Description
Det er kjent at den egentlige sentrale hostelindrende virkning av antitussive midler kan bistås av en periferisk lokal-anestetisk virkning som de samtidig oppviser. Slike antitussive midler av typen lokalanestetika er f.eks. anestesin (p-amino-benzoesyre-etyl-ester) og benzonatat (p-n-butylamino-benzoesyre-nona-etylenglykol-ester-metyleter).
Den foreliggende oppfinnelse har til oppgave å gi anvisning på en analogifremgangsmåte til fremstilling av antitussive forbindelser av typen lokalanestetika som til forskjell fra de ovennevnte midler av estertype, er anilider. Vedkommende forbindelser har følgende formel
I denne formel betegner n et helt tall, nemlig 2 eller 3,

og substituentene R og R betegner separat for seg en alkylgruppe med 1-3 karbonatomer, eller de danner sammen med nitrogenatomet de er bundet til, en piperidinring substituert med en metylgruppe.
Foruten de ovennevnte frie baser med den generelle formel (I) innbefatter oppfinnelsen også fremstilling av deres ikke-toxiske syreaddisjonssalter.
Forbindelsene med formel I fremstilles ved acylering av 4,6-dimety1-o-anisidin
med et halogen-karboksylsyre-halogenid med den generelle formel hvor Hal betegner klor eller brom og n har samme betydning som ovenfor, og ved omsetning av det således fremkommende halogen-karboksylsyre-anilid med formelen hvor n og Hal har de ovennevnte betydninger, med en aminoforbindelse med den generelle formel
hvor R 1 og R 2 har betydninger som angitt ovenfor.
Ved reaksjonen mellom halogen-karboksylsyre-anilidet med
formelen IV og aminoforbindelsen med formelen V anvendes med fordel den sistnevnte i overskudd for å binde det ved reaksjonen frigjorte hydrogenhalogenid. Det er i den forbindelse også mulig å utføre reaksjonen uten oppløsningsmiddel. Temperaturer mellom værelse-temperatur og opp til 120°C kommer i betraktning. Hvis det ønskes, kan der som oppløsningsmiddel anvendes f.eks. etanol, benzen eller xylen. Det produkt som fremkommer ved reaksjonen, er et basisk anilid med formel I som kan isoleres og renses som sådant. Fortrinnsvis blir det imidlertid ved hjelp av klornitrogengass omdannet direkte til hydroklorid, ut fra hvilket det, hvis det ønskes, også
lett er mulig å få den frie base ved å frigjøre denne ved hjelp av natriumkarbonat.
Acyleringen av 4,6-dimetyl-o-anilidet med formel II med et halogen-karboksylsyre-halogenid med formel III utføres fortrinnsvis i en buffertløsning bestående av en blanding av eddiksyre og natriumacetat under anvendelse av utgangsmaterialene med formel II og III i omtrent ekvimolekylære mengder.
4,6-dimetyl-o-anisidet som har formel II, og som benyttes som utgangsmateriale, kan fremstilles ut fra 3,5-dimetylfenol på en måte som er kjent fra litteraturen (Auwers og Borsche, Ber. 48, 198-1716 (1915)).
Forbindelsene . med formel I er nye forbindelser som foruten ved lokalanestetiske egenskaper også utmerker seg ved en kraftig antitussiv virkning. Denne gir seg til kjenne ved at forbindelsene ved intravenøs eller peroral administrering kurerer eller lindrer hoste bevirket ved elektrisk irritasjon av strupe-hovednerven (nervus laryngeus sup.) hos bedøvede katter eller ved tilførsel av ammoniaktåke eller svoveldioksyd til luft innåndet av ikke bedøvede marsvin.
