Description
Foreliggende oppfinnelse angår en ny fremgangsmåte
ved fremstilling av 2-(2-thienyl)- og 2-(3-thienyl)-ethylamin-derivater med formelen:
hvor
R^ står i 2-, 3-, 4- eller 5-stillingen og er hydrogen, halogen,nitro, carboxy, cyano, amino eller rettkjedet eller forgrenet alkyl eller alkoxy med 1-4 carbonatomer, aminoethylkjeden står i 2- eller 3-stillingen,
R2, R3 og R 4 er hydrogen, alkyl med 1-4 carbonatomer eller fenyl, og
Ar er en thienyl-, furyl-, pyridyl-, fenyl- eller nafthyl-gruppe som eventuelt er mono- eller flersubstituert med alkyl, fenyl, halogen, nitro, cyano, amino, carboxyl eller alkoxy.
En rekke av forbindelsene med formel (I) er kjent og anvendes som utgangsmaterialer ved fremstilling av forbindelser som er nyttige både i den kjemiske og farmasøytiske industri.
Blant forbindelsene som fåes ved den nye fremgangsmåte kan nevnes slike som har aminoethylkjeden i 2-stilling og
substituenten i 4- eller 5-stilling og som ved hjelp av kjente fremgangsmåter kan overfores til 4,5 , 6,7-tetrahydrothieno-(3,2-c)-pyridinderivater, og slike som har aminoethylkjeden i 3-stilling og substituenten R^ i 4- eller 5-stilling og som ved hjelp av kjente fremgangsmåter kan overføres til 4,5,6,7-tetrahydrothieno-(2,3-c)-pyridinderivater. Begge typer derivater er beskrevet i en rekke patentskrifter (FR 73 03 503;
FR 75 03 968; FR 75 20 241; FR 75 23 786; FR 75 24 486; FR 76 00 003; FR 77 21 517) hvor det redegjøres for deres terapeu-tiske anvendelser og/eller deres fremstilling.
For fremstilling av de innledningsvis angitte forbindelser er det tidligere blitt benyttet en flertrinns fremgangsmåte som er beskrevet i ovennevnte franske patentskrift FR 75 03 968. Denne fremgangsmåte har imidlertid betydelige ulemper, som unngåes ved den nye fremgangsmåte som nu fore-slåes. Ved den nye fremgangsmåte kreves det således ikke anvendelse av baser som er farlige og besværlige å arbeide med, såsom visse organolithiumbaser eller organonatriumbaser som kreves ved den kjente fremgangsmåte. Den nye fremgangsmåte krever heller ikke, slik som den kjente fremgansgmåte, bruk av ethylenoxd eller høyere derivater derav, hvis anvendelse krever omfattende sikkerhetsforanstaltninger når fremgangsmåten skal utføres i industriell målestokk.
I henhold til oppfinnelsen tilveiebringes det nu en enklere, sikrere og billigere fremgangsmåte for fremstilling av de innledningsvis angitte 2-(2-thienyl)- og 2-(3-thienyl)-ethylaminderivater, hvilken fremgangsmåte er særpreget ved at en forbindelse med den generelle formel:
hvor X og Y uavhengig av hverandre er en alkyl-, aryl-, alkoxy-, dialkylamino- eller diarylaminogruppe, kondenseres med en carbonylforbindelse med den generelle formel: hvor R1 er som ovenfor angitt, til en forbindelse med den generelle formel:
hvor X, Y og R 1 er som ovenfor angitt, som så behandles med en base med formelen NP hvor M<®> er et alkalimetall-, jordalkalimetall- eller organomagnesiumjon, og er et anion valgt blant negativt ladede grupper bestående av hydrogen, hydroxy, amino, mono- eller dialkylamino, rettkjedet eller forgrenet alkyl og rettkjedet eller forgrenet
alkoxy, for dannelse av et carbanion med den generelle formel:

alternativ (b): - i hvilket tilfelle basen ^B© anvendes i en støkiometrisk mengde - forbindelsen med formel (VI) omsettes med den halogenerte forbindelse med formel (IX) ovenfor og den erholdte forbindelse deretter reduseres, hvorved forbindelsen med formel (X) fåes direkte,
hvoretter forbindelsen med formel (X) erholdt i henhold til det ene eller det andre av de to alternativer omsettes med en syre til en forbindelse med formel (I).
Fremgangsmåten ifølge oppfinnelsen for fremstilling
av forbindelser med formel (I) kan illustreres ved følgende reaksjonsskjema (som viser alternativ (a) hva angår overfø-ringen av forbindelsen (VI) til forbindelsen (X))':
Det vil også være mulig (innenfor alternativ (a)) å bytte om rekkefølgen av trinnene d og e, slik som i det føl-gende :
Fremgangsmåten kan med fordel utføres som følger:
(a) Organofosforforbindelsene med formel (II), som er lett tilgjengelige ved velkjente fremstillingsmetoder, som f.eks. den beskrevet av I. C. Popoff og medarbeidere (J. Org.
Chem. 281,. 2898 (1963)), kan omsettes med carbonylderivater (III) i fravær'av oppløsningsmiddel og av katalysator, idet vannet som dannés i løpet av reaksjonen, fjernes ved pass-ende midler ved slutten av prosessen. Kondensasjonen kan med fordel utføres i et oppløsningsmiddel som et aromatisk hydrocarbon, f.eks-, toluen, eller en alkohol, f.eks. ethanol, fra hvilket vannet kan fjernes ved azeotrop destillasjon. Kondensasjonen kan også med fordel utføres (fra et hastig-hetssynspunkt) i nærvær av katalytiske mengder av en uorganisk eller: organisk syre som f.eks. paratoluensulfonsyre. Temperaturen ved hvilken omdannelsen utføres kan variere, men ligger mest alminnelig mellom 20°C og 120°C.
(b) (c) Ba.sen Bv-M~' anvendt i dette trinn kan være et alkalimetallhydrid, typisk natrium-, lithium- eller kaliumhydrid, et alkalimetallamid eller -alkylamid, typisk et dialkyl-
amid som lithium-diisopropylamid, en organometallforbindelse, typisk en organolithiumforbindelse som n-butyllithium, eller en organonatrium- eller organomagnesiumforbindelse. Nyttige er også alkalimetall- eller jordalkalimetall-alkoxyder som
natrium-, lithium-, kalium- og magnesium-methoxyder, kalium-t-butoxyd, nåtrium-t-amylat. Alkalimetall- eller jordalkalimetall-hydroxyder som natrium-, lithium-, kalium- og magnesium-hydroxyder kan også anvendes.
I alminnelighet anvendes basen i en støkiometrisk mengde, eller endog i et lite overskudd, f.eks. i et 10%-ig overskudd med hensyn til den ekvivalente mengde. Det er imidlertid også mulig å anvende lavere, endog betydelig lavere, mengder enn den støkiometrisk ekvivalente.
Det bør understrekes at når modifikasjonen i henhold til hvilken rekkefølgen av trinnene d og e er omvendt, anvendes, og basen anvendes i en støkiometrisk ekvivalent mengde, kan det være fordelaktig å unngå isolering av derivatet (VII) og direkte omsette forbindelse (VII) med det halogenerte derivat (IX), og således unngå anvendelse av basen (alternativ (b) ved fremgansgmåten).
Fremgangsmåten utføres i alminnelighet ved en temperatur mellom -78°C og +150°C, og ved en temperatur mere spesifikt valgt i forhold til naturen av basen B^^, innen de høyere temperaturer av området, særlig ved utførelse av trinn (c).
De foretrukne oppløsningsmidler er rettkjedede eller cycliske ethere som tetrahydrofuran;'hydrocarboner, typisk aromatiske hydrocarboner som benzen, toluen, xylener; alkoholer; amider, typisk dimethylformamid; sulfoxyder, typisk dimethylsulfoxyd. Det kan også være fordelaktig, særlig når metallhydroxyder anvendes, å arbeide i et tofase-system (vann + oppløsningsmiddel som et halogenert oppløs-ningsmiddel, f.eks. diklormethan, eller et aromatisk hydrocarbon som benzen, toluen, xylene) i nærvær av en fase-overføringskatalysator, typisk et kvartært ammoniumsalt som tetra-n-butylammoniumjodid, eller et fosfoniumsalt. Forbindelse (VII) kan isoleres ved vanlige metoder.
(d) Reduksjonen av derivatet med formel (VII) utføres med
fordel med et blandet alkalimetallhydrid, typisk et bor-hydrid som natriumborhydrid eller kaliumborhydrid. Reduksjonen utføres i et inert oppløsningsmiddel som en ether, f.eks. tetrahydrofuran eller dioxan, eller endog i en alkohol, f.eks. methanol eller ethanol.
Denne reduksjon kan også utføres ved.katalytisk hydrogenering i homogen eller heterogen fase, under velkjente betingelser. (e) Basen B anvendt i den første del av dette femte trinn, kan velges blant de under (b) og (c) ovenfor angitte. Den anvendes i alminnelighet i en støkiometrisk ekvivalent mengde, men den kan anvendes i et lite overskudd, f.eks. et 5 eller 10% overskudd, i forhold til den støkiometrisk ekvivalente mengde.
Fremgangsmåten utføres i alminnelighet mellom -20°C
og +100^C, idet temperaturene i den lavere del av området foretrekkes. De anvendte oppløsningsmidler er de som er beskrevet for trinn (b) ovenfor.
I den annen del av dette trinn omsettes den halogenerte forbindelse med formel (IX) med reaksjonsmediet som angitt ovenfor, ved en temperatur stort sett svarende til den i den første del. (f) Den syrekatalyserte spaltning av fosfor-nitrogen-bindingen i derivatet (IX) kan utføres med en uorganisk syre som en hydrohalogensyre, f.eks. saltsyre eller hydrogen-bromid, men også med en organisk syre, særlig en sterk syre som en sulfonsyre, f.eks. benzensulfonsyre eller paratoluensulfonsyre. De foretrukne oppløsningsmidler er ethere, særlig cycliske ethere som tetrahydrofuran eller dicxan, alkoholer som methanol eller ethanol, amider som dimethylformamid, og sulfoxyder som dimethylsulfoxyd. Det er mulig å arbeide i slike oppløsningsmidler i fravær av vann, men også i blandinger inneholdende vann i varierende mengder. Dessuten er det også mulig å arbeide i vann alene.
En eller to støkiometriske ekvivalenter av syre anvendes i alminnelighet.
Fremgangsmåten utføres i alminnelighet mellom 0°C og 100°C og mere spesielt mellom 30°C og 70°C.
De dannede forbindelser med formel (I) kan så isoleres og renses ved konvensjonelle metoder. I denne hen-sikt kan det være fordelaktig å overføre de frie forbindelser med formel (I) til deres salter, f.eks. til deres syreaddisjonssalter, ved omsetning med uorganiske eller organiske syrer. Forbindelsene med formel (I) kan frigjøres fra slike salter ved kjente metoder.
De følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Fremstilling av N- orthoklorbenzyl- 2-( 2- thienyl)- ethylamin-hydroklorid

