Description
Analogifremgangsmåte for fremstilling av terapeutisk
aktiv 2-L4-(2-fluorfenoksy)fenyll propionsyre og deri-
vater derav. x'
Denne oppfinnelse angår en fremgangsmåte for fremstilling av 2-[4-(2-fluorfenoksy)fenyl]propionsyre og salter, estere, amidet og alkoholen avledet derfra, som er funnet å være i besittelse av verdifulle biologiske egenskaper.
I henhold til oppfinnelsen tilveiebringes således en fremgangsmåte for fremstilling av forbindelser med den generelle formel I
hvor
Y er COOH, CONH2 eller CH2OH,
og farmasøytisk akseptable estere, uorganiske salter og organiske salter av de forbindelser hvor Y er COOH. Lignende 2-(substituert fenyl)-propionsyrederivater, som imidlertid inneholder et 4-halogenatom i fenoksy-gruppen fremstilles ifølge vår parallelle søknad 980/71. I den følgende beskrivelse skal symbolet RQ bety:
I henhold til oppfinnelsen fremstilles de nye forbindelser med formel I som følger:
Syre
1. Hydrolyse av
hvor Z er cyano, karbamoyl,
eller COOR^ hvor R. er en esterdannende gruppe, særlig lavere alkyl.
Hydrolysen kan utføres ved metoder velkjente innen tek-nikken, f.eks. ved anvendelse av syre eller alkali i vann, i et organisk, flytende reaksjonsmedium eller i en blanding derav,
og en behandlingstemperatur på 15-150°C er hensiktsmessig. Fortrinnsvis utføres hydrolysen ved tilbakeløpsbehandling i nærvær av et alkalimetallhydroksyd eller av en mineralsyre, og det organiske, flytende reaksjonsmedium er en lavere alkanol.
Utgangsmaterialene kan f.eks. fremstilles fra de substitu-erte acetofenoner Ro~C0-CH3 på vanlig måte, og andre metoder omfatter de metoder som er beskrevet nedenfor under "estere" og
"amider".
2. Dekarboksylering av
Denne kan utføres ved oppvarming av forbindelsen ved ca. 200°C.
Utgangsmaterialene kan hensiktsmessig fremstilles på vanlig måte, f.eks. ved omsetning av en alkylester av en syre Rq-CH2-<:00H med et alkylkarbonat og et alkalimetallalkoksyd for å gi et alkalimetallderivat av en forbindelse med formelen RQ-CH=(C00alkyl)2, metylering av denne og hydrolyse av produktet.
3. Oksydasjon av :
Oksydasjonen kan utføres ved anvendelse av ethvert egnet oksydasjonsmiddel så som permanganater, "kromsyre, dikromater, per-syrer, hydrogenperoksyd, salpetersyre, hypokloriter, sølvoksyd eller oksygen. En meget egnet metode omfatter oksydasjon i v.ndig etanol med alkali (f.eks. alkalimetallhydroksyd) og sølvoksyd.
Utgangsmaterialene kan fremstilles ved de fremgangsmåter som er beskrevet for beslektede forbindelser i vårt britiske pat-
ent 1.160.725.
4. Hydrogenering av
Typiske metoder omfatter hydrogenering over en vanlig katalysator så som f.eks. palladium, palladiumoksyd eller platina i et inert oppløsningsmiddel så som en lavere alkanol, benzen, toluen, xylen, tetrahydrofuran, dioksan og eddiksyre, ved en temperatur på ca. 0°C opp til tilbakeløpstemperaturen for systemet.
Utgangsmaterialene kan fremstilles på vanlig måte som f.eks. etter det følgende reaksjonsskjerna:
hvor en av gruppene A og B er OH og den annen er halogen. Fortrinnsvis er A OH og B halogen. Denne omsetning utføres normalt ved oppvarming av et metallderivat (f.eks. et alkalimetallderivat, særlig kaliumderivat) av hydroksyforbindelsen med halogenforbind-elsen (særlig en jod- eller bromforbindelse) ved 100-350 C i nærvær av et metallkatalysator, særlig kobberpulver eller kobberbronse.
6. Hydrolyse av
hvor "alkyl" fortrinnsvis er metyl. Typiske hydrolysebetingelser er beskrevet under metode (1).
Utgangsmaterialet kan fremstilles ved fremgangsmåter lik de som er beskrevet av Meyers og Temple, J.A.C.S., 1970, 92, 6644.
