Description
Denne oppfinnelse vedrører analogifremgangsmåte ved fremstilling av en ny terapeutisk aktiv forbindelse av den art som er angitt i krav 1<1>s ingress.
I henhold til oppfinnelsen frembringes forbindelser med struktur (1)
Det vil være klart at forbindelsene med struktur (1) har et chiralt senter på det karbonatom til hvilket hydroksygruppen er festet. Den foreliggende oppfinnelse omfatter alle optiske isomere av disse forbindelser i deres fullstendig oppløste eller delvis oppløste former og i form av racemiske blandinger.
Forbindelsene med struktur (1) kan fremstilles ved de følgende generelle fremgangsmåter: A. å omsette en forbindelse med struktur (2) med vann og å cyklisere produktet intramolekylært:
hvori R<3> og R<4> er hver uavhengig hydrogen, C1-4-alkyl (rettkjedet eller forgrenet), og R<5> er C1_4-alkyl (rettkjedet eller forgrenet); eller
B. å omsette en forbindelse med struktur (6)
i hvilken X er OH eller en lavere alkoksygruppe, med glycinamid.
Fortrinnsvis er R3 isopropyl og R<4> hydrogen, eller R<3> og R<4 >er begge metyl i forbindelsene med struktur (2). Fortrinnsvis er R<5> isobutyl, da forløperen 3,4-epoksybutanoatester med struktur (4) er lett å håndtere.
Avblokkering og intramolekulær cyklisering av forbindelsene med struktur (2) kan utføres ved oppvarming i nærvær, av vann ved temperaturer mellom 90° og 160°C, fortrinnsvis; mellom 100° og 130°C. Det er hensiktsmessig å anvende-vann eller blandinger av løsningsmidler og vann\ Fortrinnsvis: er løsningsmiddelet en 95% blanding av et: organisk løsningsmid-del med vann (5%). Som organisk co-løsningsmiddel kan det anvendes dimetylformamid (DMF), dimetylsulfoksid (DMSO), dimetylacetamid (DMA), acetonitril og alkoholer såsom etanol osv. Fortrinnsvis utføres reaksjonen i nærvær av en karboksylsyre, for eksempel eddiksyre eller benzosyre. Selv om katalytiske mengder av karboksylsyrene er virksomme, anvendes fortrinnsvis 0.8 til 1.2 molarekvivalenter av en karboksylsyre.
Forbindelsene med struktur (2) kan fremstilles ved å omsette en forbindelse med struktur (3) med en 3.4-epoksybutanoat ester med struktur (4).
Forbindelsene med struktur (3) kan fremstilles ved å omsette en ester med struktur (5) i hvilken R<3> og R<4> er som definert for struktur (2), og R<6> er C1_4-alkyl (rettkjedet eller forgrenet), med en amin med struktur NHR<1>R<2>. Forbindelsene med struktur (4) kan fremstilles som beskrevet i EPA 0154490 (ISF).
Omsetningen mellom forbindelsene med struktur (3) og (4) kan gjennomføres uten å anvende løsningsmidler eller i nærvær av løsningsmidler, vann eller vandige blandinger av disse i forhold på fra 1:5 til 20:1, fortrinnsvis fra 1:1 til 1:3 av organisk løsningsmiddel og vann. Som organisk løsningsmiddel kan det anvendes en rekke polare eller upol-are løsningsmidler, f.eks. acetonitril, alkoholer såsom isopropanol og pentanol og ketoner såsom aceton. Reaksjonen utføres ved en temperatur på 20° til 120°C, fortrinnsvis 70-100°C, og de to reagenser kan anvendes i støkiometriske forhold, eller et overskudd av en av dem kan anvendes, fortrinnsvis et forhold på fra 1:1 til 1:1.1 av imidazolidi-non (3) og epoksyester (4).
Forbindelsene med struktur (5) kan fremstilles fra saltene av de korresponderende syrene beskrevet av Pånetta et al. J.Org.Chem. 3_7 302 (1972), og fremgangsmåter som er analoge med dem som er beskrevet av Pånetta et al.
En forløper for glycinamid er 2-isopropyl imidazolidin-4en, andre forløpere er andre cykliske amidoacetaler og imido-ketalderivater.
