Korte aanduiding: Nieuwe aryl-piperazineverbindingen, werkwijze voor hun bereiding en hun toepassing als geneesmiddel.
De uitvinding heeft betrekking op nieuwe aryl-piperazineverbindingen, /zouten, alsmede op een werkwijze voor de bereiding en toepassing in de geneeskunde van deze verbindingen en deze zouten.
5 De nieuwe verbindingen volgens de uitvinding be antwoorden meer in het bijzonder aan de algemene formule 1 van het formuleblad waarin X: — - ofwel een propeen/y^enon-1 keten met transconfiguratie, 3 2 1 (-CH=CH-CO-)of propylenoniketen (tCH2-CH2-CO-) voorstelt 10 die gebonden zijn aan Ar door zijn koolstofatoom op plaats 3, in welk geval A voorstelt: * een carbohydroxaniiB(-C0NH0H) groep of hydroxymethyl (-CH20H) groep waarbij Ar dan de (3,4,5-trimethoxy)fenylkern voorstelt, 15 * een e+toxycarbony-lgroep (COOEt), waarbij Ar dan de para-fluorfenyl-, (3,4-dichloor) fenyl-(2,3,4 -trimethoxy) fenyl-, of (2,4,6-trimethoxy) fenylkern voorstelt, * een carboxylgroep (COOH), waarbij Ar dan de 20 parachloorfenyl-, para-hydroxyfenyl-, (3,4- methyleendioxy) fenyl- of (3,5-dimethoxy 4-hydroxy) fenylkern voorstelt, of •p * een amidogroep met de struktuur C0-N/'^2 waarin het samenstel (R2, R^) een der volgende beteke- 25 nissen bezit (Η,Η), (H, alkyl met 1 tot 4 kool- stofatomen), (alkyl met 1 tot 4 koolstofatomen, alkyl met 1 tot 4 koolstofatomen), (H, cyclo- alkyl met 3 tot 6 koolstofatomen), (H, cyclo- alkyl met 3 tot 6 koolstofatomen methyl), (H, 30 fenyl), (H, fenyl gesubstitueerd met 1 of meer halogeenatomen), (H, fenyl gesubstitueerd door een of meer methyl of methoxygroepen), (H, benzyl), (H, benzyl gesubstitueerd door 1 of meer halogeenatomen), (H, benzyl gesubstitueerd door 35 een of meer methyl of methoxygroepen) (CH^, 8300720 I f tg i - 2 - benzyl), (CH^ t benzyl gesubstitueerd door een of meer halogeenatomen), (CH3, benzyl gesubstitueerd door een of meer methyl of methoxy-groepen), (H, allyl), (H, propargyl), terwijl 5 R2r eveneeis tezamen met het stikstofatoom waaraan zij gebonden zijn een pyrrolidino-, piperidino-, morfoline-, of tropaangroep met formule 13 kunnen vormen; waarbij Ar dan een fenylkern, een fenylkern gesubstitueerd door 10 een of meer halogeenatomen, door een of meer methoxygroepen, door een methylgroep, door een trifluormethylgroep of door een of twee hydro-xylgroepen, een 1,3-benzodioxolylgroep met formule 14, een 1,4-benzodioxannylgroep met formule 15 15, een (5-methoxy) 1,4-benzodioxannylgroep, met formule 16, een naftylgroep, een (3-methoxy 4-hydroxy) fenylgroep of (3-hydroxy 4-methoxy) fenylgroep, een (3,5-dimethoxy 4-hydroxy) fenyl groep of een 4-alkoxy [met 2 tot 4 kool stof at omen]-20 3,5-dimethoxy) fenylgroep; waarbij A echter niet de volgende betekenissen kan aannemen: C0NHCH3, CONHCjHg, CONHC-^n, CONHC.3H.7iso, CON. (CH3)2, CON(C2H5)2, CON(C3H7n)2, CON (C^iso ) 2 , de groep met formule 17, met formule 18, met 25 formule 19, CONHCgH,-, de groep.met formule 20 of de groep met formule 21 wanneer Ar de (3,4,5-trimethoxy) fenyl- of (3,5-dimethoxy 4-hydroxy-fenyl) groep voorstelt; - ofwel de propeen-l-yleen keten met transconfiguratie 30 (-^Η=<?Η-^Η2), gebonden aan Ar door zijn koolstof atoom op plaats 1 of propyleenketen (-CH2-CH2-CH2~) in welk geval A een carboxamidogroep, een N-isopropylcarboxamidogroep, een pyrrolidinocarbonyl- of piperidinocarbonylgroep voorstelt, waarbij Ar dan een (3,4,5-trimethoxy) fenyl- of 35 (3,4-methyleendioxy) fenylgroep voorstelt.
Van de verbindingen met formule 1 kunnen als bijzonder belangwekkende verbindingen genoemd worden die waarin 8300720 . L * - 3 - 3 2 1 X de trans -CH=CH~CO- keten voorste It en A een COOH-groep of een CONR2R3groep en in het bijzonder de groep CONH2, de groep met formule 22, de groep met formule 23 of de groep met formule 24.
5 De uitvinding heeft eveneens betrekking op de additiezouten met minerale of organische zuren (zoals zoutzuur en maleinezuur of oxaalzuur bijvoorbeeld) van de verbindingen met formule 1, alsmede op de hydraten van deze zouten en verbindingen.
