Description
Teknisk område
Den foreliggende oppfinnelse angår en stabil salve inneholdende acetylsalisylsyre.
Nærmere bestemt angår oppfinnelsen en vannfri salve inneholdende acetylsalisylsyre hvor det som en basis anvendes hydrokarbongel og/eller vaselin (pet-rolatum) som kan bevare acetylsalisylsyre i stabil tilstand i lang tid.
Bakgrunnsteknikk
Acetylsalisylsyre er fra gammelt av blitt anvendt som et anti-infiamma-torisk, antipyretisk analgetikum. Den administreres vanligvis oralt i form av tabletter, granuler etc.
På grunn av den skadelige virkning av acetylsalisylsyre på mage- og tarmsystemet, er imidlertid den eksterne applisering av acetylsalisylsyre undersøkt i den senere tid, og dette er gjort for å forsøke å gjøre acetylsalisylsyren perkutant absorberbar. Resultatene er rapportert i japansk patentpublikasjon nr. 3-72426 når det gjelder salver for behandling av neuralgi, i japansk patentpublikasjon nr. 6-72879 når det gjelder plastre inneholdende acetylsalisylsyre, og i japansk patentpublikasjon nr. 6183980 når det gjelder en metode for stabilisering av acetylsalisylsyre i plastre. Plastre inneholdende acetylsalisylsyre er dessuten beskrevet i japansk patentpublikasjon nr. 8-113531. Teknikker, slik som forbedring av transdermal absorpsjon av acetylsalisylsyre og stabilisering av acetylsalisylsyre i plastret, er hovedsakelig beskrevet i disse publikasjoner. Teknikker for å bevare acetylsalisylsyre stabilt i salver i en lang tidsperiode, er imidlertid ikke beskrevet.
Publikasjoner hvor det beskrives metoder for stabilisering av acetylsalisylsyre i preparater, med unntak av eksterne preparater, er japansk patentpublikasjon nr. 56-32425, japansk patentpublikasjon nr. 62-89619 og japansk patentpublikasjon nr. 4-346930.
I US 4 012 508 beskrives en salve som omfatter en liten mengde kortikalt steroid intimt blandet med acetylsalisylsyre i vaselin. Salven anvendes topisk mot hudlidelser.
I GB 469 526 beskrives fremstilling av en salve ved å oppløse acetylsalisylsyre i en hovedsakelig vannfri basis som kan være glykoldioleat og/eller kamferolje eller andre planteoljer, og salven kan også inneholde andre ingredienser. Salven skal gi forbedret opptak av acetylsalisylsyre gjennom huden.
Fordi acetylsalisylsyre lett hydrolyseres, selv i nærvær av små mengder vann, og fordi hydrolysen akselereres avhengig av typen additiver, er det i disse publikasjoner, for å unngå kontakt med additivet, beskrevet å anvende et beskyttende lag bestående av sukrose, eller å anvende bindemidler hvori vann er blitt ekskludert i mest mulig grad, og å tilsette en hydrogenert olje som et smøremiddel. Det er imidlertid neppe mulig å anvende slike teknikker på salver.
For eksterne acetylsalisylsyrepreparater er det hittil ikke vist noen teknikk som sikrer stabiliteten av acetylsalisylsyre ved fremstilling av salver.
Beskrivelse av oppfinnelsen.
Den foreliggende oppfinnelse ble foretatt for å løse de ovenfor angitte problemer, og målet for den foreliggende oppfinnelse er å tilveiebringe salver som inneholder acetylsalisylsyre, som har en overlegen stabilitet og som kan lagres i lang tid.
Med den foreliggende oppfinnelse tilveiebringes således en salve som er kjennetegnet ved at den består av acetylsalisylsyre og hydrokarbongel og/eller vaselin uten noen andre additiver, hvor salven ikke inneholder noe vann for oppløsning av acetylsalisylsyren.
Hydrokarbongel og vaselin anvendes ifølge den foreliggende oppfinnelse i vesentlig vannfri tilstand. Vaselin er en vanlig anvendt basis for fremstilling av salver, slik som gul vaselin, hvit vaselin og blandinger av disse.
Mengden av acetylsalisylsyre i en salve ifølge oppfinnelsen er fra 0,001 til 30 vekt%, fortrinnsvis fra 0,01 til 20 vekt%, mer foretrukket fra 0,05 til 15 vekt%. Dersom mengden av acetylsalisylsyre er høyere enn 30 vekt%, er det umulig å opprettholde salveegenskapene, og basisens beskyttende effekt reduseres og forårsaker hydrolyse av acetylsalisylsyre. Når mengden av acetylsalisylsyre er lavere enn 0,001 vekt%, vil på den annen side acetylsalisylsyrens farmakologiske aktiviteter neppe kunne oppvises. Ingen av disse tilfeller er foretrukne.
