Abstract
La presente invención se refiere a un método para producir una proteína recombinante que comprende: (a) colocar múltiples células de mamífero recombinante en un primer medio de cultivo comprendido dentro de un recipiente para proporcionar un primer cultivo celular; (b) cultivar por lotes el primer cultivo celular hasta un intervalo de densidad celular de aproximadamente 1.0 x 106 células/mL a aproximadamente 5.0 x 106 células/mL; (c) colocar un volumen del primer cultivo celular de la etapa (b) en un segundo medio de cultivo incluido dentro de un biorreactor de perfusión para proporcionar un segundo cultivo celular con una densidad celular inicial en un intervalo de aproximadamente 0.25% x 106 células/mL a aproximadamente 0.5 x 106 células/mL; ( e) colocar un volumen del segundo cultivo celular de la etapa (d) en un tercer medio de cultivo incluido dentro de un biorreactor de producción para proporcionar un cultivo celular de producción con una densidad celular inicial en un intervalo de aproximadamente 0.25% y 106 células/mL a aproximadamente 8 x 106 células/mL; (f) cultivar por perfusión el cultivo celular de producción en condiciones que permitan que las células de mamíferos recombinantes secreten una proteína recombinante; y (g) recolectar la proteína recombinante del cultivo celular de producción.
