Description
* i -1- VO 7X42 antitumor Antibioticum.
De uitvinding heeft betrekking op een nieuw cyclisch depsipep-tide antitumor antibioticum dat hierin wordt aangeduid als sandramycine, alsmede op zijn bereiding door fermentatie van een nieuw micro-organis-me, Nocardioides species stam C49.Q09, ATCC 39419, alsmede op de farma-5 ceutische preparaten die het nieuwe antitumor antibioticum bevatten, en op werkwijzen voor het gebruikmaken van dit antitumor antibioticum als een antimicrobieel-en antitumormiddel. De uitvinding heeft ook betrekking op het nieuwe micro-organisme zelf, dat in de fermentatieve produktie van sandramycine wordt gebruikt.
10 In het Amerikaanse octrooischrift 4.360.458 van Koshiyama et al, gepubliceerd op 23 november 1982, wordt een antitumor antibacterieel complex beschreven, aangeduid als BBM-928, alsmede zijn produktie door fermentatie van een nieuwe stam van actinomyceten, aangeduid als stam G-455-101 (ATCC 31491), waarvan later werd vastgesteld dat het een nieu-15 we species was van het genus Actinomadura welke Actinomadura luzonensis nov. sp. werd genoemd[zie J. Antibiotics, 33(10), 1098 - 1102 (1980]
De produktie, isolatie, karakterisering en antitumoractiviteit van de BBM-928 componenten worden beschreven in J. antibiotics, 33(10), 1087 - 1097 (1980). De structuren van BBM-928A, B, C en D (nu respec-20 tievelijk genoemd luozopeptine A, B, C en D) die de belangrijkste componenten van het BBM-928 complex zijn, worden beschreven in het Amerikaanse octrooischrift 4.451.456 van Koshiyama et al, gepubliceerd op 29 mei 1984, en voldoen aan formule 2 van het formuleblad, waarin *1 R2 25 luzopeptine A -COCH^ -COCH^

De luzopeptines A, B, C en D bevatten structuurkenmerken die 30 3-hydroxy-6-methoxychinaldinezuur als chromofoor alsmede 4-hydroxy-2,3, 4,5-tetrahydropyridazine-3-carbonzuur omvatten, waarbij het structuurverschil tussen de drie componenten alleen gelegen is in de mate waarin de hydroxyIgroep in de tetrahydropyridazine-stukken is geacetyleerd.
De uitvinding heeft betrekking op een nieuw cyclisch depsi-35 peptide antitumor antibioticum, dat hierin als sandramycine wordt aangeduid, met formule 1 van het formuleblad, alsmede op een werkwijze • - *7 Λ o - - 'w 'w ί ί -2- voor het bereiden , isoleren en zuiveren van sandramycine in vrijwel zuivere vorm.
Het nieuwe antibioticum sandramycine wordt verkregen door fermentatie van een nieuw micro-organisme, dat voorlopig geklassificeerd is 5 als een species van het genus Nocardioides, ophoping van het door het micro-organisme geproduceerde sandramycine en verzameling van het antibioticum sandramycine uit de kweekvloeistof. Een geprefereerd sandramycine-producerend micro-organisme is de Nocardioides sp. stam C49.009, geïsoleerd uit een bodemmonster dat in Mexico is verzameld, 10 en mutanten daarvan. Deze stam is gedeponeerd in de American Type Culture Collection, Rockville, Maryland, als ATCC 39419.
Het micro-organisme.
Thans volgt een algemene beschrijving van het voor de produktie van het antitumor antibioticum sandramycine geprefereerde micro-organisme. 15 Morfologie.
De stam No. C49.009 vormt zowel substraat als luchtmycelia (met een breedte van 0,4 ym) , en het substraat-mycelium is lang, goed vertakt en verdeeld in korte draden of staven na een week. De stam No. C49.009 heeft de neiging om zijn vermogen tot de vorming van luchtmyce-20 lium te verliezen bij voortgezet kweken. De hyphae van luchtmycelium vertakken sporadisch. De microscopische waarnemingen laten zien dat de lucht hyphae min of meer zigzag-vormig zijn aan het begin en zich ontwikkelen tot lange rechte ketens van arthrosporen-achtige cilindrische segmenten (0,5 x 1 tot ongeveer 3 ym) die later gescheiden worden met 25 doorschijnende hyphae. Het oppervlak van sporen-achtige segmenten is glad. De stam No. C49.009 is grampositief en niet zuurbestendig. Sporangium, bewegelijke sporen en sclerotium worden niet waargenomen.
Celwandsamenstelling en suikercomponenten van de volledige cel.
