Description
Foreliggende oppfinnelse angår fremstilling av 1,4-disubstituerte-piperidinylforbindelser som er anvendbare som analgesiske midler og muskelrelakserende midler.
Et stort antall forbindelser er for tiden tilgjenge-lige som utviser terapeutisk aktivitet som analgesiske midler. Uheldigvis er flesteparten av de mere kraftige analgesiske midler narkotika. Narkotika er potensielt vanedannende og er derfor tilbøyelig til å fremkalle tilvending hos ømfintlige individer.
Det er også et stort antall av forbindelser tilgjen-gelige som er i stand til å lindre muskelspasmer. Mesteparten av disse forbindelser har den uønskede bivirkning at de virker sedative på pasienten eller svekker hans motoriske evne.
Det vil således være et verdifullt bidrag til faget å utvikle kraftige analgesika som er ikke-narkotiske og derfor fri for potensiell tilvending.
Det vil også være et verdifullt bidrag til faget å utvikle muskelrelakserende midler som ikke virker sedative på pasienten eller svekker hans motoriske evne.
Et mål med foreliggende oppfinnelse er således å utvikle forbindelser som utviser terapeutisk aktivitet som analgesika og som muskelrelakserende midler som utviser de ovenfor beskrevne fordeler.
Foreliggende oppfinnelse angår en analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formel
hvori R' er valgt fra gruppen bestående av hydrogen, en lavere alkanoyl med 2-4 carbonatomer, og en benzylgruppe eventuelt substituert ved 2 - 6-stilling på fenylringen med minst ett halogen; R er minst én gruppe valgt fra gruppen bestående av halogener, lavere alkylgrupper, og lavere alkoxygrupper, for-utsatt at fenylringen tilstøtende til carbonylsubstituenten alltid er substituert med minst ett fluoratom.
Som anvendt i foreliggende beskrivelse betegner: a) uttrykket carbonylgruppe en struktur svarende til b) uttrykket halogen et fluor-, klor- eller bromatom;
cj uttrykket lavere alkylgruppe en forgrenet eller rettkjedet
alkylgruppe inneholdende fra 1 til 4 carbonatomer, slik som methyl, ethyl, n-propyl, isopropyl, n-butyl og iso-butyl; d) uttrykket lavere alkoxygruppe, en rettkjedet eller forgrenet alkoxygruppe inneholdende fra 1 til 4 carbonatomer,

Uttrykket "farmasøytisk akseptable syreaddisjons-salter" er beregnet på å gjelde et hvilket som helst ikke-toksisk organisk eller uorganisk syreaddisjonssalt av basefor-bindelsene representert ved formel I. Eksempler på uorganiske syrer som danner egnede salter innbefatter saltsyre, hydrobrom-syre, svovelsyre og fosforsyre, og sure metallsalter slik som natriummonohydrogenortofosfat og kaliumhydrogensulfat. Eksempler på organiske syrer som danner egnede salter innbefatter mono-, di- og tricarboxylsyrer. Eksempler på slike syrer er for eksempel eddiksyre, glycolsyre, melkesyre, pyruvsyre, malonsyre, ravsyre, glutarsyre, fumarsyre, eplesyre, vinsyre, sitronsyre, ascorbinsyre, maleinsyre, hydroxymaleinsyre, benzoe-syre, hydroxybenzoesyre, fenyleddiksyre, kanelsyre, salicylsyre, 2-fenoxybenzoesyre, og sulfonsyrer slik som methansulfonsyre, 2-hydroxyethansulfonsyre og p-toluensulfonsyre.
For at forbindelsene fremstilt ifølge oppfinnelsen skal kunne utvise de ovenfor beskrevne terapeutiske aktivi-teter, er det nødvendig at fenylringen tilstøtende til X-substituenten alltid er substituert med minst ett fluoratom.