Til 16,7 g (0,1 mol) diethyl-aminomethylfosfonat i
200 ml absolutt ethanol tilsettes 11,2 g (0,1 mol) 2-then-aldehyd, og blandingen kokes under tilbakeløp i 30 minutter. Vannet dannet under reaksjonen, fjernes ved azeotrop destillasjon. Efter fullstendig fordampning av oppløsningsmidlet fåes 28 g (ca. 100%) av en kromatografisk ren (LPC, TLC og
GPC) gul olje.

Til en suspensjon av 11,2 g (0,1 mol) kalium-t-butoxyd i 160 ml THF (tetrahydrofuran) tilsettes dråpevis en oppløsning av .27,9 g (0,1 mol) diethyl-N-(2-thenyliden)-aminomethylfosfonat i 40 ml THF. i løpet av tilsetningen stiger temperaturen fra 20°C til 35°C. Ved avslutning av tilsetningen heves temperaturen til 40-45°C i 30 minutter, hvorefter reaksjonsmediet helles i 400 ml mettet vandig ammoniumkloridoppløsning. Den vandige fase ekstraheres med isopropyletherj de forenede etherfaser vaskes så med en mettet natriumkloridoppløsning, tørres over natriumsulfat og inndampes, hvorved man får 20,9 g (75%) av tittelproduktet, som en gul olje.