For forbindelsene med struktur (6) er eksempler på avgangs-grupper X som kan substitueres med et amin C1_4~alkoksy, amino (NH2) og azido (N3) (hvilken kan fremstilles f.eks. ved å omsette et hydrazid med et alkylnitrit). Når X er OH, vil denne gruppe kreve aktivering, f.eks. omdannelse til et syreklorid, et syreazid, et syreanhydrid (f.eks. ved omsetning med et alkylklorformat eller pivaloylklorid), en aktivert ester eller anvendelse av et peptidkoblingsmiddel.
Det vil være klart at aktiveringen av karboksylgruppen kan nødvendiggjøre den forløpige beskyttelse av hydroksygruppen. Egnede hydroksy-beskyttelsesgrupper, aktiverte estere og peptid-koblingsreagenser er velkjent, og de er beskrevet f.eks. i "Peptide Synthesis" av M. Bodansky, Y. Klausner og M. Ondetti (Wiley 1976) eller "Protective Groups in Organic Chemistry" av T.W.. Greeene (Wiley 1981) . Eksempler på hydroksy-beskyttelsesgrupper er tetrahydropyranyl, trimetyl-silyl, benzyl og t-butyl.
Eksempler på aktiverte estere er triklorofenyl-, pentakloro-fenyl-, N-hydroksysuccinimido- og 1-hydroksybenzotriazol-estere. Eksempler på peptidkoblingsreagenser er karbo-diimider (såsom N<,N'dicycloheksylkarbodiimid) , N,N'karbonyl-diimidazol, l-etoksykarbonyl-2-etoksy-l,2-dihydrokinolin (EEDQ) og isoksazoliumderivater såsom N-etyl-5-fenyl-isoksazolium3<1->sufonat (Woodward's Reagent K).
Løsningsmiddelet som velges for reaksjonen, vil være avhengig av løsligheten av reagensene og deres reaktivitet. Når X f.eks. er NH2 eller C1_4-alkoksy, kan reaksjonen utføres i en alkanol eller i en vandig blanding av en alkanol, f.eks. vandig amylalkohol. Når karboksylgruppen krever aktivering, vil reaksjonen vanligvis utføres i et dipolart aprotisk løsningsmiddel såsom tetrahydrofuran, dioksan, dimetoksyetan eller dimetylformamid.
Fortrinnvis er X metoksy eller etoksy, og forbindelsen med struktur (6) omsettes med glycinamid.
Mellomproduktene med struktur (2) og (3) er nye forbindelser.
Forbindelsene med struktur (1) har nyttig nootropisk virkning, d.v.s. de bidrar til å forbedre lære- og hukommelses-vanskeligheter som er forbundet med aldring og ulike patologier som omfatter Alzheimer's sykdom. Kunnskapsforstyrrel-ser som opptrer i slike patologier vet man henger sammen med defisitter i det kolinergiske system i hjernen som i vist både av morfologiske (B.E. Tomlinson in "Biochemistry of Dementias"; P.J. Roberts Ed.; John Wiley & Sons, New York, N.Y. P.15-22, 1980) og neurokjemiske funn (R.T. Bartus et al., Science, 217, 408, 1982). Det er også velkjent at betraktelige forstyrrelser av kunnskapsfunksjoner er de mer synlige og svekkende symptomer som kan observeres i pasienter med Alzheimer's sykdom, senil demens av Alzheimer-typen og multiinfarktual demens. På den andre side frembringer den antikolinergiske droge scopolamin både i mennesker (D.A. Drachman, Archs. Neurol., Chigaco, 3_0, 113, 1974) og i dyr (D.A. Eckerman, Pharmacol. Biochem. Behav., 12, 595, 1980) et betraktelig hukommelsestap hvilket direkte henger sammen med en nedgang av acetylkolin-konsentrasjonen i spesielle hjerneområder, såsom i cerebral cortex og hippocampus. På basis av disse forutsetninger har forbindelsene med struktur (1) vært testet i rotter både på den for-styrrende virkning av scopolamin på hukommelsesspor og på reduksjonen av acetylkolin nivåene i hippocampus. For å evaluere effekten på hukommelse og læreevne ble det anvendt et forsøkstrinn gjennom passiv unngåelsesforsøk i hann-rotter av Wistar-rotter (150-160 g). Utstyret var hovedsaklig det samme som beskrevet av Essman (Pharmacol. Res. Commun, 5, 295, 1973).
