Cytostatisch werkende farmaceutische preparaten, nieuwe isocyanuurzuurderivaten en werkwijzen ter bereiding ervan
Het is bekend, dat een reeks geallieerde produkten een cytostatische respectievelijk cytotoxische werking ontvouwen. De bekenste verbindingen worden afgeleid van het zogenaamde stikstoflost. Bovendien is ook bekend, ten min-5 ste twee epox3/groepen in het molecuul bevattende verbindingen als cancerostatica toe te passen, Dergelijke verbindingen zijn bijvoorbeeld het 4.4'-bis-^^-epoxypropjdO-di-piperidinyl-Cl .1') en het 1.2-15.16-diepoxy-4.7-10.13-te-traoxohexadecaan. Zeer zeker hebben de laatste verbindingen 10 geen wezenlijke verbetering in de cytostatische behandeling gebracht en worden nauwelijks toegepast. Slechts voor de behandeling van hersentumoren worden zij nog bij tijd en wijle gebruikt. De beperkte oplosbaarheid van de vermelde verbindingen staat ook een bredere toepassing in de weg.
15 Onderwerp van het niet vóórgepubliceerde Duitse Offen- legungsschrift 2.907.3^-9 zijn geneesmiddelpreparaten met cytostatische activiteit, die als farmacologisch werkzame stof triglycidylisocyanuraat (ÏGI) en/of zodanige BGI-deri-vaten bevatten, waarin het waterstofatoom van het koolstof-20 atoom op de 2-plaats van de glycidylgroep door een alkyl-rest met 1 tot 4 koolstofatomen vervangen kan zijn. Verbindingen van dit type onderscheiden zich, doordat de drie stikstofatomen van de isocyanuurzuurring met epoxygroepen bevattende glycidylresten gesubstitueerd zijn, die op de 25 2-plaats ook met een alkylrest met 1 tot 4. koolstofatomen gesubstitueerd kunnen zijn.
De onderhavige uitvinding gaat uit van de vaststelling, dat structuuranaloge verbindingen, die echter aan de isocyanuurzuurring in H-substitutie slechts twee glycidyl-30 groepen en aan het andere IT-atoom geselecteerde substituen-ten bevatten, eveneens een verrassend sterke cytostatische activiteit ontvouwen, die zelfs de werkzaamheid van TGI kan overtreffen.
Onderwerp van de onderhavige uitvinding zijn dienover-35 eenkomstig in een eerste uitvoeringsvorm geneesmiddelpreparaten met cytostatische activiteit, die verbindingen met de algemene formule 1 bevatten, waarin R de volgende bete- β η n * 1 δ 7

kenis kan h.ebben: alkyl, aryl, aralkyl, alkaryl, cycloalkyl, welke resten desgewenst ook van heterocyclische aard, onverzadigd en/of met ten minste een van de volgende substi-tuenten gesubstitueerd zijn kunnen: halogeen, hydroxy, 5 amino, IT-gesubstitueerd amino, mercapto, alkylmercapto, arylmercapto, alkylsulfoxy, arylsulfoxy, alkoxy, aroxy, acyloxy en een heterocyclische rest. De glycidylrest komt overeen met de algemene formule 2, waarbij in deze formule 2 R^ bij voorkeur waterstof is, evenwel ook een alkylgroep 10 met 1 tot 4· koolstofatomen kan voorstellen.
Als gesubstitueerde resten R in de verbindingen met de algemene formule 1 verdienen de geschikt gesubstitueerde alkylresten de voorkeur. Als onverzadigde resten verdienen ethenisch onverzadigde resten, in het bijzonder overeenkomstig 15 enkelvoudig onverzadigde resten, de voorkeur.
Onderwerp van de uitvinding zijn voorts de volgens formulel gekenmerkte nieuwe verbindingen, alsmede werkwijzen ter bereiding daarvan. -
Het werkingsmechanisme van de in het kader van de uit-20 vinding gebruikte verbindingen is in bijzonderheden niet opgehelderd. 7ermoedelijk zijn de ook hier, zoals bij het triglycidylisocyanuraat van het Duitse Offenlegungsschrift 2.9C7.J4-9 aanwezige glycidylgroepen voor de cytostatische werking van eruitspringende betekenis. Alle beschreven ver-25 bindingen volgens de uitvinding met de algemene formule 1 zijn gekenmerkt door de aanwezigheid van twee dergelijke glycidylgroepen. Daarnaast is de in ruim kader varieerbare rest R in de betreffende verbindingsklasse aanwezig. Eet is mogelijk, dat via deze rest R invloed op de verdeling 30 van lipofiele en hydrofiele voorkeuren wordt uitgeoefend en dat daarmee in bepaalde mate de opname van de verbindingen door het organisme gestuurd kan worden. De betekenis van de volgens de uitvinding ingevoerde nieuwe substituent R dient echter niet daartoe beperkt te worden.
35 De rest R is volgens de hiervoor gegeven definitie een koolwaterstofrest, die ook heteroatomen kan bevatten. Als heteroatomen komen in het bijzonder IT, 0, S en/of P in aanmerking. 3ij voorkeur bevat elk van deze resten in totaal niet meer dan 15 koolstofatomen, bij voorkeur niet meer 4-0 dan 12 koolstofatomen en daarbij doelmatig niet meer dan 8005187

Interessant kunnen in het bijzonder resten zijn, die tot 6 of bij voorkeur zelfs tot slechts 4 koolstofatomen bevatten, waarbij deze getalwaarden onafhankelijk van de betreffende structuur zijn op te vatten en 5 alleen betrekking hebben op de som van alle koolstofatomen in de betreffende rest.
Stelt R een aryl-, aralkyl- of alkarylrest voor, dan verdienen hier in het bijzonder 1-kernige substituenten de voorkeur. Typische vertegenwoordigers zijn fenyl, benzyl, 10 tolyl, xylyl en verwante verbindingen. Ook bij de cycloali-fatische ringen voor de rest R verdienen eenkernige ring-systemen op basis van cyclopentyl, cyclohexyl en derivaten daarvan, de voorkeur. Overeenkomstige heterocjj-clische resten, in het bijzonder eveneens eenkernige ringverbindingen 15 met 0, ïï en/of S in het systeem vallen binnen het kader van de uitvinding. Be ringsystemen kunnen daarbij bij voorkeur 1, 2 of 3 van dergelijke heteroatomen bevatten. Beze heterocyclische resten bevatten bij voorkeur 5 of 6 ringatomen. Bësgev/enst kunnen alle hier genoemde ringvormige substi-20 tuenten, hetzij van aromatische of cycloalifatische aard, op hun beurt verdere substituenten bevatten. Geschikte substituenten zijn bijvoorbeeld halogeen, hydroxy of alkoxy.
