Description
Foreliggende oppfinnelse angår en analogifremgangsmåte for fremstilling av til nu ukjente 3'-demetoksyepipodofyllotoksinglukosidderivater.
Etoposid (VP-16, IA) og tenoposid (VM-26, IB) er klinisk nyttige anticancermidler som avledes fra det naturlig forekommende lignan, podofyllotoksin (II). Numererings-
systemet som anvendes for nomenklaturformål er vist i formel
II.
Etoposid og tenoposid er 4'-dimetylepipodofyllotoksin-glukosidderivater med den generelle formel:
idet epipodofyllotoksin er eplmeren av podofyllotoksin i 4-stilling. Etoposid og tenoposid er aktive ved behandling av forskjellige kreftformer, deriblant småcellet lungekreft, ikke-lymfocytisk leukemi og ikke-seminomatøs testikkelkreft ("AMA Drug Evaluation", 5. utgave, American Medical Associaton, 1983, Chicago, Illinois, s. 1554-1555). Etoposid og tenoposid samt fremgangsmåter for fremstilling derav er beskrevet i US-PS 3 524 844. Etoposid-3',4'-kinon (Illa) er fremstilt ved elektrokjemisk oksydasjon av etoposid (J.J.M. Holthuis et al., "J. Electroanal. Chem. Interfacial Electrochem.", 1985, 184 (2): 317-329). Fremstilling av kinonet (III) ved kjemisk oksydasjon er beskrevet i US-PS 4 609 644. Epipodofyllotoksin-3',4'-kinonderivater (III), der R<*> og R<**> har den nedenfor angitte betydning i forbindelse med formel (IV), tjener som utgangsmateriale ved fremstilling av 3'-demetoksyepipodofyllotoksinglukosidderivater ifølge foreliggende oppfinnelse.
Ifølge foreliggende oppfinnelse tilveiebringes det 3'-demetoksyepipodofyllotoksinglukosidderivater med formel (IV)
der
R<2> er hydrogen, og
R<1> er blant C^^Q-alkyl, C26-alkenyl.
De angjeldende forbindelser kan anvendes for inhlbering av tumorer hos pattedyr eller mennesker.
Forbindelsene ifølge oppfinnelsen kan fremstilles ved den nedenfor anførte reaksjonssekvens I reaksjonsskjerna 1 der R<3 >er Ci_5-alkyl eller aryl-Ci_5-alkyl og uttrykket "Epipodo" representerer fragmentet:
Forbindelsene med formel I fremstilles altså ved at man omsetter en ortokinonforbindelse med formel (III) der "Epipodo" betyr fragmentet med et O-substituert hydroksylamin med formelen
der
R<3> er Ci_5-alkyl eller aryl-C^_5-alkyl eller et syreaddisjonssalt derav i et lnert organisk oppløsningsmiddel under dannelse av en 3'-oksimeter med formel (V)
der
"Epipodo" og R<3> har den ovenfor angitte betydning, hvorefter man hvis ønskelig efter isolering og rensing omdanner 3'-okslmeteren (V) ved behandling med et mildt reduksjonsmiddel eller hydrogen i nærvær av en katalysator til aminforbindelsen (VI)
der
"Epipodo" har den ovenfor angitte betydning, eller frem-stiller aminforbindelsen direkte fra ortokinonforbindelsen med formel (III) ved omsetning med ammoniakk eller et alkylamin under oppnåelse av en aminforbindelse med formel (VI), diazoterer aminforbindelsen (VI) under oppnåelse av diazoniumsaltet (VII)
og reduserer diazoniumsaltet under dannelse av den ønskede forbindelse.
Ortokinonene (III) er som nevnt tidligere kjente forbindelser og kan fremstilles ved oksydasjon av 4'-dimetylepipodofyllo-toksinglukosider ved den fremgangsmåte som er beskrevet i US-PS 4 609 644. Omsetning av ortokinonene (III) med et 0-substituert hydroksylamin eller et syreaddisjonssalt derav i et Inert organisk oppløsningsmiddel tilveiebringer den tilsvarende 3'-oksimeter (V). Omsetningen utføres fortrinnsvis ved romtemperatur i et tidsrom som er tilstrekkelig til å oppnå monooksimeteren, for eksempel i en periode fra 30 minutter til ca. 1 time. De således dannede produkter kan isoleres og renses for eksempel ved flashkromatografi, eller alternativt kan de reduseres direkte uten isolering på forhånd til den tilsvarende aminforbindelse med formel (VI). Reduksjonen av oksimeteren til den tilsvarende 3'-amino-forbindelse kan utføres på konvensjonell måte, for eksempel ved hjelp av et mildt kjemisk reduksjonsmiddel, eller ved hydrogenering i nærvær av en egnet katalysator som Pt, Pd, NI, Ru eller Rh. Fortrinnsvis benytter man seg av katalytisk hydrogenering. Aminforbindelser med formel (VI) kan også fremstilles direkte fra ortokinonet (III) ved behandling med ammoniakk eller alkylamin ved romtemperatur idet omsetning med den sistnevnte både gir aminet (VI) og det tilsvarende alkylsubstituerte amin. Den foretrukne fremgangsmåte er reduksjon av oksimeteren med formel (V). Diazotering av (VI) i et inert oppløsningsmiddel ved redusert temperatur fulgt av vandig opparbeiding gir diazoniumsaltet (VII). Reduksjon av dette med kjente reagenser som hypofosforsyre, natrium-borhydrid eller et overskudd av tlofenol gir 3'-demetoksy-4'-demetylepipodofyllotoksin med formel (IV).