Salven ifølge oppfinnelsen inneholdende acetylsalisylsyre fremstilles ved den samme metode som fremstilling av vanlige salver. Det vil si at etter smelting av en basis ved oppvarming, tilsettes finpulverisert acetylsalisylsyre under omrøring og materialene blandes til en salve.
Ved fremstilling av disse salver er det ingen tilsetning av vann for å oppløse acetylsalisylsyre.
Det er dessuten ikke tillsatt noe additiv som ødelegger stabiliteten av acetylsalisylsyre, slik som organisk syre, alkohol, flerverdig alkohol, overflateaktivt middel etc.
Beste måte for utøvelse av oppfinnelsen
Den foreliggende oppfinnelse er vist mer detaljert ved hjelp av de etterfølgende eksempler.
Eksempel 1
Hvit vaselin ble plassert i en vakuum-emulgator (T.K.ROBO MIXER produsert av Tokusyukika Kogyo) og smeltet under oppvarming ved 55 °C. Aspirinet ble tilsatt og blandingen ble omrørt under vakuum ved 2000 rpm i 15 minutter. Blandingen ble deretter avkjølt til 25 °C for å gi en salve inneholdende acetylsalisylsyre.
Eksempel 2
Gul vaselin og acetylsalisylsyre ble plassert i en rotasjonsblander og blandingen ble omrørt ved 100 rpm for å gi en salve inneholdende acetylsalisylsyre.
Eksempel 3
Hydrokarbongel og acetylsalisylsyre ble plassert i en rotasjonsblander og blandingen ble omrørt ved 80 rpm for å gi en salve inneholdende acetylsalisylsyre.
Eksempel 4
Hydrokarbongel og acetylsalisylsyre ble plassert i en rotasjonsblander og blandingen ble omrørt under vakuum ved 130 rpm i 20 minutter for å gi en salve inneholdende acetylsalisylsyre.
Eksempel 5
Ved anvendelse av de samme ingredienser og metoden beskrevet i eksempel 4, ble det fremstilt en salve inneholdende acetylsalisylsyre.
Eksempel 6
Ved anvendelse av den samme metode som beskrevet i eksempel 4, ble det fremstilt en salve inneholdende acetylsalisylsyre.
Sammenligningseksempel 1
Polyakrylsyre ble tilsatt til propylenglykol og blandingen ble smeltet ved oppvarming på et vannbad og omrørt. Aspirin ble deretter oppløst i blandingen og trietanolamin ble tilsatt til blandingen. Blandingen ble omrørt for å gi geldannende salver.
Sammenligningseksempel 2
Karboksymetylcellulose-natrium ble dispergert i etanol. Blandingen ble tilsatt til en blanding av glyserol og propylenglykol under omrøring. Aspirin ble deretter dispergert og oppløst i blandingen. Vann ble tilsatt og blandingen ble omrørt grundig for å gi en geldannende salve.
Sammenligningseksempel 3
Ved anvendelse av de følgende ingredienser ble det fremstilt hydrofil salve som beskrevet i japansk farmakope XIII, og acetylsalisylsyre ble tilsatt og blandet med denne.
Eksperiment 1
Salver ifølge foreliggende oppfinnelse fremstilt ifølge eksempler 1 og 5, og salver fremstilt ifølge sammenligningseksempler 1 til 4, ble testet for stabilitet ved 75% relativ fuktighet (RH) ved 40 °C og ved 50 °C. Testprøvene ble i hvert tilfelle lagret i 1 til 3 måneder, og etter prøveuttak ble det resterende acetylsalisylsyreinnhold i hver prøve målt og den resterende prosent av det opprinnelige innhold ble beregnet og er vist i tabell 1.
Eksperiment 2
Salver ifølge foreliggende oppfinnelse fremstilt ifølge eksempler 1 til 5 og salver fremstilt ifølge sammenligningseksempel 1, ble målt med hensyn til vann-innhold ved hjelp av et Karl-Fischer fuktighetsmålingsapparat, og resultatene er vist i tabell 2.
Industriell anvendelighet
Salvene ifølge foreliggende oppfinnelse inneholdende acetylsalisylsyre kan lagres stabilt i lang tid i form av en vesentlig vannfri salve inneholdende hydrokarbongel og/eller vaselin som en basis.
Claims (3)
1. Salve,
karakterisert ved at den består av acetylsalisylsyre og hydrokarbongel og/eller vaselin uten noen andre additiver, hvor salven ikke inneholder noe vann for oppløsning av acetylsalisylsyren.
2. Salve ifølge krav 1, hvor den består av acetylsalisylsyre og hydrokarbongel uten noen andre additiver.
3. Salve ifølge krav 1 eller 2, hvor innholdet av acetylsalisylsyre er i området fra 0,001 vekt% til 30 vekt% av totalvekten.