Der er fra før kjent aminosyre-anilider og -xylidider som har vært anvendt som lokalanestetika. Mest kjent blant disse er lidokain eller 2-dietyl-amino-2',6<1->acetoxi-xylidid. Disse kjente forbindelser skiller seg fra forbindelsene med formel I særlig når det gjelder substitueringen av den aromatiske gruppe, men delvis også hva aminosyredelen angår. Noen antitussiv virkning hiar ikke vært påvist hos disse forbindelser når unntas at lidokain har vært påvist å ha en lindrende virkning på hostereflekser ved intravenøs administrering (U. Steinhaus 6 L. Gaskin, Anesthesiology 1963, 24, 285-90). Med tanke på hostemedisiner er dette imidlertid uten betydning, siden lidokain ikke egner seg for oral anvendelse.
Med hensyn til den antitussive virkning er forbindelsene med formel I mest effektive ved oral administrering.
Ved undersøkelse av forbindelsene ifølge de nedenstående eksempler 1-3 ved Domenjoz' metode (Naunyn-Schmiedebergs Arch.exp.Pathol. Pharmacol. 1952, bind 215, side 18) viste forbindelsene en tydelig virkning ved elektrisk irritasjon av strupehode-nerven (nervus laryngeus sup.) hos bedøvede katter, men virkningen var svakere enn med kodeinfosfat.
Ved ikke bedøvede marsvin og inhalering av ammoniak (Kållqvist
& Melander, Arzneimittelforschung 1957, 1_, side 301-304) virket 10 ml/kg av forbindelsen ifølge eksempel 1 ved peroral administrering mer effektivt enn 15 mg/kg kodeinfosfat. En dose på 20 mg/kg av forbindelsen ifølge eksempel 2 hadde klart mer effektiv virkning enn 15 mg/kg kodeinfosfat, mens allerede en dose på 2,5 mg/kg av forbindelsen ifølge eksempel 3 virket mer effektivt enn 15 mg/kg kodeinfosfat.
Ved ikke bedøvede marsvin og inhalering av svoveldioksyd
virket 20 mg/kg av forbindelsen ifølge eksempel 1 ved peroral administrering mer effektivt enn 5 mg/kg kodeinfosfat, og 20 mg/kg av forbindelsen ifølge eksempel 2 virket mer effektivt enn 15 mg/kg kodeinfosfat, mens en dose på 2,5 mg/kg av forbindelsen ifølge eksempel 3 er tilstrekkelig til å gi mer effektiv virkning enn 5 mg/kg kodeinfosfat.
Den lokalanestetiske virkning av forbindelsen ifølge eksempel 1 var ved infiltrasjonsbedøvelse ca. 1,6 og ved ledningsbedøvelse ca. 1,4 ganger kraftigere enn hos lidokain. Tilsvarende verdier for forbindelsen ifølge eksempel 2 var ca. 1,6 og ca. 3 ganger kraftigere enn hos lidokain og for forbindelsen ifølge eksempel 3 ca. 4 og ca. 1,8 ganger kraftigere enn hos lidokain.
Den akutte toxisitet i mus ved peroral administrering var for
forbindelsen ifølge eksempel 1 av samme størrelsesorden som for kode-in og lidokain, for forbindelsen ifølge eksempel 2 noe høyere, og for forbindelsen ifølge eksempel 3 noe lavere enn for de nevnte referanse-stoffer.
Fremgangsmåten ifølge oppfinnelsen vil i det følgende bli belyst ved eksempler.
Eksempel 1
a) 4- klor- 4', 6'- dimetyl- o- butyranisidid
I en oppløsning av 110 g (0,73 mol) 4,6-dimetyl-o-anisidin
(Auwers & Borsche, Ber. 48, 1698-1716 (1915)) i 340 ml iseddik til-settes under kjøling og kraftig omrøring 113 g (0,8 mol) 4-klorbuty-ryl-klorid. Den fremkomne oppløsning blir under omrøring tilsatt i en annen op<p>løsning inneholdende 222 g krystallinsk natriumacetat i 665 ml vann. Omrøringen ble fortsatt i en time ved værelsestempe-ratur. Den fremkomne felling ble filtrert, vasket omhyggelig med vann og tørket. Utbytte 135,8 g (72,8%). Ved omkrystallisasjon
fra benzen fremkom smeltepunkt 108-110°C.