10 De werkwijze volgens de uitvinding voor de berei dingen met formule 1 waa^in^X de propeen-2 ylenon-1 groep met transconfiguratie (-CH=CH-C0~) of propylenon-1 keten (CH2--CH2~C0-)/ en Ar dezelfde betekenissen bezit als in formule 1 met uitzondering van de betekenissen p-hydroxy-15 fenyl, (4-hydroxy 3-methoxy)fenyl, (4-hydroxy 3,5 dimethoxy) fenyl, (3,4-dihydroxy)fenyl en (3-hydroxy 4-methoxy)fenyl bestaat uit: A) in geval dat A een etoxycarbonyl of amidogroep met de struktuur CON^ ^2 voorstelt waarin R2 en R3 dezelfde beteke-20 nissen bezitten lis in formule 1: - het vormen van het chloride van de zuren met de formule 2 of formule 2a waarin Ar^ dezelfde betekenissen bezitten als Ar in formule 1, met uitzondering van de betekenissen p-hydroxyfenyl, (4-hydroxy 3-methoxy) 25 fenyl, (4-hydroxy 3,5-dimethoxy)fenyl, (3-hydroxy 4-methoxy)fenyl en (3,4-dihydroxy)fenyl, bij voorkeur door inwerking van thionylchloride, in oplossing in tolueen, benzeen, chloroform, methyleenchloride of tetrahydrofuran op deze zuren, en het vervolgens 30 condenseren van de aldus verkregen zuurchloriden met geschikte verbindingen met formule 3, waarin A^ een etDxycarbonylgroep of amidogroep met de struktuur C0N^^2 voorstelt waarin R2 en R^ dezelfde betekenissen belitten als in formule 1, 35 - het condenseren van de zuren met formule 2 of 2a en de geschikte verbindingen met formule 3 volgens de zogenaamde "BOISSONNAS" reaktie, of 8300720 - 4 - - behandelen van zuren met formule 2 of 2a met het trifenylfosfine-tetrachloor>koolstofcomplex, bij voorkeur in oplossing in dioxan, en het vervolgens condenseren van de verkregen tussenprodukten met 5 de aminen met formule 3; B) in het geval A een eitoxycarbonyl, carboxyl of amidogroep met de struktuur C0Nt^R2 voorstelt, waarin R2 en dezelfde betekenissen bezitten als infbrmulel, door condenseren van de verbindingen met formule 4, of 4a, waarin Ar^ dezelfde 10 betekenissen bezit als in formule 2, met de geschikte verbindingen met formule 5, waarin A« een carboxylgroep, ethoxy-

carbonylgroep, of 'amidogroep met de struktuur CON -£_.2 voor- R3 stelt waarin R2 en R^ dezelfde betekenissen bezitten als m formule 1, waarbij deze condensatie bij voorkeur uitgevoerd 15 wordt in aprotisch milieu,(acetonitril, aceton, tetra-hydrofuran, dimethylformamide of tolueen bijvoorbeeld) en in tegenwoordigheid van kalïumcarbonaat. Wat de verbindingen met de formule 4 en formule 4a betreft, deze worden respek-tievelijk verkregen door condensatie in aijnzuur milieu van 20 de zuurchloriden met de verbindingen met formule 2 en formule 2a met het chloorhydraat van piperazine met formule 6, C) in het geval dat A een amidogroep met een struktuur CON<^R2 voorstelt waarin R2 en R^ dezelfde betekenissen 25 bezittln als in formule 1: - ofwel condenseren volgens de "BOISSONNAS" methode van de zuren met formule 7 of formule 7a waarin Ar^ dezelfde betekenissen bezitten als in formule 2 met de geschikte aminen met formule 8, waarin R2 en R3 30 dezelfde betekenissen bezitten als in formule 1, ter wijl de verbindingen met formule 7 en formule 7a op zichzelf verkregen worden volgens de werkomstandigheden beschreven onder voorgaand punt B, - condenseren van de verbindingen met formule 9 of 9a 35 waarin Ar^ dezelfde betekenissen bezitten als in formule 2, met geschikte verbindingen met formule 8, in tegenwoordigheid van ammoniumchloride, waarbij de 8300720 * * * - 5 - verbindingen met formule 9 of 9a op zichzelf bereid worden volgens de werkomstandigheden beschreven onder de voorgaande punten A en B.-
De werkwijze volgens de uitvinding voor de berei-5 ding van de verbindingen met formule 1/ waarin X de trans-propeen-2 ylenon-1 keten of de propylenon-1 keten voorstelt,· A een eiboxy carbonyl groep of amidogroep met de struktuur CONR^R^ waarin R2 en R^ dezelfde betekenissen bezitten als in formule 1 en Ar een parahydroxyfenylkern, (4-hydroxy 10 3-methoxy)fenylkern, (3-hydroxy 4-methoxy)fenylkern, (4-. hydroxy 3,5-dimethoxy)fenylkern of (3,4-dihydroxy)fenylkern, bestaat uit het condenseren in tegenwoordigheid van dicyclo-hexylcarbodiimide (DCCX) en bij voorkeur in oplossing in chloroform, van de zuren met formule 2b of 2c waarin Ar2 15 een 4-hydroxy fenylkern,(4-hydroxy 3-methoxy)fenylkern, (3-hydroxy 4-methoxy) fenylkern, (4-hydroxy 3,5-dimethoxy) fenylkern of (3,4-dihydroxy) fenylkern voorstelt met de verbindingen met formule 3.
De werkwijze volgens de uitvinding voor de berei-20 dig van de verbindingen met formule 1 waarin X de trans propeen-2-ylenon-l keten of propylenon-1 keten voorstelt, A de carboxylgroep en Ar een 4-hydroxy fenylkern of (4-hydroxy 3,5-dimethoxy) fenylkern, bestaat uit het hydroly-seren, bij voorkeur met waterige natronloog van de verbindin-25 gen van formule 1 met de bijzondere struktuur met formule la of formule lb waarin Armeen parahydroxyfenylgroep of (4-hydroxy 3,5-dimethoxy)fenylgroep voorstelt.
De verbindingen met formule la en lb worden op zichzelf bereid volgens de hierbovengenoemde werkwijze uit-30 gaande van de verbindingen met formule 2b of 2c en van de verbindingen met formule 3. De werkwijze volgens de uitvinding voor de bereiding van de verbindingen met formule 1, waarin A de hydroxymethylgroep voorstelt, bestaat uit het condenseren van de verbindingen met formule 4 of 4a waarin 35 Ar^ de (3,4,5,-trimethoxy)fenylkern voorstelt met ethyleen-oxyde, bij voorkeur in oplossing in ethanol.
De werkwijze volgens de uitvinding ter bereiding van de verbindingen met formule 1, waarin A de carbohydroxaP
8300720 - 6- ‘ f ‘ * groep (-CONHOH) voorstelt, bestaat uit het condenseren van hydroxylamine, bij voorkeur in de vorm van/0¾oorhy-draat en in tegenwoordigheid van natriummethylaat met de verbindingen met formule 1 met bijzondere struktuur met 5 formule lc of formule ld.
