Description
Foreliggende oppfinnelse vedrører fremstilling av
et terapeutisk aktivt derivat av fenyleddiksyre samt dets salter. Mer spesielt angår oppfinnelsen en fremgangsmåte til fremstilling av 4-cyklopropylmetylenoksy-3-klorfenyleddiksyre og de ikke-toksiske, uorganiske og organiske farmasøytisk akseptable salter derav. Disse forbindelser har meget interes-sante anti-inflammatoriske, anti-pyretiske og analgetiske egenskaper. Noen derivater av fenyleddiksyre som er kjent fra litteraturen har biologisk aktivitet som kvalitativt ligner den til den nye forbindelsen, 4-cyklopropylmetylenoksy-3_klor-fenyleddiksyre. Sammenliknet med de kjente forbindelser viser imidlertid den nye forbindelse ikke bare forbedret kjemisk stabilitet, men også en betydelig forøkning av den biologiske aktivitet og en konstant høyere terapeutisk indeks.
Ifølge foreliggende oppfinnelse fremstilles 4-cyklopropylmetoksy-3-klorfenyleddiksyre samt salter derav ved at man omsetter en lavere alkylester av 3-klor-^-hydroksy-fenyleddiksyre i oppvarmet tilstand under vannfrie betingel-
ser med et cyklopropylmetylhalogenid og forsåper den oppnådde ester, hvoretter man eventuelt overfører den oppnådde 4-cyklo-propylmetoksy-3-klorfenyleddiksyre med metallsalter eller baser, spesielt organiske baser, til det tilsvarende salt.
Med betegnelsen "lavere alkyl" menes et mettet,
rettkjedet radikal med opptil 3 karbonatomer.
Saltene av 4-cyklopropylmetylenoksy-3-klorfenyl-eddiksyre omfatter salter med f.eks. alkali- og jordalkali-metaller slik som natrium, kalium, kalsium og magnesium, ammoniumsaltet og kobbersaltet, samt saltene med ikke-toksiske organiske baser, f.eks. basisk aminosyre slik som ornitin,
lysin, arginin og histidin.
Det er ifølge oppfinnelsen foretrukket at man
som metallsalt anvender et kobbersalt, og at man som base be-nytter en basisk aminosyre, fortrinnsvis DL-lysin.
Nedenstående eksempler illustrerer oppfinnelsen.
Eksempel
En blanding av 10,6 g metyl-3-klor-3-hydroksyfenyl-acetat, 13,5 g cyklopropylmetylenbromid og 13,8 g vannfritt kalium-karbonat i 200 ml aceton, -oppvarmes under tilbakeløp og med omrøring i 20 timer. Oppløsningsmidlet fjernes deretter ved destillasjon i vakuum på et vannbad og resten opptas i 60 ml destillert vann og ekstraheres to ganger med dietyleter. De kombinerte eterekstrakter vaskes med en kald, fortynnet vandig oppløsning av natriumhydroksyd og deretter med vann og tilslutt blir det tørket med vannfritt magnesiumsulfat.
Etter filtrering og inndampning av oppløsningsmidlet oppnås 12,7 g metyl-4-cyklopropylmetylenoksy-3-klorfenylacetat i form av en stråfarget olje. Den således oppnådde forbindelse tilsettes til en oppløsning av 27,5 ml etanol og 27,5 ml 2N vandig natriumhydroksyd og kokes i 2 timer hvoretter reaksjonsblandingen fordampes til tørrhet i vakuum og resten oppløses i vann, avkjøles og 50% svovelsyre tilsettes inntil sur reaksjon til Kongo-rødt.
Det således oppnådde hvite bunnfall ekstraheres to ganger med dietyleter og de kombinerte eterekstrakter vaskes med vann og tørkes over vannfritt magnesiumsulfat. Etter filtrering fordampes oppløsningsmidlet og resten 'omkrystalliseres fra cyklo-heksan til 11,4 g 4-cyklopropylmetylenoksy-3-klorfenyleddiksyre i form av et krystallinsk hvitt fast stoff som smelter ved 105-106°C.
