Description
ENZYMATISK SYNTESE AV KIRALE AMINER SAMT FREMGANGSMÅTE FOR FREMSTILLING AV (S)-1-METOKSY-2-AMINOPROPAN
Foreliggende oppfinnelse angår forbedringer ved den enzymatiske syntese av kirale forbindelseir inneholdende en aminogruppe, f.eks. kirale aminer. Oppfinnelsen vedrører også en fremgangsmåte for fremstilling av (S)-l-metoksy-2-aminopropan.
US 4 950 606, 5 169 780, 5 300 437 og 5 360 724 beskriver enantiomeranrikning av kirale aminer ved bruk av en aminosyre-transaminase. Aminosyre-transaminaser er kjente pyridoksalfosfat-avhengige enzymer som finnes i forskjellige mikro-organismer inkludert Pseudomonas, Escherichia, Bacillus, Saccharomyces, Hansenula, Candida, Streptomyces, Aspergillus og Neurospora. To aminosyre-transaminaser, EC 2.6.1.18 og EC 2.6.1-19 er krystallisert og karakterisert av Yonaha et al., se "Agric. Biol. Chem.", 47 (10), 2257-2265 (1983).
US 4 950 606, 5 169 780 og 5 300 437 beskriver at individuel-le stammer av transaminaseholdige organismer kan isoleres ved kjemostatkultur, det vil si dyrking i en konstant, men be-grenset kjemisk omgivelse, med en aminoakseptor og et amin som eneste nitrogenkilde. Den typiske stamme som isoleres på denne måte i de angitte patenter, ble karakterisert {av "Ame-rican Type Culture Collection") som Bacillus megaterium. Van-ligvis metaboliserer ca-aminosyre-transaminaser aminosyrer hvori aminogruppe er på et terminalt, kiralt (ikke-kiralt) karbonatom, og aminet som benyttes som nitrogenkilde i en slik kjemostatkultur, kan være av den samme type, nemlig akirale aminer som n-oktylamin, cykloheksylamin, 1,4-butandiamin, 1,6-heksandiamin, 6-aminoheksansyre, 4-aminosmørsyre, tyramin og benzylamin. Det angis også imidlertid i disse patenter at aminet som benyttes som nitrogenkilde i en slik kjemostatkultur kan være et kiralt amin som 2-aminobutan, a-fenetylamin og 2-amino-4-fenylbutan. Kirale aminosyrer som L-lysin, L-ornitin, p-alanin og taurin kan også benyttes.
I tillegg til enantiomer-anrikning beskriver US 4 950 606,
5 169 780 og 5 300 437 den stereoselektive syntese av en kiral form av et amin ved innvirkning av en aminosyre-transaminase på et keton med formelen R<x>COR<2>, der R<1> og R<2> er forskjellige alkyl- eller arylgrupper, i nærvær av en aminodonor. Aminodonorene som beskrives, tilsvarer de aminer som benyttes som nitrogenkilder i kjemostatkulturene; dvs. akirale aminer hvori aminogruppen er på et terminal karbonatom som propylamin og benzylamin, kirale aminer der aminogruppen er på et terminalt karbonatom som (S)-2-aminobutan og kirale aminosyrer som L-alanin og L-aspartinsyre.
Foreliggende oppfinnelse er basert på den oppdagelse at det akirale amin 2-aminopropan uventet er overlegen som en amindonor ved slike transaminase-aminsynteser sammenlignet med enten akirale aminer der aminogruppen er på et terminalt karbonatom, eller kirale aminer der aminogruppen er på et ikke-terminalt karbonatom. Oppfinnelsen utgjør således en forbedring i den kjente stereoselektive syntese av et kiralt amin der et keton bringes i kontakt med en transaminase i nærvær av en aminodonor ved anvendelse av 2-aminopropan som amindonor .
