Abstract

La presente invención se refiere a una método para preparar una proteína por medio de un microorganismo, caracterizado porque comprende las etapas de: (a) introducir en un microorganismo una primera construcción de expresión que codifica la proteína; (b) introducir en el microorganismo una segunda construcción de expresión que codifica una proteasa auxiliar que difiere de la proteína y que comprende una secuencia de aminoácidos que es por lo menos 90% idéntica a la secuencia de aminoácidos indicada en la SEC ID NO. 1, en donde la proteasa auxiliar tiene actividad proteolítica y en donde la segunda construcción de expresión se introduce en una secuencia de ácido nucleico cromosómico que codifica una proteasa del microorganismo de manera que la proteasa codificada por el cromosoma está funcionalmente inactiva; y (c) expresar la proteína y la proteasa auxiliar en el microorganismo, en donde la proteína es una enzima, particularmente una proteasa, amilasa, celulasa, hemicelulasa, mananasa, tanasa, xilanasa, xantanasa, xiloglucanasa, betaglucosida, pectinasa, carragenasa, perhidrolasa, oxidasa, oxidoreductasa o una lipasa.
