Foreliggende oppfinnelse angår en ny og forbedret fremgangsmåte ved fremstilling av 5-fluor-2-methy1-1-(p-methylsulfiny1 - benzyliden)-ind-2-en-3-y1-3-eddiksyre.
5-f luor-2-methyl -1 - (p-methylsalf inylbenzyliden) -ind-2-en-3" yl-eddiksyre er en kjent forbindelse med antiinflammatorisk aktivi-tet beskrevet i US patent 3 654 3'- i9 • Tidligere er denne forbindelse blitt fremstilt ved kondensering av et passende substituert benzaldehyd med en eddiksyreester i en Claisen-reaksjon eller med en a-halogenert propionsyreester i en Reformatsky-reaksjon. Den dannede unettede ester ble redusert og hydrolysert for å få en 3-arv 1-propionsyre som derpå ringsluttes under dannelse av indanonet. Den alifatisxe sidekjede ble så innført ved en Reformatsky- eller Wittig-reaksjon, og 1-substituenten ble inn-ført i den dannede indenyleddiksyre eller -ester ved omsetning av eddiksyrederivatet med et aromatisk aldehyd eller keton med den ønskede strukturformel og dehydratisering for å danne den ønskede indenyledri iksyre.

Mellomproduktet, 5-fluor-2-methyl-indan-l-ol-3-yl-eddiksyre, og dets fremstilling er beskrevet i norsk patentansøkning 791267.
Utgangsmaterialet, 5-fluor-2-methyl-indan-l-ol-3-yl-eddiksyre (V), er en ny forbindelse, som ved foreliggende fremgangsmåte dehydratiseres ved syrekatalyse under dannelse av 5-fluor-2-methyl-ind-l-en-3-yl-eddiksyre (VI), eller ved basekatalyse under dannelse av 5-fluor-2-methyl-ind-2-en-3-yl-eddiksyre (Via). Inden-3-yl-eddiksyren (VI) eller (Via) kondenseres med p-raethylthiobenzaldehyd fulgt av oxydasjon, eller med p-methylsulfinylbenzaldehyd, fulgt av surgjøring for å danne cis-5-fluor-2-methyl-1-(p-methylsulfinyl - benzyliden)-ind-2-en-3-yl-eddiksyre (VII). p-methylsulfinylbenz-aldehydet kan fremstilles ved å oxydere p-methylthiobenzaldehyd med et passende oxydasjonsmiddel, som hydrogenperoxyd.
5-fluor-2-methyl-indan-l-ol-3-yl-eddiksyre (V) kan dehydratiseres under sure eller basiske betingelser, fortrinnsvis sure betingelser ved oppvarmning for å danne 5-fluor-2-methyl-ind-l-en-3-yl-eddiksyre (VI), som er en ny forbindelse,. Både organiske og uorganiske syrer kan anvendes, fortrinnsvis mineralsyrer, alkylsulfonsyrer og arylsulfonsyrer som p-toluensulfonsyre, saltsyre, svovelsyre og methansulfonsyre. Syrékonsentrasjonen er fra 0,001 til 10 mol pr. mol indanol (V), fortrinnsvis 0,01 til 0,2 mol.
Den sure -dehydrat iser ing kan utføres i et inert organisk oppløsningsmiddel som hydrocarboner eller halogenerte hydrocarboner eller blandinger derav. Foretrukne oppløsningsmidler innbefatter ethylbenzen, toluen og xylen, og særlig benzen.
