Foreliggende oppfinnelse angår hyperforin- og adhyperforinderivater samt anvendelse derav i det farmasøytiske og/eller ernæringsmessige område, spesielt til behandling av depresjon og Alzheimers sykdom.
TEKNISK BAKGRUNN
De blomstrende topper av Hypericum perforatum inneholder en rekke klasser struk-turelt forskjellige substanser som virker direkte eller indirekte på sentralnervesys-temet. Virkningsmekanismene for disse forbindelser er forskjellige og omfatter anti-MAO-virkning (Suzuki OR. et al. Planta Med., 272-4, 1984), virkning på sero-tonin-frigjøring og gjenopptak (Muller W. E. et al., Pharmacopsychiatry, 30, 102-107, 1997) og benzodiazepin-liknende aktivitet (Coot J. M., Pharmacopsychiatry 30, 108-112, 1997).
Hyperforin, som er et floroglukin-derivat, er en av hovedkomponentene av den lipo-file fraksjon fra de blomstrende topper av Hypericum perforatum, hvor nevnte fraksjon også inneholder adhyperforin som er en høyere homolog av hyperforin, om enn i lavere konsentrasjon (Erdelmeier C. A. J., Pharmacopsychiatry, 31, 2-6, 1998).
Hyperforin har nylig vært gjenstand for en mengde studier som etablerer dens vik-tige rolle som et antidepressivt middel (Pharmacopsychiatry, 31, Suppl. 1, 1-60, 1998). Videre er det etablert at ekstraktene av Hypericum perforatum kan bli brukt til profylakse og behandling av neurodegenerative sykdommer, blant annet Alzheimers sykdom (WO/9940905, WO 0057707). Spesielt ble hyperforin- og adhyperforin-salter med uorganiske kationer eller ammoniumsalter beskrevet for dette formål (WO9941220).
Det er kjent fra litteraturen at hyperforin er lite stabilt under vanlige ekstraksjons-og lagringstilstander; i henhold til WO 97/13489 faller hyperforininnholdet i et St. John's Wort vann-alkoholisk ekstrakt allerede etter noen uker. WO 97/13489 angir videre at for å oppnå stabile hyperforinekstrakter bør antioksydanter være til stede under hele opparbeidelsen (ekstraksjon, opprenskning og lagring). Det er følgelig tydelig at den store ustabilitet av hyperforin gjør fremstillingen av farmasøytiske hyperforinformuleringer relativt vanskelig. For å fjerne nevnte ulempe har forbindelser som er mer stabile enn hyperforin, så som saltene beskrevet i WO 99/41220 og de hydroksyfunksjonaliserte derivater (WO 99/64388) angitt ovenfor, nylig blitt fremstilt.
Det er videre kjent (Bystrov et al., Bioorg. Khim, 1978) at hyperforin og adhyperforin kan bli omdannet til de tilsvarende octahydroderivater, octahydrohyperforin (Ia) og octahydroadhyperforin (Ib) ved katalytisk reduksjon av isopren-sidekjedene. eller til de tilsvarende tetrahydroderivater, tetrahydrohyperforin (Ic) og tetrahydroadhyperforin (Id) ved reduksjon av keto-gruppene ved 1- og 10-posisjonene til hydroksygrupper med metallhydrider.
DETALJERT BESKRIVELSE AV OPPFINNELSEN
Det har nå blitt funnet at hyperforin- og adhyperforinderivater som kan fremstilles ved reduksjon av alle dobbeltbindinger av isoprenkjedene av ketogruppene ved 1-og 10-posisjonene til hydroksygrupper, ikke bare har høy stabilitet, men har også antidepressive, anxiolytiske og anti-neurodegenerative aktiviteter som overrasken-de er høyere enn hyperforin og adhyperforin.
Målet for foreliggende oppfinnelse er følgelig anvendelse av hyperforin- og adhyperforinderivater av formel (I)
hvor R er metyl eller etyl, R2 er hydrogen, et farmasøytisk akseptabelt uorganisk eller organisk base-kation eller et rett eller forgrenet C2-C5 acyl-residuum, og hvor, alternativt: a) Ri er 3-metyl-but-l-yl og okso-grupper er til stede ved 1- og 10-posisjonene; b) Ri er 3-metyl-2-buten-l-yl og hydroksygrupper er til stede ved 1- og 10-posisjonene; c) Ri er 3-metyl-but-l-yl og hydroksygrupper er til stede ved 1- og 10-posisjonene
ved fremstilling av medikamenter, spesielt ved fremstilling av medikamenter til behandling av depresjon og Alzheimers sykdom.

Dodecahydrohyperforin (le), dodecahydroadhyperforin (If), acetyloktahydrohyperforin (li) og acetyloktahydrohyperforin (II) er nye forbindelser og er også del av foreliggende oppfinnelse.
