Abstract

"MéTODO PARA DETECTAR ALTERAçõES EM UM GENE ATRAVéS DE PCR DE AMPLA ABRANGêNCIA USANDO ELEMENTOS HUMANOS MóVEIS". Os métodos descritos são para detectar uma alteração em um gene de interesse, de modo que a deleção no gene C4A (por exemplo, para detecção de C4AQ0), através da execução da amplificação através da reação em cadeia por polimerase de ampla abrangência em uma amostra teste compreendendo o DNA genómico. Os métodos amplificam o DNA alvo compreendendo todo ou uma parte de um elemento humano móvel (por exemplo, uma inserção retroviral no intron 9 do gene C4A), usando primers construídos de modo que os produtos de PCR sejam formados apenas se a amostra teste compreender o DNA genómico apresentando a alteração no gene de interesse; alternativamente, os métodos amplificam o DNA alvo usando primers construídos de modo que os produtos de PCR sejam formados apenas se a amostra teste compreender o DNA genómico que não apresente a alteração no gene de interesse.