Passering fra en lys boks inn i en mørk boks ble straffet med unngåelige elektriske fotsjokk. Dyrene må lære å unngå etter en eneste læreseksjon å krysse fra den lyse til den mørke boks. Tredve minutter etter første sesjon (læresesjon) ble læreeffekten kvantifisert (gjentatt forsøkssesjon) ved hjelp av latens (i sekunder) mellom adgang for dyrene i lysboksen og inngang i den mørke. Læreeffekten forstyrres betydelig ved en behandling med scopolamin (0,63 mg/kg i.p.) seksti minutter før læresesjonen. Saltoppløsning eller forsøksforbindelsene ble administrert i.p. tredve minutter før scopolamin. Kontrollgruppen ble behandlet på samme måte, men kun med saltoppløsning. Som et eksempel er resultatene for forbindelse A (1, R<1>=R<2>=H) oppført i tabell 1.
For å evaluere acetylkolin-nivåene i "hippocampus" ble fremgangsmåten beskrevet av Jenden et al., ("Choline and Acetylcholine : Handbook of Chemical Assay Methods", i.Hanin Ed., Raven Press, pp. 135-150, 1974) anvendt på Wistar-rotter. Dyrene ble forbehandlet med saltoppløsning eller en forsøksforbindelse tredve minutter før scopolamin (0,63 mg/kg i.p.). Kontrollgruppen ble behandlet på samme måte med de respektive bærere. Nitti minutter etter administrering av scopolamin ble rottene drept ved mikrobølgebestråling; hippocampus ble dissekert ut av hjernen og homogenisert i 0,4 N perklorsyre. Til den klare supernatanten oppnådd etter sentrifugeringen ble det tilsatt butyrylkolin (innvendig standard) og kaliumacetat etter hverandre. Etter en ytter-ligere sentrifugering ble det tilsatt tetrametylammonium klorid og ammonium-reineckat (Reinecke salt) til prøver for å utfelle acetylkolin. De utfelte pellets ble behandlet som beskrevet (Jenden et al, ibidem). Til slutt ble prøvene ekstrahert med 0,1 ml etylacetat; 1 ml av prøvene ble injisert i en HP Sigma 3B gasskromatograf utstyrt med en nitrogen-fosfordetektor.
Som eksempel er resultatene for forbindelse A (1, R<1>=R<2> =H) oppført i tabell 2.
Det er overraskende at forbindelse A skulle ha så god aktivitet, de polare og hydrofile substituenter tatt i betrakt-ning, hvilke substituebter reduserer dets biotilgjenglighet.
For å anvende en forbindelse med struktur (1) for den tera-peutiske behandling av mennesker og dyr, vil den vanligvis formuleres i henhold til standard farmasøytisk praktisk som en farmasøytisk blanding.
Forbindelsene med struktur (1) kan administreres på standard måte for behandling av de antydede sykdommer, for eksempel oralt, parenteralt, rektat, transdermalt eller ved trans-mucosal (for eksempel sub-lingual, eller buccal eller insufflatorisk) administrering.
Forbindelsene med struktur (1) som er aktive når de administreres oralt eller via sub-lingual eller buccal administrering, kan formuleres som siruper, tabletter, kapsler og piller. En sirupformulering vil generelt bestå av en suspensjon eller oppløsning av forbindelsen eller et salt i en flytende bærer for eksempel etanol, glycerin eller vann med et smaksstoff eller fargestoff. Når blandingen forelig-ger i form av en tablett, kan enhver rutinemessig farma-søytisk anvendt bærer anvendes for fremstillingen av de faste formuleringer. Eksempler på slike bærere omfatter magnesiumstearat, stivelse, lactose og sucrose. Når blandingén er i form av en kapsel, kan enhver rutinemessig anvendt innkapsling anvendes, for eksempel anvendelsen av en forrut nevnt bærer i et hardgelatinkapselskjell. Når blandingen er i form av et mykgelatinkapselskjell, kan enhver farmasøytisk bærer som rutinemessig anvendes for fremstilling av dispersjoner eller oppløsninger, anvendes, for eksempel flytende gummi, cellulose, silikater eller oljer og innlemmes i et mykgelatinkapselskjell.
Typiske parenterale blandinger består av en oppløsning eller en suspensjon av forbindelsene med struktur (1) i en steril vandig eller ikke-vandig bærer, eventuelt innehol-dende en parenteral akseptabel olje, for eksempel polyetyl-englycol, polyvinylpyrriolidon, lecitin, arachisolje eller sesamolje.
