Abstract

La presente invención se refiere a un procedimiento para recuperar antígenos con superficie de la hepatitis B, a partir de células de Pichia pastoris, caracterizado porque comprende: a) lisar las células de levadura en presencia de un compuesto caotrópico amortiguado, y separar el sobrenadante que contiene el antígeno de superficie de la hepatitis B de las células lisadas; b) someter el sobrenadante que contiene el antígeno de superficie de la hepatitis B obtenido en la etapa (a) a condiciones adecuadas para precipitar los lípidos y proteínas contaminantes del sobrenadante ; c) someter el sobrenadante que contiene el antígeno de superficie de la hepatitis B obtenido en la etapa (b) a diafiltración; d) poner en contacto el material retenido que contiene el antígeno de superficie de la hepatitis B obtenido en la etapa (c) con sílice; e) lavar las proteínas contaminantes a partir del antígeno de superficie de la hepatitis B absorbido en sílice resultante, con un amortiguador que tiene un pH en la gama de 6-8; f) eluir el antígeno de superficie de la hepatitis B de la sílice con un eluyente amortiguado que tiene un pH dentro de la gama de 9.5-11.0, que contiene urea de 0.5 a 8 molar; g) someter la fracción que contiene el antígeno de superficie de la hepatitis B obtenida en la etapa (f) a una etapa de filtración en gel, adecuado para separar el antígeno de superficie de la hepatitis B de las proteínas contaminantes; y h) someter la fracción que contiene el antígeno de superficie de la hepatitis B, obtenida en la etapa (g) a una etapa de cromatografía de intercambio aniónico, adecuada para separar el antígeno de superficie de la hepatitis B de las proteínas contaminadas.