De celwand van stam No. C49.009 bevat LL-diaminopimelinezuur 30 en glycine als karakteristieke aminozuurcomponenten in de celwand volgens de methoden, beschreven door B. Becker et al in Appl. Microbiol., 13, 236 - 243 (1965) en T. Yamaguchi in J. Bacteriol, 89, 441 - 453 (1965). Een hydrolysaat van de volledige cel laat de aanwezigheid zien van glucose, mannose en ribose, maar mist diagnostische suikers 35 zoals werd bepaald volgens de procedures beschreven door Μ. P. Lecheva-lier et al. in Biol. Actinomycetes Related Organisms, 11. 78-92 (1976).

De bovenstaand vermelde celwandsamenstelling en verder componenten van de volledige cel laten zien dat stam No. C49.009 een actinomycete species van celwand type I is.
Cultuur- en fysiologische eigenschappen.
5 De stam No. C49.009 is een obligaat aerobe actinomyceet en groeit goed in de meeste beschrijvende media. Het luchtmycelium wordt overvloedig of in bescheiden mate gevormd op ISP medium Nrs. 3, 5 en 7 en Czapek's sucrose-nitraat agar, maar spaarzaam op voedingsrijke organische media zoals ISP Medium Nrs. 2 en 6, of Bennett's agar. De kleur 10 van het luchtmycelium is wit. Melanoide en andere afwijkende pigmenten werden in alle tot dusverre onderzochte beschrijvende media niet geproduceerd. De achterkantkleur van het vegetatieve mycelium is slechts gelig. Het vertoont een optimale groei bij 28°C, een matige groei bij 15°C en 37°C, maar geen groei bij 5°C en 45°C. Gelatine en zetmeel wor-15 den afgebroken. De tyrosinase reactie is negatief. De groei wordt geremd in tegenwoordigheid van 7 % NaCl of 0,01 % lysozyme. De kweek- en fysiologische-eigenschappen van stam No. C49.Q09 worden respectievelijk in de tabellen A en B getoond. De stam No. C49.009 gebruikt voor groei de meeste suikers, en het gebruik van koolstofbronnen wordt in tabel C 20 getoond.

Taxonomy.
De stam No. C49.009 wordt gekarakteriseerd door zijn gramposi-tieve reactie, niet-zuurbestendige aard, fragmentatie van sübstraat-mycelium, vorming van wit luchtmycelium, arthrosporen-achtige segmenta-5 tie in hele delen luchtmycelium en afwezigheid van het vermogen om een apart pigment te vormen. Deze eigenschappen samen met de aanwezigheid van LL-diaminopimelinezuur en glycine, maar de afwezigheid van diagnostische suikers in de celwand, plaatsen de stam No.C49.009 in het genus Nocardioides [H. Prauser, Inti. J. Syst. Bacteriol., 26, 58 - 65 (1976)] 10 De stam No. C49.009 onderscheidt zich van de luchtmycelium-vormende nocardioform actinomyceten waaronder Nocardiopsis dassonvillei, Saccha-ropalyspora hirsuta en alle heterologe species van het genus Nocardia in het diagnostische aminozuur of suiker van celwand en diagnostische fysiologische eigenschappen zoals beschreven door R.E. Gordon et al., 15 J. Gen. Microbiol., 109, 69 - 78 (1978) en zoals getoond in tabel D.
De stam No. C49.009 onderscheidt zich verder van het genus Streptomyces in zijn sporogene nocardiae-type morfologie; bijvoorbeeld fragmenterende substraat hyphae van stam No. C49.009 tot korte draden of staven. De arthrosporen-achtige segmenten van stam No. C49.009 kunnen worden onder-20 scheiden van de sporen van Streptomyces in de plaats van vorming, de ligging in de hyphale schede en de afwezigheid van een aparte sporen-wand.

Het gevoeligheidsprofiel voor een reeks taxon-specifieke fagen onderscheidt de stammen van het genus Nocardioides van verwante genera zoals Streptomyces of Nocardia [zie H. Prauser: Host-phage relationships in nocardioform organisms, "In the Biology of the Nocardiae", 5 Edit. M. Goodfellow et al·., London, New York en San Francisco, Academie Press (1976)]. De gevoeligheid van stam No. C49.009 voor deze actino-fagen werd niet onderzocht omdat de fagen niet beschikbaar waren. Gebaseerd op de kweek- en fysiologische eigenschappen, werden de 17 stammen van het genus Nocardioides die geïsoleerd waren uit bodemmonsters die 10 in verschillende gebieden van de wereld zijn verzameld, ondergebracht in een enkele species, Nocardioides albus. De stam No. C49.009 verschilt van Nocardioides albus in zijn gebruik van L-arabinose en inositol, maar lijkt op de laatstgenoemde in de totale kweek- en fysiologische eigenschappen. Derhalve werd stam No. C49.009 voorlopig geklassificeerd als 15 een species van het genus Nocardioides.