In een uitvoeringsvorm van de uitvinding, die in het bijzonder de voorkeur verdient, stelt de rest R een even-25 tueel gesubstitueerde alkylrest voor. Beze alkylrest kan recht of vertakt en verzadigd of onverzadigd zijn en bevat - onder uitsluiting van zijn substituenten - bij voorkeur niet meer dan 10, in het bijzonder niet neer dan 8 kool-stofatomen. In deze uitvoeringsvorm van de uitvinding ver-30 dienen in het bijzonder die verbindingen met de algemene formule 1 de voorkeur, waarin de rest R niet gesubstitueerd alkyl met 1 tot 6 koolstofatomen of een overeenkomstige alkylrest voorstelt, die met halogeen, hydroxy, amino, N-gesubstitueerd amino, mercapto, alkyimercapto, arylmer-35 capto, alkylsulfoxy, arylsulfoxy, alkoxy, aroxy en/of acyloxy gesubstitueerd is, waarbij de substituent ook van heterocyclische aard kan zijn.
Eierbij kunnen dergelijke gesubstitueerde resten een-of meermalen met de genoemde groepen gesubstitueerd zi^n.
40 Bij voorkeur zijn 1 tot 3 van de genoemde substituenten in ft η n 5 1 r 7 τ- de betreffende rest R aanwezig, waarbij in een bijzonder geval, dat de voorkeur verdient, dergelijke gesubstitueerde alkylresten van bet genoemde type bevattende verbindingen met de algemene formule 1 in de geneesmiddelpreparaten 5 volgens de uitvinding gebruikt worden.
Zijn in de gesubstitueerde alkylrest R substituerende groepen aanwezig, die op hun beurt koolwaterstofresten bevatten - in het bijzonder dus in het geval van de resten N-gesubstitueerd amino, alkylmercapto, arylmercapto, alkyl-10 sulfoxy, arylsulfoxy, alkoxy, aroxy en acyloxy - dan bevatten deze substituerende groepen bij voorkeur niet meer dan 10, doelmatig niet neer dan 8 koolstofatomen. De grens, die in het bijzonder de voorkeur verdient, ligt hier bij 6 koolstof atomen, in het bijzonder bij niet meer dan 4- koolstof-15 atomen. Deze substituerende koolwaterstofresten kunnen op hun beurt aryl, aralkyl, alkaryl, cycloalkyl en/of alkyl-resten zijn, die desgewenst ook substituenten zoals halogeen, hydroxy, alkoxy en dergelijke kunnen bevatten. Ook hier zijn heteroatomen bevattende resten van het hiervoor 20 geschetste type, dus bijvoorbeeld heterocyclische ringsy-stemen met 1 tot 3 heteroatomen van het hiervoor geschetste type - in het bijzonder eenkernige ringen met N, 0 en/of S als heteroatomen - mede inbegrepen. De voorkeur verdienen de overeenkomstige heterocyclische 5- of 6-ringen.
25 In een uitvoeringsvorm, die in het bijzonder de voor keur verdient, worden verbindingen met de algemene formule 1 toegepast, waarin de rest R een mono- of digesubstitueer-de alkylrest van het genoemde type voorstelt, die uit de volgende groepen is gekozen: monohydroxyalkyl, dihydroxy-30 alkyl, halogeenhydroxyalkyl, N-gesubstitueerd aminohydroxy-alkyl, alkylmercaptohydroxyalkyl, gesubstitueerd alkylmer-captohydroxyalkyl, de overeenkomstige alkylsulfoxyhydroxy-alkylresten, eventueel gesubstitueerd alkoxyhydroxyalkyl en eventueel gesubstitueerd acyloxyhydroxyalkyl. 3ij voor-35 keur kunnen daarbij de alkylresten tot 7» bij voorkeur 3 tot 7 on in het bijzonder 3? 4 of 5 koolstofatomen bevatten.
In het kader van de uitvinding kunnen verbindingen met de algemene formule 1 toepassing vinden, waarin de rest R recht of vertakt, niet-gesubstitueerd alkyl met ten hoogste 40 6, bij voorkeur ten hoogste 4 koolstofatomen voorstelt.

In aanmerking komen in het bijzonder de resten methyl, ethyl, propyl, isopropyl alsmede de overeenkomstige C -resten en hun etherisch onverzadigde analogen. Toorts verdienen zeer in het bijzonder die verbindingen met de alge-5 mene formule 1 de voorkeur, waarin de rest R een mono- of digesubstitueerde alkylrest van het genoemde type met in het bijzonder 3 koolstofatomen is en daarbij voorkeur ten minste een hydroxylgroep bevat. Hier is dus dan steeds ten minste een hydroxylgroep - bij voorkeur naast een andere 10 substituent aan deze rest - aanwezig.
Deze substituerende groepen zijn daarbij volgens een verder de voorkeur verdienende uitvoeringsvorm van de uitvinding over de 2- en de 3-plas-tsen van de betreffende rest R verdeeld. Daarbij kan de hydroxylgroep hetzij op de 2-15 plaats hetzij op de 3-P-aats aanwezig zijn. Toorts in het bijzonder de voorkeur verdienen geschikt gesubstitueerde verbindingen met formule 1, die in de rest R naast de hydroxylgroep geen andere substituerende groep of als verdere substituenten hydroxy, halogeen, een ^-gesubstitueerde 20 aminorest, een eventueel gesubstitueerde alkoxyrest, een eventueel gesubstitueerde alkylmercaptcrest respectievelijk alkylsulfoxyrest of een eventueel gesubstitueerde acylcxy-rest bevatten. Als halogeen verdienen in het bijzonder chloor en/of broom de voorkeur, fluor en jcod zijn echter 25 niet uitgesloten. De IT-gesubstitueerde aminoresten komen overeen met de formules 3. Hierbij zijn de rest ^ respectievelijk R^ koolwaterstofresten, die op hun beurt gesubstitueerd kunnen zijn. Bij de uitvoeringsvorm van de uitvinding, die de voorkeur verdient, bevatten deze resten R2 30 en eventueel R^ ten hoogste 12 koo1stofatomen, waarbij bij de disubstitutie aan het stikstofatoom de som van de resten R2 en R^ bij voorkeur niet de 12 koolstofatomen overschrijdt. In het bijzonder bij voorkeur bevatten deze substituenten R0 en Rz tezamen ten hoogste 8 en daarbij in het bijzonder 35 niet meer dan 5 koolstofatomen. De resten Rp en R- kunnen ook tot een verzadigde of onverzadigde, eventueel aromatische en/of heterocyclische ring zijn verbonden. Bij voorkeur kunnen Ii0 en eventueel R-, cck alkylresten zijn. Dijn c— ^ deze resten op hun beurt nogeens gesubstitueerd, dan worden 40 in het kader van de uitvinding als zodanige substituenten 8005187 6 in het bijzonder hydroxy, alkoxy respectievelijk halogeen, bij voorkeur chloor of broom, beoogd. Is naast de hydroxyl-groep in H een acyloxyrest, een alkoxyrest of een alkyl-mercapto- respectievelijk alkylsulfoxyrest aanwezig, dan 5 bevat bij voorkeur deze rest eventueel ten hoogste 10 koolstof atomen, waarbij ook hier de voorkeursgrens bij 8 kool-stofatomen ligt en in het bijzonder de voorkeur verdient het niet meer dan 5 koolstofatomen op deze plaats in het molecuul in te voeren. De voorkeur verdienen ook bij stig 10 acyloxyresten alkylresten met het overeenkom aantal kool- Π16Ϊ stofatomen, waarbij ook arylresten/uitgesloten zijn. De acyloxyresten zijn bij voorkeur afgeleid van monocarbonzu-ren met het genoemde aantal koolstofatomen en de genoemde structuur.