Estere
1. Forestring av syrene på vanlig måte, f.eks. 2. Ved hjelp av metodene (4) og (5) som beskrevet under "syre", men ved å starte med den ønskede ester istedenfor syren. 3. Ved alkoholyse av oksazolinet beskrevet under "syre 6" . Amider 3. Ved hjelp av metode (4) som beskrevet under "syre", men ved å starte med amidet istedenfor syren.
Salter
1. Omsetning av syren med organiske eller uorganiske
baser.
2. Alkalisk hydrolyse av
Alkohol
1. Reduksjon av syrene eller fortrinnsvis av estrene (særlig alkylestrene). Anvendelse av litiumaluminiumhydrid i et egnet oppløsningsmiddel, f.eks. eter, fulgt av surgjøring; er et
eksempel. Alternativt kan hydrogenering i nærvær av en kobber/ kromoksyd-katalysator anvendes. Estere kan reduseres med natrium i en lavere alkanol. 2. Ved hjelp av metode (5) som beskrevet under "syre", men 'ved å starte med en beskyttet alkohol istedenfor syren. Alkoholen kan være beskyttet ved hjelp av en konvensjonell, lett utskiftbar gruppe, f.eks. benzyl, som fjernes etter de tidligere syntesetrinn.
Forbindelsene med den generelle formel I er i besittelse av anti-inflammatorisk aktivitet og er nyttige til behandling av inflammatoriske tilstander. De er også i besittelse av smertestillende og anti-pyretiske egenskaper og er nyttige for behandling av smertetilstander og pyretiske tilstander. De er nyttige for behandling av disse tre lidelser hver for seg eller i hvilken som helst kombinasjon. Deres aktivitet er bestemt på forsøksdyr ved anvendelse av farmakologiske undersøkelser som er kjent for å kunne karakterisere forbindelser som er i besittelse av aspirins tera-peutiske egenskaper, nemlig anti-inflammatorisk, smertestillende og antipyretisk aktivitet.
En foretrukket forbindelse med formel I er den i hvilken Y er COOH. Det antas at når salter, estere, amidet eller alkoholen avledet fra syren anvendes istedenfor syren, metaboliseres disse derivater av kroppen og omdannes der til den tilsvarende syre.
Ved en prøve utført ved metoden ifølge Adams og Cobb, Nature, 1958, 181, 773 ble det funnet at den kjente forbindelse 2-(2-fluor-4-fenoksyfenyl)propionsyre hadde en anti-inflammatorisk aktivitet, uttrykt i aktiviteten av aspirin, på 20 til 40, mens forbindelsen 2-[4-(2-fluorfenoksy)fenyljpropionsyre fremstilt ifølge oppfinnelsen hadde en aktivitet på 64 til 128.
De følgende eksempler skal tjene til å illustrere oppfinnelsen ytterligere.
Eksempel 1
2-fluorfenol (2,35 g) ble smeltet med kaliumhydroksyd
(1,7 g) og noen få dråper vann ved oppvarmning til 180°C. 2-(4-jodfenyl)propionsyre (2,76 g) og kobberpulver (0,1 g) ble deretter tilsatt, og smeiten ble oppvarmet ved 160-170°C i 1 times-Det avkjølte, faste stoff ble fordelt med metylenklorid (50 ml) og 2,5N natriumhydroksyd (50 ml). Det vandige lag ble surgjort med saltsyre, den resulterende gummi ble ekstrahert inn i eter, og eterekstrakten ble ekstrahert med mettet kaliumkarbonatoppløsning. Denne vandige ekstrakt ble surgjort med saltsyre, og produktet ble isolert i eter. Etter inndampning ble det resulterende, faste stoff renset ved preparativ sjiktkromatografi under anvendelse av 5% eddiksyre/petroleter, k.p. 62-68°C og eluering med etylacetat. Omkrystallisering fra petroleter k.p. 62-68°C ga 2-[4-(2-fluor-fenoksy) fenyllpropionsyre, sm.p. 106-108°C.
Eksempel 2
2- 14-(2-fluorfenoksy)fenyllpropionsyre (2,32 g) i etanol (16 ml) inneholdende konsentrert svovelsyre (0,5 ml) ble tilbake-løpsbehandlet i 5 timer, og alkoholen ble fjernet. Etter for-tynning med vann ble produktet isolert i eter og destillert for å gi etyl-2-[4-(2-fluorfenoksy)fenyllpropionat, sm.p. 176-177°C/ 2,5 ira Hg.
