Anti-tandaanslag/antigingivitis methode.
De uitvinding betreft de toepassing van niet-antibacteriële middelen en orale preparaten voor het bevorderen van de menselijke mondhygiëne/ en speciaal een methode voor het behandelen, tegengaan of remmen van zowel tandaanslag als gingivitis, welke laatste gepaard 5 gaat met symptomen als ontsteking, bloeden, terugtrekking en/of zwellen van het tandvlees. Bepaalde typen gingivitis omvatten afunktionele gingivitis, marginale gingivitis en katoen-kussen gingivitis.
Gingivitis leidt tot periodontitis.
Het tandvlees wordt duidelijk geschaad door afzetting van 10 tandsteen en tandaanslag, een combinatie van mineralen en bacteriën in de mond. De bacteriën, die gepaard gaan met tandsteen kunnen enzymen en endotoxinen afgeven, welke het tandvlees irriteren en een ontste-kings-gingivitis veroorzaken. Naarmate het tandvlees sterker wordt geïrriteerd door dit proces, heeft het de neiging te gaan bloeden, 15 zijn taaiheid en veerkracht te verliezen en zich van de.tanden los te maken, waardoor periodontale ruimten ontstaan, waarin afzettingen, secreties, meer bacteriën en toxinen verder kunnen accumuleren. Het is ook mogelijk, dat zich voedsel in deze ruimten ophoopt, waardoor een voedingsbodem voor een verhoogde groei van bacteriën en daarmede 20 een produktie van endotoxinen en destructieve enzymen wordt geleverd.
Actinomyces viscosus, een gram positief staafje, is geïdentificeerd als deel hebbend in de etiologie van gingivitis, Loeche et al: "Bacteriology of human experimental gingivitis: effects of plaque and gingivitis sores". Infection and Immunity 21, 830-839 (1978). Dit 25 organisme hecht zich aan het tandoppervlak en vormt daar aanslag en tandsteen.
Een reeks stoffen is eerder voorgesteld en gebruikt voor het bestrijden van tandsteen en gingivitis, maar geen daarvan is volledig bevredigend gebleken. Bijv. bleken enkele van deze stoffen instabiel bij 30 aanwezigheid van de anionogene oppervlakte-actieve middelen, die gewoonlijk aanwezig zijn in gebruikelijke orale preparaten. Een aantal van dit soort stoffen, zoals de kationogene kwatemaire ammoniumverbindingen oefenen een antibacteriële activiteit uit, die op een ongewenste wijze de neiging heeft om de normale microflora van de mond en/of het spijs-35 verteringssysteem te verstoren of te doden.

Amerikaans octrooischrift 3.429.963 vermeldt onder een reeks andere in water oplosbare polyelektrolyten het gebruik van polyvinyl-fosfonzuur (VPA polymeer) of zouten daarvan voor het complexvormig binden van calcium en het remmen van orale afzettingen, maar de enige 5 in vivo proef, daarin beschreven, omvatte het geven van drinkwater met een gehydrolyseerd copolymeer van etheen en maleinezuuranhydride voor het ad libitum drinken door ratten gedurende 5 dagen. Deze proef is er een, waarbij calciumstoechiometrisch complexvormig wordt gebonden en heeft geen verband met het drempeleffekt optredend bij actueel oraal 10 gebruik, voor het behandelen van tandoppervlakken 1 tot 3-maal per dag gedurende tenminste twee weken of het gehele leven. Bij toepassing in een dergelijke praktijkproef bleek polyvinylfosfonzuur ongeschikt voor het goed remmen van orale afzettingen.
Amerikaans octrooischrift 4.342.857 beschrijft anti-gingivitis-15 preparaten met een vinylfosfonzuur/vinylfosfonylfluoridecopolymeer, maar een reeks wetenschapsmensen, autoriteiten en/of regelingen hebben bezwaar tegen een toediening van gefluoreerde materialen aan mensen.
De uitvinding betreft nu een anti-tandsteen/anti-gingivitis methode, die niet onderhevig is aan een of meer van de boven weergegeven 20 gebreken en bezwaren. Verdere doeleinden en voordelen van de uitvinding volgen hieronder.