15 De geneesmiddelen volgens de uitvinding kunnen bij voorkeur afzonderlijke gedefinieerde verbindingen met de algemene formule 1 bevatten, het is evenwel gebleken, dat ook mengsels van werkzame stoffen van meerdere onder de 5T= gemene formule 1 valleêeverbindingen sterk werkzame cyto-20 statica zijn. In het kader van de uitvinding kan het voorts de voorkeur verdienen afzonderlijke bepaalde of een mengsel van enkele verbindingen met de definitie volgens de uitvinding volgens formule 1 gemengd met de 1GI-verbindingen volgens de vermelde oudere Duitse octrooiaanvrage 2.907.34-9 25 toe te passen.
De bereiding van de werkzame stoffen met formule 1 is een verder onderwerp van de uitvinding. In principe kan de bereiding op op zichzelf bekende wijze plaats hebben. Daarbij worden de volgende mogelijkheden opgesomd: 30 1. 'Omzetting van triglycidylisocyanuraat met een overmaat water, alcohol, primaire en/of secundaire aminen, mercaptanen, iminen, imiden, carbonzuren, waterstof-halogenide en dergelijke of waterstof.
Op grond van de equivalentie van de drie glycidylgroe-35 pen in het ‘TGI leidt deze reactie eerst steeds tot produktmengsels, die op hun beurt therapeutisch werkzaam kunnen zijn. Eet is evenwel ook mogelijk en een deel van de in het volgende geschetste werkwijze van de uitvinding, uit deze mengsels de overeenkomstige 40 verbindingen met de algemene formule 1 door geschikte 8005137 7 f scheidingsmethoden, bijvoorbeeld preparatieve dunne-laag-chromatografie of kolonchromatografie te scheiden. Binnen het kader van deze omzettingen wordt een glyci-dylgroep tot de rest E van de verbindingen met de alge-5 mene formule 1 omgezet. Bij de reductieve behandeling van de glycidylgroep met waterstof respectievelijk waterstof voortbrengende verbindingen ontstaat een mono-hydroxyalkylrest E. Als waterstof voortbrengende verbindingen kunnen bijvoorbeeld hydrideverbindingen, 10 bijvoorbeeld complexe boorhydriden zoals natriumboor- hydride toepassing vinden. In de andere genoemde gevallen wordt de triglycidyluitgangsverbinding met een nu-cleofiele verbinding ΗΦΑΘ in overmaat omgezet, waardoor een gedisubstitueerde rest E ontstaat, die naast een 15 hydroxylgroep de rest Αθ als tweede substituent gewoon lijk aan het aangrenzende atoom ten opzichte van het gehydroxyleerde koolstofatoom van de rest H bevat.
In principe is de omzetting van de glycidylgroepen van het 23-1 met dergelijke nucleofiele reactieparrners be-20 kende stand der techniek en bijvoorbeeld beschreven m
Angew. Chemie SC, (1968), 85^. 3eze reactie wordt echte-” ir de stand der techniek gericht on meer dan slechts een eooxt—srroen van het 201 uitsevoerd en κι Ό - w dient bijvoorbeeld op industriële schaal voer ae ver-25 knoping van epoxyharssystemen. Bij de werkwijze vol gens de uitvinding werden daarentegen bij voorkeur werkwijze-omstandigheden gekozen, die een zo ver mege-lijk gaande opbrengsttoename gericht op 1 : 1 reaetie-produkten toelaten, alsmede de aansluitende isolering 30 en winning van deze 1 : 1 reactieprodukten onder af scheiding van niet-omgezette delen van het uitgangsmateriaal en verder voerende onzettingsproaukren, aie door reactie van meer dan slechts een epoxygroep met de nucleofiele reactiepartner ontstaan zijn.
33 Bij de omzetting van ÏG-I-verbindingen met nucleofiele reactiepartners van het hiervoor geschetsre type kan het moeilijk zijn de gewenste 1 t 1 reac ^leprod^x— ten in voorkeursopbrengsten te verkrijgen, omaar ae drie epoxygroepen van het molecuul van ae uitgangsv^r-40 binding bij de reactie ongeveer gelijkv/aaraig zijn en 8005187 o daarom vaak de gewenste diglycidylverbinding niet als voorkeursreactieprodukt gevormd wordt. Ook de poging door omzetting van het triglycidylisocyanuraat met een overmaat nucleofiele reactiepartner de gewenste verbin-5 ding te verrijken, blijkt slechts in weinig gevallen werkelijk gangbaar, omdat hierbij in vele gevallen zeer gemakkelijk een polymerisatie van het triglycidylisocyanuraat optreedt.
Verhoudingsgewijze probleemloos gelukt de winning van 10 het 1 : 1 reactieprodukt als regel enkel bij de keuze van mercaptanen, aminen en hydriden als reagentia. Hier kunnen partiële ringopeningsprodukten van de gewenste constitutie met uitgangsmengsels verkregen worden, die de reagentia in de verhouding 1 : 1 of in slechts een 15 geringe overmaat van een van de reagentia bevatten.
ïloeilijker is reeds de winning van de overeenkomstige omzettingsprodukten van het DGI met bijvoorbeeld car-bonzuren, water of alcoholen.