Alternativt kan dehydrat iser ingen utføres under basisk?e betingelser for å danne ind-2-enet. Passende baser innbefatter natriumalkox<y>der , nat r-iumamid, kaliumalkoxyder, allcalimeta 11-hydroxyder og jordalkalimetallhydroxyder, som natriummethoxyd, natriumethoxyd, kaliumhydroxyd, natriumhydroxyd, kaliummethoxyd
og kaliumethoxyd. Basekonsentrasjonen er fra 0,1 mol til 100 mol pr. mol indanol (V), fortrinnsvis 3,0 til IO mol. Den basiske dehydratisering kan utføres i et inert organisk oppløsningsmiddel som alkoholer, etnere og blandinger derav. Passende oppløs-ningsmidler innbefatter glyme, di-glyme , triglyme, met nano 1, ethanol, isopropanol og t-butanol, idet de foretrukne oppløsnings-midler er ethanol og isopropanol.
Dehydratiseringen lean utføres mellom 20 og l5d°C, fortrinnsvis mellom 6o og lOO°C. Reaksjonshastigheten er avhengig av den temperatur ved hvilken den utføres. Reaksjonstiden er ikke kritisk, og i alminnelighet utføres reaksjonen inntil den er i det vesentlige fullstendig. Trykket er ikke kritisk ved denne reaksjon, skjønt atmosfæretrykk eller lavere foretrekkes da et mol vann elimineres. I alminnelighet utføres reaksjonen ved atmosfæretrykk i et åpent system. Dehydratiseringen under sure betingelser gir ind-l-enet (VI), en ny forbindelse, mens dehydratisering under basiske betingelser gir ind-2-enet (Via), en kjent forbindelse.
Innføringen av benzylidensubstituenten på. indenyl-eddiksyre-forbindelsen (VI eller Via) utføres bekvemt ved omsetning med p-methylsulfinyl- (eller p-methylthio)-benzaldehyd i nærvær av en sterk base ved en temperatur fra 0° til 100°C, og fortrinnsvis fra 20° til 80°C i nærvær av et oppløsningsmiddel. Forholdet av aldehyd til inden er passende 1 til 2:1, men fortrinnsvis ca. 1,5:1, og forholdet mellom base og inden kan variere fra en kata-lytisk mengde til ekvimolar eller mere. Sterke baser som alkali-og jordalkalihydroxyder eller C1_^-alkoxyder som NaOH, KOCH^, KOtBu, tet ra-C^_^-alkyl-ammoniumhydroxyder eller benzyl-tri--alkyl-ammoniumhydroxyder, som benzyltrimethylammoniumhydroxyd, og alkalimetallhydrider kan passende anvendes. Passende oppløsningsmidler er polare oppløsningsmidler som dimethoxyethan, methanol, pyridin, dimethylformamid og lignende, og upolare opp-løsningsmidler som benzen, toluen, xylen o.l.
Det således fremstilte benzyliden-inden-eddiksyresalt over-føres til den frie syre under opparbeidelsen. Overføringen kan utføres under alminnelig kjente sure betingelser som i nærvær av sterkt organiske syrer (p-toluensulfonsyre, trifluoreddiksyre) eller mineralsyrer (saltsyre, svovelsyre), særlig saltsyre.
Når p-methylthiobenzaldehyd anvendes ved kondensasjonen,
kan oxydasjonen av methylthiogruppen til den ønskede methylsul-finylgruppe utføres ved en rekke standardmetoder, som oxydasjon med hydrogenperoxyd, basiske perjodater og hypohalogenitter, fortrinnsvis alkali- eller jordalkali-perjodater eller -hypohalogenitter) eller organiske persyrer, som pereddiksyre og monoperfthai-syre, men særlig med hydrogenperoxyd. Reaksjonen utføres fortrinnsvis i nærvær av et oppløsningsmiddel. Til slike formål kan C1_^-alkansyrer (eddiksyre), halogenerte hydrocarboner (kloro-
form, 1,2-diklorethan) , ethere (dioxan), C-^^-alkanoler (isopropanol) eller blandinger derav, anvendes. Molforholdet av oxyderingsmiddel til inden-eddiksyreforbindelse kan være fra 0,5 til 10, men fortrinnsvis fra 0,8 til 1,5. Reaksjonstiden og temperaturen er ikke kritisk, idet reaksjonen utføres til den i det vesentlige er fullstendig. Trykket er ikke kritisk ved denne reaksjon. i alminnelighet utføres reaksjonen under atmosfæretrykk i et åpent system.