Forbindelsene av formel (Ia) og (Ib) blir oppnådd ved reduksjon av isopren-sidekjedene ved katalytisk hydrogenering ved å bruke for eksempel palladium på trekull eller Nikkel/Raney.
Forbindelsen av formel (Ic) og (Id) blir oppnådd ved reduksjon av ketogruppene ved 1- og 10-posisjonene med hydrider valgt for eksempel fra NaBH4, Redal<®>, Vitri-de<®>, UAIH4.
Forbindelsene av formel (le) og (If) blir oppnådd ved først å redusere isopren-sidekjedene og så ketogruppene ved 1- og 10-posisjonene i henhold til hva som er beskrevet ovenfor.
Forbindelser av formel (I) hvor R2 er et uorganisk eller organisk basekation eller et acyl-residuum, kan bli fremstilt fra forbindelser av formel (I) hvor R2 er hydrogen ved salifisering eller esterifisering med konvensjonelle metoder.
Fremgangsmåten for fremstilling av forbindelsene ifølge oppfinnelsen ved å starte fra de blomstrende topper av Hypericum perforatum, kan bli oppsummert som føl-ger: De blomstrende topper av Hypericum perforatum kan bli ekstrahert med alkoholer eller alifatiske ketoner som enten er rene eller i en blanding derav med vann eller med gass under superkritiske forhold, hvor det resulterende ekstrakt blir fordelt mellom n-heksan og vandige oppløsninger av alifatiske alkoholer. Heksanoppløs-ningen ekstraheres med alkalisk metanol for å ekstrahere hyperforin og adhyperforin. Den metanoliske oppløsning surgjøres og behandles med et svakt basisk io-nebytterresin som selektivt holder tilbake hyperforin og adhyperforin. Resinet elueres med sur metanol og eluatet konsentreres til et lite volum, fortynnes så med vann og tilbake-ekstraheres med n-heksan. Heksanoppløsningen konsentreres til et lite volum og det resulterende konsentrat er klart for derivatisering. Residuet tas opp i klorinerte oppløsningsmidler og den egnede reaktant tilsettes til dette i henhold til fremgangsmåter som er beskrevet i eksemplene.
Forbindelsene ifølge oppfinnelsen har vist antidepressiv effekt som ble evaluert i rotte ved tvungen svømmetest som evaluerer parametrene: Kjemping, flyting og svømming i henhold til hva som er beskrevet av Cervo et al. i Neuropharmacology, 26, 14969-72, 1987. Forbindelsene ble administrert i 3 doser; 30 minutter etter pre-testen, 5 timer og 30 minutter før testen. Resultatene angitt i tabellen nedenfor viser at forbindelsene ifølge oppfinnelsen er mer aktive enn det opphavelige hyperforin.
Forbindelsene ifølge oppfinnelsen viste seg også spesielt aktive mot Alzheimers sykdom grunnet deres egenskap å øke APPs, som er den oppløselige, harmløse form av Alzheimer Precursor Protein (APP). Det er faktisk kjent at proteolytisk spaltning av Alzheimers Precursor Protein (APP) blir fremmet både av p- og y-secretase som induserer en øket produksjon av amyloid peptid Abl-42 (som også spiller en sentral rolle i opptreden av Alzheimers sykdom) samt a-secretase som gir opphav til oppløselige APPs som ikke har noen patogen aktivitet (Eslr W. P., Wolfe M. S., Science, 293, 1449-54, 2001).
Effekten av forbindelsene ifølge oppfinnelsen, på frigjøringen av APPs dannet av a-secretase ble vurdert i dyrkningsmediet av en neuroblastoma-cellelinje (SH-SY5Y) i henhold til fremgangsmåten beskrevet av Galbete J. L, et al. i Biochem J. 348, 307-313, 2000.
Resultatene rapportert i den følgende tabell viser at de undersøkte forbindelser ak-tiverer a-secretase-mediert APP-metabolisme innbefattende en økning i APPs utskilt i dyrkningsmediet:
Forbindelsene ifølge oppfinnelsen kan bli formulert i henhold til konvensjonelle tek-nikker, for eksempel i henhold til hva som er beskrevet i Remington's Pharmaceuti-cal Sciences Handbook, XVII Ed. Mack Pub., N.Y., U.S.A., i form av mykgelatinkapsler, hardgelatinkapsler, tabletter, suppositorier; fortrinnsvis er ekstraktet ifølge oppfinnelsen formulert i mykgelatinkapsler eller i formuleringer for kontrollert fri-gjøring. Dosene ligger i området fra 10 til 100 mg per enhetsdose i vanlige formuleringer og opp til 200 mg i formuleringer for kontrollert frigjøring, hvor i dette til-fellet, den foreslåtte dose er 200 mg per dose/dag. Videre kan forbindelsene bli administrert via den transdermale rute med kontrollert frigjøring ved å tilføre for-muleringen i det proksimale området til delingen av den cerebrale karotide arterie. Dosene av forbindelse i disse formuleringer ligger i området fra 10 til 100 mg per dose/dag.