Het bereiken van het gegeven doel wordt mogelijk gemaakt door de ontdekking, dat VPA-polymeer en zijn zouten de aanhechting van Actinomyces viscosus aan met speeksel bedekte hydroxyapatite (HAP) 25 korrels stoort of remt. Dit is een betrouwbare indicatie, dat dit middel de aanhechting van dit organisme aan tandoppervlakken remt, daardoor tandaanslag en tandsteen vermindert en aldus gingivitis tegengaat. Een dergelijke anti-gingivitis activiteit is in feite bevestigd door een in vivo proef op bio-honden, die hieronder meer uitgebreid is beschreven. 30 De uitvinding betreft dus een methode voor het remmen van men selijk tandaanslag, tandsteen en gingivitis door in de menselijke orale mondholte een preparaat aan te brengen met daarin een voor de tanden geschikte orale drager en een effectieve tandsteen- en gingivitis remmende hoeveelheid polyvinylfosfonzuur of een oraal geschikt zout 35 daarvan.
Het VPA polymeer volgens de uitvinding moet bij voorkeur een 84 Q 3 9 4 f

* -3- getalsgemiddelde molecuulgewicht (berekend uit de viscositeit of lichtverstrooiingsmetingen) van ca.6000 tot ca.100.000, liefst 8000 tot ca.20.000 bezitten, en kan op een op zichzelf bekende wijze worden bereid door een polymerisatie van vinylfosfonyldichloride onder nage-5 noeg watervrije voorwaarden bij aanwezigheid van een vrije radicaal-katalysator, gevolgd door mengen van het verkregen polymeer met water waardoor de vinyifosfonyldichloride-eenheden in het polymeer hydroly-tisch in VPA-eenheden worden omgezet. Het verkregen polymeer is dan in de vrije zuurvorm en kan desgewenst in een zoutvorm worden omgezet 10 door een behandeling met elke oraal geschikte kationen leverende base, zoals alkalimetaalhydroxyden (bijv. natrium of kalium), ammonia,
Cl mono-, di- en tri-gesubstitueerd ammonium, bijv. alkanol gesubstitueerd ammonium, zoals mono-, di- en tri-ethanolammonium), organische aminen enz.
15 Het zal duidelijk zijn, dat de mono- en di-zoutvormen van het polymeer de equivalenten zijn van de vrije zuurvorm en dat de uitdrukking "in water oplosbaar", die toepasbaar is voor al deze vormen, ook s'nel in water dispergeerbare vormen daarvan in de gebruikelijke concentraties omvat.
20 Het zal tevens duidelijk zijn, dat het VPA polymeer ook onder geschikte hoeveelheden, d.w.z. minder dan 50 gew.%, bij voorkeur minder dan ca. 10 gew.%, liever nog minder dan ca. 5 gew.% en liefst minder dan 2 gew.% aan eenheden afgeleid van andere niet-gefluoreerde etheen-onverzadigde monomeren kan bevatten, die in type en hoeveelheid niet 25 giftig zijn en de gewenste in water oplosbaarheid en anti-gingivitis activiteiten van het polymeer niet storen. Dit soort andere monomeren omvatten bijv. alkenen, zoals etheen, propeen, isopropeen, buteen en isobuteen, vinylalkylethers, zoals vinylmethyl, ethyl en isobutyl-ethers, alpha, beta-onverzadigde carbonzuren en hun alkyl- en gesüb-30 stitueerde alkylesters, zoals acryl-, methacryl-, aconiet, maléine-en fumaarzuren en hun methyl, ethyl, isobutyl en dimethylaminoethyl-esters, voorts allylalkohol en acetaat, vinyl en vinylideenhalogenlden, vinylalkaanzuuresters, zoals vinylacetaat en butyraat, acrylamide en methacrylamide en N-alkyl en Ν,Ν-dialkyl gesubstitueerde derivaten 35 daarvan enz.
De concentratie van het polymere VPA-anti-gingivitis middel 8403941 » o -4- in orale preparaten kan sterk variëren, meestal van ca.0,01 gew.% af tot geen bovengrens, behalve als bepaald door kosten of onverenigbaarheid met de drager..Gewoonlijk worden concentraties van ca.0,01-10%, bij voorkeur 0,1-8,0%, en liefst 0,5-5,0 gew.% toegepast. Orale 5 preparaten, die bij een gebruikelijke toepassing per ongeluk kunnen worden ingeslikt, bevatten bij voorkeur concentraties in de lagere regionen van de genoemde trajecten.