Biologisk aktivitet
3'-demetoksyetoposid er bedømt med henblikk på antitumorvirk-ning mot transplanterbar murin-P388-leukemi.
I GDF^-hunnmus implanteres lntraperitonealt et tumorinokulum bestående av 10^ ascittceller fra P388-murinleukemi og de behandles med forskjellige doser av en prøveforbindelse idet det for hvert dosisnivå anvendes 4 mus og for en med saltvann behandlet kontrollgruppe ble det anvendt 10 mus. Forbindelsene ble administrert ved intraperitoneal injeksjon den 5. og 8. dag (dag 1 er tumorimplanteringsdagen). Antitumor-aktiviteten uttrykkes som % T/C, noe som betyr forholdet mellom den midlere overlevelsestid for den med preparat behandlede gruppe og den midlere overlevelsestid for den med saltvann behandlede kontrollgruppe. En forbindelse med en T/C-Sé-verdi på 125# eller mer må ansees vanligvis å ha signifikant antitumoraktivitet ved P388-prøven. Forsøket varte i 31 dager hvorefter antall overlevende dyr ble tellet. Tabell I viser resultatene av denne vurdering idet kun den maksimale T/C-#-verdi og dosen som viser den maksimale virkning er angitt.
Det fremgår av resultatene av det ovenfor angitte dyreforsøk at forbindelsene med formel (IV) er i besittelse av en effektiv, inhiberende virkning overfor tumorer i pattedyr. De angjeldende forbindelser kan som følge av dette anvendes for inhlberlng av tumorer 1 pattedyr og mennesker.
De følgende eksempler tjener til å belyse oppfinnelsen.
I de følgende eksempler ble smeltepunktene registrert på et Thomas-Hoover-kapillær-smeltepunktsapparat og er ikke korrigert. ^H-NMR-spektra ble registrert enten på et Bruker "KVM-360" eller Varian "VX2-200"-spektrofotometer (under anvendelse av CDCI3 som intern referanse). Kjemiske skift ble registrert i S-enheter og koblingskonstanter i Hertz. Oppdellngsmønstre gis følgende betegnelser: s = singlett, d = dublett, t - triplett, q = kvartett, m = multiplett, bp = bred topp og dd = dublett av dublett. IR-spektra ble bestemt enten på Beckman "Model 4240"- eller Perkin-Elmer "1800" Infrarødt fouriertransformasjons-spektrofotometer og er angitt i resiproke centimeter (cm-<1>). Tynnsjiktskromatografi, TLC, ble gjennomført på forbelagte silikagelplater (60F-254) under anvendelse av UV-lys og/eller joddamper som Visuali-seringsmidler. Massespektra med høy og lav oppløsning ble registrert på et "Kratos MS 50"- henholdsvis "Kratos MS 25RFA"-spektrofotometer. "Flashkromatografi" angir den metode som er beskrevet av W.C. Still et al. i "J. Org. Chem.", 1978, 43:2923, og ble utført under anvendelse av enten E. Merck silikagel (200-400 mesh) eller Woelm silikagel (32-63 pm). Avdamping av oppløsningsmidler ble gjennomført under redusert trykk.
Eksempel 1
Etoposid- orto- klnon- 3'- O- metvloksim ( Va)
En oppløsning av 350 mg (0,611 mmol) etoposid-orto-kinon (Illa) i 20 ml pyridin ble behandlet med en oppløsning av 350 mg (4,19 mmol) metoksylamin.hydroklorid i 10 ml pyridin. Den resulterende oransjefarvede oppløsning ble omrørt i 30 minutter ved romtemperatur og derefter ble pyridin fjernet under vakuum. Resten ble oppløst i 50 ml CH2CI2 og ble fordelt mellom den organiske fase og en blanding 20 ml H2O og 10 ml IN HC1. Det vandige sjikt ble ekstrahert ytterligere med 25 ml CItøClg og de kombinerte organiske ekstrakter tørket over MgS04. Oppløsningsmidlet ble dampet av i vakuum og man oppnådde en mørk oransjefarvet olje. Flashkromatografi på 14 g silikagel med 5% CH3OH i CH2C12 ga 243 mg tilsvarende 6656 av tittelforbindelsen som et oransjefarvet faststoff. Tri-turering med Et20 ga en analytisk prøve. Dette oksim ble generelt ikke renset i større skala, men hydrogenert direkte til aminet (Via) i et totalt utbytte på ca. 70%.

