Farmasøytiske og kosmetiske sammensetninger for beskyttelse av hud fra skader indusert ved solstråling, inneholdende ingredienser av vegetabilsk opphav i tillegg til konvensjonelle solfiltre og eksipienser.
Description
FARMASØYTISKE OG KOSMETISKE SAMMENSETNINGER FOR BESKYTTELSE AV HUD MOT SOLSTRÅLER
Sammenfatningsvis er oppfinnelsen definert slik som gjort rede for i vedføyde krav-sett.
Foreliggende oppfinnelse vedrører farmasøytiske og kosmetiske sammensetninger for beskyttelse av huden mot skader forårsaket av solstråling, inneholdende ingredienser av vegetabilsk opphav, i tillegg til konvensjonelle solfiltre og eksipienser.
Foreliggende oppfinnelse vedrører nærmere bestemt farmasøytiske og kosmetiske sammensetninger for beskyttelse av hud fra skader indusert av solstråling, inneholdende fosfolipidkomplekser av flavanolignaner ekstrahert fra Silybum marianum; fosfolipidkomplekser av Vitis vinifera standardisert ekstrakt og fosfolipidkomplekser av triterpener ekstrahert fra Glycyrrhiza glabra, i tillegg til konvensjonelle solfiltre og eksipienser.
Formuleringen av preparater inneholdende høybeskyttelse, spesifikke solfiltre har vært en av de ledende sektorene av dermokosmetisk forskning. Dette er på grunn av en økende etterspørsel på markedet på grunn av både en evig økende oppmerk-somhet blant publikum av de farlige effektene av solstråling og problemet med en reduksjon av ozonnivåene i atmosfæren.
Forskning på dette feltet har tillatt formuleringen av varierte preparater, avhengig av behovene: produkter for intens eksponering til sollys (for eksempel ved sjøen eller på fjellet), for daglig bruk (antialdring) eller ved spesielle anvendelser slik som tilfellet hvor eksponering for UV-stråling i arbeidssammenheng, eller ved sykdom-mer av pigmentsystemet (albinisme, vitiligo).
Det har videre nylig vært en vekst i kunnskapen om de molekylære mekanismene ansvarlig for konsekvensene av mangel på beskyttelsen fra sollys. Disse tillater formuleringen av preparater i stand til å motstå, ved ethvert nivå, de alvorlige konsekvensene som kan forårsakes av ikke-beskyttet eksponering til sollys.
Det er godt kjent at lang eksponering for sollys, så vel som ultrafiolett stråling, kan forårsake dermatologiske forstyrrelser, til og med i alvorlig grad. Solemisjonsandelen som i dag erkjennes å være årsaken til hudendring, består av ikke-ioniserende elektromagnetisk stråling av UV-spektrumet. UV-strålingene som når jordens overflate er UV-A (320 til 400 nm) og UV-B (290 til 320 nm).
Forstyrrelser indusert av UV-strålinger er heri generelt henvist til som "fotodermatose".
Mediert (toksiske eller allergiske reaksjoner til sollys) og idiopatiske (urticaria sola-ris, polymorf dermatitt, aktinisk retikuloid) fotodermatose, så vel som de utsatt for forverring ved sollys (pellagra, lupus erytematose, pemfigus erytematose, xero-derma pigmentosum, dermatomyositt) kan anses å være noe sjeldne konsekvenser av eksponering for lys. I motsetning er direkte fotodermatose mye hyppigere.
Fotodermatose kan forekomme som erytem/ødem vesiculose reaksjon (akutt skade), fotoaldring og fotokarsinogenese (kronisk skade).
Ervtem
Erytem er den hyppigste hudreaksjonen til UV-stråling. Den er doseavhengig og kan variere fra beskjeden, asymptomatisk hudrødme til alvorlig erytem som kan være assosiert med en smerte, ødem og dannelse av vesikler. Erytem resulterer fra en perifer kappilær vasodilatasjon og er konsekvensen av både en direkte interaksjon med blodårer og en fotokjemisk reaksjon utløst av solstråler på vevkromo-forer (aromatiske aminosyrer, nitrogenøse baser, umettede fettsyrer, etc). Et an-tall formidlere er tilsynelatende involvert i disse interaksjoner: prostaglandiner av E- og D-seriene, interleukiner og primært frie radikaler. Sistnevnte frigjøres som følge av energien overført fra kromoforene til molekylær oksygen, og er ansvarlig for det meste av vaskulær skade forårsaket av eksponering til sollys.