De tre siste trinn, nemlig reduksjonen av indanonet, dehydrat iser ingen av indanolen og kondenseringen av indenet kan ut-føres uten å isolere produktene, i det samme reaksjonskar. De tre trinn kan alle utføres under basiske betingelser eller det er mulig å syre reaksjonen før dehydratiseringstrinnet.
De følgende eksempler er gitt for å belyse oppfinnelsen.
Eksempel 1
5 - fluor- 2- methyl- ind- 1- en- 3~ yl- eddiksyre ( VI)
En oppløsning av 387 mg 5-fluor-2-methyl-indanol-3-eddiksyre (V) i IO ml benzen til hvilken der tilsettes 50 mg p-toluensulfonsyre (p-TsOH-H^) , oppvarmes under tilbakeløp,8 85 minutter i løpet av hvilken tid 3 ml oppløsningsmiddel tillates å avdestil-lere. Oppløsningen får lov til å avkjøles, fortynnes med methylendiklorid og vaskes med vann. Den organiske oppløsning tørres og inndampes, hvorved man får 5-fluor-2-methy1-ind-1-en-3-yl-eddiksyre (VI).
På lignende måte fåes, når en ekvivalent mengde saltsyre, svovelsyre eller methansulfonsyre anvendes istedenfor p-toluensulfonsyre i ovenstående eksempel, tilsvarende resultater.
Eksempel 2
5- fluor- 2- methyl- ind- 2- en- 3- yl- eddiksyre ( Via)
En oppløsning av 390 mg 5~fluor-2-methyl-indan-1-ol-3-yl-eddiksyre (V) i IO ml vann til hvilken der tilsettes 400 mg kaliumhydroxyd, oppvarmes under tilbakeløp i 5 timer. Oppløsningen får lov til å avkjøles, syre med 2N saltsyre og ekstraheres med methylendiklorid. Den organiske oppløsning tørres og inndampes, hvorved man får 5-fluor-2-methyl-ind-2-en~3-yl-eddiksyre (Via).
på. lignende måte fåes, når en ekvivalent mengde natriumhydroxyd eller nat riumethoxyd anvendes istedenfor kaliumhydroxyd 1 ovenstående eksempel, tilsvarende resultater.
Eksempel 3
5-fluor-2-methyl-1-(p-methylthiobenzyliden)-ind-2-en-3-yl-eddiksyre ( Vila)
Til O,01 mol 5-fluor-2-methyl-ind-l-en-3-yl-eddiksyre (VI) og 0,01 mol p-metbylthiobenzaldehyd tilsettes 2 ekvivalenter 25%-ig methanolisk nat riummethoxyd. Blandingen kokes under tilbakeløp i 2 timer, avkjøles, nøytraliseres med eddiksyre og fortynnes med vann. Reaksjonsblandingen ekstraheres i ethylacetat som vaskes med vann og inndampes for å få råproduktet (Vila).
på tilsvarende måte fåes, når en ekvivalent mengde natriumhydroxyd, kaliumhydroxyd, natriumhydrid, kaliumhydrid, natriumethoxyd eller benzyltrimethylammoniumhydroxyd anvendes istedenfor nat riummethoxyd i ovenstående eksempel, tilsvarende resultater.
-Eksempel 3
5-fluor-2-methyl-l-(p-metnylsulfinylbenzyliden)-ind-2-en-3-y1 - eddiksyre { VII)
0,0422 mol natriummetaperjodat-trihydrat i 8,5 ml vann tilsettes til 0,Ol mol 5-fluor-2-methyl-l-(p-methylthiobenzyliden)-ind-2-en-3-yl-eddiksyre (Vila) i 240 ml methanol og IO ml aceton ved værelsetemperatur. Blandingen omrøres over natten og inndampes så til lite volum, fortynnes med vann og filtreres. Bunnfallet vaskes med vann og tørres i luft.