Eksempel 3
Etyl-2-[4-(2-fluorfenoksy)fenyljpropionat (1,3 g) i tørr eter (5 ml) ble satt dråpevis til litiumaluminiumhydrid (200 mg)
i tørr eter (5 ml). Etter tilbakeløpsbehandling i 1 time ble ove. skudd av hydrid spaltet med fortynnet svovelsyre, og eterlaget ble destillert for å gi 2-[4-(2-fluorfenoksy)fenyl]propanol,
k.p. 178-180°C/2,5 mm Hg.
Eksempel 4
2- 14-(2-fluorfenoksy)fenyllpropionsyre (0,2 g) og tionylklorid (1,5 ml) ble tilbakeløpsbehandlet i 1 time. Overskudd av tionylklorid ble destillert, og residuet ble satt til ammonium-hydroksyd (sp.v. 0,88, 2 ml) avkjølt i is. Bunnfallet ble oppsamlet og omkrystallisert fra benzen/petroleter k.p. 60-80°C for å gi 2-[4-(2-fluorfenoksy)fenyllpropionamid, sm.p. 106-107,5°C.
Eksempel 5
2-[4-(2-fluorfenoksy)fenyllpropionsyre (160 mg) og benzylamin (72 mg) ble blandet i eter. Det utfelte, faste stoff ble
omkrystallisert fra alkohol/eter for å gi benzylamin-saltet av 2-[4-(2-fluorfenoksy)fenyllpropionsyre, sm.p. 122-126°C.
E ksempel 6
1,96 g etyl-2-[4-(2-fluorfenoksy)fenyl]propionat ble hydrolysert ved tilbakeløpsbehandling i 1 time ved 12 ml 2,5N NaOH og 6 ml 95% etanol. Det ble derefter avkjølt og surgjort med HCl. Det erholdte, faste stoff ble ekstrahert inn i eter, vasket med vann og tørret over vannfritt natriumsulfat. Eteren ble fjernet, og residuet ble omkrystallisert fra bensin. Utbytte av 2-[4-(2-fluorfenoksy)fenyllpropionsyre 1,53 g (87%), sm.p. 103-105°C.
Eksempel 7
1,1 g 2-[4-(2-fluorfenoksy)fenyl]propionamid i 7 ml vann ble behandlet under tilbakeløp med 1 ml natronlut. Blandingen ble tilbakeløpsbehandlet og omrørt i 24 timer, avkjølt, surgjort og ekstrahert inn i eter. Eterlaget ble vasket med vann, ekstrahert med 2,5% I^CO^-oppløsning som derefter ble surgjort, ekstrahert på ny inn i eter, vasket med vann og tør-ret over vannfritt natriumsulfat. Eteren ble fjernet, og produktet ble isolert i bensin med kokepunkt 62-68°C, avkjølt, oppsamlet, vasket med bensin og tørret i vakuum. Utbytte av 2-[4-(2-fluorfenoksy)fenyllpropionsyre 1,04 g (88%), sm.p. 103-105°C.
E ksempel 8
1,5 g 2-[4-(2-fluorfenoksy)fenyl]propionitril i 10 ml vann ble behandlet under tilbakeløpskjøling med 1,5 ml natronlut, omrørt og tilbakeløpsbehandlet i 24 timer. Blandingen ble derefter avkjølt, surgjort og ekstrahert på vanlig måte med 2,5% K-jCO-j-oppløsning. Den endelige eterekstrakt ble tørret over vannfritt natriumsulfat. Efter fjernelse av eteren ble det faste residuum omkrystallisert fra bensin som koker i om-rådet 62-68°C. Utbytte av 2-[4-(2-fluorfenoksy)fenyllpropionsyre 600 mg (38%), sm.p. 103-105°C.
Eksempel 9
4,82 g etyl-2-[4-(2-fluorfenoksy)fenyl]-2-metyl malonate ble tilbakeløpsbehandlet i 1 time med 28 ml 5N NaOH og 14 ml 95% etanol. Det ble derefter avkjølt, surgjort med 5N HCl, ekstrahert inn i eter, vasket med vann og tørret over vannfritt
natriumsulfat. Eteren ble fjernet, og residuet ble dekarboksyl-ert ved oppvarmning ved 200°C i 20 minutter. Residuet ble av-kjølt og omkrystallisert fra 5 ml benzen og 10 ml bensin med kokeområde 62-68°C, avkjølt, og det nesten fargeløse produkt ble oppsamlet, vasket og tørret i vakuum. Utbytte 2,42 g
(70%) , sm.p. 98-l00°C.