Fotoaldring
Hudaldring er en kompleks biologisk prosess som påvirker forskjellige hudlag inklu-dert dermisen. Aldringsprosessen til de hudområdene som er eksponert for atmosfæriske agenser er, i tillegg til den såkalte medfødte eller indre aldring, nemlig den irreversible, degenerative prosessen assosiert med aldring som påvirker både huden og de indre organene, også grunnet ytre eller fotobestemt aldring som er primært koplet til ultrafiolett eksponering.
Ifølge nyere undersøkelser bestemmes 80 % av den totale skaden i områder eksponert for atmosfæriske agenser ved eksponering til sollys. Fra et kosmetisk og farmasøytisk synspunkt er det imidlertid klart at tiltak kan kun utøves mot den andre typen av aldring.
Tilveiebringelse av oksygenfrie radikaler som de nevnt over, fremmer videre oksidativt stress som skader slike vevsmessige strukturer som endotele membraner, proteiner, nukleinsyrer og bindevevstråder. Konsekvensene er: overdrevet mikrosirkulasjonspermeabilitet som forårsaker vevhypoksi og
ødem;
proteindenaturering som er særlig farlig i tilfelle av strukturelle proteiner og
enzymer;
lipidperoksidering som fører til celledød;
bindevevssklerose.
En videre skade indusert av UV-stråling er en økning i transkripsjons- og transla-sjonsprosessene til et bestemt DNA-segment som koder for noen metalloproteiner som utøver en effektiv fordøyningsaktivitet på kollagen. Dette skjer ved en hastig-het som er markant høyere enn ved reparasjonsprosessene som innvirker på bin-devevsmatrisene, resulterende i hudskade og dannelse av rynker.
Fotokarcinooenese
Epidemiologisk og klinisk bevis har nå vist hvor dypt solstråling, særlig UV-stråling, er involvert i utviklingen av noen kreftforstadier (aktinisk keratoser) som kan utvik-les til neoplastiske lesjoner, slik som basalcelleepiteliom, spinocellulær epiteliom og melanom, som er den mest skadelige.
Hittil er det tre mekanismer som tilsynelatende er involvert i utviklingen av neoplasmer forårsaket av UV-stråling: mutasjon indusert ved direkte interaksjon med nitrogenbaser, interaksjon med frie radikaler og DNA, og UV-mediert immun-hemning indusert ved hudnivået på nøkkelceller i immunresponsen, nemlig dendrit-tiske celler som ikke lenger er i stand til å utøve en effektiv immun respons som kan undertrykke tumoral lesjon.
Som en konklusjon er det tydelig at et verktøy rettet mot å tilveiebringe en full-stendig beskyttelse av huden mot enhver UV-mediert skade, bør være i stand til å filtrere UV-stråling så vel som å fungere mot oksidativt stress, fordøyelse av binde- vevsmukopolysakkaridfibrene, mutagenetisk aktivitet og UV-mediert immunhem-ning. Imidlertid tilfredsstiller ikke preparatene på dagens marked alle de oven-nevnte siterte behovene.
Foreliggende oppfinnelse tilfredsstiller behovene antydet ovenfor og tilveiebringer farmasøytiske og kosmetiske sammensetninger som, i tillegg til konvensjonelt solf-ilter og eksipienser, også inneholder minst to av følgende ingredienser av vegetabilsk opphav:
De farmasøytiske og kosmetiske sammensetningene ved foreliggende oppfinnelse inneholder foretrukket minst to av de følgende ingrediensene av vegetabilsk opphav i følgende prosentandeler:
i kombinasjon med konvensjonelle solfiltre og eksipienser.
Fosfolipidkompleksene av flavanolignanene ekstrahert fra Silybum marianum (også henvist til som "bestanddelene a") er beskrevet i EP 0209038.
De er særpreget ved svært gode antioksideringer og antiinflammatoriske egenskaper. I undersøkelser ved anvendelse av CCU som en prooksidant stimulans og he-patiske mikrosomer som et mål, tilsvarer LC50 (mikrogram per ml) av bestanddelene a) 25,0, mens det for vitamin E (som referanseforbindelse) er 30,0. Med hensyn til antiinflammatorisk aktivitet, viste bestanddelene a) seg å være i stand til hemming ved 75 % ødem indusert i den eksperimentelle dyrepoten gjen nom inokulasjon av krotonolje. Hos mennesker viste bestanddelene a), undersøkt i 20 friske frivillige mot placebo, seg å være i stand til å hemme ved 25 % eryte-matosereaksjon fremmet ved eksponering av hud for ultrafiolett stråling.
Fosfolipidkompleksene av standardisert ekstrakt av Vitis vinifera (også henvist til som "bestanddel b") er beskrevet i EP 0275224.