Pa lignende måte fåes, når en ekvivalent mengde hydrogenperoxyd, et hypohalogenitt eller en organisk persyre som pereddiksyre eller monoperfthalsyre anvendes istedenfor natriummetaperjodat i ovenstående eksempel, tilsvarende resultater.
Eksempel 4
5-fluor-2-methyl-i-(p-methylsulf inylbenzyliden)-ind-2-en-3-yl - eddiksyre ( Vllj
Til 0,01 mol 5-fluor-2-methyl-ind-l-en-3-yl-eddiksyre {VI) og 0,01 mol p-methylsulfinylbenzaldehyd tilsettes 2,0 ekvivalenter 25%-i<3 metbanolisk nat r iummethoxyd. Blandingen kokes under til-bakeløp i 2 timer, avkjøles og nøytraliseres med eddiksyre og fortynnes med vann. Reaksjonsblandingen ekstraheres i ethylacetat som vaskes med vann og inndampes for å få råproduktet (VII).
På lignende måte fåes, når en ekvivalent mengde natriumhydroxyd, kaliumhydroxyd, natriumhydrid, kaliumhydrid, natriumethoxyd eller benzyltrimethylammoniumhydroxyd anvendes istedenfor natriummethoxyd i ovenstående eksempel, tilsvarende resultater.
Eksempel 5
5-fluor-2-methyl-(p-methylsulfinylbenzyliden)-ind-2-en-3-yl-eddiksyre ( VII)
En oppløsning av 0,01 mol 5-fluor-2-methyl-indan-1-on-3-eddiksyre (IV) i 6 ml isopropanol gjøres alkalisk ved tilsetning av 2,5M natriumhydroxyd (til ca. pH 9 - 10). Til denne oppløsning tilsettes 0,015 mol natriumborhydrid. Oppløsningen omrøres ved værelsetemperatur i 80 minutter og ved 60°C i 45 minutter. Opp-løsningen gjøres sur ved tilsetning av 6m saltsyre (ca. pH 1 -2), og oppvarmes under tilbakeløp i 30 minutter. Oppløsningen får lov til å avkjøles og gjøres alkalisk med natriumhydroxyd til en pH på ca. 13,0. Til oppløsningen tilsettes 0,01 mol p-methylsulfin - ylbenzaldehyd og 2,0 ekvivalenter natriumhydroxyd. Blandingen kokes under tilbakeløp i 2 timer, avkjøles, nøytraliseres med eddiksyre og fortynnes med vann. Reaksjonsblandingen ekstraheres i ethylacetat som vaskes med vann og inndampes, hvorved man får råproduktet (VII).
På lignende måte fåes, når en ekvivalent mengde kaliumbor-hydrid eller lithiumborhydrid anvendes istedenfor natriumborhydrid eller svovelsyre, p-toluensulfonsyre eller methansulfonsyre anvendes istedenfor saltsyre, eller kaliumhydroxyd anvendes istedenfor natriumhydroxyd i ovenstående eksempel, tilsvarende resultater.
Claims (1)
- Fremgangsmåte ved fremstilling av 5-fluor-2-methyl-1-(p-methylsulfinylbenzyliden)-ind-2-en-3~yl-eddiksyre, karakterisert ved trinnene: a) 5-f luor-2-met hyl-indan-l'-ol-3-y 1-eddiksy re dehydrat is ere s til en 5-fluor-2-methyl-inden-3-yl-eddiksyre, og b) den erholdte indenyl-3-eddiksyre kondenseres med p-methylthiobenzaldehyd fulgt av oxydasjon, eller med p-methylsulfinylbenzaldehyd, fulgt av surgjøring, for å danne det ønskede produkt.