En typisk suppositorieformulering består av en forbindelse med struktur (1) som er aktiv når administrert på denne måte, med et binde- og/eller smøremiddel, for eksempel poly-mere glycoler, gelatiner, kokos-smør eller andre lavtsmel-tende vegitabilske vokser eller fetter.
Typiske transdermale formuleringer består av en konvensjo-nell vandig eller ikke-vandig bærer, for eksempel en krem, salve, lotion eller pasta eller kan foreligge i form av et medisinsk plaster eller membran.
Fortrinnsvis er blandingen i en enhetsdoseform, for eksempel en tablett eller kapsel, slik at pasienten kan admini-strere en enkelt dose til seg selv.
Piracetam er en forbindelse som anvendes ved behandlingen av senil demens og beslektede sykdommer. Forbindelsene med struktur (1) kan administreres i lignende oppsetninger som er etablert for piracetam med enhver hensiktsmessig tilpassing i dosenivået eller hyppighet i doseringen idet man tar hensyn til den større aktivitet og bedre far-makologiske profil av forbindelsene med struktur (1).
Hver enhetsdoseform for oral administrering innholder hensiktsmessig fra 0,5 mg/kg til 50 mg/kg og fortrinnsvis fra 1 mg/kg til 8 mg/kg og hver doseenhet for parenteral administrering inneholder hensiktsmessig fra 0,1 mg/kg til 10 mg/kg av en forbindelse med struktur (1).
Den daglige doseoppsetning for oral administrering er hensiktsmessig fra 0,5 mg/kg til 100 mg/kg, mer hensiktsmessig fra 1 mg/kg til 25 mg/kg av en forbindelse med struktur (1). Den aktive ingrediens kan administreres fra 1 til 6 ganger daglig. Forbindelsene med struktur (1) kan co-administreres med andre farmasøytisk aktive forbindelser, for eksempel i kombinasjon, parallelt eller sekvensielt, særlig med andre forbindelser som anvendes ved behandling av eldre pasienter, for eksempel beroligende midler, diuretika, antihypertensive midler, vasodilatorer og inotropiske midler.
Oppfinnelsen illustreres ved de følgende eksempler.
Fremstilling 1
2- metylpropvl 3- karbamoYlmetyl- Beta- hydroksy- 2- f1- metyl-etvl)- 4- oxo- l- imidazolidinbutanoat
1) Til 50 ml vannfri etanol rørt ved 0-5°C tilsettes 2 ml tionylklorid. Ved den samme temperatur tilsettes 2,1 g (0,01 mol) natrium-(1-metyletyl)-5-oxo-l-imidazolidin-acetat. Den dannede suspensjon ble rørt ved 0°C i 1 time og ved romtemperatur i 2 timer. Løsningsmiddelet ble fordampet under redusert trykk, og resten ble tatt opp med etylacetat. Den faste rest ble filtrert fra og løsningsmiddelet fordampet. Resten ble oppløst i en mettet oppløsning av natriumhydrogenkarbonat og ekstrahert med 3 x 50 ml av diklormetan. De organiske lag ble tørket og fordampet for å gi etyl-2-(1-metyletyl)-5-oxo-l-imidazolidinacetat (0,9 g) som en blek-gul olje (42%) (Rf 0,6, etyl acetat/diklormetan 6:4; kiselgel-plater). Hydrokloridsalt, s.m.p. 148149°C (metanol/etylacetat). 2) En iskald oppløsning av 3,8 g (0,018 mol) av etyl-2-(1-metyletyl)-5-oxo-l-imidazolidinacetat i 100 ml metanol ble mettet med gassformig ammoniakk. Oppløsningen ble rørt ved romtemperatur over natten, og løsningsmiddelet ble fordampet under redusert trykk for å gi 2-(1-metyletyl)-5-oxo-l-imi-dazolidinacetamid (3,4 g) som en viskos olje (Rf 0,33; etylacetat/metanol 6:4; kiselgelplater). Monohydratet av sulfatsaltet s.m.p. 64°C ble fast igjen med en endelig spaltning ved 114-118°C. 3) En oppløsning av 1,2 g (6,4 mmol) 2-(1-metyletyl)-5oxo-l-imidazolidinacetamid og 2 g 2-metylpropyl 3,4-epoksybutanoat i 40 ml vandig pentanol ble rørt ved 80°C 8 48 timer. Løsningsmiddelet ble fordampet under redusert trykk. Resten ble oppløst i 15 ml 15% HC1, vasket med 2x25 ml diklormetan, gjort basisk med natriumkarbonat og ekstrahert med 3x35 ml diklormetan. Det organiske lag ble tørket og fordampet for å gi 1,08 g av tittelforbindelse som en fargeløs olje (blanding av to diastereoisomerer, Rf 0,27 og 0,32 (diklormetan/metanol 9:1; kiselgelplater). Massespekt-rum (E.I., 70 eV, 1,5 mA), m/z = 300 (M<+> C3H7). IR-spektrum (ren): lambdamaks 3400, 3200, 1730 og 1690 cm"l.
