Abstract
Sammendrag DNA-amplifisering og hybridisering blir vellykket utført i et reaksjonssystem som inneholder primere for DNA-amplifiseringen og hybridiseringsprober, etterfulgt av påvisning av hybriden i reaksjonsløsningen ved hjelp av affinitetskromatografi, der minst én av primerne som skal bli benyttet i DNA-amplifiseringen, er merket med et første merkemiddel, slik at det amplifiserte DNA vil være merket med det første merkemidlet, en hybridiseringsprobe er merket med et andre merkemiddel og inneholdt i en reaksjonsløsning for å utføre DNA-amplifiseringen. Basesekvensen til hybridiseringsproben er designet slik at den ikke inhiberer DNA-amplifiseringen, og en hybrid blir påvist ved hjelp av affinitetskromatografi ved hjelp av det første og det andre merkemidlet.