Bij bepaalde sterk geprefereerde vormen van de uitvinding kunnen de orale preparaten vloeibaar zijn, zoals mondwas of spoelvloei-10 stoffen. Dergelijke preparaten bevatten gewoonlijk een bevochtigings-middel en de drager is meestal een water-alkoholmengsel. Gewoonlijk ligt de verhouding van alkohol tot water in het traject van ca.l:l tot ca. 20:1, bij voorkeur van 3:1 tot 20:1 en liefst bij ca.17:3 gew.dln. De totale hoeveelheid water-alkohol-mengsel van dit type preparaat 15 ligt meestal tussen 70 en 99,9 gew.% van het preparaat. De pH van dit soort vloeibare en andere onderhavige preparaten ligt gewoonlijk tussen 4,5 en ca. 9 en meestal tussen ca. 5,5 en 8. De pH ligt bij voorkeur tussen 6 en ca.8,0. Opgemerkt wordt, dat de onderhavige preparaten * oraal kunnen worden toegediend bij een vrij lage pH zonder dat het 20 tandemaille sterk wordt ontkalkt.
Dergelijke vloeibare orale preparaten kunnen tevens een opper- vlakteactief middel en/of een fluorleverende verbinding bevatten.
In bepaalde andere gewenste vormen volgens de uitvinding kan het orale preparaat nagenoeg vast of pastavormig zijn, zoals een tand- 25 poeder, een tandtablet, een tandpasta of een tandcrëme. De drager van dit soort vaste of pastavormige orale preparaten bevat gewoonlijk een polijstmateriaal. Voorbeelden van polijstmaterialen zijn in water onoplosbaar natrium-metafosfaat, kaliummetafosfaat, tricalciumfosfaat, calciumpyrofosfaat, magnesiumorthofosfaat, trimagnesiumfosfaat, cal- 30 ciumcarbonaat, alumina, gehydrateerd alumina, aluminium silicaat, zircoonsilicaten, silica, bentoniet of mengsels daarvan. Voorkeurs polijstmaterialen omvatten kristallijne silica met deeltjesgrootten tot 5 micron en een gemiddelde deeltjesgrootte tot 1,1 micron en een 2 specifiek oppervlak tot 50.000 cm /g, silicagel, complex amorf alkali-35 metaal aluminosilicaat, gehydrateerde alumina en dicalciumfosfaat.

Alcoa als C333, met een aiuminagehalte van 64,9gew.%, een silicagehalte van 0,008%, een ferri-oxydegehalte van 0,003% en een vochtgehalte van 0,037 % bij 110°C en met een soortelijk gewicht van 2,42 en een deeltjesgrootte zodanig, dat 100% van de deeltjes kleiner is dan 50 micron 5 en 84% van de deeltjes kleiner zijn dan 20 micron, is een bijzonder gewenst materiaal.
Wanneer visueel heldere gelen worden toegepast, zijn polijst middelen van colloidale silica, zoals verkocht onder hethandelsmerk SYLOID als Syloid 72 en Syloid 74 of onder het handelsmerk SANTOCEL 10 als Santocel 100 en alkalimetaal aluminosilicaatcomplexen bijzonder geschikt, aangezien zij brekingsindices bezitten, die zeer nabij de brekingsindices van de gelmiddel-vloeistof (met inbegrip van water en/ of bevochtigingsmiddel) systemen liggen, die gewoonlijk in tandpasta's worden toegepast.
15 Veel van de zogenaamde "onoplosbare” polijstmaterialen zijn anionogeen van karakter en bevatten ook kleine hoeveelheden oplosbaar materiaal. Zo kan onoplosbaar natriummetafosfaat op elke passende wijze worden gevormd, als weergegeven in Thorpe's Dictionary of Applied Chemistry, deel 9, vierde druk, blz.510-511.' De vormen van 20 onoplosbaar natriummetafosfaat, bekend als Madrell's zout en Kurrol's zout zijn verder voorbeelden van geschikte materialen. Deze metafosfnatzouten bezitten een minimale oplosbaarheid in water, en worden daarom gewoonlijk aangeduid als onoplosbare metafosfaten. Hierin is een ondergeschikte hoeveelheid oplosbaar fosfaatmateriaal als onzuiverheden 25 aanwezig, gewoonlijk enkele procenten, bijv. ten hoogste 4 gew.%. De hoeveelheid oplosbaar fosfaatmateriaal, welke naar men aanneemt een oplosbaar natriumtrimetafosfaat bij onoplosbaar metafosfaat omvat, kan desgewenst worden verminderd door te wassen met water. Het onoplosbare alkalimetaalmetafosfaat wordt gewoonlijk toegepast in poedervorm met 30 een deeltjesgrootte, zodanig, dat niet meer dan ca.1 % van het materiaal groter is dan 37 micron.