Uttrykket kiralt amin benyttes her i bredeste forstand. Som beskrevet i patentene ovenfor, kan de kjente stereospesifikke synteser anvendes ved fremstilling av et vidt spektrum av alifatiske og alisykliske forbindelser av forskjellig og blandede funksjonelle typer, og som karakteriseres kun ved nærværet av en primær aminogruppe bundet til et sekundært karbonatom som, i tillegg til hydrogenatomet, bærer enten (i) en toverdig gruppe som danner en kiral, syklisk struktur, eller (ii) to substituenter (andre enn hydrogen) som skiller seg fra hverandre i struktur eller kiralitet.
Toverdige eller divalente grupper som danner en kiral, syklisk struktur inkluderer f.eks. 2-metylbutan-l,4-diyl, pen-tan-1,4-diyl, heksan-1,4-diyl, heksan-1,5-diyl, 2-metyl-pentan-1,5-diyl. Således kan den her beskrevne forbedring ved anvendelse av 2-aminopropan som amindonor brukes ved den ste-reospesif ikke syntese av l-amino-2-metylcyklopentan fra 2-metylcyklopentanon, l-amino-3-metylcyklopentan fra 3-metyl-cyklopentanon, l-amino-2-metylcykloheksan fra 2-metylcyklo-heksanon, osv.
De to forskjellige substituenter på det sekundære karbonatom (R<1> og R2 ovenfor) kan også variere innen vide grenser og inkluderer alkyl, aralkyl, aryl, halogen, hydroksy, lavereal-kyl, laverealkoksy, laverealkyltio, cykloalkyl, karboksy, karbalkoksy, karbamoyl, mono- og di-(laverealkyl)substituert karbamoyl, trifluormetyl, fenyl, nitro, amino, mono- og di-
(laverealkyl) substituert amino, alkylsulfonyl, arylsulfonyl, alkylkarboksamido, arylkarboksamido, osv., så vel som alkyl, aralkyl eller aryl substituert med de ovenfor nevnte.
Således kan den her beskrevne forbedring ved anvendelse av 2-aminopropan som amindonor, også benyttes ved stereospesifikk syntese av 2-aminobutan fra butanon, 2-amino-l-butanol fra 1-hydroksybutan-2-on, alanin fra pyruvinsyre, 1-amino-l-fenyl-etan fra acetofenon, 1-amino-l-(2-metoksy-5-fluorfenyl)etan fra 2-metoksy-5-fluoracetofenon, y-amino-pentansyre fra levu-linsyre, 1-amino-l-fenylpropan fra 1-fenylpropan-l-on, 1-amino-l- (4 -bromf enyl) propan fra 1-(4-bromfenyl)propan-l-on, 1-amino-1-(4-nitrofenyl)-propan fra 1-(4-nitrofenyl)propan-1-on, 1-fenyl-2-aminopropan fra 1-fenylpropan-2-on, valin fra 2-okso-3-metylbutansyre, 1-(3-trifluormetylfenyl)-2-aminopropan fra 1-(3-trifluormetylfenyl)propan-l-on, 2-amino-propanol fra hydroksypropanon, 1-metoksy-2-aminopropan fra metoksyoksypropanon, 1-amino-l-fenylbutan fra 1-fenylbutan-1-on, 1-fenyl-2-aminobutan fra 1-fenylbutan-2-on, l-(2,5-di-metoksy-4-metylfenyl)-2-aminobutan fra 1-(2,5-dimetoksy-4-metylfenyl)butan-2-on, 1-(4-hydroksyfenyl)-3-aminobutan fra 1- (4-hydroksyfenyl)butan-3-on, 1-amino-l-(2-naftyl)etan fra 2- acetylnaftalen, fenylalanin fra fenylpyruvinsyre, glutamin-syre fra 2-keto-glutarsyre, aspartinsyre fra 2-ketoravsyre og lignende.