Representative eksempler på de mest foretrukne forbindelser ifølge oppfinnelsen er de valgt fra gruppen bestående av a-(4-fluorfenyl)-4-(hydroxymethyl)-l-piperidin-ethanol, eller
1-(4-fluorfenyl)-2-[4-[(1-oxopropoxy)methyl]-1-piperidinyl]-ethanon, eller
1- (4-fluorfenyl) -2-[ 4-(hydroxymethyl) -1-piperidinyl]-ethanon, eller
1-(4-fluorfenyl)-2-[4-(benzyloxymethyl)-1-piperidinyl]-ethanon, eller
a-(4-fluorfenyl)-4-(benzyloxymethyl)-1-piperidinethanol, eller 1-(4-fluorfenyl)-2-[4-(4-fluorbenzyloxymethyl)-1-piperidinyl]-ethanon, eller
oc- (4-fluorfenyl) -4- (4-fluorbenzyloxymethyl) -1-piperidin-ethanol.
Analogifremgangsmåten ifølge oppfinnelsen er kjennetegnet ved at
et 4-substituert piperidin hvori R' er som ovenfor angitt,
omsettes med
et 2-halo-substituert acetofenon hvori Y er et halogenatom, fortrinnsvis klor, og R er som ovenfor definert,
Utgangsmaterialer er et 4-substituert piperdin som vist i formel II hvori R' er som definert i formel I; og et 2-halo-substituert acetofenon som vist i formel III, hvori R er som definert i'formel I og Y er et halogen, fortrinnsvis klor.
Det 4-substituerte piperidin skal tilsvare strukturelt dets motpart i det ønskede 1,2-disubstituerte ethanon, da alle av dets substituenter vil forbli i sluttproduktet. Likeledes skal det 2-halo-substituerte acetofenon svare strukturelt til dets motpart i det ønskede 1,2-disubstituerte ethanon, da alle av dets substituenter med unntak av halogen-atomer ved Y-stillingen vil være bibeholdt i sluttproduktet.
Hvis for eksempel den ønskede forbindelse er l-(4-fluorfenyl)-2-[4-(hydroxymethyl)-1-piperidinyl]-ethanon, er de foretrukne utgangsmaterialer: (a) 4-hydroxymethylpiperidin og, (b) 2-klor-4<1->fluoracetofenon.
Hvis det foretrukne utgangsmateriale av formel III
er utilgjengelig (dvs. et 4-substituert piperidin hvori R' tilsvarer strukturelt dets motpart i det ønskede 1,2-disubstituerte ethanon), kan R' tilsettes til strukturen etter at det 1,2-disubstituerte ethanon er dannet. Dette kan utføres etter vel kjente teknikker innen faget.
Det er for tiden foretrukket at tilnærmet ekvimolare konsentrasjoner av det 4-substituerte piperidin og det 2-halo-substituerte acetofenon anvendes i syntesen. Et svakt overskudd av den ene eller den andre av reaktantene er ikke skade-lig for syntesen.
Det er også foretrukket at reaksjonen utføres i nærvær av enten en organisk eller uorganisk base. Triethylamin er for tiden foretrukket. Basen er fortrinnsvis tilstede i et molart overskudd i forhold til det 4-substituert piperidin
Det foretrekkes også at reaksjonen utføres i nærvær av en alkalijodkatalysator. Natriumjodid er for tiden foretrukket. Alkalijodkatalysatoren er fortrinnsvis tilstede i en mengde på 0,1 til 1 mol% basert på mengden av 4-substituert-piperidin tilstedeværende i reaksjonssonen.
Det 4-substituerte piperidin og det 2-halo-substituerte acetofenon omrøres fortrinnsvis i et tidsrom varierende fra 1 til 30 timer. Det foretrekkes at reaksjonen utføres ved et temperaturområde fra 25 til 115° C. Det er også foretrukket at reaksjonen utføres i et organisk løsningsmiddel. Eksempler på egnede løsningsmidler innbefatter diklormethan, methanol, tetrahydrofuran, toluen eller kloroform og lignende.
Det 1,2-disubstituerte ethanon fremstilt som ovenfor angitt kan gjenvinnes fra reaksjonsblandingen ved teknikker vel kjent innen faget. En egnet teknikk er å ekstrahere reaksjonsblandingen med et organisk løsningsmiddel etter at vann er blitt tilsatt til reaksjonsblandingen. Det ønskede 1,2-disubstituerte ethanon vil være i den organiske fase.