Het polijstmateriaal is gewoonlijk aanwezig in hoeveelheden . tussen ca.10 en ca.99gew.% van het orale preparaat. Bij voorkeur is het aanwezig in hoeveelheden tussen 10 en ca.75% in tandpasta en tussen 35 70 en ca.99% in tandpoeder.

* de diverse vaste bestanddelen in passende hoeveelheden en deeltjesgrootten mechanisch te mengen, bijv. door te malen.
Bij pastavormige orale preparaten moet de boven weergegeven combinatie van antigingivitis middel en polijst materiaal verenigbaar 5 zijn met de overige bestanddelen van het preparaat. Zo kan in een tandpasta de vloeibare drager water en een bevochtigingsmiddel bevatten, meestal in een hoeveelheid tussen ca.10 en ca.90 gew.% van het preparaat. Glycerol, sorbitol of polyethyleenglycol kan eveneens als bevochtigingsmiddel of bindmiddel aanwezig zijn. Bijzonder gunstige 10 vloeibare bestanddelen zijn polyethyleenglycol en polypropyleenglycol. Ook gunstig zijn vloeistofmengsels van water, glycerol en sorbitol.
Bij heldere gelen, waar de brekingsindex een belangrijk punt van overweging is, wordt ca.3-30 gew.% water, 0 tot ca.80 gew.% glycerol en ca.20-28 gew.% sorbitol bij voorkeur toegepast. Een gelmiddel, 15 .zoals natuurlijke of synthetische gommen of gomachtige materialen, zoals Iers mos, natriumcarboxymethylcellulose, methylcellulose, hydroxy-• ethylcellulose, tragacanth gom, polyvinylpyrrolidon, stijfsel, en bij voorkeur hydroxypropylmethylcellulose en de carbopolen (bijv. 934,940 en 941) , enz., is gewoonlijk in de tandpasta in een hoeveelheid van 20 ten hoogste 10 gew.% aanwezig, liefst in een hoeveelheid vein ca.0,5 tot 5 gew.%. In een tandpasta of gel worden de vloeistoffen en vaste materialen zo op elkaar afgestemd, dat een crêmevormige of gelvormige massa wordt gevormd, welke extrudeerbaar is uit een onder druk staande houder of uit een uitdrukbare tube van bijv. aluminium of lood.
25 De vaste of pastavormige orale preparaten, die gewoonlijk een pH, gemeten bij een 20%'s suspensie van ca.4,5-9, bij voorkeur 5,5-8 en liefst 6-8,0 bezitten, kunnen tevens een oppervlakteactief middel en/of een fluor leverende verbinding bevatten.
Het is duidelijk, dat als gebruikelijk de onderhavige orale 30 preparaten worden verkocht of anderszins gedistribueerd in geschikt gemerkte verpakkingen. Zo zal een flesje mondspoelvloeistof een aanduiding hebben, die het als mondvloeistof of mondwasvloeistof beschrijft en aanwijzingen bevatten voor zijn gebruik; een tandpasta,zal gewoonlijk aanwezig zijn in een uitdrukbare tube, speciaal van aluminium of 35 gevoerd lood, dan wel een andere uitdrukhouder voor het uitpersen van de inhoud met een aanduiding daarop, die hem als tandpasta of tandcrême 8403941 -7- aanduidt.
De onderhavige orale preparaten kunnen een niet-zeep-synthetisch, voldoende in water oplosbaar organisch an-ionogeen of noh-ionogeen bevochtigingsmiddel bevatten en wel in concentraties gewoonlijk tussen 5 ca.0,05 en 10, bij voorkeur 0,5-5 gew.%, voor het bevorderen van de bevochtigings, detergens- en schuimeigenschappen. Amerikaans octrooi-schrift 4.041.149 beschrijft-dit soort geschikte anionogene middelen in kolom 4, regels 31-38 en dit soort non-ionogene middelen in kolom 8, regels 30-68 en kolom 9, regels 1-12, waarnaar hier wordt verwezen.