Gevonden werd, dat de bereiding van de Ί : 1 reactie-20 produkten verrassenderwijze eenvoudig dan mogelijk wordt, wanneer men het triglycidylisocyanuraat met een overmaat en wel bij voorkeur een grote overmaat van het nucleofiele reagens ΗβΑθ omzet, de reactie daarbij echter voortijdig afbreekt en de overmaat van de nu-25 cleofiele reactiepartner, niet-omgezet TGI en mede ge vormde di- en triadditieprodukten afscheidt. Het achterblijvende onzuivere diglycidylprodukt kan dan op gebruikelijke wijze, bijvoorbeeld door kolomchromatografie, gezuiverd worden. Bij voorkeur wordt bij deze 30 werkwijze de nucleofiele reactiepartner in een 3- tot 30-voudige overmaat, in het bijzonder in een 5- tot 20-voudige overmaat ten opzichte van de vereiste hoeveelheid gebruikt. De omzetting kan in oplosmiddelen worden uitgevoerd, desgewenst kan echter ook de over-35 maat van de nucleofiele reactiepartner als oplosmiddel dienen. V/ordt met oplosmiddelen te werk gegaan, dan dienen deze doelmatig in hoofdzaak polair, echter onder de gekozen werkwijze-omstandigheden bij voorkeur niet reactief te zijn. Eventueel is het oplosmiddel 40 ook niet met water mengbaar. Door de keuze van derge- 8005187 o lijke oplosmiddelen wordt de polymerisatie-neiging van het I'C-I onderdrukt en de vorming van nevenreactie door additie van het oplosmiddel aan de epoxygroepen vermeden. Tenslotte wordt de omwerking van het reactiemeng-5 sel onder afscheiding van de reagentia en de ongewenste reactieprodukten op deze wijze vergemakkelijkt. Bijzonder geschikte oplosmiddelen zijn bijvoorbeeld gehalo-geneerde koolwaterstoffen, in het bijzonder chloorkool-waterstoffen. De reactietemperatuur ligt gewoonlijk 10 tussen ongeveer 30 en 120°C, bij voorkeur tussen 40 en 1C0°G en wordt daarbij bij een bijzonder doelmatige uitvoeringsvorm zodanig gekozen, dat binnen 4 tot 5 uren het epoxygehalte van het reactiemengsel met de helft is verminderd.
13 Bij de keuze van deze reactieparameters is de zuivering van het onzuivere reactieprodukt vaak op zeer eenvoudige wijze mogelijk. De overmaat van de nucleofiele reac-tiepartner, een deel van de monoglycidylverbinding, alsmede het in alle drie epox3'grcepen omgezerte deel van 20 het reactieprodukt kan vaak door uitschudden van de organische fase mem water verwijderd worden. Biet omgezet TG-I wordt door opneming van het achterbiljvende reactiemengsel in methanol bij asantitatief neergeslagen, Sr blijft tenslotte de onzuivere diglycidylverbin-25 ding achter (1 : 1 reactieprodukt), die door eenvoudi ge fractionering bijvoorbeeld koiomchromatografisch gezuiverd kan worden. Als scheidingsmiddel is bijvoorbeeld kiezelgei geschikt. -kLs loopvioeistof kunnen bijvoorbeeld dichloornethaan/ethylacetaat of dichloorme-30 thaan/aceton worden toegepast. De zuivere bereiding en de winning van het twee epox3^groepen bevattende 1 : ^ reactieprodukt uit het mengsel van de reagentia is hier en bij de andere hierna geschetste werkwijzer, als regel een essentiële trap van de werkwijze volgens de 35 uitvinding. Voor de bereiding van de sulfoxyverbindin- gen uit de overeenkomstige mercartoverbindingen si® ?Iouben-,«7e3rl aaO, Bend 9 (*955), 207-21?, alsmede hakromol.Chen. 16^, (1970), 305.
2. Sen zeer elegante algemene werkwijze ter bereiding ”sn 40 de verbindingen met de algemene formule 1 berust op de O Λ Λ K 1 fl 7 10 omzetting van de mono-N-gesubstitueerde cyanuurzuren met epih.alogeenh.ydrienen. De bereiding van mono-ÏT-gesubstitueerde cyanuurzuren kan volgens in de literatuur bekende methoden plaats hebben. Voor de desbe-5 treffende literatuur wordt bijvoorbeeld verwezen naar 77.J. Close, J. Am. Chem. Soc. 22» 0953)? 3617. Beschreven wordt hier naast oudere in aanmerking komende methoden een algemeen toepasbare werkwijze, waarbij mono-gesubstitueerde biureetverbindingen met alkyl-10 carbonaten, in het bijzonder ethylcarbonaat, bij aan wezigheid van een alkal^^anolaat, in het bijzonder natriumethanolaat, tot mono-gesubstitueerde cyanuurzuren worden omgezet. De op deze wijze ingevoerde substituent komt daarbij als regel overeen met de rest H uit 15 de verbindingen met de algemene formule 1. In de hier na volgende reactie werden dan de beide glycidylgroepen ingevoerd. Hiertoe set men op een op zichzelf bekende 7/ijze de mono-gesubstitueerde cyanuurzuren om se-t-de geschikte epihalogesnhydrienverbinding, bijvoorbeeld 20 met epichlcorhydrien. Deze omzetting heeft eveneens op een on zichzelf bekends wijze plaats. ίΊ<~ kan bi^j aan— wezisheid van een geringe hoeveelheid van een kwater-naire ammoniumverbinding als katalysator worden uirge-voerd (zie hiertoe bijvoorbeeld Houben-7/eyl, Methoden 25 der organischen Chemie, Band 14/2 (1963)» 497, 5^-7).
3ij een modificatie van deze weg wordt het mono-gesubstitueerde cyanuurzuur niet onmiddelli^k met c.e epoxy-verbinding omgezet. In plaats daarvan heeft eerst de omzetting ervan plaats met allylhalogeniaen, die met de 2o glycidylrest van de verbindingen met de algemene formu le 1 overeenkomen, echter in plaats van de epoxygreep een ethenische dubbele binding bezitten, waarop de ontstane diallylgesubstitueerde isocyanuraten geëpoxy-deerd worden. De epoxydatie kan op een op zichzelf oe-^2 kende wijze met perzuren worden uitgevoerd. De omzeutirg van cyanuurzuur met allylhalogeniden is oi,jvooroeeid beschreven in het Amerikaanse octrooischrift 3.376.301, de eroxydatie van aliylisocyanuraten met perzuren is bijvoorbeeld in Houben-V/eyl aaC, Band 6/3» 365 ff he-40 schreven. Dij kan bijvoorbeeld bij aanwezigheid van een 8005187 11 geringe hoeveelheid var. een kwaternaire annoniunverbm- ding als katalysator worden uitgevoerd.