Det 1,2-disubstituerte ethanon kan deretter renses ved teknikker vel kjent innen faget. En slik egnet teknikk er omkrystallisering fra et egnet løsningsmiddelsystem. Eksempler på egnede løsningsmiddelsystemer som for tiden anvendes innbefatter metanol/2-butanon, methanol/ethylacetat, kloroform/benzen og ethylacetat/hexan hvis den ønskede forbindelse er tilstede som et syreaddisjonssalt. Kloroform/benzen, methanol/vann og ethylacetat/hexan er representative eksempler på egnede løsningsmiddelsystemer som for tiden anvendes hvis den ønskede forbindelse er tilstede som en fri base. Andre egnede løsningsmiddelsystemer kjent innen faget kan også anvendes .
Som ovenfor angitt kan forbindelsene av formel I anvendes som analgesiske midler. Forbindelsene utviser et styrkenivå tilstrekkelig til å inhibere følelsen av de sterke nivåer av smerte som vanligvis er forbundet med tilstander slik som metastatisk carcinoma, myocardiale infarkt eller traumetisk skade.
Til tross for dette høye styrkenivå er forbindelsene ikke-narkotiske. Dette betyr at de er fri for det potensielle misbruk som ledsager de fleste analgesiske midler.
En måte på å demonstrere den analgesiske anvendelig-het av disse forbindelser er å utføre de følgende testproto-koller.
Fem til ti mus administreres med fra 0,1 til 200 mg/ kg av forbindelsen, enten subcutant eller intragastrisk. 30 minutter etter administreringen av testforbindelsen administreres musen 0,4 ml av en 0,25 % v/v løsning av eddiksyre intraperitonealt.
Fem minutter etter administreringen av eddiksyren observeres musen med hensyn til tegn på sprelling og kramper som er en indikasjon på smerte.
En forbindelse betraktes å utvise signifikant analgesisk aktivitet hvis musen som administreres angitte forbindelse ikke utviser tegn på smerte under testen (dvs. sprelling og vridning).
En måte på å vise de ikke-narkotiske egenskaper av disse forbindelser er følgende testprotokoll.
Tre mus administreres med opp til 800 mg/kg av den ønskede forbindelse subcutant. 30 minutter senere anbringes musen på en varm plate som er blitt oppvarmet til en tempera-tur på 55° C.
Forbindelsen betraktes å være ikke-narkotisk hvis musen hopper av i løpet av de første 20 sekunder etter at den er -blitt anbragt på den varme plate.
En måte på å vise anvendeligheten av disse forbindelser som muskelrelakserende midler er ved deres evne til å antagonisere den langvarige kontraksjon av sacrococcygeus dorsalis-muskelen i mus, som administrering av morfin typisk fremkaller (Straub Tail-test). Denne kan vises på følgende måte. 5 til 10 mus administreres fra 0,1 til 200 mg/kg av forbindelsen. 30 minutter senere administreres musen 60 mg/kg morfin-subcutant.
Musen observeres i 30 minutter etter administrering av morfin for å bestemme hvorvidt testforbindelsen har blokkert morfihets evne til å forårsake den langvarige kontraksjon av sacrococcygeus dorsails-muskelen i musen. Kontraksjon av denne muskel forårsaker at halen av musen bøyes oppover til en vinkel på minst 9 0°. Hvis således en forbindelse er muskelrelakserende vil halen av musen ikke bli hevet.
En måte på å demonstrere at forbindelsene fremstilt ifølge oppfinnelsen ikke svekker motorisk evne eller forårsaker sedasjon er følgende testprotokoll.