10 In bepaalde vormen van de uitvinding is een fluorleverende verbinding in de orale preparaten aanwezig. Deze verbindingen kunnen vrij slecht of volledig in water oplosbaar zijn. Zij worden gekarakteriseerd door hun vermogen fluorionen in water af te geven en doordat zij nagenoeg geen reaktie geven met andere bestanddelen van het orale 15 preparaat. Tot deze materialen behoren anorganische fluorzouten, zoals de oplosbare alkalimetaal, aardalkalimetaal en zwaarmetaalzouten, bijv. natriumfluoride, kaliumfluoride, ammoniumfluoride, calciumfluoride, een koperfluoride, zoals Cuf, zinkfluoride, een tinfluoride, zoals % stannifluoride of stannochloorfluoride, bariumfluoride, natriumfluor-20 silicaat, ammoniumfluorsilicaat, natriumfluorzirconaat, natriummono- fluorfosfaat, aluminium mono- en di-fluorfosfaat en gefluoreerd natrium-calciumpyrof osf aat. Alkalimetaal en tinfluoriden, zoals natrium en stannofluoriden, natriummonofluorfosfaat en mengsels daarvan verdienen de voorkeur.
25 De hoeveelheid fluor leverende verbinding is in zekere mate afhankelijk van de soort verbinding, zijn oplosbaarheid en het type van hetorale preparaat, maar de hoeveelheid ervan moet niet giftig zijn.
Bij een vast oraal preparaat, zoals een tandpasta of een tandpoeder wordt een hoeveelheid van dit soort verbindingen als bevredigend be-30 schouwd, welke maximaal ca.1 gew.%, berekend op het preparaat, af geeft. Elke passende minimale hoeveelheid van dit soort verbindingen kan worden gebruikt, maar bij voorkeur wordt voldoende verbinding toegepast om 0,005-1% en bij voorkeur 0,1% fluorideion vrij te geven. Gewoonlijk is bij alkalimetaalfluoriden en stannofluoriden deze verbinding in een 35 hoeveelheid tot 2 gew.% aanwezig, berekend op het gewicht van het preparaat, en bij voorkeur in het traject tussen 0,05 en 1%. Bij natrium- 8 4 0 ó 8 4 1 Ö 3· <*· -8- monofluorfosfaat kan de verbinding in een hoeveelheid tot 7,6 gew.% aanwezig zijn, maar gewoonlijk wordt 0,76% gebruikt.

In een' bloeibaar oraal preparaat, zoals een mondwasvloeistof, is de fluorleverende verbinding gewoonlijk aanwezig in een hoeveelheid 5 voldoende om ca.0,13%, bij voorkeur ca.0,0013 tot 0,1% en liefst ca.0,0013gew.% fluoride af te geven.
Diverse andere stoffen kunnen in de onderhavige orale prepa- t raten worden verwerkt. Voorbeelden zijn witmakers, conserveermiddelen, siliconen, chlorofylverbindingen en ammonium afgevend materiaal, 10 zoals ureum, diammoniumfosfaat en mengsels daarvan. Deze hulpstoffen worden indien aanwezig in de preparaten verwerkt in hoeveelheden, die de eigenschappen en kenmerken daarvan niet ongunstig beïnvloeden.
Elk passend aroma of zoetmiddel kan eveneens worden toegevoegd, eveneens rekening houdend met het bovenstaande. Voorbeelden van ge-15 schikte aromastoffen zijn aromatische oliën, zoals muntolie, pepermunt-olie, wintergroenolie, sassafrasolie, kruidnagelolie, salie-olie, eucalyptusolie, majoramolie, kaneelolie, citroenölie en sinaasappelolie · alsmede mêthylsalicylaat. Geschikte zoetmiddelen omvatten saccharose, lactose, maltose, sorbitol, natriumcyclamaat, perillartine, APM 20 (aspartylfenyl-alanine, methylester) en saccharine. Meestal maken aroma-stof en zoetmiddel tezamen 0,1-5% of meer van het preparaat uit.
Bij de uitvoering van de uitvinding kan een oraal preparaat volgens de uitvinding, zoals een mondwasvloeistof of: tandpasta met daarin het anti-gingivitis middel in een oraal aanvaardbare drager wor-25 den bereid door samenvoegen van de bestanddelen op een gebruikelijke wijze, en op de gingiva en de tanden op een gebruikelijke wijze worden aangebracht, liefst dagelijks, bijv. 1-3 maal dagelijks of elke tweede of derde dag, enz. en wel bij een pH van ca.4,5-9, gewoonlijk 5,5-8,5 en liefst 6-8, bij voorkeur gedurende tenminste 2 weken tot 8 weken of 30 meer of gedurende het gehele leven.