6,07 (d, 1H), 6,01 (d, 1H), 5,75 (d, 1H), 4,92 (d, 1H), 4,76 (q, 1H), 4,66 (d, 1H), 4,50 (dd, 1H), 4,38 (dd, 1H), 4,27 (d, 1H), 4,22-4,17 (m, 1H), 4,15 (s, 3H), 3,79 (s, 3H) , 3,78-3,74 (m, 1H), 3,63-3,58 (m, 1H), 3,44 (dd, 1H), 3,38-3,30 (m, 3H), 2,95-2,87 (m, 1H), 1,40 (d, 3H).
Analyse for C2<gH>3iN013:
Eksempel 2 3- amino- 3'- dernetoksv- etoposid ( Via)
Den urene oksim (Va) oppnådd fra 4,1 g (7,2 mmol) etoposid-orto-kinon (Illa) og 4,1 g (49 mmol) metoksylamin.hydroklorid ved fremgangsmåten som angitt i eksempel 1 ble oppløst i 275 ml alkohol av reagenskvalitet og behandlet med 290 mg 20 %- lg palladiumhydroksyd-på-karbon og 1,6 g 10 %- ig palladium-på-karbon. Blandingen ble hydrogenert ved 276-345 kPa. Efter 16 timer ble blandingen filtrert gjennom "Celite", vasket med etylacetat og oppløsningsmidlet dampet av. Det urene produkt ble renset ved flashkromatografi på 300 g E. Merck 230-400 mesh silikagel under eluering med EtOAciheksan 8:2 hvorved man oppnådde 2,89 g eller 70% totalt utbytte av tittelforbindelsen som et hvitt faststoff. Omkrystallisering fra etanol ga den analytiske prøve.

Analyse for C28<H>3iN0^2:
Eksempel 3 Indre- 3'- diazonlumhvdroksvd- etoposldsalt ( VIIa)
3,0 ml (26,2 mmol) lseddikk fulgt av 0,15 g (2,17 mmol) NaN02 ble satt til en oppløsning av 0,22 g (0,384 mmol) 3'-amino-etoposid (produkt fra eksempel 2) i 17 ml tørr THF under omrøring ved _0°C under N2. Reaksjonsblandlngen ble omrørt i 3,4 timer ved 0°C og helt i 150 ml CH2CI2. Det mørkerøde organiske sjikt ble vasket med 100 ml vandig NaHC03. De kombinerte organiske ekstrakter ble vasket med 100 ml mettet NaHCC"3, tørket over MgSC-4 og konsentrert i vakuum for å oppnå 0,177 g tilsvarende 79# av et rødlig oransjefarvet faststoff med smeltepunkt 150°C under langsom dekomponer ing.

Eksempel 4
3'- demetoksv- etoposid ( IV)
Det urene etoposid-diazoniumsalt i eksempel 3 (1,03 g, 1,76 mmol) ble oppløst 1 100 ml absolutt metanol og behandlet med 400 mg natrlumborhydridpulver fulgt 5 minutter senere av tilsetning av 5 ml iseddik. Blandingen ble omrørt ved romtemperatur i 2 timer, oppløsningsmidlet dampet av i vakuum og resten behandlet med 100 ml H20 og ekstrahert med 100 ml og derefter 2 x 50 ml CH2C12. De kombinerte ekstrakter ble vasket med 25 ml mettet vandig natriumbikarbonat og 75 ml saltvann og tørket over MgS04. Rotasjonsfordamping fulgt av flashkromatografi på 32 g silikagel og eluering med 3-436 CH3OH i CH2C12 ga 400 mg tilsvarende 41% av tittelforbindelsen som et farveløst faststoff med smeltepunkt 190-195°C.

- Analogifremgangsmåte for fremstilling av et terapeutisk aktivt 3'-demetoksyepIpodofyllotoksinglukosidderivat med den generelle formel derR<2> er hydrogen; og R<1> er Ci_6-alkyl, karakterisert ved at man omsetter en ortoklnonforbindelse med formel (III) der "Epipodo" betyr fragmentet med et O-substituert hydroksylamin med formelenderR<3> er Ci_5-alkyl eller ar<y>l-Ci_5~alkyl eller et syreaddisjonssalt derav i et inert organisk oppløsningsmiddel under dannelse av en 3'-oksimeter med formel (V)der"Epipodo" og R<3> har den ovenfor angitte betydning, hvorefter man hvis ønskelig efter isolering og rensing omdanner 3'-oksimeteren (V) ved behandling med et mildt reduksjonsmiddel eller hydrogen i nærvær av en katalysator til aminforbindelsen (VI) der "Epipodo" har den ovenfor angitte betydning, eller frem-stiller aminforbindelsen direkte fra ortokinonforbindelsen med formel (III) ved omsetning med ammoniakk eller et alkylamin under oppnåelse av en aminforbindelse med formel (VI), diazoterer aminforbindelsen (VI) under oppnåelse av diazoniumsaltet (VII) og reduserer diazoniumsaltet under dannelse av den ønskede forbindelse.