Opgemerkt wordt dat de onderhavige uitvinding niet beperkt is tot het gebruik van de bijzondere stam No. C.49.009 of op organismen die volledig aan de bovenstaande beschrijving beantwoorden. Het is in het bijzonder de bedoeling dat andere sandramycine-producerende stammen 20 of mutanten van het organisme die uit het beschreven organisme kunnen worden verkregen met behulp van bekende middelen zoals röntgenbestraling, ultraviolette bestraling, behandeling met stikstofmosterd, blootstelling aan fagen, en dergelijke,worden omvat.
Produktie van het antibioticum.
25 Sandramycine wordt geproduceerd door Nocardioides sp. stam No.
C49.009 (ATCC 39419), of een mutant daarvan in een conventioneel voe-dingsmedium te kweken. Men laat het organisme groeien in een voedings-medium dat bekende voedingsbronnen bevat, dat wil zeggen, assimileerbare bronnen van koolstof en stikstof plus optionele anorganische zouten en 30 andere bekende groeifactoren. Ondergedompelde aerobe condities worden bij voorkeur gebruikt voor de produktie van grote hoeveelheden antibioticum, hoewel voor de produktie van beperkte hoeveelheden ook opper-vlakte-culturen en flessen kunnen worden gebruikt.
Het voedingsmedium dient één of meerdere assimileerbare koolstof-35 bronnen zoals glycerol, glucose, sucrose, mannose, fructose, maïszetmeel, maltose, mannitol, melasse en dergelijke in gezuiverde of in ruwe toestand te bevatten. Het voedingsmedium dient ook één of meerdere assimi- 1,¾ , ί ^ (y) Ki * ·' v ^ • ΐ -11- leerbare stikstofbronnen zoals bijvoorbeeld sojabonenmeel, vismeel, moutextract, gistextract, pepton, glutenmeel, katoenzaad embryomeel, sojabloem, lijnzaadmeel, katoenzaadbloem, caseïne, gehydrolyseerde proteïne materalen, nitraten, ammoniumzouten, ureum en dergelijke 5 te bevatten. Anorganische voedingszouten zoals natriumchloride, kalium-fosfaat, magnesiumsulfaat, calciumcarbonaat en spore hoeveelheden van zware metaalzouten zoals koper, zink, mangaan, ijzer en dergelijke, kunnen eveneens aan het voedingsmedium worden toegevoegd.
De fermentatietemperatuur dient in het bereik van 15°C tot on-10 geveer 37°C te liggen, en ligt bij voorkeur in het bereik van ongeveer 25°C tot ongeveer 30°C. De pH van het fermentatiemedium dient in het bereik van ongeveer 5 tot ongeveer 10,5 te liggen, en het geprefereerde gebied is van ongeveer 6 tot ongeveer 8,5. Gewoonlijk wordt een optimale produktie van sandramycine verkregen in ongeveer 2-9 dagen, afhanke-15 lijk van de temperatuur. Wanneer tankfermentatie moet worden uitgevoerd, is het gewenst dat een vegetatief inoculum wordt geproduceerd in een voedingsvloeistof, door het voedingsmedium te enten met een hellende-of bodejncultuur, of een gelyofiliseerde cultuur van het micro-organisme.
Nadat op deze wijze eem actief inoculum is verkregen, wordt het op 20 aseptische wijze overgebracht in het fermentatietankmedium.
Isolatie van sandramycine.
Wanneer de fermentatie volledig is, wordt sandramycine uit het kweekmedium gewonnen en geïsoleerd in nagenoeg zuivere vorm volgens de meerstapsprocedure, die in het in de tekening weergegeven stroomschema 25 is afgebeeld.
Tot nadere uitwerking van het stroomschema, wordt de volledige vloeistof van de fermentatie van Nocardiodes sp. stam No. C49.Q09 eerst geëxtraheerd met een niet met water mengbaar organisch oplosmiddel en bij voorkeur met ethylacetaat. Bij voorkeur wordt aan het 30 extractiemengsel een filtreer-hulpmiddel zoals Dicalite (merk van
Grefco Inc., Torrance, Ca., voor diatomeeën aarde) toegevoegd en wordt het mengsel daarna gefiltreerd om onoplosbare stoffen te verwijderen.