Bij kauwgomsoorten en andere produkten kunnen de VPA-actieve stoffen op elke geschikte wijze tijdens de gebruikelijke vervaardiging van het produkt daarin worden verwerkt. Bijv. kunnen zij worden verwerkt in een warme gombasis onder roeren, waardoor zij daarin uniform worden 35 verdeeld. Zij kunnen ook worden toegevoegd aan de buitenkant of het buitenoppervlak van een gombasis om daarmede deze basis te bekleden.

De gebruikelijke gombases kunnen worden gebruikt, voorbeelden zijn jelutone, rubberlatex, vinylietharsen enz., in combinatie met andere gebruikelijke stoffen, zoals weekmakers of plastificeermiddelen, suikers of andere geschikte koolhydraten, zoals glucose, sorbitol enz.
5 De volgende voorbeelden lichten de uitvinding nader toe, maar beogen deze niet te beperken. Alle hoeveelheden en verhoudingen zijn gewichtshoeveelheden en gewichtsverhoudingen, de temperaturen zijn in graden Celsius tenzij anders aangegeven. Het in deze voorbeelden gebruikte VPA-polymeer had een molecuulgewicht van ca. 10.700 en werd 10 toegepast in de vorm van het dinatriumzout.
Voorbeeld I
Adsorptie van natriumpolyvinylfosfonaat aan tandemaille:
De adsorptie van het polymeer aan emaille oppervlakken werd in vitro gemeten. Zij extraheerde mensentanden, die niet door caries 15 waren aangetast of waren gevuld, werden met puimsteen schoongemaakt. Vervolgens werden zij gepolijst met een rubber cup en een polijst-middel. Zij werden vervolgens bevestigd op een rubber stop met een nichroom draad, die door een holte in de wortels was aangetrokken. Een natriumzout van polymeeroplossingen met pH 7,0 werd in polyetheen 20 buizen toegevoerd. De tanden werden een uur bij 37°C in deze oplossing onder continu roeren ondergedompeld. Er werd speciaal op gelet, contact tussen oplossing en tandwortel te vermijden. Na 1 uur incuberen werden de tanden verwijderd en de oplossingen geanalyseerd pp de hoeveelheid polymeer, overgebleven in de oplossing. De adsorptie werd berekend uit 25 het verschil tussen de aanvankelijk toegevoerde hoeveelheid minus de hoeveelheid overgebleven na contact met de tanden.
De concentratie van het polymeer in de oplossing werd bepaald door turbidimetrische metingen onder toepassing van 5M CaCl.oplossing 3 3 ^ met pH 4,5. 1 cm CaCl2 werd toegevoegd aan 1 cm van de polymeerop- 30 lossing. De troebeling van de verkregen colloidale suspensie bij 500 nm bleek daarbij evenredig met de polymeerconcentratie tussen 1 en 8 mg/cm^.
Een ijkkromme werd opgesteld uit bekende waarden van het polymeer.

De cijfers duiden aan, dat een duidelijke adsorptie van VPA-polymeer op het emaille oppervlak is opgetreden.
Voorbeeld II
^ Effekt van het polymeer op de adsorptie van Actinomyces Viscosus T14 op met speeksel beklede hydroxyapatiet (HAP) korrels:
80 mg HAP korrels werden voorbekleed met menselijk speeksel (bloedtype A) en wel gedurende 12 'uren. De korrels werden gewassen en voorbehandeld met de oplossing van het polymeer bij pH 7,0 en wel 2Q gedurende 5 minuten. De behandelde korrels werden gewassen met een buffer, bestaande uit 0,05 M KC1, ImM POImM CaC^ en 0,1 mM MgClj met pH
6,0. Deze buffer stimuleert de anorganische bestanddelen van speeksel.