Fremstilling 2
2- metylpropYl 3- karbamoYlmetyl- Beta- hYdroksy- 2-( i- metyl-etyl)- 4- oxo- 4- l- imidazolidinbutanoat
En oppløsning av 1.91 g (0,01 mol) glycylglycinamidacetat og 0,72 g (0,01 mol) isobutyraldehyd i 40 ml metanol ble rørt i 24 timer ved romtemperatur. Løsningsmiddelet ble fordampet under redusert trykk, og resten ble oppløst i 30 ml isopropanol og behandlet med 0,106 g natriumkarbonat, 1,8 g (0,0115 mol) 2-metylpropyl 3,4-epoksybutanoat og 2 dråper vann. Blandingen ble kokt under tilbakeløp i 3 6 timer. Etter fordampingen av løsningsmiddelet ble resten kromatografert på en kiselgelkolonne, eluert med diklormetan/metanol 9:1. Tittelforbindelsen (0,48 g) bestod av av to diastereoisomerer.
Eksempel 1
2-( 4- hvdroksv- 2- oxo- l- pvrrolidinacetamido) acetamid
En oppløsning av 11,8 g (34,3 mol) 2-metylpropyl 3-karbamoylmetyl-Beta-hydroksy-2-(1-metyletyl)-4-oxo-l-imidazolidin-butanoat i 90 ml dimetylformamid og 10 ml vann ble forsiktig kokt under tilbakeløp (utvendig temperatur 130°C) i 24 timer. Løsningsmiddelet ble fordampet under redusert trykk. Den oljeaktige rest ble tatt opp med 40 ml etanol og rørt ved 0°C i 30 minutter. Bunnfallet ble samlet og krystalli-sert fra etanol/isopropanol 1:1 for å gi 2,5 g av tittelforbindelsen som et hvitt pudder, s.m.p. 158-160°C. Massespekt-rum (E.I., 70 eV, 1,5 mA), m/z = 197 (M<+->H20), 180. IR-spektrum (olje mol): lamdamaks 3420, 3320, 3220, 1680, 1655 og 1630 cm~l.
Eksempel 2
2-( 4- hYdroksy- 2- oxo- l- pvrrolidinacetamido) acetamid
En blanding av etyl 4-hydrosky-2-oxo-l-pyrrolidinacetat (0,5 g) og glycinamid (0,6 g) ble rørt under nitrogen ved 80°C i 4 timer. Etter avkjøling ble blandingen kromatografert på en kiselgelkolonne ved eluering med diklormetan/metanol 7:3 og gav tittelforbindelsen (250 mg).
Eksempel 3
2-( 4- hydroksy- 2- oxo- l- pYrrolidinacetamido) acetamid
Til en iskald blanding ble 4-hydroksy-2-oxo-l-pyrrolidin-acetsyre (1 g) og glycinamid (700 mg) 8 dimetylformamid (20 ml), dicycloheksylkarbodiimid (1,3 g) raskt tilsatt. Røringen fortsatte i 5 timer ved romtemperatur, deretter ble løsningsmiddelet fjernet under vakuum. Resten ble suspen-dert i vann (20 ml) og rørt i 30 minutter ved romtemperatur. Det uoppløselige materiale ble filtrert fra, og filtra-tet ble inndampet til tørrhet. Resten ble findelt med 2-propanol for å gi tittelforbindelsen (700 mg).

karakterisert ved: A. å omsette en forbindelse med struktur (2) med vann og å cyklisere produktet intramolekylært:
hvori R<3> og R<4> er hver uavhengig hydrogen, C^_4-alkyl (rettkjedet eller forgrenet), og R<5> er C1_4-alkyl (rettkjedet eller forgrenet); eller B. å omsette en forbindelse med struktur (6)
i hvilken X er OH eller en lavere alkoksygruppe, med glycinamid.