Eksemplene angitt nedenfor illustrerer oppfinnelsen i større detalj.
EKSEMPLER
Eksempel 1 - Fremstilling av hyperforin
10 kg blomstrende topper av Hypericum perforatum og 30 I metanol ekstraheres i et 50 I ekstraksjonsoppsett og massen blir oppbevart ved romtemperatur i 3 timer, idet ekstraksjonen gjentas 3 ytterligere ganger, og derpå blir de kombinerte ekst-rakter konsentrert under vakuum til 5 kg og konsentratet ekstraheres med 3x51 heksan. Vann-metanol-oppløsningen kastes mens heksanet blir tilbakeekstrahert med alkalisk metanol (KOH) inntil utarming av hyperforin og ad hyperforin.
Oppløsningen nøytraliseres og filtreres gjennom et svakt basisk Amberlite-resin som selektivt holder tilbake hyperforin og adhyperforin, hvor det tilbakeholdte produkt elueres igjen med metanol surgjort med fosforsyre, det metanoliske eluat konsentreres under vakuum ved 25 °C, derpå fortynnes med vann og tilbakeekstra-heres med n-heksan inntil utarming av hyperforin.
De kombinerte organiske lag avfarges med 0,3 % trekull, tørkes derpå over Na2S04 og konsentreres til en olje under 40 °C under vakuum. Etter solidifisering gir oljen en voks (0,52 kg) inneholdende omtrent 90 % av hyperforin.
Eksempel 2 - Fremstilling av oktahvdrohvperforin- dicvkloheksvl-ammoniumsalt
50 g hyperforin fremstilt som beskrevet i eksempel 1, oppløses i 500 ml etylacetat ved nærvær av 2 g 5 % palladium på trekull og hydrogeneres inntil fullstendig hy-drogenabsorpsjon. Katalysatoren filtreres fra, oppløsningen konsentreres til tørrhet under vakuum og residuet oppløses i n-heksan. Oppløsningen tilsettes en støkio-metrisk mengde av dicykloheksylamin for å oppnå en tilstrekkelig selektiv krystallisering av det tilsvarende salt. 62 g av oktahydrohyperforin-dicykloheksylammonimsalt blir oppnådd og som har de følgende karakteristika: <1>H-NMR (300 MHz CDCI3): 5 3,03 (2H, m, CH-DCHA), 2,55-2,30, 2,10-1,76 (20H, m, CH2-DCHA), 1,70-1,10 (22H, m, H-4, H-ll, CH2-5, CH2-15, CH2-16, CH2-17, CH2-21, CH2-22, CH2-26, CH2-27, CH2-31, CH2-32), 0,97-0,83 (24H, d, CH3-19, CH3-20, CH3-24, CH3-25, CH3-29, CH3-3O, CH3-34, CH3-35), 1,19, 1,12 (6H, d, J = 6,5 Hz, CH3-I2, CH3-13), 0,91 (3H, s, CH3-14).

Eksempel 3 - Fremstilling av tetrahydrohyperforin
2 g hyperforin (MW = 536,01) oppløses i 20 ml THF under magnetisk omrøring, oppløsningen tilsettes LiAIH4 i stort overskudd (1 g, 0,026 mol, MW = 38). Reak-sjonsforløpet overvåkes med TLC (elueringsmiddel petroleumeter/EtOAc 9:1). Etter ti minutter er reaksjonen fullstendig.
Na2SO410H2O støttet på Celitt (3:1 per vekt) tilsettes for å ødelegge det reaktive overskudd; reaksjonen er sterkt eksoterm og følgelig bør den bli avkjølt med is. Deler av oppløsningsmiddelet avdampes på grunn av den utviklede varme. Blandingen filtreres gjennom Celitt og filtratet vaskes tre ganger med 20 ml AcOEt. Oppløsningen plasseres i en 150 ml rundbunnet kolbe med hals og oppløsningsmid-delet dampes bort fullstendig.
Den resulterende blanding renses med kolonnekromatografi ved å bruke en 200 ml kolonne pakket med 100 ml silikagel og petroleumeter/EtOAc 95:5 som eluerings-blanding. Eluatfraksjoner på omtrent 20 ml samles opp og innholdet undersøkes med TLC (petroleumeter/EtOAc 9:1). Produktet det er mest av (1,5 g) krystallisert fra metanol har de følgende spektroskopiske egenskaper: <1>H-NMR (300 MHz CDCI3): 5 5.11 (1H, m, H-22), 5,00 (3H, m, H-17, H-27, H-32), 3,11 (1H, dd, J = 14,0, 7,4 Hz, CH2-26), 2,92 (1 H, dd, J = 14,0, 7,0 Hz, CH2-26), 2,50-1,35 (12H, m, H-4, H-ll, CH2-5, CH2-15, CH2-16, CH2-21, CH2-31), 1,80-1,52 (24H, s, CH3-19, CH3-2O, CH3-24, CH3-25, CH3-29, CH3-3O, CH3-34, CH3-35), 1,19-0,95 (9H, d, CH3-I2, CH3-I3, CH3-14).

