskrevne test. Insulinresponsen er svak.
De samme dyr behandles deretter oralt i 5 dager med L^pyroglutamyl-L-prolinamid ved en dose på 4 0 mg/kg løst i 1 ml vann. Glucosetoleransetesten utføres deretter igjen som ovenfor beskrevet.
Resultatene er oppført i den etterfølgende tabell:

Det skal observeres at insulingrunnhastigheten før et overskudd av glucose gis (tid 0) ikke modifiseres ved behandlingen. Dette viser derfor at forbindelsen i seg selv ikke er et insulinf rigivningsmiddel .■
Hunner ble underkastet samme test. Ved en dose på 5 mg/kg ble den høyeste respons på L-pyroglutamyl-L-prolinamid
erholdt hos hunner som har de høyéste grunn glycemi og insulinemi. Dyr med en lav insulinhastighet men normal glycemi gir faktisk ingen respons på denne behandling. Ved lavere; doser
(1 mg/kg) hadde behandlingen faktisk ingen innflytelse på glucose og insulinhastighetene, med unntak for et drektig dyr som utviste høy insulinhastighet og normal glycemi. Etter behandling var både glycemi og insulinemi redusert til verdier nøyaktig sammenlignbare med de for de andre dyr.

2. C glucoseinnarbeidelsestest
Under aldring observeres en generell nedsettelse av cellulær metabolisme, dvs. både energiproduserte systemer (glycolyse, mitochondrisk oxydasjon etc.) og makromolekylære biosyntesesystemer (RNA og proteinsyntese etc).
Formålet med denne test er å følge innarbeidelsen av 14
C glucose i de makromolekylære deler av aldrede dyr. Den ble utført på ti 20-måneders gamle hun-NMRI-mus med en midlere vekt på 38 g + 5 g.
Etter 10 dagers behandling med L-pyroglutamyl-L-14 prolinamid (0,1 mmol/kg) og administrering av en dose av C glucose, ble dyrene avlivet 5 timer senere for å bestemme

Resultatene viser at innarbei-delse av den radioaktive forløper stimuleres til forskjellige grader i de organiske og proteine ekstrakter og i de fraksjo-ner som er rike på RNA og DNA. Under de anvendte eksperimen-telle betingelser ble de mest uttalte resultater erholdt i

hjernen (målt 5 timer etter C glucoseadministrering).
3. Dyrenes adferd
Under tester utført på mus ble det observert at aldrede mus som var 'blitt behandlet med L-pyroglutamyl-L-prolinamid var mer vaktsomme enn ubehandlede mus. Denne økning i vaktsomhet kunne beregnes ved å passere et likt an-tall behandlede og ubehandlede mus i et bur og deretter notere graden av inntagelse av dyrene. Fortolkningen ble statistisk gjort med mediantesten (W.J. CONOVER, Practical Non parametric Statistics, Wiley & Sons, 1971, s. 167). Det ble observert at denne forskjell i årvåkenhet mellom behandlede og ubehandlede dyr bare kunne finnes ved inntagelsesbevegelsen idet deres adferd ved hvile var identisk.
Farmasøytiske preparater inneholdende den nye forbindelse ifølge oppfinnelsen kan administreres intravenøst, intraperitonéalt, intramuskulært, subcutant og oralt f.eks. i doser på fra 1 til 200 mg/kg pr. dag.
Det etterfølgende er et eksempel på en farmasøytisk komposisjon ifølge oppfinnelsen:
En nr. 1 gelatinkapsel inneholder:
2 5 mg L-pyroglutamyl-L-prolinamid
400 mg lactose og
10 mg magnesiumstearat.
Claims (3)
1. Fremgangsmåte for fremstilling av L-pyroglutamyl-L-prolinamid, karakterisert ved at L-pyroglutamyl-syre omsettes med L-prolinamid i nærvær av en koblingsreaktant.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at koblingsreaktanten er valgt.fra gruppen bestående av dicyclohexylcarbodiimid og l-ethoxycarbonyl-2-ethoxy-1,2-dihydrokinolin.
3. Fremgangsmåte ifølge krav 1, karakterisert ved at reaksjonen utføres i et organisk medium valgt fra grupper, bestående av tetrahydrof uran, dimethylf ormamid, dioxan, ethylacetat og diklormethan.