De werkwijze volgens de uitvinding voor de bereiding van de verbindingen met formule 1 waarin X de trans- 12 3 propeen-1 yleen (-CH=CH-CH2~) voorstelt: - condenseren van aldehyden met formule 10, waarin Ar 10 een (3,4,5-trimethoxy) fenyl of (3,4-methyleendioxy) fenylgroep voorstelt met de verbindingen met formule 3, waarin A^ een carboxamidogroep, N-isopropylcarboxa-midogroep, pyrrolidino-carbonylgroep of piperidino-carbonylgroep voorstelt, gevolgd door reduceren van de 15 aldus verkregen tussenprodukten bij voorkeur met natriumboriumhydride in oplossing in methanol, waarbij de verbindingen met formule 10 ofwel bekend zijn ofwel bereid worden volgens de werkwijze beschreven in Indian J. Chem. Vol. 13, blz. 10-15 (1975).
20 - ofwel condenseren van halogeenverbindingen met formule 11 waarin Ar^ dezelfdjèbetekenissen bezit als in formule 10 en hal een halogeen atoom voorstelt, met de verbindingen met formule 3 waarin A^ een carboxamidogroep, n-isopropylcarboxamidogroep, pyrrolidinocarbonyl- of 2.5 piperidinocarbonylgroep voor stelt, bij voorkeur in tolueen en in tegenwoordigheid van een base zoals triethylamine, waarbij de verbindingen met formule 11 bekend zijn of bereid worden volgens de werkwijze beschreven in Duits octrooischrift 2.252.080.
30 De werkwijze volgens de uitvinding voor de berei ding van de verbindingen met formule 1, waarin X een propy-leenketen voorstelt bestaat uit het halogeneren van de verbindingen met formule 12, waarin Ar4 dezelfde betekenissen bezit als in formule 10, bij voorkeur met fosfortribromide, 35 en het vervolgens laten reageren van het verkregen halogeen-tussenprodukt met de verbindingen met formule 3 waarin A^ een carboxamidogroep, een N-isopropylcarboxamidogroep, een pyrrolidinocarbonylgroep of piperidinocarbonylgroep voor- 8300720 i * * - 7 - stelt.
De verbindingen met formule 12 zijn op hun beurt bekend of worden bereid door reductie met AlLiH^ in tetra-hydrofuran van overeenkomstige ethyl cinnamoaten, die op 5 zichzelf bekend zijn of bereid worden volgens de werkwijze beschreven in Indian J. Chem. Vol. 13, blz. 10-15 (1975).
De verbindingen met formule 1 waarin X de propyle-non-1 (-C^-C^-CO) groep voorstelt, A een ethoxycarbonyl-groep of de struktuur CONI^R^ waarin en dezelfde bete-10 kenissen bezitten als in formule 1 en Ar dezelfde betekenissen bezit als Ar^ in formule 2, kunnen eveneens verkregen worden door katalytische reductie, bij voorkeur in tegenwoordigheid van 10%'s palladium op kool en in alcoholisch milieu, van de verbindingen met formule 1 met de bijzondere 15 struktuurformule le waarin Ar^ dezelfde betekenis bezit als in formule 4 en Ar^ dezelfde betekenissen bezit als in formule 3.
De verbindingen met formule 1 kunnen volgens gebruikelijke werkwijzen omgezet worden in zouten. De omzet-20 ting in het zout kan bijvoorbeeld verkregen worden door inwerking op deze verbindingen van een mineraalzuur zoals zoutzuur of van een organisch zuur zoals oxaalzuur of malei-nezuur, waarbij deze bewerking bij voorkeur uitgevoerd wordt in een oplosmiddel of een mengsel van oplosmiddelen zoals 25 aceton, ethanol of water.
De volgende voorbeelden lichten de uitvinding toe. VOORBEELD 1 : trans 1-cinnamoyl 4-pyrrolidinocarbonylmethyl piperazine, chloorhydraat (formule 1),
Codenummer : 36 30 Aan een oplossing van 38,5 gram trans cinnamoyl- chloride in 200 ,ml tolueen, voegt men langzaam, bij kamertemperatuur, 22,1 g 4-pyrrolidinocarbonylmethyl piperazine (formule 3) toe. Vervolgens roert men 4 uren bij kamertemperatuur, filtreert het neerslag af en herkristalliseert in 35 alcohol. Men isoleert op deze wijze 15 g van de gewenste verbinding (rendement ^ 42 %).
Volgens dezelfde werkwijze, doch uitgaande van 8300720 - 8 - r f t i overeenkomstige reaktiemiddelen, verkrijgt men de verbindingen met formule 1 en codenummers 3 tot 6, 10 tot 16, 18 tot 24, 27 tot 34, 37 tot 57 en 78 tot 88 die weergegeven zijn in de navolgende tabel A.
5 VOORBEELD 2 : trans l-(3,4-methyleendioxy) cinnamoyl 4-pyrrolidinocarbonyl piperazine, chloorhydraat (formule 1)

Aan een tot 5°C afgekoelde oplossing van 19,2 g 10 (3,4-methyleendioxy) kaneelzuur, transvorm (formule 2) in 200 ml dimethylformamide, voegt men 14 ml triethyl amine toe, en vervolgens 9,6 ml ethylchloorformiaat. Men laat 30 minuten bij 5°C staan, waarna men langzaam 39,4 g 4-pyrrolidino-carbonylmethyl piperazine (formule 3) toevoegt. Men laat 15 vervolgens 12 uren bij kamertemperatuur, waarna men het gevormde neerslag affiltreert. Men verkrijgt op deze wijze 13 g (rendement ^ 36%) van de gewenste verbinding.
Volgens dezelfde werkwijze doch uitgaande van overeenkomstige reaktiemiddelen, verkrijgt men de verbindin-20 gen met formule 1 met de codenummers: 3 tot 6, 10 tot 16, 18 tot 24, 27 tot 34, 36 en 38 tot 57 die. weergegeven zijn in tabel A.