Na filtratie wordt de organische fase afgescheiden van het filtraat van het extractiemengsel en door verdamping geconcentreerd waarbij het 35 ruwe extract van genoemd antibioticum wordt verkregen. Het ruwe extract wordt verdeeld over een alifatische koolwaterstof-oplosmiddel zoals Skellysolve B (merk van Skelly Oil Co. voor isomere hexanen) en onge-

De waterige methanol-fase wordt verdund met een extra hoeveelheid water en de resulterende oplossing van ongeveer 25 % water in methanol wordt geëxtraheerd met een organisch oplosmiddel zoals koolstoftetrachloride. De waterige methanolfase wordt verder ver-5 dund met water en de resulterende oplossing van ongeveer 35 % water in methanol wordt geëxtraheerd met een organisch oplosmiddel zoals chloroform. De koolstoftetrachloride-extracten worden bijeengebracht en onder verminderde druk verdampt waarbij residu A wordt verkregen. De chloro-form-extracten worden bij elkaar gebracht en onder verminderde druk ver-10 dampt waarbij residu B wordt verkregen. De residuen A en B kunnen worden gecombineerd en chromatografisch worden behandeld op een kolom die dia-tomeeën aarde zoals Dicalite bevat, onder toepassing van organische oplosmiddelen van lage tot hoge polariteit. De juiste fracties die het antibioticum bevatten, worden gecombineerd en onder verminderde druk 15 verdampt waarbij residu C wordt verkregen. Een verdere zuivering kan worden gerealiseerd met behulp van silicagel lage-druk vloeistofkolom-chromatografie van residu C of middelmatige druk HPLC (high performance liquid chromatography) van de residuen D, D’, E en E' zoals onderstaand in voorbeeld III, stap C wordt beschreven, onder toepassing van een 20 lineaire gradiënt van chloroform tot 5 % methanol in chloroform als het eluens· De juiste fracties worden drooggedampt waarbij sandramycine wordt verkregen. Herkristallisatie uit een geschikt oplosmiddelsysteem geeft zuiver kristallijn sandramycine.
Structuur van sandramycine.
25 Sandramycine is een cyclisch depsipeptide antitumor antibioticum dat een 3-hydroxychinoline-kem als het chromofoor en een pipecoline-zuurstuk bevat. Uit de spectraal analyse en chemische analyse is bepaald dat sandramycine beantwoordt aan de structuurformule 1 van het formuleblad.
30 Sandramycine is een witte kristallijne vaste stof met een smelt punt van 208 - 212°C, een molecuulformule van C..H_.04/.N,en een mole- 60 76 16 12 cuulgewicht van 1221,3312. Het is samengesteld uit de elementen koolstof, waterstof, stikstof en zuurstof. Het ultraviolette absorptiespectrum van sandramycine werd bepaald in methanol (0,01798 g/1) onder neutrale, zure en basische condities,

De structurele kemeigenschappen van sandramycine waardoor dit verschilt van de luzopeptines A, B en C zijn de aanwezigheid van 3-hy-droxychinaldinezuur-en pipecolinezuurstukken in plaats van respectievelijk de 3-hydroxy-6-methoxychinaldinezuur en 4-gesubstitueerde-tetra-5 hydropyridazine-3-carbonzuurgroepen.
Biologische activiteit van sandramycine- ♦
De antibacteriele werkzaamheden van sandramycine werden bepaald door middel van een seriegewijze tweevoudige agar-verdunningsmethode.
De resultaten worden getoond in tabel E in vergelijking met de activi-10 teiten van luzopeptine A en echinomycine. Zoals men uit tabel E waarneemt, is sandramycine een sterke remmer van de grampositieve organismen zoals Bacillus subtilis en Staphylococcus aureus alsmede Streptococcus faecalis.

f S f i ƒ f; £ -17- zoals aangetoond door de bovenstaand gegeven antimicrobiële en muis-tumorgegevens, is sandramycine bruikbaar als antibioticum en ook als antitumormiddel voor inhibitie van kwaadaardige zoogdiertumoren zoals P-388 leukemie.
S Tot de omvang van de uitvinding behorend farmaceutische preparaten die een pffectieve antimicrobiële of tumor-inhibitie gevende hoeveelheid van sandramycine bevatten in combinatie met een inerte farmaceutisch aanvaardbare drager of verdunningsmiddel. Dergelijke preparaten kunnen ook andere actieve antimicrobiële of antitumormiddelen bevatten en kunnen 10 in elke farmaceutische vorm worden gebracht die geschikt is voor de gewenste toedieningsroute. Voorbeelden van dergelijke preparaten omvatten vaste preparaten voor orale toediening zoals tabletten, capsules, pillen, poeders en granules, vloeibare preparaten voor orale toediening zoals oplossingen, suspensies, siropen of elixers en preparaten voor parenterale 15 toediening zoals steriele oplossingen,suspensies of emulsies. Ze kunnen ook in de vorm van steriele vaste preparaten worden vervaardigd, die opgelost kunnen worden in steriel water, fysiologische zoutoplossing of een ander steriel injecteerbaar medium, onmiddellijk voor het gebruik.