Mus sorteres først for anvendelse i testen ved å anbringe disse<*> på en horisontal stav som roterer ved 15 omdr./ min. De mus som faller fra under en 120 sekunders periode utelukkes fra ytterligere testing. ;Musene som tilfredsstiller det ovenfor beskrevne kriterium administreres deretter opp til 800 mg/kg enten subcutant eller intragastrisk av testforbindelsen. 30 minutter senere anbringes musene tilbake på den roterende horisontale stav og observeres i 9 0 sekunder. ;For å bestemme om en forbindelse er ikke-sedativ og ikke svekker motorisk evne, er det nødvendig å tolke resulta-tene av denne test i lys av de ED^-verdier erholdt ved Straub Tail-testen angitt tidligere. En forbindelse betraktes å være ikke-sedativ og ikke å svekke motorisk evne hvis for-holdet mellom dosen ved hvilket ca. halvparten av musene faller av den roterende stav, og dosen ved hvilken ca. halvparten av musene ikke opplever morfin-^fremkalt kontraksjon av sacrococcygeus dorsails-muskel er ca. 2:1 eller større. ;Forbindelsene fremstilt ifølge oppfinnelsen kan administreres på et utall måter. De er effektive hvis de administreres oralt. Forbindelsene kan også administreres parenteralt (dvs. subcutant, intravenøst, intramuskulært eller intraperitonealt). ;Det er for tiden foretrukket at forbindelsene administreres parenteralt. Mengden av administrert forbindelse vil variere avhengig av pasienten, administreringsmåten og strengheten av den tilstand som skal behandles. Gjentatt dag-lig administrering av forbindelsen kan være Ønskelig og vil variere med pasienttilstanden og administreringsmåten. ;Selv om den nødvendige dose vil variere fra pasient til pasient er det generelt foretrukket at forbindelsene ifølge oppfinnelsen administreres innen et doseringsområde på fra 0,1 til 200 mg/kg kroppsvekt pr. dag, enten de administreres oralt eller parenteralt. Dette doseringsområde er anvend-bart enten forbindelsene anvendes som et analgesisk middel eller som et muskelrelakserende middel. ;Som anvendt i foreliggende beskrivelse angir uttrykket pasient et varmblodig dyr. Forbindelsene er således effektive for lindring av smerte og muskelspasmer i fugler slik som kyllinger og kalkuner, eller pattedyr, slik som mennesker, primater, sauer, hester, kveg, griser, hunder, katter, rotter og mus. ;For oral administrering kan forbindelsene formuleres i faste eller væskeformige preparater slik som kapsler, piller, ;tabletter, pastiller, smelter, pulvere, suspensjoner eller emulsjoner. Faste enhetsdoseringsformer kan være kapsler av vanlig gelatintype, inneholdende for eksempel overflateaktive midler, smøremidler og inerte fyllstoffer slik som lactose, sucrose og maisstivelse, eller de kan være i form av preparater med forlenget frigivelse. I en annen utførelsesform kan forbindelsene av formel I tabletteres med konvensjonelle tablett-baser slik som lactose, sucrose og maisstivelse i kombinasjon med bindemidler slik som acacia, maisstivelse eller gelatin, oppbrytende midler slik som potetstivelse eller alginsyre, ;og et smøremiddel slik som stearinsyre eller magnesiumstearat. Væskeformige preparater fremstilles ved oppløsning av den aktive bestanddel i et vandig eller ikke-vandig farmasøytisk akseptabelt løsningsmiddel som også kan inneholde suspenderings-midler, søtningsmidler, smaksgivende midler og konserveringsmidler, hvilket er vel kjent innen faget. ;For parenteral administrering kan forbindelsene opp-løses i en fysiologisk akseptabel farmasøytisk bærer og administreres enten som en løsning eller suspensjon. Eksempler på egnede farmasøytiske bærere er vann, saltvann, dextroseløsnin-ger, fructoseløsninger, ethanol eller oljer av animalsk, vegetabilsk eller syntetisk opprinnelse. Den farmasøytiske bærer kan også inneholde konserveringsmidler, buffere etc. hvilket er vel kjent innen faget. ;De etterfølgende eksempler illustrerer oppfinnelsen. ;Eksempel 1 ;Formålet