Volgens deze werkwijze, doch uitgaande van de verbindingen met formule 7 of 7a en van de aminen met formule 25 8 verkrijgt men de verbindingen met formule 1 die voorkomen in tabel A en de codenummers: 10 tot 16, 18 tot 24, 27 tot 34, 36 tot 57 en 78 tot 88 bezitten.
VOORBEELD 3 : trans 1-(2,4,6-trimethoxy) cinnamoyl 4-N-iso-propylaminocarbonylmethyl piperazine, oxa-30 laat (formule 1).

Men houdt een oplossing van 26 g trifenylfosfine in 100 ml tetrachloor koolstof en 300 ml dioxan 1 uur op 70°C. Vervolgens koelt men af tot 5 tot 10°C, voegt 14,9 35 g trans (2,4,6-trimethoxy) kaneelzuur toe, laat 30 minuten bij 5 tot 10°C, voegt vervolgens 55,5 g 4-N-isopropylamino-carbonylmethyl piperazine (formule 3) toe en verwarmt de 8300720 »| ' ^ - 9 - oplossing 2 uren op 70°C. Vervolgens filtreert men, dampt het filtraat in, neemt het residu op in chloroform, waarna men extraheert met een verdunde oxaalzuuroplossing, wast e de waterfase met ether, neutraliseert met natriumbicarbonaat, 5 filtreert het gevormde neerslag af (base, smeltpunt : 93°C), lost het op in aceton, voegt een acetonoplossing van oxaalzuur toe, filtreert het gevormde neerslag af en herkristalli-seert het in n-butanon. Men isoleert op deze wijze 9 g (rendement ^ 36%) van de gewenste verbinding.
Volgens dezelfde werkwijze, doch uitgaande van overeenkomstige reaktiemiddelen, verkrijgt men de verbindingen met formule 1 met de codenummers : 3 tot 6, 10 tot 15, 18 tot 24, 27 tot 34, 36 tot 57 en 78 tot 88 die weergegeven zijn in tabel A.
15 VOORBEELD 4 : trans l-(3,4,5-trimethoxy) cinnamoyl 4-amino carbonylmethylpiperazine (formule 1)

Men houdt een suspensie van 15,3 g trans (3,4,5-trimethoxy) cinnamoyl piperazine (formule 4), 9,3 g chloor- 20 aceetamide en 20,7 g kaliumcarbonaat in 200 ml ace toni tril 3 uren onder terugvloeikoeling. Vervolgens filtreert men, dampt het filtraat in en herkristalliseert het residu in ethanol. Men isoleert op deze wijze 13 g (rendement ^ 72%) van de gewenste verbinding.
25 Volgens dezelfde werkwijze, doch uitgaande van overeenkomstige reaktiemiddelen verkrijgt men de verbindingen met formule 1 die voorkomen in tabel A met de codenummers : 3 tot 10, 12 tot 16, 18 tot 24, 27 tot 34, 36 tot 57 en 78 tot 88.
50 VOORBEELD 5 : trans l-(3,4,5-trimethoxy) cinnamoyl 4-N-cyclo- pentylaminocarbonylmethyl piperazine (formule 1)

Men houdt een mengsel van 19,6 g trans 1-(3,4,5- 55 trimethoxy) cinnamoyl 4-ethoxycarbonylmethyl piperazine (formule 9)^42 g cyclopentylamine (formule 8) en 2 g ammo-niumchloride, 7 uren op 90 tot 100°C. Vervolgens dampt men 8300720 V « * -lO- de overmaat amine in vacuo af en neemt het residu op in water, extraheert met chloform, wast met water, droogt boven natriumsulfaat, filtreert en dampt het filtraat in. Het verkregen produkt wordt gekristalliseerd in ether en her-5 kristalliseerèl in ethylacetaat en vervolgens in isopropyl- alcohol. Men verkrijgt op deze wijze 2,6 g van de gewenste verbinding (rendement ^12 %).
Volgens dezelfde werkwijze doch uitgaande van overeenkomstige reaktiemiddelen, verkrijgt men de verbin-10 dingen met formule 1 die voorkomen in tabel A met de codenummers : 10 tot 16, 18 tot 24, 27 tot 34, 36 tot 57 en 78 tot 88.
VOORBEELD 6 : trans l-(4-hydroxy 3siethoxy) cinnamoyl 4-N-isopropylaminocarbonylmethyl piperazine 15 (formule 1)

Aan een tot 0°C afgekoelde oplossing van 9,7 g trans (4-hydroxy 3-methoxy) kaneelzuur (formule 2b) en 9,2 g N-isopropylaminocarbonylmethyl piperazine (formule 3) 20 in 250 ml dioxan voegt men 10,3 g DCCI toe. Vervolgens laat men 70 uren bij kamertemperatuur,filtreert, dampt het filtraat in en chromatografeert het residu op een siliciumoxyde kolom. Door elutie met een mengsel vanmethyleenchloride (95%) - methanol (5%) verkrijgt men 6 g (rendement =^33%) 25 van het gewenste produkt dat men herkristalliseert in iso-propylether.
Volgens dezelfde werkwijze doch uitgaande van overeenkomstige reaktiemiddelen verkrijgt men de verbinding met formule 1 die weergegeven is in tabel A met codenummer 30 25,en 74 tot 77 alsmede de verbinding met formule la: trans-para1-hydroxycinnamoyl 4-ethoxycarbonmethyl piperazine, chloorhydraat met de volgende eigenschappen: . Men behandelt een oplossing van 3/5 g 1-para hydroxycinnamoyl 4-ethoxycarboi4m^iihyl piperazine (formule la) in 30 ml waterige IN natronloog 90 minuten op 60°C/ 10 zuurt vervolgens aan tot pH 6/5 met behulp van waterig IN zoutzuur/, filtreert het verkregen neerslag af en wast het filter met methanol. Men verkrijgt 1/6 g (rendement ^ 55%) van de gewenste verbinding.
VOORBEELD 8 : trans l-(3,4,5-trimethoxy) cinnamoyl 4-(2-15 hydroxy)ethyl piperazine, maleaat (formule 1).