I motsetning til de amindonorer som er beskrevet i den kjente teknikk og i særdeleshet majoritet av aminoalkan-aminodonorer som teoretisk er tilgjengelige, har 2-aminopropan den rela-tivt unike kombinasjon av (i) å være akiral og (ii) ha aminogruppen på et ikke-terminalt, alifatisk karbonatom. Uansett bruken av en (O-aminosyre-transaminase som av natur virker på en aminogruppe i den terminale eller (O-posisjon på en aminosyre, er det funnet at bruken av en aminodonor med en aminogruppe på et ikke-alifatisk karbonatom gir en termodynamisk fordel. Uten å ønske å være bundet av noen spesiell teori, synes det som om denne forbedring er en konsekvens av at biproduktet ved den enzymatiske reaksjon i et slikt tilfelle er et keton i motsetning til ved bruken av en aminodonor med en aminogruppe på et terminalt karbonatom som etylamin, n-propylamin, n-oktylamin, 1,4-butandiamin, 1,6-heksandiamin, 6-amino-heksansyre, 4-aminosmørsyre, tyramin eller benzylamin som danner aldehyd ved en aminosyre-transaminasereaksjon. I reaksjoner som involverer aminosyrer fra ketosyrer, resulterer den termodynamiske fordel ved bruk av isopropylamin som aminodonor i en likevektskonstant på rundt 1000. Fordi den termodynamiske fordel stammer fra den kjemiske omgivelse av den reagerende karbonylgruppe, gjelder dette på samme måte syntesen av alle kirale a-aminosyrer fra de respektive ketosyrer, uansett naturlig eller ikke-naturlig.
Uansett denne termodynamiske fordel, resulterer nærværet av
en aminogruppe på et ikke-terminalt, alifatisk karbonatom generelt i kiralitet i motsetning til substitusjon på et terminalt karbonatom som, nødvendigvis med to hydrogenatomer, for-hindrer kiralitet. Fordi transaminasen er stereoselektiv,
betyr bruken av en kiral amindonor at kun halvparten av et slikt amin er tilgjengelig som donor.
Uheldigvis er den alt overveiende majoritet av amino (lavere)alkaner som tilfredsstiller det første krav, nemlig å ha en aminogruppe på et ikke-terminalt karbonatom, i seg selv kirale. Hvis man således begrenser betraktningene til aminoalkaner med ikke mer enn 8 karbonatomer, anslås det at det teoretisk er minst 130 mulige homologer og isomere aminer, hvori aminogruppen ikke er på et trisubstituert karbonatom (for å være en aminodonor, må forbindelsen også bære minst ett tilgjengelig hydrogenatom på karbonatomet hvortil aminogruppen er bundet). Av disse 130 mulige aminodonorer, har mindre enn halvparten (54) en aminogruppe på et ikke-terminalt karbonatom, og av disse er 93% (50) kirale. Kun 4 av alkylaminene med en aminogruppe på et ikke-terminalt karbonatom er akirale, og av disse er 3 prohibitive med henblikk på omkostninger og tilgjengelighet og igjen uegnet som amindonorer: 3-aminopentan, 2,2-dimetyl-3-aminopentan og 4-amino-heptan. Av alle amino(lavere)alkaner som teoretisk er egnet som aminodonorer, har således kun 2-aminopropan (i) en aminogruppe på et terminalt karbonatom og er således termodynamisk begunstiget i forhold til aminoalkaner der aminogruppen er et terminalt karbonatom, (ii) er akiral og således fullstendig tilgjengelig for reaksjonen, og (iii) er akseptabel uttrykt ved omkostninger og tilgjengelighet. Som en ytterligere fordel genererer 2-aminopropan også et biprodukt, aceton, som lett kan gjenvinnes, og som i seg selv er en kommer-siell artikkel.
Den virkelige enzymatiske omdanning kan påvirkes ved konven-sjonelle dyrkingsteknikker med isolerte, men ikke-voksende celler, eller med et oppløselig aminosyre-transaminaseprepa-rat. Aminosyre-transaminasen kan foreligge i fri form, enten som en cellefri ekstrakt eller som et fullee1lepreparat, eller immobilisert på en egnet bærer eller matriks som fornet-tet dekstran eller agarose, silika, polyamid eller cellulose. Den kan også innkapsles i polyakrylamid, alginater, fibere eller lignende. Metoder for slik immobilisering er beskrevet i litteraturen, og det henvises f.eks. til "Methods of Enzy-mology", 44, 1976.
Selv om det ikke er nødvendig, er det generelt fordelaktig å tilsette en kilde for pyridoksamin som pyridoksalfosfat.