Voor gebruik als antimicrobieel middel wordt het sandramycine 20 of farmaceutische preparaten daarvan op zodanige wijze toegediend, dat de concentratie aan actieve ingrediënt groter is dan de minimum inhibitore concentratie voor het desbetreffende behandelde organisme. Voor gebruik als antitumormiddel kunnen de optimale doseringen en regimes van sandramycine voor een bepaalde zoogdiergastheer gemakkelijk door de deskundigen 25 worden vastgesteld. Het zal natuurlijk duidelijk zijn, dat de feitelijk toegepaste dosis van sandramycine zal variëren met het desbetreffende samengestelde preparaat, de wijze van toediening en de desbetreffende plaats, gastheer en ziekte van behandeling. Vele factoren die de werking van het geneesmiddel beïnvloeden zullen in aanmerking worden genomen met 30 inbegrip van leeftijd, gewicht, sexe,dieet, toedieningstijd, toedieningsroute, snelheid van uitscheiding, conditie van de patiënt, geneesmiddel-combinaties, reactie-gevoeligheden en ernst van de ziekte.
De volgende voorbeelden worden slechts ter toelichting gegeven en zijn niet bestemd om de omvang van de uitvinding te beperken. Skellysolve 35 B is een in de handel verkrijgbaar petroleum-oplosmiddel (Skelly Oil Co.), dat isomere hexanen omvat en een kookpunt heeft van 60 - 69°C. Dicalite w Ü 3 J y _ _ ^ -18- is diatomeeën aarde, vervaardigd door Grefco, Inc., Torrance, California. Tenzij anders is aangegeven, zijn alle temperaturen uitgedrukt in °C. Voorbeeld I.
Fermentatie van sandramycine.
5 A. Schudkolf fermentatie.
De Nocardioides sp. stam No. C49.009, ATCC 39419 werd gehandhaafd en overgebracht in kweektestbuizen op agar-hellingen van gist-moutextract agar. Dit medium bestond uit 4,0 g glucose, 4,0 g gistex-tract, 10,0 g moutextract en 20,0 g agar, met gedeioniseerd water aan-10 gevuld tot 1 liter. Bij elke overbrenging werd de agar-helling-cultuur gedurende 7 dagen bij 27°C gelncubeerd. Teneinde een inoculum voor de productiefase te bereiden, werd myceliumgroei van de hellingcultuur overgebracht in een 500 ml Erlenmeyer kolf,die 100 ml steriel medium bevatte, bestaande uit 30,0 g glucose, 10,0 g sojabloem, 10,0 g katoen-15 zaad-embryomeel en 3,0 g CaCO^, met gedeioniseerd water aangevuld tot 1 liter. Deze vegetatieve cultuur werd gedurende 48 uur bij 27 °C gelncubeerd op een Gyrotory-tier-schudinrichting (model G53, New Brunswick Scientific Co., Ine.), ingesteld op 210 omw/min waarbij een cirkel werd beschreven met een diameter van 5,1 cm. Er werd 4 ml vegetatieve 20 cultuur overgebracht in een 500 ml Erlenmeyer kolf die 100 ml bevatte van een steriel produktie medium, bestaande uit 20,0 g sucrose, 10,0 g sojabloem, 10,0 g lijnzaadmeel en 5,0 g CaCO , met gedeioniseerd water 3 aangevuld tot 1 liter. De produktiecultuur werd bij 27°C gelncubeerd op een schudinrichting als gebruikt werd voor de vegetatieve cultuur, inge-25 steld op 250 omw/min. Na 96 uur werd de produktiecultuur geoogst voor het isoleren van sandramycine.
B. Bench-top fermentatie.
Voor de produktie van sandramycine in een bench-top fermentor, werd 400 ml vegetatieve cultuur zoals beschreven in voorbeeld IA, over-30 gebracht in een fermentor (Microgen Model SF-116, New Brunswick Scientific Co., Inc.) met 10 liter produktiemedium, bestaande uit 40 g maïszetmeel, 20 g lijnzaadmeel, 1 g (NH ) SO en 5 g CaCO per liter gedelo-

niseerd water. De temperatuur werd op 27°C gehouden, de agitatiesnel-heid bedroeg 300 omw/min en de luchtstroomsnelheid bedroeg 8 1/min.