De omzetting van de monogesubstitueerde cyanuurzuren met epihalogeenhydrienen respectievelijk allylhalcgeniden 5 heeft doelmatig plaats in een temperatuurtrajeet van ongeveer 50 tot 150°0, bi^j voorkeur van ongeveer 70 tot ongeveer 125°c. Allylhalogenide respectievelijk epihalcgeenhy-drien wordt in een molverhouding tot cyanuurzuurverbincing van ten minste 2 : 1 gebruikt, waarbij' echter ook met een 10 aanzienlijke overmaat te 7/erk kan worden gegaan, bijvoorbeeld tot een molverhouding van 10 : 1. Het werken met nol-verhoudingen in het traject van 2 tot d nol allylhalogenide respectievelij'k epihalogeenhydrien per mol cyanuurzuurver-binding kan bijzonder doelmatig zijn. De allylhalogemaen 15 respectievelijk epihalogeenhydrienen, die de voorkeur verdienen, bevatten chloor of eventueel broom als halogeen.
De reactie kan in polaire aprorische oplosmiddelen worden uitgevQsrd, die ten minste een van de reaotiepartners ten dele oplossen en ten opzichte van de reagentia niet 20 reactief zijn. Den bijzonder doeimarig oplosmiddel is de groep van de dialkylfermamiden, in het bijzonder oe dialkyu-formamiden met een klein aantal koolstofaromen, zoals dime-thylformamide. De cyanuurzuurverbinding kan als zodanig of als zout worden gebruikt. De reactietijd, die de voorkeur 25 verdient, bedraagt 1 tot 1C uren, in het bijzonder 2 tot 5 uren.
Ook de epoxydatie van de allylgroepen door middel van perzuren wordt bij voorkeur in oplosmiddelen uitgsvoerd. Geschikt zijn ook hier polaire oplosmiddelen, bijvoorbeeld 50 haiogeenkoolwaterstoffen of alcoholen. De geschikte reacrie-temperatuur ligt gewoonlijk in het traject van 0 tot 50°C in het bijzonder tussen ongeveer "O en 30 0. Her perzuur wrordt doelmatig in vrijwel equivalente hoeveeineio. of slechts in een geringe overmaat gebruikt. m-Chloorperben-35 zoezuur is als handelsprcdukt gemakkelijk toegankelijk en voor de uitvoering van de reactie geschikt. De reactieduur li™t a’' - T>ee:ei in het traject van 2d uren of meer, bijvoor-beeld tot dS uren.
Cnderwern van de onderhavige uitvinding is dientenge-dQ volge in een verdere uitvoeringsvorm de werkwijze ter be-Q η Λ *% 1 fl 7 reiding van N-gesubstitueerde diglycidyl-isocyanuurzuur-verbindingen met de algemene formule 1, waarin H alkyl, aryl, aralkyl, alkaryl of cycloalkyl voorstelt, welke resten eventueel ook van heterocyclische aard, onverzadigd 5 en/of met ten minste een van de volgende substituenten gesubstitueerd kunnen zijn: halogeen, hydroxy, amino, N-ge-substitueerd amino, mercapto, alkylmercapto, arylmercapto, alkylsulfoxy, arylsulfoxy, alkoxy, aroxy, acyloxy en een heterocyclische rest, en waarin glycidyl een rest met de 10 algemene formule 2 voorstelt, waarin R^ bij voorkeur waterstof is, maar ook een alkylrest met 1 tot 4 koolstofatomen kan voorstellen. Deze werkwijze wordt gekenmerkt, doordat men in een met de rest R mono-N-gesubstitueerd cyanuurzuur de beide glycidylresten met de algemene formule 2 in M-sub-15 stitutie invoert of een triglycidylisocyanuurzuur met glycidylresten met de algemene formule 2 aan een partiele omzetting met water, alcoholen, verbindingen met een primaire of secundaire aminogroep, mercaptanen, zwavelwaterstof, carbonzuren, waterstofhalogeniden of waterstof respectie-20 velijk waterstof voortbrengende verbindingen onderwerpt, eventueel gevormde mercaptoverbindingen tot overeenkomstige sulfoxyverbindingen omzet en de gevormde reactieproduk-ten met de algemene formule 1 uit het reactiemengsel afscheidt en als zodanig wint.
25 Worden bij deze werkwijze de beide glycidylresten met de algemene formule 2 in mono-ïT-gesubstitueerde cyanuurzu-ren ingevoerd, dan kan dit plaats hebben, doordat cnmiddel-lijk het met de rest R gesubstitueerde cyanuurzuur met epi-halogeenhydrienen wordt omgezet, waarbij deze epihaligeen-50 hydrienen met de glycidylresten met de algemene formule 2 overeenkomen ofwel worden eerst de mono-gesubstitueerde cyanuurzuurverbindingen met geschikte allylhalogeniden tot reactie gebracht, waarna door epoxydatie met bij voorkeur perzuren de allylresten respectievelijk met R^ gesubsti-55 tueerde allylresten tot de glycidylgroep worden omgezet. Gp de hoedanigheid van de verbindingen met de algemene formule 1, de resten R en glycidyl en de reagentia voor het vormen van deze resten zijn alle hiervoor gegeven aanwijzingen van toepassing.
40 De verbindingen met de algemene formule 1 in gezuiver- 8005187 13 - de en in de massa gewonnen vorm zijn nieuwe verbindingen.
Zij vallen als zodanig en in liet bijzonder in de geïsoleerde, voor de toepassing als geneesmiddel geschikte vorm binnen het kader van de onderhavige uitvinding. Ook voor dit 5 aspect van de uitvinding gelden de hiervoor gegeven algemene aanwijzingen voor de definitie van de verbindingen met de algemene formule 1 met hun resten E en glycidyl. De li-teratuurplaats Budnowski, Angewandte Chemie 80 (1968), 851 formuleert als tussentrap van de omzetting van een grote 10 overmaat epichloorhydrien met z.yanuurzuur als tussentrap een reactieprodukt, dat naast twee. epoxygroepen in h-sub-stitutie een 2-hydroxy-3-chloorpropylsubstituent bevat. De isolering van deze verbinding 15 als zodanig in de massa wordt echter niet beschreven, bovendien ontstaat de verbinding volgens deze literatuurplaats langs een weg, die met de hiervoor geschetste werkwijze niets gemeen heeft.

In principe zijn al deze verschillende vormen - voor de doeleinden van de uitvinding geschikt. Zij kunnen daarbij gemengd of ook in de vorm van bepaalde geïsoleerde isomeren worden gebruikt. .
Voor de toepassing als cancerostatica moeten de werk-35 zame produkten door middel van geschikte dragers worden toegepast. Hiertoe geschikt zijn de gebruikelijke hulpstoffen respectievelijk dragers voor farmacologische preparaten. In het onderhavige geval is de toepassing van waterige systemen, eventueel tezamen met verenigbare glycolethers, zoals 4-0 glycolnoncethylether of butyleenglycolmethylePher of pro-.