Kobbersalt - 7,2 g 4-cyklopropylmetylenoksy-3-klor-fenyleddiksyre oppløses i 30 ml av en natriumbikarbonatoppløsning. Til nevnte oppløsning tilsettes dråpevis og under omrøring en oppløsning av kobberacetat. Blandingen omrøres i \ time, det faste stoff oppsamles ved filtrering, vaskes med varmt vann inntil nøytral reaksjon og tørkes på et vannbad til konstant vekt. 4,5 g kobber 4-cyklopropylmetylenoksy-3-klorfenylacetat som smelter ved 198-200°C oppnås. DL-lysinsalt - 2,4 g 4-cyklopropylmetylenoksy-3-klorfenyleddiksyre oppløses i 20 ml absolutt alkohol. Til opp-løsningen tilsettes 3 g av en 50% DL-lysin-vandig oppløsning. Blandingen hensettes i \ time hvoretter det oppnådde gelatinøse hvite bunnfall filtreres og tørkes i vakuum. Dette gir 3,80 g DL-lysin 4-cyklopropylmetylenoksy-3-klorfenylacetat som om-krystallisert fra 95% etylalkohol smelter ved 175-176°C.
4-cyklopropylmetylenoksy-3-klorfenyleddiksyre og dens salter har anti-inflammatorisk, anti-pyretisk og analgetisk aktivitet samt lav toksisitet. Disse aktiviteter har blitt bestemt ved sammenligningsforsøk utført med forbindelsen fremstilt ifølge oppfinnelsen og kobber- og DL-lysin-saltet derav ved en dosering uttrykt som syren, med fenyl-butazon (4-butyl-l,2-difenyl-3,5-dioksopyrazolidin) som er et kjent anti-inflammatorisk, anti-pyretisk og analgetisk middel, og med 4-allyloksy-3-klorfenyleddiksyre kjent fra britisk patent nr. 1.174.535 og er en utbredt kjent forbindelse med struktur i likhet med den ifølge oppfinnelsen fremstilte forbindelse. 4-allyloksy-3_klorfenyleddiksyre er den mest representative av de forbindelser som omfattes av nevnte britiske patent.
Analgetisk aktivitet
Denne ble bestemt ved hjelp av følgende metoder:
a) Randall og Selitto
Trykket på en rottepote, som på forhånd var gjort
oppsvulmet ved injeksjon i poten av 0,1 ml av en 20% vandig suspensjon av gjær til frembringelse av en smertefull reaksjon, ble målt i mm Hg. Smerte-terskelverdien ble bestemt 30 minutter før og 30, 60, 90 og 120 minutter etter administrasjon av forsøksforbindelsene.
10 Wistar hannrotter med en legemsvekt på 170-190 g ble benyttet for hvert doseringsnivå og de tilsvarende midlere verdier gjort gjeldende for hver dose. Den prosentvise økning av smerte-terskelverdien sammenlignet med basisverdiene, anvendes som indeks for analgetisk aktivitet. De oppnådde resultater er angitt i følgende tabell I.
b) Siegmund
Antagonismen mot abdominale sammentrekninger indusert
ved fenylbenzokinon ble bestemt. Sveitsiske hanmus med en legemsvekt på 19-21 g ble behandlet oralt med forsøksforbindelsene 30 minutter før den endoperitoneale administrasjon av 0,25 ml av en 0, 02% vandig fenylbenzokinonoppløsning. Dyrene ble holdt under observasjon i 30 minutter etter behandling med fenylbenzokinon og de abdominale sammentrekninger for hvert dyr ble telt. 10
mus ble benyttet for hvert doseringsnivå og de tilsvarende midlere verdier bestemt. Den prosentvise nedsettelse av antall abdominale sammentrekninger hos dyrene behandlet med forsøksforbindelsene
sammenlignét med kontrolldyr behandlet med vann, ble benyttet som indeks for analgetisk aktivitet. De oppnådde resultater er angitt i følgende tabell II.