Eksempel 10
7,3 g sølvnitrat ble oppløst i 8,6 ml destillert vann og behandlet dråpevis med 3,26 ml 13, 2N KOH i 7,5 ml destillert vann. En eksoterm reaksjon som gav en temperatur på 30°C, ble iakttatt. Blandingen ble fortynnet med 9,8 ml 95% etanol, og en oppløsning på 5 g 2-[4-(2-fluorfenoksy)fenyljpropional-dehyd i 9,8 ml 95% etanol ble tilsatt dråpevis, fulgt av vask-ing med 8 ml 95% etanol. En eksoterm reaksjon som gav en temperatur på 40°C, fant sted. Efter omrøring i 15 minutter ble 1,95 ml 13,2N KOH i 2 ml vann tilsatt langsomt i løpet av 1 time mens temperaturen ble holdt fra 43-45°C. Blandingen ble omrørt ved 40°C i 30 minutter, og derefter ble sølvet frafiltrert gjen-nom en "celite"-pute og vasket med vann. Etanolen ble fjernet, og kaliumsaltoppløsningen ble avkjølt og surgjort med 4 ml HCl. Produktet ble ekstrahert inn i metylenklorid og vasket med vann. Det organiske lag ble ekstrahert med kaliumkarbamat-oppløsning, surgjort og ekstrahert på ny inn i metylenklorid, vasket med
vann og tørret over vannfritt natriumsulfat. Oppløsningsmid-delet ble fjernet, og residuet ble omkrystallisert fra 5 ml benzen og 10 ml bensin med kokeområdet 62-68°C, avkjølt, oppsamlet og vasket med kold bensin og tørret i vakuum. Utbytte av 2-[4-(2-fluorfenoksy)fenyllpropionsyre var 3,1 g (58%), sm.p. 104-106°C.
Eksempel 11
0,62 g 2-[4-(2-fluorfenoksy)fenyl]akrylsyre ble hydro-genert i .etylacetat under anvendelse av 100 mg palladium på tre-kull (10%) . Hydrogeneringen var ferdig på 2 minutter. Kata-lysatoren ble frafiltrert med "celite", og filtratet ble inn-dampet til tørrhet. Det blekgule, gummiaktige residuum ble omkrystallisert fra bensin med kokeområde 62-68°C, avkjølt, oppsamlet, vasket med bensin og tørret i vakuum. Utbytte 57o mg (91%), sm.p. 105-107°C.
Eksempel 12
0,6 g 2-[l-(4-2'-fluorfenoksyfenyl)etyl]-4,4-dimetyl-2-oksazolin ble suspendert i 3 ml 3N HCl- og tilbakeløpsbehandlet i 15 minutter. Efter avkjøling ble oljen ekstrahert inn i eter, vasket med vann og derefter ekstrahert med 2,5% K2C03-oppløsning. De alkaliske ekstrakter ble surgjort og ekstrahert på ny med eter, vasket med vann og tørret over vannfritt natriumsulfat. Eteren ble fjernet, og det resulterende, faste residuum ble omkrystallisert fra bensin med kokeområde 62-68°C, avkjølt, oppsamlet, vasket i kold bensin og tørret i vakuum. Utbytte 320 mg (65%) , sm.p. 105-10 7°C.
Claims (1)
- Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser med den generelle formel Ihvor Y er COOH, C0NH2 eller C^OH, og farmasøytisk akseptable estere, uorganiske salter og organiske salter av de forbindelser hvor Y er COOH, karakterisert ved(1) hydrolyse av en forbindelse med den generelle formel II hvor Z er cyano, karbamoyl, eller C00R4 hvor R4 er en esterdannende gruppe, eller (2) dekarboksylering av forbindelsen med formel III eller (3) oksydasjon av forbindelsen med formel V eller (4) hydrogenering av forbindelsen med formel VII eller en ester eller et amid derav, eller (5) omsetning av en forbindelse med den generelle formel IX med en forbindelse med den generelle formel X eller en ester eller alkohol avledet derfra, i hvilke formler en av gruppene A og B (fortrinnsvis A) er OH og den annen (fortrinnsvis B) er halogen, eller (6) hydrolyse eller alkoholyse av en forbindelse med den generelle formel XI hvorefter de forbindelser med formel I hvor Y er COOH eventuelt omdannes til ester på vanlig måte, eller eventuelt omdannes til salter på vanlig måte, eller eventuelt omdannes til de tilsvarende alkoholer hvor Y er CH2OH ved reduksjon på vanlig måte, eller eventuelt omdannes til de tilsvarende amider hvor Y er CONH2 ved overføring av syren til syrehalogenid og omsetning med ammoniakk.