In wfro-assayer viste svært kraftfull antioksidant aktivitet av bestanddelene b). I undersøkelsene som benyttet Fe<+++>/ADP som en stimulans og liposomale preparater som målet for oksidering, viste bestanddelene b) seg å være 35 ganger mer aktive enn katekin og 50 ganger mer aktive enn vitamin E. Det bør også under-strekes at disse kompleksene hemmer ved 95 % lipid peroksidering indusert ved ultrafiolett stråling, selv ved konsentrasjoner så lave som 10 mikromol.
Bestanddelene b) viste seg også å være i stand til å hemme, selv på en ikke-konkurrerende måte, enhver elastase, kollagenase og hyaluronidase til stede i be-tent vev: denne egenskapen er svært nyttig for et produkt utviklet for å motvirke hudfotoaldring.
I nyere mutageneseassayer viste bestanddelene b) svært bra antimutageniske egenskaper. Undersøkelser på spontan mutasjon i Saccharomyces cerevisiae (gjær og eukaryotiske celler) viste at disse kompleksene kan snu mutasjonen av mito-konder og nukleær DNA ved henholdsvis 60 % og 90 % (0,5 mg/ml).
Anti-immunhemmende egenskaper av bestanddelene b) ble også vurdert. Bestanddelene b), applisert topisk på huden av nakne mus og undersøkt mot placebo, hemmet UV-mediert hud immunsuppresjon ved ca 50 %. Hos mennesker viste en undersøkelse hos 18 friske frivillige en hemming på 30 % av erytem reaksjon frem-stilt ved eksponering av hud til ultrafiolett stråling.
Fosfolipidkompleksene av triterpenene ekstrahert fra Glycyrrhiza glabra (også vist til som "bestanddelene c") er beskrevet i EP 0283713.
De utøver kraftfull anti-inflammatorisk virkning ved topisk administrasjon og hemmer omdannelsen av kortisol fra dets aktive form til den inaktive formen gjennom inhibisjon av vevsmessig lip-hydroksysteroiddehydrogenase, noe som forlenger den anti-inflammatoriske virkningen av kortisol frigjort etter en inflammatorisk stimulans. Bestanddelene c) viste seg å være hemmende ved 95 % ødem indusert i eksperimentelle dyrepoter gjennom inokulasjon av krotonolje, og viste en større effektivitet enn vanlige ikke-stereoide anti-inflammatoriske stoffer (i dette tilfellet indometacin).
Bestanddelene a), b) og c) ble også toksikologisk testet. Resultatene av akutt tok-sisitetstestene utført i rotter er sammenfattet i følgende tabell.
Akutte og kronisk kutane irritasjonstester i øyet utført på kaniner påviste videre at bestanddelene a), b) og c) ikke utøvde irritasjonsvirkninger.
Verken sensitisering- eller intoleransesymptomer ble observert i hudtester på friske frivillige.
Sammensetningene ved foreliggende oppfinnelse vil bli administrert topisk i form av egnede formuleringer, både likvid (slik som gel, lotions, melkelignende preparater, emulsjoner, skum og lignende) og faste eller semi-faste (slik som kremer, salver, leppestifter og lignende). Formuleringene vil fremstilles ifølge konvensjonelle me-toder slik som de beskrevet i "Remington's Pharmaceutical Handbook", Mack Pub-lishing Co, NY, USA, sammen med egnede eksipienser slik som bløtgjøringsmidler, fuktmidler, tykningsagenser, emulgeringsmidler, fargestoffer, smaksstoffer og lignende.
Claims (3)
1. Farmasøytiske og kosmetiske sammensetninger for beskyttelse av hud fra skader indusert ved solstråling, inneholdende minst to av de følgende ingrediensene av vegetabilsk opphav:
i kombinasjon med konvensjonelle solfiltre og eksipienser.
2. Farmasøytiske og kosmetiske sammensetninger ifølge krav 1 inneholdende minst to av de følgende ingrediensene av vegetabilsk opphav:
i kombinasjon med konvensjonelle solfiltre og konvensjonelle eksipienser.
3. Anvendelse av minst to av de følgende bestanddelene: a) fosfolipidkomplekser av flavanolignaner ekstrahert fra Silybum marianum; b) fosfolipidkomplekser av Vitis vinifera polyfenolsk fraksjon; c) fosfolipidkomplekser av triterpener ekstrahert fra Glycyrrhiza glabra;
for fremstillingen av farmasøytiske og kosmetiske sammensetninger for be skyttelse av hud fra skader indusert ved solstråling.