35 Na 202 uur incubatie, werd sandramycine uit de cultuur geïsoleerd.
C. Tankfermentatie.
Voor produktie op pilotschaal van sandramycine werd 2 liter '4/ m w ü ij ö -19- vegetatieve cultuur (bereid volgens de algemene procedure van voorbeeld IA) overgebracht in een fermentor die 30 1 produktiemedium bevatte, bestaande uit 40 g maïszetmeel, 20 g lijnzaadmeel, 1 g (NH^^SO en 5 g CaCO^ per liter gedeloniseerd water. De incubatietemperatuur bedroeg 5 26,5°C, de luchtstroomsnelheid 80 1/min, de agitatiesnelheid 375 omw/min en de tegendruk 1 atmosfeer. Na 183 uur werd de tankfermentatie voor het isoleren van sandramycine geoogst.
Voorbeeld II.
Isolatie en zuivering van sandramycine.
10 Stap A: Extractie.
Ruwe volledige fermentatievloeistof (ongeveer 8 liter) werd overgebracht in een 20-liter polyetheen tank (topdiameter 48 cm, bodem-diameter 44 cm, hoogte 55 cm), met een bodem voorzien van een kraan.
Er werd een gelijk volume ethylacetaat toegevoegd. Het mengsel werd 15 gedurende 30 minuten met een lucht aangedreven roerder met een goede mengsnelheid geroerd. Ongeveer 6 liter (2 kg) Dicalite werd toegevoegd en er werd gemengd. Het mengsel werd gefiltreerd met een Dicalite kussen dat in een No. 12 Buchner trechter werd gehouden. Het filtraat werd verzameld in een 19-liter oplossingfles, voorzien van een vacuum-20 afvoer. De mat werd met 2 liter ethylacetaat gewassen. Het filtraat werd overgebracht in een 20-liter scheitrechter en de fasen liet men scheiden. Het ethylacetaatextract werd verwijderd en geconcentreerd tot ongeveer 1 liter in een glazen circulerende verdamper van laboratorium-afmetingen, voorzien van een continue toevoer. Het concentraat werd j 25 onder verminderde druk verder verdampt tot een visceuze olie in een j roterende verdamper, waarbij 1,94 g ruw extract werd verkregen. · j
Stap B: Vloeistof-vloeistof verdeling van het ruwe extract.
Het ruwe extract van stap A (1,94 g) werd opgelost in een mengsel · van 200 ml methanol en 200 ml Skellysolve B. De bi-fasische oplossing 30 werd overgebracht in een 1-liter scheitrechter en verdund met 22 ml water. Het mengsel werd geschud en de resulterende fasen liet men scheiden. De waterige methanolfase (onderste fase) werd overgebracht in een tweede 1-liter scheitrechter en nog twee keer geëxtraheerd met hoeveelheden Skellysolve B van 200 ml. Het Skellysolve B was tevoren 35 verzadigd met een gelijk volume 10 % water in methanol. De waterige methanolfase werd verdund met 44 ml water en drie keer geëxtraheerd met 200 ml porties van koolstof tetrachloride. Het koolstof tetra- -ai * 1 . ? -20- chloride was tevoren verzadigd met een gelijk volume van 25 % water in methanol. De waterige methanolfase werd verdund met 41 ml water en drie keer geëxtraheerd met 200 ml porties van chloroform. Het chloroform was tevoren verzadigd met een gelijk volume van 35 % water in methanol. De 5 koolstof tetrachloride extracten werden bij elkaar gevoegd en onder verminderde druk drooggedampt in een roterende verdamper waarbij 595 mg van residu A werd verkregen. De chloroformextracten werden bij elkaar gebracht en onder verminderde druk drooggedampt in een roterende verdamper waarbij 458 mg van residu B werd verkregen.