Voor het adsorptieonderzoek bevatte het mengsel (1,0 cm ) 7 5 x 10 met 3H thymidine gemerkte bacteriën (Actinomyces viscosus), 22 30 mg met speeksel beklede korrels (S HAP) en de buffer. Het mengsel werd bij kamertemperatuur 2 uur continu geschud. Men liet de korrels een minuut uitzakken en de bovenstaande vloeistof die niet geadsorbeerde cellen bevatte, werd verwijderd. De radioactiviteit werd via vloei-stof-scintillatietellingen gemeten. Porties bekende met 3H gemerkte 2q cellen werden op een zelfde wijze geteld, zodat de tellingen per minuut evenredig zijn met de bacteriecellen. Controle-bacterie-suspensies werden met S-HAP-korrels geincubeerd.

Deze resultaten tonen aan, dat een voorbehandeling van S-HAP
met 1% polyvinylfosfonaat duidelijk effectief is bij het tegengaan van baeterie-aanhechting.
s Voorbeeld III
Dit onderzoek bij 20 bio-honden geeft het effekt weer van een placebo en een spoeloplossing met 1% natriumzout van polyvinylfosfonzuur gedurende vier weken op tandsteen/gingivitis. De honden kregen een volledige profylaxe ter verwijdering van zachte en harde tandaf-2Q zettingen. Een ontsluitende oplossing werd toegepast om een volledige verwijdering van tandaf zetting te garanderen. De bio-honden werden 4 weken op een zacht dieet gehouden. Groep 1 CIO honden)' werden hierna behandeld met de placebo-spoelvloeistof, terwijl groep 2 werd behandeld met de spoelvloeistof met het polymeer. De behandeling werd eenmaal 3 22 per dag gedurende 5 dagen per week toegepast, waarbij 5-6 cm van de spoelyloeistof op alle tanden werd aangebracht. Het onderzoek was dubbel blind. Noch de beoordelaar noch de mensen, betrokken bij de behandelingen wisten welke spoelvloeistof het polymeer bevatte. De tandaanslag en de gingivitis werden bepaald volgens de Loe en Silness index (Acta 2Q Odontologica Scandinavica, 21, 551-555 (1963)..

De uitvinding is hiermede beschreven wat betreft zijn voorkeursuitvoeringsvormen, maar het zal duidelijk zijn, dat vele modificaties en variaties mogelijk zijn zonder buiten het kader van de 30 uitvinding te treden.

Claims (10)
1. Werkwijze voor het remmen van tandaanslag, tandsteen en gingivitis bij mensen,met het kenmerk, dat men in de mondholte van mensen een preparaat brengt met een dentaal geschikte orale drager en 5 een effectieve tandaanslag, tandsteen en gingivitis remmende hoeveelheid van een polyvinylfosfonzuur of een oraal geschikt zout daarvan.
2. Werkwijze volgens conclusie 1, met het kenmerk, dat het polyvinylfosf onzuur een getalsgemiddelde molecuulgewicht van ca.6000-100,000 bezit.
3. Werkwijze volgens conclusie 1, met het kenmerk, dat het preparaat nagenoeg dagelijks gedurende tenminste 2 weken wordt toegepast.
4. Werkwijze volgens conclusie 1, met het kenmerk, dat het preparaat ca.0,01-10 gew.% van het polyvinyIfosfonzuur of zout daarvan 15 bevat.
5. Werkwijze volgens conclusie 1, met het kenmerk, dat het preparaat ca. 0,5-5 gew.% aan polyviny Ifosfonzuur of zout daarvan bevat.
6. Werkwijze volgens conclusie 1, met het kenmerk, dat het preparaat een mondvloeistof is met een pH van ca.4,5-9 op basis van 20 een water-alkoholdrager.
7. Werkwijze volgens conclusie 1, met het kenmerk, dat het preparaat een tandpasta is met een pH van 4,5-9, een vloeibare drager, een gelmiddel en een dentaal geschikt polijstmiddel.
8. Werkwijze voor het remmen van menselijk tandaanslag, tand- 25 steen en gingivitis, met het kenmerk, dat men aan de menselijke mondholte een preparaat toedient met daarin een dentaal geschikte orale drager en een effectieve tandsteen, tandaanslag en gingivitis remmende hoeveelheid tussen ca. 0,01 en 10 gew.% van een polyviny Ifosfonzuur met een getalsgemiddelde molecuulgewicht van ca.6000 tot 100,000, of een 30 oraal geschikt zout daarvan.
9. Werkwijze volgens conclusie 8, met het kenmerk, dat dit preparaat nagenoeg dagelijks gedurende tenminste 2 weken wordt toegediend.