Men houdt een oplossing van 15 g trans -(3,4,5-trimethoxy) cinnamoyl piperazine (formule 4) en 2 ml pipera-dine in 40 ml alcohol. Vervolgens leidt men een ethyleen-20 oxyde gasstroom door (tot absorptie van 0,05 mol) en laat 1 uur bij 60°C, en vervolgens nog 12 uren bij kamertemperatuur staan. Vervolgens dampt men in en chromatografeert het residu op een siliciumoxydekolom. Men elueert met een methyleen-chloride (95%) - methanol (5%) en lost de verkregen zuivere 25 verbinding op in aceton. Men voegt een acetonoplossing toe van maleinezuur en filtreert het gevormde neerslag af. Men isoleert op deze wijze de gewenste verbinding met een rendement van 76%.
VOORBEELD 9 : trans [4-(3,4,5-trimethoxy cinnamoyl) 1-30 piperazinyl·] aceethydroxaminezuur (formule 1).
Aan een tot 5°C gekoelde oplossing van 13,8 natrium in 300 ml methanol voegt men een oplossing toe van 20,8 hydroxylamine chloorhydraat in 200 ml methanol. Men filtreert en aan het filtraat voegt men bij kamertemperatuur 68,8 g 35 .athylester van trans [4-(3,4,5-trimethoxy cinnamoyl) 1-piperazinyl] azijnzuur toe. Na 20 uren reaktie, dampt men het oplosmiddel af, neemt het residu op in water, wast met 8300720 - 12 - ether, voegt vervolgens een oplossing toe van 18 g azijnzuur in 30 ml water. Men filtreert het verkregen neerslag af, wasthet met water, droogt het in vacuo en herkristalliseert het met 280 ml 70%'s aceton. Men isoleert 37 g van de ge-5 wenste verbinding (rendement : ^ 47%) VOORBEELD 10 : trans ^-^3,.4,5-trimethoxy) cinnamyl 4-N-isopropylaminocarbonylmethyl piperazine (formule 1)
Codenummer : 58 10 ' Men houdt een mengsel van 8,8 g trans (3,4,5- trimethoxyj cinnamaldehyde (formule 10) en 7,4 g 4-N-iso-propylaminocarbonylmethyl piperazine) (formule 3) in 300 ml methanol en voegt vervolgens langzaam 6 g natrium-borium-hydride toe. Na 1 uur dampt men het oplosmiddel af en neemt 15 het residu op in chloroform, wast het met water, droogt boven natriumsulfaat, filtreert en dampt het filtraat in.
Men herkristalliseert het verkregen produkt in isopropyl-ether en vervolgens in ethylacetaat. Men isoleert op deze wijze 2 g (rendement ^ 16%) van de gewenste verbinding.
20 VOORBEELD 11 : trans 1-(3,4-methyleendioxy) cinnamyl 4- pyrrolidinocarbonylmethyl piperazine, chloorhydraat (formule 1).

Aan een oplossing van 2,6 g 4-pyrrolidinocarbo-25 nylmethyl piperazine (formule 3) in 16 ml tolueen en 0,9 ml triethylamine voegt men langzaam bij kamertemperatuur 1,6 g trans 3-(3,4-methyleendioxy)fenyl 1-broom propeen-2 (formule 9) toe. Vervolgens laat men 12 uren bij kamertemperatuur staan, filtreert, dampt het filtraat in, kristalli-30 seert het residu in ether. Men verkrijgt 1,5 g (rendement - 65%) van de verbinding die overeenkomt met de gewenste verbindingan de vorm van de base (smeltpunt : 110°C).
Deze verbinding wordt vervolgens opgelost in ethanol, en men voegt een oplossing toe van ^6,5 N zoutzuur in ethanol 35 en filtreert het verkregen neerslag af wat overeenkomt met de gewenste verbinding.
VOORBEELD 12 : 1-[3-(3,4,5-trimethoxy)fenyl propyl] 4- 8300720 - 13 - N isopropylaminocarbonylmethyl piperazine dioxalaat (formule 1).

Aan een oplossing van 24 g 3-{3,4,5-trimethoxy) 5 fenyl propanol-1 (formule 12) in 200 ml chloroform, voegt men langzaam 57,2 g fosfortribromide (PBr^V^^^rvolgens roert men 1 uur bij kamertemperatuur, verdunt met water, decanteert de organische fase, wast haar met behulp van een verzadigde natriumbicarbonaatoplossing, droogt boven 10 natriumsulfaat, filtreert en dampt het filtraat in. 15 Vervolgens filtreert men het onoplosbaar af, dampt het filtraat in, lost het residu op in chloroform, wast met water, droogt boven natriumsulfaat, filtreert, dampt het filtraat in, lost het residu op in chloroform, wast met water, droogt boven natriumsulfaat, filtreert, dampt het 20 filtraat in, lost het residu op in ethanol, voegt een etha-— nolische oplossing van oxaalzuur toe, filtreert het verkregen neerslag af en herkristalliseert in absolute alcohol.
Men isoleert op deze wijze 4 g (rendement * 20%) van de gewenste verbinding. De verbinding met formule 1 en code-25 nummer 63 als weergegeven in tabel A kan eveneens bereid worden onder toepassing van deze zelfde werkwijze doch uitgaande van overeenkomstige reaktiemiddelen.
VOORBEELD 13 : 3-(3,4,5-trimethoxy)fenyl l-[4-(3,4,5 N-trimethoxy) benzylaminocarbonylmethyl 1-30 piperazinyl] propanon (formule 1).

De verbindingen met formule 1 en hun zouten en hydraten zij[n onderzocht bij laboratoriumdieren en blijken farmacologische werking te bezitten, in het bijzonder stimulerende, beschermende en/of corrigerende werkingen ten 5 opzichte van de cerebrale functies.
Deze werkingen zijn in het bijzonder aangetoond nines i s olie door de/proef van het behoud van het exploratievermogen uitgevoerd volgens het volgende protocol.