Funnet Cl 13,52; N 5,57
b) 4- ( dietylamino)- 4', 6'- dimetyl- o- butyranisidid
En.blanding av 61,8 g (0,24 mol) 4-klor-4',6'-dimetyl-o-butyranisidid, 360 ml dietylamin og 1 g kaliumjodid ble oppvarmet i 48 timer ved 65°C. Overskuddet av dietylamin ble avdestillert/og i resten ble der tilsatt vann og natriumkarbonat (25 g). Blandingen ble ekstrahert med diklormetan. Ekstrakten ble inndunstet til tørrhet, resten ble oppløst i vannfri aceton og mettet med HCl-gass. Produktet ble her-under utfelt i form av hydroklorid, som ble filtrert og vasket med vannfri aceton. Utbytte 52,7 g (66%). Ved omkrystallisasjon av råproduktet fra isopropanol fikk man 50,0 g (62,5%) rent hydroklorid, smeltepunkt 185-186°C.

Funnet C 62,45; H 8,62; N 8,39
For istedenfor hydrokloridet å få den frie base behandlet man det førstnevnte med en sodaoppløsning. Den frigjorte base ble ekstrahert med diklormetan, oppløsningen ble tørket med kaliumkarbonat og filtrert, og oppløsningsmiddelet drevet av. Ved omkrystallisasjon fra petroleter (kokepunkt 40°C) forsynt med en tilsetning av noe benzen, fikk man den frie base, smeltepunkt 92-93°C.

Eksempel 2
a) 3- klor- 4', 6'- dimetyl- o- propionanisidid
Ved anvendelse av 92,8 g (0,615 mol) 4,6-dimetyl-o-anisidin og
86 g (0,68 mol) 3-klorpropinyl-klorid som utgangsmateriale fikk man ved å gå frem som i eksempel la) 115,3 g (77,4%) 3-klor-4',6'-dimetyl-o-propionanisidid , smeltepunkt 123-125°C (krystallisert fra benzen).
b) 3- @ipropylamino)- 4', 6'- dimetyl- o- propionanisidid
En blanding av 50 g (0,206 mol) 3-klor-4<1>,6'-dimetyl-o-propionanisidid, 310 ml di-n-propylamin og 1 g kaliumjodid ble varmet opp i 48 timer ved 110°C. Ved å behandle reaksjonsblandingen på samme måte som i eksempel lb) fikk man 57,7 g (81,4%) av råproduktets hydroklorid. Ved omkrystallisasjon fra isopropanol fikk man 48 g (67,5%) rent hydroklorid, smeltepunkt 174°C.

Funnet C 63,41; H 8,95; N 8,28
Eksempel 3
3-( 2- metylpiperidyl)- 4', 6'- dimetyl- o- anisidid
En blanding av 40 g (0,165 mol) 3-klor-4',6'-dimetyl-o-propionanisidid fremstilt som beksrevet i eksempel 2a), 54,3 g 2-metyl-piperidin og 330 ml vannfritt benzen ble varmet opp i 72 timer ved 65°C. Ved å behandle reaksjonsblandingen på samme måte som i eksempel lb) fikk man 47,7 g (84,5%) 3-(2-metylpiperidyl)-4',6<1->dimetyl-o-propionanisidid-hydroklorid. Ved omkrystallisasjon av råproduktet fra en blanding av isopropanol og aceton fikk man 32,1 g (57%) rent hydroklorid, smeltepunkt 192-193°C.
Ved å gå frem som beskrevet i eksempel 1 omdannet man det fremkomne hydroklorid til den frie base, smeltepunkt 77-78°C.

Claims (1)
- Analogifremgangsmåte til fremstilling av nye anilider med antitussiv virkning og ikke-toxiske syreaddisjonssalter herav, hvilke forbindelser har den generelle formelhvor n betegner et helt tall 2 eller 3 og R 1 og R <2>hver for seg betegner en lavere alkylgruppe med 1-3 karbonatomer eller sammen med nitrogenatomet de er bundet til, danner en piperidinring substituert med en metylgruppe, karakterisert ved at 4,5-dimetyl-o-anisidinacyleres med et halogen-karboksylsyre-anhydrid med den generelle formelhvor Hal betegner klor eller brom og n har samme betydning som ovenfor, hvoretter detfremkomne halogen-karboksylsyre-anilid med den generelle formelhvor n og Hal har samme betydning som ovenfor, omsettes med en aminoforbindelse med den generelle formelhvor R 1 og R 2 har samme betydning som ovenfor, og at det som reak-sjonsprodukt dannede basiske anilid (1)^ hvis det ønskes, omdannes til et ikke-toxisk syreaddisjonssalt.