med dette eksempel er å vise en måte for fremstilling av forbindelsen 1-(4-fluorfenyl)-2-[4-(hydroxy-methyl)-1-piperidinyl]-ethanon-hydroklorid. ;Til en løsning av 4,0 g (34,8 mmol) 4-hydroxymethylpiperidin i 150 ml tetrahydrofuran ble tilsatt 9,6 ml (69,6 mmol) triethylamin, 6,5 g (38,3 mmol) 2-klor-4'-fluoraceto-fenon og en katalyttisk mengde av natriumjodid. Reaksjonsblandingen ble omrørt ved romtemperatur i 24 timer. Etter omrøring ble reaksjonsblandingen helt over i en mettet løsning av vandig natriumbicarbonat. Løsningen ble deretter ekstrahert med ethylacetat og det resulterende organiske lag ble fraskilt. ;Det organiske lag ble deretter tørket med magnesiumsulfat, ble filtrert og konsentrert på en rotasjonsfordamper. Den resulterende tykke røde olje ble deretter fortynnet med methylenklorid og gassformig hydrogenklorid ble boblet gjennom løsningen. Denne blanding ble deretter konsentrert på en rotasjonsfordamper. 1-(4-fluorfenyl)-2-[4-(hydroxymethyl)-1-piperidinyl]-ethanon-hydrokloridet ble renset ved omkrystallisering fra et løsningsmiddelsystem inneholdende methanol og 2-butanon. 7,18 g (25 mmol) av det ønskede produkt ble erholdt, og som hadde et smeltepunkt på 250° C. ;Eksempel 2 ;Formålet med dette eksempel er å vise en måte for fremstilling av en forbindelse ifølge oppfinnelsen hvori den ønskede substituent på den 4-substituerte piperidinylring (dvs. R<1>) ikke er lett tilgjengelig som utgangsmateriale. ;I en slik situasjon kan substituenten tilsettes etter at det 1,2-disubstituerte ethanon er blitt dannet. ;Den ønskede forbindelse i denne situasjon er l-(4-fluorfenyl)-2-[4-[(1-oxypropoxy)methyl]-1-piperidinyl]-ethanon. Imidlertid var 4-(1-oxopropoxy)methylpiperidin ikke tilgjengelig som utgangsmateriale. Det ønskede 1,2-disubstituerte ethanon ble derfor fremstilt på følgende måte. ;2,3 g (8,0 mmol) 1-(4-fluorfenyl)-2-[4-(hydroxy-methyl)-1-piperidinyl]-ethanon ble fremstilt på samme måte som beskrevet i eksempel 1. ;Dette materiale ble deretter oppløst i 200 ml tetrahydrofuran. Til denne løsning ble tilsatt 1,6 g (12,0 mmol) propionsyreanhydrid, 5 g (59 mmol) natriumbicarbonat, og en katalyttisk mengde av 4-dimethylaminopyridin. Denne blanding ble deretter omrørt ved romtemperatur i 24 timer. ;Reaksjonsblandingen ble deretter blandet med destil-lert vann. • Den resulterende løsning ble ekstrahert med ethylacetat. ;Det organiske lag erholdt som ovenfor angitt ble fraskilt og ble deretter vasket med en 10 % w/w løsning av vandig hydrogenklorid. Det resulterende vandige lag ble fraskilt og nøytralisert med fast natriumbicarbonat. Denne løs-ning ble deretter ekstrahert med ethylacetat, og det resulterende organiske lag ble oppsamlet. ;Det organiske lag ble deretter tørket med magnesiumsulfat, ble filtrert, og gassformig hydrogenklorid ble boblet gjennom det resulterende filtrat. ;Produktet, 1-(4-fluorfenyl)-2-[4-[(1-oxypropoxy)-methyl]-1-piperidinyl]-ethanon-hydroklorid ble gjenvunnet ved omkrystallisering fra et løsningsmiddelsystem inneholdende kloroform og benzen. ;1,1 g (3,2 mmol) 1-(4-fluorfenyl)-2-[4-[(1-oxyprop-oxy ) methyl ] -1-piperidinyl ] -ethanon-hydroklorid ble erholdt, med smeltepunkt på 170 - 174° C. ;Eksempel 3 ;Formålet med dette eksempel er å vise en metode for fremstilling av en forbindelse av formel I hvori R' er en substituert benzylgruppe. ;Til en løsning av 20,0 g (150 mmol) isonipecotin-syre i 200 ml vann ble tilastt til 20,0 g (190 mmol) natrium-carbonat og 37,2 g (170 mmol) di-t-butyl-dicarbonat. Suspensjonen ble omrørt i 24 timer ved romtemperatur og ble ekstrahert med ether. Det vandige lag ble surgjort til pH a* 4 med vandig 10 % hydrogenklorid og ble ekstrahert med ethylacetat. Det organiske lag ble tørket med magnesiumsulfat og ble konsentrert under dannelse av 31,4 g 4-carboxy-piperidin-l-carboxylsyre-t-butylester som et hvitt fast materiale som smeltet ved 146 - 150° C.