10 Stap C; Fijnwrijven van de residuen A en B.
Het resdu A (547 mg) en residu B (389 mg),verkregen in stap B, werden samengevoegd en opgelost in 30 ml van 2 delen chloroform-1 deel methanol. Men voegde 17,4 g Dicalite toe aan de oplossing, en daarna werd een suspensie geproduceerd door toevoeging van 200 ml Skellysolve B. 15 De oplosmiddelen werden onder verminderde druk verdampt met behulp van een roterende verdamper. Het overblijvende poeder werd in 500 ml tolueen gesuspendeerd en gepakt in een 4,1 cm inwendige diameter x 45,7 cm flash chromatografiekolom. Het Dicalite werd gepakt in een bed met een onder druk gebrachte (1^-5,7 psi, dat is 39 kPa) stroming. Toen het 20 pakkingsoplosmiddel het oppervlak van het bed bereikte, werd de stroming beëindigd en werd de druk van de kolom gelaten. Aan het oppervlak werd een laag Ottawa zand (ongeveer 2 cm) toegevoegd. De kolom werd daarna geëlueerd met onder druk gebrachte stikstofstroming volgens de hiernavolgende elutrope reeks: 500 ml tolueen; 500 ml diethylether; 500 ml 25 methyleenchloride; 500 ml chloroform; 500 ml ethylacetaat; 500 ml ace tonitrile; 500 ml tetrahydrofuran en 500 ml methanol. Het tolueeneluens en het pakkingsfiltraat werden gecombineerd (ongeveer 1 liter) en onder verminderde druk drooggedampt in een roterende verdamper waarbij 0,699 g . van residu C werd verkregen.
30 Stap D:, Kolomchromatografie van residu C.
Een 2,0 cm inwendige diameter x 30 cm Glenco kolom werd gepakt met een suspensie van 37 g Woelm silicagel (0,060 - 0,200 mm, 70 - 230 mesh) in chloroform. Residu C (0,699 g) uit stap C werd opgelost in 5 ml chloroform en op de top van de kolom aangebracht. Het monster liet 35 men in het gepakte bed sijpelen. De lege ruimte tussen de top van de kolom en het silicagelbed werd gevuld met standaard Ottawa zand. De kolom werd verbonden met een Glenco gradiënt-elutie-apparaat en elutie

-21- werd gestart met een 2-liter lineaire gradiënt van chloroform tot 5 % methanol in chloroform, waarbij 20 x 100 ml fracties werden verzameld.
Elke fractie werd onder verminderde druk in een roterende verdamper drooggedampt. Het residu werd opgelost in 3 mm van 2 dln chloroform-1 dl 5 methanol. Van elke fractie werden hoeveelheden van 2 yl aangebracht op Analtech silicagel GHLF dunne-laag chromatografie (TLC) platen. De platen werden geëlueerd met 5 % methanol in chloroform en zichtbaar gemaakt met 254 nm en 366 nm ultraviolet licht. Fractie 6 werd als homogeen beoordeeld. Het kristallijne residu van fractie 6 werd herkristalliseerd 10 uit chloroform-methanol, waarbij 215 mg sandramycine werd verkregen.
Dit materiaal werd herkristalliseerd uit chloroform-methanol, waarbij 188 mg zuiver sandramycine werd verkregen met een smeltpunt van 208 -212°C.

Voorbeeld III.
Isolatie en zuivering van sandramycine op grotere schaal.
Stap A: Extractie en vloeistofverdeling van het ruwe extract.
Bij herhaling van de algemene isolatie-procedures beschreven in 20 voorbeeld II, stappen A en B, werden 1,12 g van residu A en 3,03 g van residu B verkregen uit de volledige fermentatievloeistof (ongeveer 8 -liter).
Stap B: Fijnwrijven van de residuen A en B.
Het in stap A verkregen residu A (1,12 g) werd opgelost in onge- 1 25 veer 300 ml van 2 dln chloroform-1 dl methanol. Men voegde aan de oplossing 20 g Dicalite toe. Het oplosmiddel werd in een roterende verdamper onder verminderde druk drooggedampt. Het residu werd in 300 ml tolueen gesuspendeerd en opnieuw drooggedampt. Dit werd een tweede keer herhaald. Het resulterende residu werd in 300 ml Skellysolve B gesuspen-30 deerd en in een roterende verdamper drooggedampt. Dit werd een tweede keer herhaald. Het Dicalite residu werd in 300 ml Skellysolve B gesuspendeerd en gepakt in een 4,1 cm inwendige diameter x 45,7 cm flash chromatografie kolom. Het Dicalite werd gepakt in een bed met een onder druk gebrachte (^-5,7 psi, dat wil zeggen 39 kPa) stroming. Op het bed 35 werd een laag van Ottawa zand gepakt (ongeveer 2 cm). Elutie werd gestart met onder druk gebrachte stikstofstroming volgens de volgende elutrope reeks: 500 ml Skellysolve B; 500 ml tolueen; 500 ml diëthyl- - i w * -22- ether; 500 ml methyleenchloride; 500 ml chloroform; 500 ml ethylacetaat; 500 ml acetonitrile; 500 ml tetrahydrofuran en 500 ml methanol. Het tolueeneluens werd onder verminderde druk in een roterende verdamper drooggedampt, waarbij 912,7 mg van residu D werd verkregen.