EKSEMPEL 1
Foreliggende oppfinnelse kan eksemplifiseres ved fremstilling av (S)-1-metoksy-2-aminopropan, et kjemisk mellomprodukt for syntese av landbrukskjemikalier, der metoksyaceton bringes i kontakt med en transaminase i nærvær av 2-aminopropan som amindonor, og tillate reaksjonen å fortsette inntil en vesentlig mengde metoksyaceton er omdannet til (S)-l-metoksy-2-aminopropan (og 2-aminopropan samtidig til aceton) og så iso-lere det (S)-1-metoksy-2-aminopropan som er dannet. Den totale enzymatiske transformering kan angis som følger:
5 mmol monobasisk natriumfosfat og 250 ml konsentrert saltsyre ble satt til 1000 ml vann. Blandingen ble avkjølt til 5-10aC i et isvannbad, og 258 ml 2-aminopropan ble tilsatt, fulgt av 206 ml 98 %-ig metoksyaceton. Denne blanding ble blandet og pH-verdien justert til 7,5 med enten natriumhydroksid eller saltsyre, etter behov. Blandingen ble overført til en 3 liters rundkolbereaktor med temperaturkontroll og omrøringsapparatur. Etter at temperaturen i reaksjonsblandingen var stabilisert til 30 ± 1<S>C, ble det tilsatt 0,2 mM pyridoksal-5'-fosfat. pH-verdien ble, hvis nødvendig, re-justert til 7,5, og en liten mengde vann kan tilsettes for å bringe volum i blandingen til 1800 ml.
Enzymoppløsningen ble fremstilt separat. Til 200 ml 5 mM nat-riumfosfatoppløsning med pH 7,5 ble det satt 0,2 mM pyridok-sal-5'-fosfat og 2 g (tørrvekt) Bacillus-celler inneholdende en (S)-transaminase. Etter at cellene var fullstendig suspen-dert, ble enzymoppløsningen avgitt til reaksjonsblandingen beskrevet ovenfor.
Den endelige reaksjonsbuljong inneholdt 1,5 M 2-aminopropan og 1,0 M metoksyaceton. Reaksjonen fortsatt i 8 timer ved 30 ± 1<B>C og pH 7,5, ved hvilket tidspunkt (S)-1-metoksy-2-aminopropan var til stede i reaksjonsblandingen i en konsentrasjon på 0,6 M, det vil si en ee på over 99 %.
Reaksjonen ble avsluttet ved tilsetning av 5 ml konsentrert saltsyre, fulgt av flashdestillasjon for å fjerne ikke-omsatt metoksyaceton og biproduktet, aceton, i et enkelt kutt. En separat kolonnedestillasjon av et destillat kan gjennomføres senere for å separere metoksyaceton og aceton. 270 ml 50 %-ig vandig natriumhydroksid ble satt til reaksjonsblandingen for å deprotonere aminene. Aminene ble så fjernet fra blandingen ved destillasjon som et enkelt kutt og (S)-1-metoksy-2-aminopropan separert fra gjenværende 2-aminopropan ved separat destillasjon, og man oppnådde 125 g (S)-1-metoksy-2-aminopropan inneholdende 50 % vann. Produktet er mer enn 99 % kjemisk og enantiomert rent, bestemt ved gasskromatografisk ana-lyse .
EKSEMPEL 2
Oppfinnelsen kan eksemplifiseres ytterligere ved syntese av L-alanin, en brukbar aminosyre, der pyruvinsyre bringes i kontakt med en transaminase i nærvær av 2-aminopropan som aminodonor, og tillate reaksjonen å fortsette inntil en vesentlig mengde pyruvinsyre er konvertert til L-alanin og 2-aminopropan samtidig er konvertert til aceton. Den totale, enzymatiske transformering kan angis som følger: 50 mM tilsvarende 0,165 g natriumpyruvat og 50 mM tilsvarende 0,23 ml 6,5 molar oppløsning av isopropylaminhydroklorid ble oppløst i 29,0 ml 50 mM natriumdihydrogenfosfatoppløsning, hvoretter pH-verdien ble justert til 7,5. 1,0 mM tilsvarende 8,0 mg pyridoksalfosfat ble tilsatt, fulgt av 8 mg E.coli-celler inneholdende en (S)-transaminase, slik at sluttvolumet var 30 ml og slutt-pH-verdien 7-5. Etter inkubering ved 30<fi>C i 24 timer ble konsentrasjonene av isopropylamin, aceton og L-alanin målt ved HPLC og GC og L-alaninkonsentrasjonen bestemt til 45,6 mM, ekvivalent med en Keq for reaksjonen ovenfor på over 100.