Men meet in een apparaat ACTIMETRE APELAB [BOISSIER 10 en SIMON, Arch. Inter. Phrmacodyn. 158, 212, (1965)], het exploratievermogen gedurende 5 minuten van mannelijke SWISS-WEBSTER muizen, waarna de dieren hun intraperitoneale (of orale) injectie ontvangen van de verbindingen met formule 1 of hun zouten of een fysologische zoutoplossing. Na -een 15 week bepaalt men opnieuw het exploratievermogen van de behandelde dieren en het effect op het behoud wordt gemeten via een gewenning , dat wil zeggen een significante statiiiische vermindering (t van STUDENT bij- gelijke groepen)van het exploratievermogen.

De voorgaande resultaten tonen dat de verbindingen volgens de uitvinding een opmerkelijke farmacologische wer- 2 0 — - king en een geringe giftigheid bezitten. Deze verbindingen vinden derhalve hun toepassing in de geneeskunde. Zij kunnen in het bijzonder gebruikt worden voor het stimuleren van het intellectuele vermogen van de normale persoon voor het behouden van de cerebrale funkties bij de ouder wordende 25 persoon en voor het behandelen van stoornissen van de
Werkzaamheid en van het memoriseren, volgend op ver- ;S Chede 1 - schillende ziekten, in het bij zonder/traumatismen, cerebrale commoties of acute of subacute cerebrovasculaire stoornissen.
De uitvinding heeft eveneens betrekking op farma-30 ceutische samenstellingen die als werkzaam bestanddeel tenminste een van de verbindingen met formule 1 of hun zouten en hydraten bevatten, waarbij deze verbindingen in het bijzonder geformuleerd kunnen zijn met hun oog op orale of parenterale toediening. Zij kunnen derhalve bijvoorbeeld 35 oraal toegediend worden m de vorm van beklede tabletten, pillen, tabletten of in de vorm van drinkbare oplossingen in doses van 2,5 g per dag in meerdere keren per dag (tot 8300720 - 37 - 6 keer) of parenteraal in de vorm van injecteerbare ampullen die tot 1 gram werkzaam bestanddeel (1 tot 3 dagelijkse injecties) bevatten.
Bij orale toediening in de vorm van beklede 5 tabletten, pillen en tabletten, kunnen deze laatstgenoemde met voordeel een drager (zoals celluloseverbindingen, fenyl-polymeren of gommen) bevatten die een modulering van het vrijmaken van het werkzame bestanddeel mogelijk maken. De drinkbare oplossingen zijn met voordeel waterige oplossin-10 gen of waterige suspensies (drager = water) of gedeeltelijk waterige oplossingen of suspensies (drager = water + alcohol, water + glycerine of water + propyleenglycol). Tenslotte kan bij parenterale toediening het werkzame bestanddeel ingespoten worden in de vorm van injecteerbare suspensies 15 of oplossingen uit lyophilisaten die dit werkzame -bestanddeel bevatten.

Claims (16)
1. Nieuwe piperazineverbindingen die beantwoorden aan de algemene formule 1 van het formuleblad waarin X voorstelt: - ofwel eenpropeen-ylenon-1 keten met transconfiguratie, 3 2 1 (-CH=CH-CO-) of propyl enon keten (-CE^-CE^-CO-) 5 die gebonden zijn aan Ar door zijn koolstofatoom op plaats 3, in welk geval A voorstelt: . Λ * een carbohydroxa/. .(-CONHOH) groep of hydroxymethyl (-CE^OH) groep waarbij Ar dan de (3,4,5-trimethoxy_fenylkern voorstelt, 10. een e^oxycarbonylgroep (COOEt), waarbij Ar dan de para-fluorfenyl-, (3,4,5-dichloor)fenyl-(2,3,4-trimethoxy)fenyl-, of (2,4,6-trimethoxy) fenylkern voorstelt, * een carboxylgroep (COOH), waarbij Ar dan de 15 parachloorfenyl-, para-hydroxyfenyl-, (3,4- methyleendioxy)fenyl- of (3,5-dimethoxy 4-hy-droxy) fenylkern voorstelt, of R * een amidogroep met de struktuur CO-N' 2 waarin het samenstel (Ε^,ϊΙ^) een der volgende beteke- 20 nissen bezit (Η,Η), (H, alkyl met 1 tot 4 kool- * « stofatomen,(alkyl met 1 tot 4 koolstofatomen, alkyl met 1 tot 4 koolstofatomen), (H, cyclo- alkyl met 3 tot 6 koolstofatomen), (H, cyclo- alkyl met 3 tot 6 koolstofatomen methyl), (H, 25 fenyl), (H, fenyl gesubstitueerd met 1 of meer halogeenatomen), (H, fenyl gesubstitueerd door een of meer methyl of methoxygroepen), (H, benzyl, (H, benzyl gesubstitueerd door 1 of meer halogeenatomen), (H, benzyl gesubstitueerd door 30 een of meer methyl of methoxygroepen) (CH^, benzyl), (CH^, benzyl gesubstitueerd door een of meer halogeenatomen), (CH^, benzyl gesubstitueerd door een of meer methyl of methoxygroepen), (H, allyl), (H, propargyl), terwijl 35 r^, r^ eveneens tezamen met het stikstofatoom waaraan zij gebonden zijn een pyrrolidino-, 8300720 - 39 - piperidino-, morfoline- of tropaangroep met formule 13 kunnen vormen; waarbij Ar dan een fenylkern, een fenylkern gesubstitueerd door een of meer halogeenatomen, door een of meer 5 methoxygroepen, door een methylgroep, door een trifluormethylgroep of door een of twee hydro-xylgroepen, een 1,3-benzodioxolylgroep met formule 14, een 1,4-benzodioxannylgroep met formule 15, een (5-methoxy) 1,4-benzodioxannylgroep, 10 met formule 16, een naftylgroep, een (3-methoxy 4-hydroxy) fenylgroep of (3-hydroxy 4-methoxy) fenylgroep, een (3,5-dimethoxy 4-hydroxy) fenyl groep of een 4- alkoxy [met 2 tot 4 koolstofato-men]- 3,5-dimethoxy) fenylgroep; waarbij A echter 15 niet de volgende betekenissen kan aannemen: C0NH2 C0NHCH3, C0NHC2H5, C0NHC3H7n, CONHC^iso, CON (CH3)2, C0N(C2H5)2, CON(C3H7n)2, CONiC^iso)2, de groep met formule 17, met formule 18, met formule 19, CONHCgH^, de groep met formule 20 20 of 3e groep met formule 21 wanneer Ar de (3,4,5- trimethoxy) fenyl- of (3,5-dimethoxy 4-hydroxy-fenyl) groep voorstelt; en = CON(C2H5)2 wanneer Ar de 2-hydroxy fenylgroep en X = CH2CH2C0 groep voorstelt, 25. ofwel 3e propeen-l-yleen keten met transconfiguratie (-CH=CH-CH2), gebonden aan Ar door zijn koolstofatoom op plaats 1 of propyleenketen (-CH2-CH2-CH2~) in welk geval A een carboxamidogroep, een N-isopropylcarboxamidogroep, een pyrrolidinogroep, een N-isopropylcarboxamidogroep, 30 een pyrrolidinocarbonyl- of piperidinocarbonylgroep voorstelt, waarbij Ar dan een(3,4,5-trimethoxy) fenyl- of (3,4-methy-leendioxy) fenylgroep voorstelt, waarbij echter A geen pyrrolidinocarbonylgroep kan voorstellen als Ar een (3,4,5— trimethoxy) fenylgroep voorstelt, alsmede een zuur additie-35 zout en de hydraten van deze verbindingen en zouten.