800518? 14 pyleenglycolmethylether, geschikt gebleken, in het bijzonder wanneer de werkzame stof parenteraal moet worden toegediend. Bij orale toediening zijn de farmaceutisch gebruikelijke hulpstoffen respectievelijk dragers toepasbaar, voor 5 zover zij een geschikte verenigbaarheid met de glycidyl·verbindingen bezitten.
Bij de dierproef is de toepassing van pas bereide, waterige oplossingen, die intraperitoneaal of intraveneus worden toegediend, doelmatig gebleken.
10 De volgens de uitvinding toegepaste verbindingen zijn werkzaam tegen verschillende vormen van leukemie, alsmede kwaadaardige neoplasmata, zoals longcarcinoom, darmcarci-noom, melanomen, ependymoblastoom en sarcoom. Gebleken is, dat in enkele gevallen een duidelijk betere werking in ver-15 gelijking met cyclofosfamide en fluoruracil is vast te stellen.
Een combinatie therapie tezamen met andere cytostatica, zoals derivaten van het stikstoflost of ook fluoruracil is mogelijk.
20 Zeer in het algemeen geldt voor de in het kader van de uitvinding gebruikte verbindingen met de algemene formule 1 met hun rest H, dat deze rest ten minste onder normale omstandigheden geen of geen wezenlijke reactiviteit met de epoxygroepen van de glycidylsubstituent(ön) aan het nngsy-25 steem met de algemene formule 1 toont respectievelijk zal tonen.
Cp deze wijze is veilig gesteld, dat de volgens de uitvinding toegepaste werkzame stoffen voldoende houdbaar zijn en geen ongewenste omzetting onder vernietiging van de 30 epoxy—groepen plaats heeft. Deze regel dient in het bijzonder ook bij de keuze van eventueel aanwezige substituen-ten in de rest R in acht genomen te worden.

20 Voorbeeld I
Bereiding van diglyciayl-2.5-dihydroxynronyli3ocyanuraat 10C g Driglycidylisocyanuraat morden in 1000 ml water met een pH van 7 tij 7Q°0 3 uren geroerd. Het onopgeloste achtergebleven uitgangsprodukt wordt na het afkoelen van 23 de oplossing gefiltreerd en de oplossing met behulp van verminderde druk opgewekt met een oliepcmp voorzichtig tot droog ingedampt. Het mengsel bevat 9,1 gew.% epoxyzuurstof en bestaat in hoofdzaak uit een mengsel van diglycidyi- 2.3-dihydroxypropylisocyanuraat en glycidyl-di(2.3-dihydro-30 . xypropyl)-iso cyanuraat.
Het mengsel wordt kolomchromatografisch gefractioneerd.
De kolom met een diameter van 3.cm wordt met 170 g kiezel-gel van 0,063 tot 0,2 mm gevuld. Als elueermiddel wordt een mengsel van dichloormethaan/ethylacetaat 3 · 2 + 5 % metha-33 wol gebruikt.
Bij een begingewicht van 38 g worden 18 fracties van telkens 100 ml geïsoleerd. De fracties 10 tot 18 gaven 21 g van de gewenste verbinding.

3ereiding van diglycidyl-2-hydroxy-3-chloorproOylisocyanu-raat.
5 1 mol Cyanuurzuur, 60 mol epichloorhydrien en 0,02 mol tetraethylammoniumbromide worden 3 uren onder terugvloei-koeling gekookt. De reactie is in verloop van 1/2 tot 3/4- h na oplossing van tiet cyanuurzuur beëindigd, De overmaat epichloorhydrien wordt met verminderde druk opgewekt met de 10 waterstraalpomp gedestilleerd. Vervolgens destilleert men met verminderde druk opgewekt met een oliepomp (0,013 kPa) op het oliebad (70 tot 100°C) het dichloorhydroxypropaan. Deze destillatie dient zeer zorgvuldig te worden uitgevoerd, omdat zelfs kleine hoeveelheden niet verwijderd dichloor-15 hydroxypropaan de opbrengst doen dalen.
Uit de achterblijvende, lichtgele hars kristalliseert na toevoeging van methanol gedurende een nacht in de koelkast triglycidylisocyanuraat; opbrengst 35tot-40 %.
De methanolfase wordt bij 50°C met behulp van de olie-20 pomp geconcentreerd en bestaat uit een mengsel van ongeveer 70 % di-glycidyl-2-hydroxy-3-chloorpropylisocyanuraat en 20 % glycidyl-di(2-hydroxy-3-chloorpropyl/-isocyanuraat.

Bereiding van diglycidyl-methyl-isocyanuraat 1 mol Kethylcyanuurzuur (W.J. Close, Journal of the American Chemical Society, 21 (1953) 3618) wordt met 90 mol epichloorhydrien en 0,1 mol tetramethylanmoriumchloride 40 4 uren onder terugvloeikoeling gekookt.

Het reactieprodukt wordt bij 40 tot 50°C onder een verminderde druk intensief op koken onder terugvloeikoeling gehouden. Daarbij worden 3 niol HaOH (4-0 % in water) langzaam onder intensief roeren druppelsgewijze toegevoegd en 5 tegelijkertijd het water continu azeotroop gedestilleerd. Men roert nog 30 minuten na, filtreert het keukenzout af en concentreert onder een verminderde druk tot droog.
De ontstane lichtgele hars heeft een epoxygehalte van 11.6 % (berekend12,5 %). De structuur werd. door het massa-10 spectrum bevestigd.
Voorbeeld IY .
Bereiding: van diglycidyl-(S-hvdroxv-^-diethvlaminooroOvl·)-isocvanuraat 10 g a-Triglycidylisocyanuraat en 12 g diethylamine 15 worden in 250 ml absolute tolueen opgelost en 6 uren onder uitsluiting van vocht tot 60°G verwarmd. Men destilleert de tolueen onder een verminderde druk en mengt het harsachtige residu met 50 ml methanol. Gedurende de nacht kristalliseert het niet-omgezette ïGI (ongeveer 5 g) nagenoeg vol-20 ledig. De moederloog werd ingedampt en bestaat uit een mengsel van de hiervoor genoemde verbindingen, waarbij de gly-cidylverbinding duidelijk overweegt. Het epoxygehalte van het mengsel bedraagt 6,3 c/o. Het dunne-laag-chromatogram (kiezelgel, dichloormethaan:ethylacetaat:dimethylamine 25 55 ‘ 4-5 ‘ 2) vertoont na verkoling met zwavelzuur twee vlekken. EE =0,5 diglycidylverbinding, H? = 0,2 monoglyci-dylverbinding, intensiteitsverhouding 5 : 1.
Het onzuivere produkt wordt via een kiezelgelkolom gefractioneerd (lengte 50 cm, diameter 5 cm). Als elueer-50 middel wordt dichloormethaan : ethylacetaat 3 : 2 + 15 % methanol gebruikt. -
Het massaspectrum en het H-MiE-spectrum ondersteunen de structuur van het geïsoleerde diglycidyl-(2-hydroxy-3-diethy1aminopropyl)-isocyanuraat.