5 De bovenstaand aangegeven algemene procedure werd herhaald, be halve dat het daarin gebruikte residu A werd vervangen door 3,03 g van residu B, en daarmee werd 756,7 mg van residu E uit het tolueeneluens geproduceerd.
De bovenstaand in voorbeeld III, stappen A en B beschreven alge-10 mene procedures werden herhaald, behalve dat de daarin verkregen en gebruikte residuen A en B werden vervangen door 1,5 g respectievelijk 1,97 g van de residuen A' en B', waardoor uit het tolueeneluens respectievelijk 993,9 mg van residu D' en 593 mg van residu E' werd geproduceerd.
15 Stap C; Kolomchromatografie van de residuen D en E.
Een 2,0 cm inwendige diameter x 30 cm Glenco kolom werd gepakt met een suspensie van 40 g Woelm silicagel (0,063 - 0,200 mm, 70 - 230 mesh) in chloroform. De kolom werd ingepast in het middelmatige druk HPLC-systeem en geëquilibreerd met chloroform. De residuen D, D', E 20 en E' van stap B werden bij elkaar gevoegd (ongeveer 3,366 g) en opgelost in 6 ml chloroform. De oplossing werd in een 15-ml monsterlus getrokken. De monsterlus werd in het middelmatige druk HPLC-systeem ingepast, en het monster werd met 200 ml chloroform op de kolom gepompt. Elutie begon met een 2-liter lineair gradiënt van chloroform tot 5 % 25 methanol in chloroform, waarbij 17 x 117 ml fracties werden verzameld. Van elke fractie werden hoeveelheden van 6 μΐ met behulp van TLC, waarbij 5 % methanol in chloroform als eluens werd gebruikt, geanalyseerd, en zichtbaar gemaakt met 366 nm licht. De fracties 5 en 6 werden samengevoegd en drooggedampt, waarbij 2,166 g van residu F werd verkregen.
30 De fracties 7-12 werden samengevoegd en onder verminderde druk met behulp van een roterende verdamper drooggedampt. Het residu (953 mg) werd opgelost in 6 ml chloroform en opnieuw chromatografisch behandeld als bovenstaand uiteengezet, waarbij een 2-liter lineaire gradiënt van chloroform tot 1,5 % methanol in chloroform werd gebruikt en 20 x 100 ml 35 fracties werden verzameld. De fracties 9 - 20 werden samengevoegd en in een roterende verdamper drooggedampt, waarbij 860 mg van residu G werd verkregen.

De residuen F en G werden gecombineerd en herkristalliseerd uit chloroform-methanol, waarbij 1,985 g zuiver sandramycine werd verkregen, dat identiek was aan het produkt dat in voorbeeld II was geïsoleerd.

Claims (8)
1. Antitumor antibioticum sandramycine met structuurformule 1 volgens het formuleblad.
2. Werkwijze voor het bereiden van het antitumor antibioticum sandramycine, omvattende het kweken van een sandramycine-producerende 5 stam van Nocardioides sp. nov. met de identificerende eigenschappen van ATCC 39419, of een mutant daarvan, onder ondergedompelde aerobe condities in een kweekmedium dat assimileerbare bronnen van koolstof en stikstof bevat, totdat een substantiële hoeveelheid sandramycine is geproduceerd en opgehoopt in het kweekmedium.
3. Werkwijze volgens conclusie 2, omvattende de extra stap van het isoleren van het sandramycine uit het kweekmedium.
4. Biologisch zuivere cultuur van het micro-organisme Nocardioides sp. ATCC No. 39419, dat in staat is tot het produceren van het antitumor antibioticum sandramycine in een winbare hoeveelheid na kweken in een 15 kweekmedium dat assimileerbare bronnen van koolstof en stikstof bevat onder ondergedompelde aerobe condities.
5. Werkwijze voor het therapeutisch behandelen van een dierlijke gastheer, die aangetast is door een bacteriële infectie, omvattende het toedienen aan de gastheer van een effectieve antibacteriële dosis van 2. sandramyc ine.
6. Werkwijze voor het therapeutisch behandelen van een dierlijke gastheer, die aangetast is a»r een kwaadaardige tumor die gevoelig is voor sandramycine, omvattende het toedienen aan de gastheer van een dosis sandramycine die tumor-inhibitie geeft.
7. Farmaceutisch preparaat, omvattende een effectieve antibacteriële 25 hoeveelheid van sandramycine in combinatie met een farmaceutische drager of verdunningsmiddel.