Til en 0° C løsning av 15,0 g (65,5 mmol) av 4-carboxy-piperidin-l-carboxylsyre-t-butylesteren fremstilt som ovenfor angitt i 200 ml tørr tetrahydrofuran, ble under nitrogengass tilsatt 9 8,2 ml (98,2 mmol) 1,0M boran i tetrahydrofuran i løpet av 10 minutter. Reaksjonsblandingen ble omrørt i ytterligere 4 timer ved romtemperatur, ble tilsatt 500 ml vandig mettet natriumbicarbonat og ble ekstrahert med ethylacetat. Det organiske lag ble tørket med magnesiumsulfat og konsentrert under dannelse av 14,1 g 4-hydroxymethylpiperidin-l-carboxylsyre-t-butylester som et hvitt fast materiale med smeltepunkt 75 - 7 6° C.
Til en løsning av 5,0 g (23,2 mmol) av 4-hydroxy-methyl-piperidin-l-carboxylsyre-t-butylesteren fremstilt som ovenfor beskrevet i 100 ml dimethylformamid, ble tilsatt 1,2 g (25,6 mmol) av en 50 % natriumhydrid-oljedispersjon. Etter omrøring i 1/2 time ble 5,0 g (34,9 mmol) 4-fluorbenzylklorid tilsatt, etterfulgt av en katalyttisk mengde av tetrabutyl-ammoniumjodid. Suspensjonen ble omrørt i ytterligere 24 timer, ble tilsatt mettet vandig natriumklorid og ble ekstrahert med ether. Det organiske lag ble tørket med magnesiumsulfat og ble konsentrert. Den resulterende olje ble behandlet med methanolisk hydrogenklorid i 4 timer. Et hvitt fast materiale ble erholdt etter konsentrering, som ble filtrert og vasket med hexan. 5,1 g 4-(4-fluor-benzyloxymethyl)-piperidin-hydroklorid ble erholdt som hadde et smeltepunkt på 142 - 145° C.
Til en løsning av 1,4 g (6,3 mmol) av det ovenfor fremstilte 4-(4-fluorbenzyloxymethyl)-piperidin i 125 ml methanol, ble tilsatt 1,3 g (12,6 mmol) triethylamin og 1,4 g (9,4 mmol) 2-klor-4-fluoracetofenon. Etter 24 timer ved romtemperatur ble reaksjonsblandingen fortynnet med mettet vandig natriumbicarbonat og ble ekstrahert med kloroform. Det organiske lag ble tørket med magnesiumsulfat og ble konsentrert. Residuet ble kromatografert på silicagel under anvendelse av 30 % ethylacetat/hexan som elueringsmiddel. Den resulterende olje ble isolert som dets hydrokloridsalt som ble omkrystalli-sert fra ethylacetat under dannelse av 0,90 g 1-(4-fluorfenyl)-2-[4-(4-fluorbenzyloxymethyl)-1-piperidinyl]-ethanon-hydro-klorid som et hvitt fast materiale med smeltepunkt på 155 - 158° C.

1. Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formel
hvori R' er valgt fra gruppen bestående av hydrogen, en lavere alkanoyl med 2-4 carbonatomer, og en benzylgruppe eventuelt substituert ved 2 - 6-stilling på fenylringen med minst ett halogen; R er minst én gruppe valgt fra gruppen bestående av halogener, lavere alkylgrupper, og lavere alkoxygrupper, for-utsatt at fenylringen tilstøtende til carbonylsubstituenten alltid er substituert med minst ett fluoratom,karakterisert ved at
et 4-substituert piperidin hvori R' er som ovenfor angitt,
omsettes med
et 2-halo-substituert acetofenon hvori Y er et halogenatom, fortrinnsvis klor, og R er som ovenfor definert,