35 Voorbeeld V

15 Bereiding van diglycidyl-Γ 2-h:njroxv-3-(2-hydroxyethyl-thio)-prouyl1-isocyanuraat 29,7 g (0,1 mol) cc-ïriglycidylisocyanuraat en 8,0 g (0,1 mol) mercaptoethanol, alsmede 1,5 nil triethyiasine in 500 ml dichloormethaan worden 3 uren op koken onder terug- 20 vloeikoeling gehouden. 'Daarna wordt met 3 x telkens 60 ml water uitgeschud, op natriumsulfaat gedroogd en vervolgens geconcentreerd.

Bereiding van diglycidylallylisocyanuraat 20 g (0,08 mol) Triallylisocyanuraat, 30 g (0,176 mol) 3-chloorperoxybenzoëzuur in 600 ml dichloormethaan laat men 100 uren bij 5°C en 24 uren big omgevingstemperatuur staan. 40 Daarna wordt de oplossing met een natriumcarbonaatoplossing 8005187 21 . . β 3χ uitgewassen en de dichloormethaanfase geconcentreerd. Gewicht van het onzuivere produkt: 26 g olieachtige vloeistof.
Het onzuivere produkt wordt in methanol opgenomen en 5 gefractioneerd neergeslagen. Het eerste precipitaat bevat triglycidylisocyanuraat met een smeltpunt van 70 tot 115°C. Het tweede precipitaat hevat 6,4 g van een reactieprodukt met een smeltpunt van 50 tot 55°0, % SpO: 9,0 (theorie 11,4·), joodgetal: 14-3 (theorie 90).
10 Dit onzuivere produkt wordt over een kiezelgelkolom (diameter 5 cm, lengte 4-0 cm), die met kiezelgel 60 is gevuld, met dichloormethaan : ethylacetaat 3 : 2 als elueer-middel gefractioneerd en uit methanol herkristalliseerd.

Daarna wordt de op-40 lossing geconcentreerd en in 200 ml methanol opgenomen. Het A η n R 1 7 neergeslagen TGI wordt afgezogen en de methanolfase weer geconcentreerd.

De troebele oplossing wordt met verdund zoutzuur aangezuurd, gefiltreerd en aan-20 sluitend 3x met telkens 200 ml dichloormethaan geëxtraheerd, op magnesiumsulfaat gedroogd en opnieuw geconcentreerd.
Gewicht van het onzuivere produkt: 8 g % EpO: 8,5 (theorie 10,7).
25 Set onzuivere reactiemengsel wordt door middel van een kiezelgelkolom gefractioneerd. De kolom heeft een diameter van 5 cm en een lengte van 4-5 cm en is met kiezelgel 60 gevuld. Als elueerniddel wordt dichloormethaan : ethylacetaat : methanol 3 · 2 : 1 toegepast.
30 Gewicht van de diglycidylverbinding: 3,7 g % EpO: 10,6 (theorie 10,7).
De verbinding heeft in het dunne-laag-chromatagram op kiezelgel een RF-waarde van 0,4-5 (dichloormethaan : ethylacetaat : methanol 3:2:1).
35 Voorbeeld XI
Bereiding van diglycidyl-2-hydroxv-3(21.3'-dihydroxyoro-pylthio)-OrooyIisocyanuraat 10 g (0,034- mol) Iriglycidylisocyanuraat, 3,3 g (0,034-mol) 1,2-dihydroxy-3-mercaptopropaan en 0,1 g triethylamine 40 werden in 200 ml methanol opgelost respectievelijk gesuspen- 8005187 23 - y deerd en 3*5 uren bij 40°C geroerd. Na voorzichtige destillatie van het oplosmiddel blijven 13 g produktmengsel achter, dat door kolomchromatografie met kiezelgel 60 (Birma Merck) en ethylacetaat/methanol 80/20 als elueeriüiddel werd 5 afgescheiden. Opbrengst aan mono-additieproduktί 1,6 g (11,6 %)\ % epoxyzuurstof: 7,92 (theorie 7,89)·
Voorbeeld XII .
Bereiding van dis:lycidvl-2-hydroxy-3-n-octvlthioOroryl-isocyanuraat 10 10 g (0,034 mol) Triglycidylisocyanuraat, 5,^ g (0,034 mol) n-ootylmercaptan. en 0,1 g triethylaffiine werden in 200 ml methanol opgelost respectievelijk gesuspendeerd en 3,5' uren bij 4Q°C geroerd. Na destillatie van het oplosmiddel blijven 16,1 g produktmengsel achter, dat door kolom-13 sbheiding met kiezelgel 60 (Birma Merck) en ethylacetaat : dichloormethaan : n-hexaan 45 ·* 45 J 10 als elueermiddel werd gescheiden. 3r werden 1,4 g van een 83-procents pro-dukt (op.brengst 7,7-fa^an de theorie) met 6,21 % epoxyzuurstof geïsoleerd. De structuur komt overeen met het hier-20 voor genoemde mono-additieprodukt.
Voorbeeld XIII
Bereiding van diglycid?l-2-hydroxy-5(benzthiazol-21-yl- . thio )-orou7/lisocyanuraat 10 g (0,054 mol) Triglycidylisocyanuraat, 5,7 g 25 (0,034 mol) 2-mercaptobenzthiazool en 0,1 g triethylamine werden in 200 ml methanol opgelcst respectievelijk gesuspendeerd en 2,5 uren bij 40°C geroerd. Na destillatie van het oplosmiddel bleven 12,7 g van een lichtgeel vast pro-dukt achter, dat met kiezelgel 60 (Birma Merck) en ethyl-30 acetaat : dichloormethaan 60 : 40 kolomchromatografisch werd gescheiden. Sr werden 2,0 g van een 92-procents mono-additieprodukt van de hiervoor genoemde structuur geïsoleerd (opbrengst 11,4 % van de theorie), 6,31 % epoxyzuur-stof, De verbinding is vast en kleurloos.
35 Voorbeeld XIV
Bereiding van. diglycidyl-2-hydroxy-5(benzimidazol-2’-yl-thio )-orooTlisocTanuraat 10 g (0,034 mol) 'Triglycidylisocyanuraat, 5,1 g (0,034 mol) 2-mercaptohenzinidazool· en 0,1 g triethylamine 40 werden in 200 ml methanol opgelost respectievelijk gesus- 8005187 24 pendeerd en 2 uren bij 40°C geroerd. Na destillatie van het oplosmiddel bleven 13,0 g van een vast produkt achter, dat met kiezelgel 60 (Firma Merck) en ethylacetaat kolom-chromatografisch gescheiden werd. Fr werden 1,8 g van een 5 89-procents produkt verkregen, dat vast en kleurloos is en een epoxyzuurstofwaarde van 5,35 bezit. Opbrengst 10,5 '1° van de theorie.