3. Verbinding volgens conclusie 2, m e t het k :e n m e r k, dat het samenstel (Ar, A) een van de volgende betekenissen bezit met de formules 118 tot en met 178.
54. Verbindingen volgens conclusie 1, m e t het kenmerk, dat X de propeen-2-ylonen-l groep met transconfiguratie voorstelt, A de carboxylgroep en Ar een der groepen voorstelt met de formules 30, 31 of 32.
5. Geneesmiddelen met stimulerende, beschermende 10 en/of corrigerende werkingen van de cerebrale funkties, m et het kenmerk, dat zij bestaan uit verbindingen volgens conclusie 1 tot 4.
6. Farmaceutisch preparaat, met het kenmerk, dat zij als werkzaam bestanddeel tenminste een van 15 de geneesmiddelen volgens conclusie 5 tezamen met een farmaceutisch verdraagbare drager bevat.

Werkwijze ter bereiding van verbindingen met formule 1 en van hun zuuradditiezouten waarin X de propeen-2- 3 2 1 ylonen-1 groep^met^transconfiguratie (-CH=CH-CO) of propyle-20 non-1 groep (-C^^C^-CO) voorstelt, Ar deze-lfde betekenissen bezit als in conclusie 1, met uitzondering van de 4- ~ hydroxyfenyl-, (4-hydroxy 3-methoxy) fenyl-, (3-hydroxy 4-methoxy)fenyl-, (4-hydroxy 3,5-dimethoxy)fenyl-, en 3,4-dihydroxyfenylgroep en A een ethoxycarbonyl of amidogroep 25 met de struktuur CONR^R^ waarin R£ en R^ dezelfde betekenissen bezitten als in conclusie 1, m e t het kenmerk, dat deze bestaat uit (a) condenseren van zuren met formule 2 of 2a waarin Ar^ dezelfde betekenissen bezit als Ar in conclusie 1, met uitzondering van de 4-hydroxyfenyl-, (4-30 hydroxy 3-methoxy)fenyl-, (3-hydroxy 4-methoxy)fenyl-, (4-hydroxy 3,5-dimethoxy)fenyl- en 3,4-dihydroxyfenylgroep, respektievelijk met de geschikte verbindingen met formule 3 waarin: A^ een ethoxycarbonyl of amidogroep met de struk-35 tuur CONR2R2 voorstelt,, waarin R^ en R^ dezelfde betekenissen bezitten als in conclusie 1, waarbij de zuren met formule 2 of 2a toegevoegd worden aan de verbindingen met formule 3, 8300720 » ί β 4 - 41 - ofwel in de vorm van het zuurchloride of gemengd met het trifenylfosfine-tetrachloor koolstofcomplex, ofwel volgens de zogenaamde BOISSONNAS reaktie; eventueel gevolgd door (b) omzetten in zouten van de aldus verkregen verbindingen.
8. Werkwijze ter bereiding van verbindingen met for mule 1 en van hun zuuradditiezouten waarin X de propeen-2- 3 2 1 ylenon-1 groep met transconfiguratie (-CH=CH-CO~) of propylenon-1 groep (CI^-C^-CO) voorstelt, Ar dezelfde betekenissen bezit als in conclusie 1, met uitzondering van de 10 4-hydroxyfenylgroep, (3-hydroxy.4-methoxy)fenyl, (4-hydroxy 3-methoxy)fenyl-, (3,4-dihydroxy)fenyl en (4-hydroxy 3,5-dimethoxy)fenylgroep en A een ethoxycarbonylgroep, carboxyl-groep of amidogroep met de struktuur CONR2R3 waarin R2 en R^ dezelfde betekenissen bezitten als in. conclusie 1, m e t 15 h e t k e n m e r k, dat deze bestaat uit (a) condenseren van de verbindingen met formule 4 of 4a waarin Ar^ dezelfde betekenis bezit als in formule 2, en omschreven in conclusie 7, met de geschikte verbindingen met formule 5, waarin A2 een carboxylgroep, ethoxycarbonylgroep of amidogroep met 20 de struktuur CONR2R3~-voorstelt, waarin R2 en R3 dezelfde betekenissen bezitten als in conclusie 1, in tegenwoordigheid van een base zoals kaliumcarbonaat; eventueel gevolgd door (b) omzetten van de aldus verkregen verbindingen in zouten.