Eksempel 4 - Fremstiling av oktahvdroadhvperforin- litiumsalt
15 g oktahydrohyperforin-dicykloheksylammoniumsalt elueres på et surt resin (Do-wex 50X8, 300 g) med 600 ml metanol. 11,01 g av oktahydrohyperforin blir oppnådd, som blir tilsatt 0,8745 g LiOH-monohydrat oppløst i vann. Blandingen inndampes til tørrhet for å oppnå 11,41 g litiumsalt som har de følgende spektroskopiske karakteristika: <1>H-NMR (300 MHz CDCI3): 5 1,93-1,00 (22H, m, H-4, H-ll, CH2-5, CH2-15, CH2-16, CH2-17, CH2-21, CH2-22, CH2-26, CH2-27, CH2-31, CH2-32), 1,00-0,80 (24H, d, CH3-I9, CH3-2O, CH3-24, CH3-25, CH3-29, CH3-3O, CH3-34, CH3-35), 1,20, 1,06 (6H, d, J = 6,3 Hz, CH3-I2, CH3-13), 0,91 (3H, s, CH3-14).

Eksempel 5 - Fremstilling av dodekahvdrohvperforin
1,72 g dicykloheksylammonium-oktahydrohyperforinat (MW = 716; 2,41 mmol) oppløses i 20 ml TH F under magnetisk omrøring; oppløsningen tilsettes et stort overskudd (3,5 g) av LiAIH4 (MW = 38, 0,092 mol). Forløpet av reaksjonen overvåkes med TLC (elueringsmiddel petroleumeter/EtOAc 9:1). Etter ti minutter er reaksjonen fullstendig.
Det reaktive overskudd ødelegges som beskrevet i eksempel 5. Blandingen filtreres og residuet vaskes grundig med etylacetat. Oppløsningen inndampes til tørrhet, reaksjons-grovproduktet oppløses i 15 ml petroleumeter/etyleter 3:1 og oppløs-ningen plasseres i en 150 ml separasjonstrakt. Den organiske fase vaskes tre ganger med 2N svovelsyre og derpå med saltlake. Den vandige fase kastes, den organiske tørkes over Na2S04 og konsentreres til tørrhet. Det resulterende produkt renses med kolonnekromatografi på 75 g silikagel under eluering av den ønskede forbindelse med petroleumeter/etylacetat 99:1. 0,9 g av dodekahydrohyperforin blir oppnådd som har de følgende spektroskopiske karakteristika:
EIMS m/ z 548[M]<+>.
Eksempel 6 - Fremstilling av acetyloktahydrohyperforin
300 mg acetylhyperforin (MW = 578; 0,52 mmol) oppløses i 3 ml MeOH i en tohal-set rundbunnet kolbe, derpå tilsettes katalysatoren (5 % Pd på trekull). Reaksjonen overvåkes med TLC (petroleumeter/EtOAc 95:5 Rfp = 0,43; Rfa = 0,52). Etter fire timer er reaksjonen fullstendig. Katalysatoren filtreres fra gjennom et lag av Celitt, derpå avdampes metanolen.
Reaksjonsproduktet renses med kolonnekromatografi på 30 g silikagel under eluering med en petroleumeter/etylacetat 9:1 blanding. Krystallisering fra metanol gir 150 mg av den ønskede forbindelse som har de følgende spektroskopiske karakteristika:
EIMS m/ z 586 [M]<+>.

1. Hyperforin- og adhyperforin-derivater av formel (I)
hvor R er metyl eller etyl, R2 er hydrogen, et farmasøytisk akseptabelt uorganisk eller organisk basekation eller et rett eller forgrenet C2-C5 acylresiduum, og hvori alternativt: a) Ri er 3-metyl-but-l-yl og oksogrupper er til stede ved 1- og 10-posisjonene; b) Ri er 3-metyl-2-buten-l-yl og hydroksygrupper er til stede ved 1-og 10-posisjonene; c) Ri er 3-metyl-but-l-yl og hydroksygrupper er til stede ved 1- og
10-posisjonene;
for anvendelse som medikamenter.
2. Derivater ifølge krav 1, for fremstilling av medikamenter for anvendelse ved behandling av depresjon eller Alzheimers sykdom.