Da en analog reaksjon ble gjennomført ved bruk av (R)-transaminase inneholdende 0,3 g E.coli-celler, forløp omdanningen til en D-alaninkonsentrasjon som ble bestemt til 46 mM.
EKSEMPEL 3
Syntese av L- alanin
I et separat eksempel på syntese av L-alanin ble 1 M tilsvarende 110,0 g natriumpyruvat og 1 M tilsvarende 153 ml av en 6,5 molar oppløsning av isopropylaminhydroklorid oppløst i 800 ml 50 mM natriumdihydrogenfosfatbuffer og pH-verdien justert til 7,5. 1 mM tilsvarende 265 mg pyridoksalfosfat ble tilsatt, fulgt av 5 g E.coli-celler inneholdende en (S)-transaminase, slik at sluttvolumet var 1 liter og slutt-pH-verdien var 7,5. Etter inkubering ved 30aC i 24 timer ble konsentrasjonene av isopropylamin og L-alanin bestemt ved HPLC og konsentrasjonen av aceton ved GC. Konsentrasjonen for den produserte L-alanin ble bestemt til 970 mM, ekvivalent en likevektskonstant for reaksjonen på rundt 1000.
EKSEMPEL 4
Syntese av L- 2- aminosmørsyre
50 mM tilsvarende 186 mg natriumketobutyrat og 50 mM tilsvarende 0,23 ml av en 6,5 molar oppløsning isopropylamin ble oppløst i 29 ml 50 mM natriumdihydrogenfosfatbuffer og pH-verdien justert til 7,5. 1 mM tilsvarende 8,0 mg pyridoksalfosfat ble tilsatt, fulgt av 100 mg E.coli-celler inneholdende en (S)-transaminase, slik at sluttvolumet var 30 ml og slutt-pH-verdien var 7,5. Etter inkubering ved 30aC i 24 timer ble konsentrasjonene av isopropylamin og L-2-aminosmør-syre bestemt ved HPLC og konsentrasjonen av aceton ved GC. Konsentrasjonen for den fremstilte L-aminosmørsyre ble bestemt til 48 mM, ekvivalent en likevektskonstant for reaksjonen på over 500.
EKSEMPEL 5
Syntese av ytterligere aminosyrer
Ved å folge i det vesentlige samme prosedyre som beskrevet i eksempel 4, ble syntesen av L-glutamat, L-methionin og L-norvalin demonstrert fra natriumsaltene av de tilsvarende ketosyrer: 2-ketoglutarsyre (50 mM 252 mg), 4-metyltio-2-okso-smørsyre (50 mM, 255 mg) og 2-ketovaleriansyre (50 mM, 207 mg). I alle tilfeller produserte (S)-transaminasen utelukken-de L-isomeren av aminosyren i konsentrasjoner på respektivt 45, 47 og 46 mM.
Claims (1)
- Stereoselektiv syntese av et kiralt amin der et keton bringes i kontakt med en transaminase i nærvær av en aminodonor, karakterisert ved at det anvendes 2-aminopropan som amindonor. Syntese ifølge krav 1, karakterisert ved at det kirale amin er en kiral aminosyre. Fremgangsmåte for fremstilling av (S)-l-metoksy-2-aminopropan, karakterisert ved at den omfatter å bringe metoksyaceton i kontakt med en transaminase i nærvær av 2-aminopropan som amindonor inntil en vesentlig mengde metoksyaceton er konvertert til (S)-l-metoksy-2-aminopropan og 2-aminopropan er konvertert til aceton, med etterfølgende isolering av (S)-l-metoksy-2-aminopropan.