9. Werkwijze ter bereiding van verbindingen met formule 1 en van hun zuur-additiezouten waarin X de propeen- 3 2 1 2- ylenon-l groep met^transconfiguratie (-CH=CH-CO) of propylenon-1 groep (CH2-CH2~CO) voorstelt en Ar dezelfde betekenissen bezit als in conclusie 1, met uitzondering van 30 de 4-hydroxyfenyl,(3-hydroxy 4-methoxy)fenyl, (4-hydroxy 3- methoxy)feny1, (3,4-dihydroxy)feny1 en (4-hydroxy 3,5~ dimethoxy)fenylgroep en A een amidogroep voorstelt met de struktuur CONR2R3 waarin R2 en R^ dezelfde betekenissen bezitten als in conclusie 1, met het kenmerk, 35 dat deze bestaat uit (a) condenseren van aminen met formule 8 waarin R2 en R3 dezelfde betekenissen bezitten als hierboven, ofwel met de zuren met formule 7 of 7a waarin Ar^ de- 8300720 - 42 - * l ♦ *· zelfde betekenis bezit als in formule 2 als omschreven in conclusie 7, volgens de zogenaamde BOISSONNAS methode, ofwel met de verbindingen met formule 9 of 9a waarin Ar^ dezelfde betekenis bezit alsin de formules 7 en 7a, in 5 tegenwoordigheid van ammoniumchloride;en eventueel gevolgd door.(b) omzetten in zouten van de aldus verkregen verbindingen.
10. Werkwijze ter bereiding van verbindingen met for mule 1 en hun zuur additiezouten waarin X de propeen-2-10 ylenon-1 groep met transconfiguratie of propyleno.n-1 groep voorstelt, A een ethoxycarbonylgroep of amidogroep met de struktuur CONR2R3 waarin R2 en R3 dezelfde betekenissen bezitten als in conclusie 1, en Ar een 4-hydroxyfenyl-, (3-hydroxy 4-methoxy)fenyl-, (4-hydroxy 3-methoxy)fenyl-, 15 (3,4-dihydroxy)fenyl-, of(4-hydroxy 3,5-dimethoxy)fenylkern voorstelt, met het kenmerk, dat deze bestaat uit (a) condenseren in tegenwoordigheid van dicyclohexyl carbodiimide (DCCI) van de zuren met formule 2b of 2c waarin Ar2 een 4-hydroxyfenyl-, (3-hydroxy 4-methoxy)fenyl-, (4-20 hydroxy 3-methoxy)fenyl-, (3,4-dihydroxy)fenyl- of (4-hydroxy 3,5-dimethoxy)fenylkern voorstelt met de verbindingen met formule 3 als omschreven in conclusie 7 gevolgd door eventueel (b) omzetten van de aldus verkregen verbindingen in zouten.
11. Werkwijze ter bereiding van verbindingen met formu le 1 en van hun zuur additiezouten waarin X de transpropeen-2-ylenon-l of propylenon-1 groep voorstelt, A de carboxyl-groep en Ar een 4-hydroxyfenyl- of (4-hydroxy 3,5-dimethoxy) fenylkern voorstelt, met het kenmerk, dat deze 30 bestaat uit het hydrolyseren, bij voorkeur met waterige natronloog, van de verbindingen met formule la of lb waarin Ar^ een 4-hydroxyfenyl-, of (4-hydroxy 3,5-dimethoxy) fenyl-groep voorstelt, eventueel gevolgd door (b) omzetten in zouten van de aldus verkregen verbindingen.
12. Werkwijze ter bereiding van verbindingen met formule 1 en van hun zuur additiezouten, waarin A de carbohydroxamine groep voorstelt, met het kenmerk, dat deze be- 8300720 - 43 - staat uit (a) condenseren van hydroxylamine, bij voorkeur in de vorm van zijn chloorhydraat en in tegenwoordigheid van een base >zoals natriummethylaat, met de verbindingen met formule lc of ld eventueel gevolgd door (b) omzetten 5 van de aldus verkregen verbindingen in zouten.
13. Werkwijze ter bereiding van verbindingen met formule 1, en van hun zuur additiezouten waarin X de transpropeen-1 yleengroep voorstelt, met het kenmerk, dat deze bestaat uit (a) condenseren van verbin- 10 dingen met forule 3 waarin A een carboxamidogroep, N-iso-propylcarboxamidogroep, pyrrolidinocarbonyl of piperidino-carbonylgroep voorstelt, ofwel met de aldehyden met formule i 10 waarin Ar^ de (3,4,5-trimethoxy)fenyl- of (3,4-methyleen-dioxy)fenylgroep voorstelt, in welk geval de verkregen ver-15 bindingen vervolgens bij voorkeur met natrium-boriumhydride gereduceerd worden, ofwel met halogeenverbindingen met de formule 11 waarin Ar^ dezelfde betekenis bezit als hierboven en hal een halogeenatoom aangeeft, bij voorkeur in tegenwoordigheid van een base eventueel gevolgd door (b) omzetten 20 van de aldus verkregen verbindingen in zouten.
14. Werkwijze ter bereiding van verbindingen met formule 1 en van hun zuur additiezouten, waarin X de propyleen-groep voorstelt, met het kenmerk, dat deze bestaat uit (a) condenseren van het overeenkomstige halogenide 25 met de alcohol met formule 12, waarin Ar^ dezelfde betekenis bezit als in conclusie 13, met de verbindingen met formule 3 waarin A^ dezelfde betekenis bezit als in conclusie 13, eventueel gevolgd door (b) omzetten van de aldus verkregen verbindingen in zouten.
15. Werkwijze ter bereiding van verbindingen met formule 1 en van hun zuuradditiezouten, waarin X de propyle-non-1 groep voorstelt, A een ethoxycarbonylgroep of amido-groep met de struktuur CONR2R3 waarin R2 en R^ dezelfde betekenissen bezitten als in conclusie 1, en Ar dezelfde be-35 tekenis bezit als in Ar^ in formule 2 zoals omschreven in conclusie 7, met het kenmerk, dat deze bestaat uit (a) katalytisch reduceren van verbindingen met formule 8300720 - 44 - ·* k, * >- le waarin A^ dezelfde betekenissen bezit als in formule 4 als omschreven in conclusie 8 en A^ dezelfde betekenis bezit als in formule 3 als omschreven in conclusie 7, en het vervolgens eventueel (b) omzetten van de aldus 5 verkregen verbindingen in zouten. 8300720 * * ,-, trans-Ar -.