Anti- inflammatorisk aktivitet
Denne aktivitet ble bestemt ifølge Winter's metode som bestemmer den inhiberende effekt på ødem indusert ved hjelp av karragenin.
Hanrotter med en legemsvekt på 170-190 g ble behandlet med forsøksforbindelsene og etter 60 minutter ble 0,05 ml av en 1% vandig oppløsning av karragenin injisert i den plantare sone i en bakpote på hvert dyr. Volumet av den behandlede pote ble bestemt umiddelbart etter injeksjonen og 3 timer senere. 10 dyr ble benyttet for hvert doseringsnivå. Forskjellen i økning i volumet av poten hos behandlede dyr sammenlignet med kontrolldyrene, representerer et indeks for anti-inf lammatorisk aktivitet. De oppnådde resultater uttrykt som prosent inhibering av volumet av ødemet sammenligning med kontrolldyrene og bestemt som en midlere verdi for hver dose,
er angitt i følgende tabell III.
Anti- pyretisk aktivitet
Denne aktivitet ble bestemt ved bedømmelse av
2 typer hyperpyrexia.
Hyperpyrexia indusert ved
a) baktopepton
1 ml av en 5% vandig oppløsning av baktopepton
preinkubert i 18 timer ved 37°C, ble injisert subkutant i Wistar hannrotter med en legemsvekt på 170-190 g. 4 timer etter behandling med det pyrogene middel, ble dyrene oppdelt i grupper på 10 dyr og behandlet oralt med forsøksforbindelsene. Rektaltemperaturen ble målt ved begynnelsen av forsøket, 4 timer etter injeksjon av pyrogenforbindelsen og deretter 1, 2 og 3 timer etter administrasjonen av forsøksforbindelsene. Forskjellen mellom temperaturen hos behandlede dyr og hos kontrolldyrene er et indeks for anti-pyretisk aktivitet.
b} gjær
Albinokaniner ble injisert intravenøst med
1 ml/kg av en 0, 2% vandig suspensjon av gjær. Rektaltemperaturen ble målt ved begynnelsen av forsøket og 60 minutter etter behandling med pyrogenforbindelsen. Umiddelbart etter at temperaturen var målt, ble forsøksforbindelsene administrert oralt til dyrene og den rektale temperatur deretter målt hos hvert dyr etter 30, 60, 120 og 180 minutter. 10 dyr ble benyttet for hvert doseringsnivå.
Forskjellen mellom temperaturene hos de behandlede dyr og hos kontrolldyrene er et indeks for anti-pyretisk aktivitet. De oppnådde resultater med de to metoder som er be-skrevet ovenfor for bestemmelse av anti-pyretisk aktivitet, er angitt i følgende tabeller IV og V, respektivt, og bestemt som en middelverdi for hver dose.
Claims (3)
1. Analogifremgangsmåte til fremstilling av terapeutisk virksom 4-cyklopropylmetoksy-3-klorfenyleddiksyre samt salter derav, karakterisert ved at man omsetter en lavere alkylester av 3-klor-4-hydroksyfenyleddik-syre i oppvarmet tilstand under vannfrie betingelser med et cyklopropylmetylhalogenid og forsåper den oppnådde ester, hvoretter man eventuelt overfører den oppnådde 4-cyklopropyl-metoksy-3-klorfenyleddiksyre med metallsalter eller baser, spesielt organiske baser, til det tilsvarende salt.
2. Analogifremgangsmåte ifølge krav 1, karakterisert ved at man som metallsalt anvender et kobbersalt.
3. Analogifremgangsmåte ifølge krav 1, karakterisert ved at man som base anvender en basisk aminosyre, fortrinnsvis DL-lysin.