Voorbeeld XV
De volgende proeven werden uitgevoerd volgens proef-10 voorschriften van het National Cancer Institute Bethesda, Maryland 200014, gepubliceerd in "Cancer Chemotherapy Reports" deel 3? september 1972, Vol. 3» Nr. 2. Als werkzaam produkt werden de verbindingen volgens de uitvinding toegepast. De produkten werden als waterige 1-procents in-15 jectie-oplossingen onmiddellijk voor de toepassing vers bereid.
Bij muizen werd volgens Protokol 1200 (blz. 9 l.c.) r de tumorsoort P 388 (leukemie) i.p. net 10° cellen/muis geplaatst. De gemiddelde overlevingsduur van de aldus voor-20 behandelde dieren werd in een controlegroep (C) bepaald.
Bij proefgroepen van op dezelfde wijze voorbehandelde dieren worden de werkzame verbindingen volgens de uitvinding telkens in 3 giften toegediend. Daarbij wordt bij de verschillende proefreeksen net ^wisselende hoeveelheden van 25 de betreffende werkzame stof te werk gegaan. De gemiddelde levensduur van de betreffende proefgroep wordt bepaald (I). Vergelijking met de gemiddelde overlevingsduur van de aldus behandelde proefgroep ten opzichte van de onbehandelde controle-groep geeft de mate van verlenging T/C, bepaald in 30 %. Deze mate van verlenging T/C is een mate voor de werk zaamheid van de bij de proef gebruikte verbinding. In de volgende tabel zijn de T/C-waarden afhankelijk van de toegediende concentratie van de betreffende ^werkzame stof sa-nengevat.

Claims (10)
- 2. Preparaat volgens conclusie 1, met het 15 kenner k, dat een of meer verbindingen met de algemene formule 1 aanwezig zijn, waarin de rest R niet neer dan lp koolstofatomen, hij voorkeur niet meer dan 12 koe1stof-atomen en in het bijzonder niet meer dan 8 koolstofatomen bevat.
- 3. Preparaat volgens conclusies 1 en 2, met het kenmerk, dat het preparaat een of meer verbindingen met de algemene formule 1 bevat, waarin R een eventueel Se~ . substitueerde alkylrest met ten hoogste 1C koolstofatomen, bij voorkeur met ten hoogste δ koclstofatomen en in heb 25 bijzonder met ten hoogste 3 koolstofatomen is. 4-, Preparaat volgens conclusies 1 tct 3, met h et kenmerk, dat het preparaat een of meer verbindingen met formule 1 bevat, waarin R een met een ten minste een hydroxylgroep gesubstitueerde alkylrest voorste 30 p. Preparaat volgens conclusies 1 tot 4, net het kenmerk, dat in de verbindingen met de mene formule 1 de rest R een hydroxyalkylrest is, die minste een andere van de volgende substituenten bevat: hydroxy, halogeen, amino, --gesubstitueerd amino, 35 alkylnercapto, arylmercapto, alkylsulfoxy, arylsulfoxy» alkoxy, aroxy, acyloxy.
- 6. Preparaat volgens conclusies 1 tot 5, met her kenmerk, dat het nreraraat een of meer de verbindingen met formule 1 bevat, die als rest R 40 gesubstitueerd of monogehydroxyleerd alkyl met 1 tot - 8005187 ^ 27 - «- stofatomen of digesubstitueerd alkyl net 3 tot 7 koolsrof-atomen zoals dihydroxyalkyl, halogeenhydroxyalkyl, IT-ge-substitueerd sjninohydroxyalkyl, alkoxyhydroxyalkyl, alkyl-mercaptohydroxyalkyl, alkylsulfoxyhydroxyalkyl en acyloxy-3 hydroxyalkyl bevat.
- 7· Preparaat volgens conclusie 1 tot 6, met het kenner k, dat het preparaat een of meer verbindingen met formule 1 bevat, waarbij 3 een gedisubstitueerde alkylrest met 3 koolstofatomen is, die de hydroxy*Igroep op 10 de 2- of 3-513-3^13 en. de andere substituent op de andere van de beide genoemde plaatsen in de alkylgroep met 3 koolstof-atomen bevat.
- 8, Preparaat volgens conclusies 1 tot 7, e e t het kenmerk, dat het preparaat mengsels van meer 15 verbindingen met de algemene formule 1 en/of mengsels van deze verbindingen met triglycidylisocyanuraat (T'GI) bevat.
- 9. Preparaat volgens conclusies 1 tot 8, met het ken m e r k, dat het preparaat als verbindingen met de algemene formule 1 omzettingsprodukten van 2Gj. mer 20 waterstófhalogenide, water, verbindingen met een primaire en/of secundaire aminogroep, carbonzuren, alcoholen, fenolen en/of alifatische en/of aromatische thiolen respectievelijk met waterstof of waterstof leverende verbindingen bevat .
- 10. Preparaat volgens conclusies 1 tor 8, m e o het k e n merk, dat het preparaat omz'éttingsprodur-ten van mono-N-gesubstitueerde cyanuurzuren met epina^c-geenhydrienen of net allylhalogeniae en daaropvolgende epoxiderinm van de dubcele bindingen tan de aj-n/lg-oe.. b^ 30 vat.
- 11. Preparaat volgens conclusies 1 not 10, m e t. het kenner k/dat het preparaat verbindingen met de algemene formule 1 gemengd, met gebruikelijke nulp»to.L-fen en/of dragers bevat. 35 . 12. C-lycidylisocyanuurzuurverbindingen ne„ ze sug^m^e formule 1, waarin 3 alkyl, aryl, aralkyl, alkaryl of cyc.o-• alkyl vo.orstelt, welke resten ook van heterocyclisch onverzadigd en/of met ten minste ^=n vSxx ~ stituenten gesubstitueerd kunnen ’ 4-0 amino,, rï-gesubstitueerd amino, mercaprc, 8005187 arylmercapto, alkylsulfoxy, arylsulfoxy, alkoxy, aroxy, acyloxy en een heterocyclische rest, en waarin glycidyl een rest set de algemene formule 2 voorstelt, waarin waterstof of een alkylrest met 1 tot d koolstofatomen is.
- 13. Werkwijze ter bereiding van isocyanuurzuurverbin- dingen, met het kenmerk, dat men verbindin gen met de algemene formule 1, waarin de symbolen de bij conclusie 1 vermelde betekenissen bezitten, op een op zichzelf bekende wijze bereidt.
- 14.