De 20,9 g (0,075 mol) diethyl-(3- (2-thienyl) -N-vinyl-fosforamidat erholdt ovenfor tilsettes til en oppløsning av 2,85 g (0,075 mol) natriumborhydrid i 200 ml ethanol.
I løpet av tilsetningen øker temperaturen og blir så stabil ved 30°C. Efter omrøring i ytterligere 2 timer heves temperaturen av mediet til 45-50°C i 1 time, hvorefter ethanolen avdampes og det dannede residuum taes opp i isopropylether-vann.
Den vandige fase tilbakeekstraheres gjentatte ganger med isopropylether, og de forenede organiske faser vaskes med vann, tørres over natriumsulfat og inndampes, hvorved man får 21 g (ca. 75% med hensyn på aminomethylfosfonat-utgangs-materialet) av tittelproduktet, som en gul olje,

Til en suspensjon av 3,6 g (0,075 mol) natriumhydrid (50% i olje) i 150 ml toluen tilsettes dråpevis under omrør-ing ved værelsetemperatur 21 g (0,075 mol) diethyl-N-[2-(2-thienyl)-ethyl]-fosforamidat i 20 ml toluen. Reaksjonsmediet blir rødt; det oppvarmes til 80°C i 1 time, hvorefter 16,1 g (0,1 mol) orthoklorbenzylklorid tilsettes dråpevis dertil. Reaksjonsblandingen omrøres kontinuerlig ved 80°C i 3 timer og blir efter avkjøling vasket med vann. Toluenfasen dampes så av, hvorved man får fosforamidat-tittelforbindelsen (forurenset med overskudd av orthoklorbenzylklorid) som en olje som anvendes som sådan i det påfølgende trinn.
En ren prøve ble erholdt ved kolonnekromatografi (siliciumoxyd; elueringsmiddel: ethylacetat).

Til en suspensjon av 11,2 g (0,1 mol) kalium-t-butoxyd i 160 ml THF (tetrahydrofuran) tilsettes dråpevis en oppløs-ning av 27,9 g (0,1 mol) diethyl-N-(2-thenyliden)-aminomethylfosfonat i 40 ml THF. I løpet av reaksjonen økes temperaturen fra 20°C til 35°C. Ved avslutning av tilsetningen heves temperaturen til 40-45°C i 30 minutter, hvorefter 1,6 g (0,1 mol) orthoklorbenzylklorid tilsettes dråpevis til reaksjonsmediet. Efter avsluttet tilsetning kokes reaksjonsmediet under tilbakeløp i 3 timer, og THF fordampes så. Det erholdte residuum taes opp i isopropylether; etherfasen vaskes med vann, tørres og inndampes, hvorved man får 32,7 g (85%) av tittelproduktet, som en orangegul olje som anvendes som sådan i det påfølgende trinn.
En prøve renset ved kolonnekromatografi (siliciumoxyd; elueringsmiddel: ethylacetat) oppviser de følgende kjenne-tegn:

Det urene produkt erholdt i det foregående trinn tilsettes til en suspensjon av 6,5 g (0,17 mol) natriumborhydrid i 100 ml dioxan. Til reaksjonsmediet avkjølt til 0°C tilsettes dråpevis 14,3 g (0,17 mol) trifluoreddiksyre. Efter avsluttet tilsetning kokes det erholdte materiale under til-bakeløp i 1 time og hydrolyseres efter avkjøling ved tilsetning av 200 ml vann. Reaksjonsmediet ekstraheres med methylenklorid. Den isolerte organiske fase tørres over natriumsulfat og inndampes, hvorved man får 33 g (85% med hensyn på aminomethylfosfonat-utgangsforbindelsen) av fosforamidat-tittelforbindelsen som en gul olje. En prøve renset ved kolonnekromatografi over siliciumdioxyd oppviser eqen-skaper identiske med dem for forbindelsen erholdt i eksempel 1.

1. Fremgangsmåte ved fremstilling av 2-(2-thienyl)- og 2-(3-thienyl)-ethylaminderivater med den generelle formel:

hvoretter forbindelsen med formel (X) erholdt i henhold til det ene eller det andre av de to alternativer omsettes med en syre til en forbindelse med formel (I).
