Abstract

본 발명은, (a) 휘발성의 약제학적으로 활성인 화학적으로 알칼리성인 성분 또는 이것의 약리학적으로 허용 될 수 있는 염이 분포되어 있고 접착제를 함유하는 매트릭스, (b)(a)가 염을 함유한 경우에, 염으로부터 유리 염기를 방출시키는 알칼리성 기를 함유하는 접착제 조성물의 요소, (c)(a) 및 (b)에서 언급한 확산할 수 있는 성분에 불투과성인 뒷층, 및 (d)(a) 및 (b)에서 언급한 확산할 수 있는 성분에 불투과성인 풀링-오프(pulling-off) 박으로 이루어지는 다중 요소 시스템을 갖는, 휘발성의 약제학적으로 활성인 화학적으로 알칼리성인 성분의 경피투여용 플라스틱에 관한 것이다. 매트릭스 또는 요소(b)가 풀링-오프 박(d)에 접촉하는 지의 여부에 의존하여, 매트릭스(a) 또한 최소한 요소(b) 부분은 피부에 플라스터를 고정시키기에 충분한 접착력을 갖는다. 매트릭스(a)와 풀링-오프 박(d) 사이에 배열된 각각의 (b)부분은 활성성분, 바람직하게는 데프레닐 및/또는 이것의 염중 하나에 대해 투과성이다. 본 발명은 또한 이러한 플라스터의 제조방법에 관한 것이다.

휘방성이고, 약제학적으로 활성이고 화학적으로 알칼리성인 성분의 경피투여를 위해 사용되는 플라스터, 및 이것의 제조방법(PLASTER FOR THE TRANSDERMAL ADMINISTRATION OF VOLATILE PHARAMACEUTICALLY ACTIVE, CHEMICALLY ALKALINE INGREDIENS, AND PROCESS FOR PRODUCING THE SAME)

제5도는 요소(b) 및 매트릭스(a)를 둘러싸고 있는 뒷층의 측면에 도포되어 있는, 본 발명에 따르는 패치의 추가의 구현의 단면도이다.

Claims (10)

  1. (a)화학적으로 염기성인 휘발성의 약제학적 활성 성분 또는 이것의 약리학적으로 허용될 수 있는 염이 약제로서 분포되어 있고, 압력-민감성 접착제를 함유하는 매트릭스, (b)(a)가 염을 함유할 경우에, 그 염으로부터 유리 염기를 유리시키기 위해 아미노기를 함유하는 압력-민감성 접착제 조성물의 요소, (c)(a) 및 (b)의 확산할 수 있는 성분에 불투과성인 뒷층, 및 (d)(a) 및 (b)의 확산할 수 있는 성분에 불투과성인 릴리이스 라이너로 구성되는 다중 요소 시스템으로 이루어지며, 매트릭스(a) 또는 (b)의 최소한 일부는 릴리이스 라이너(d)와 접촉하게 되면, 패치를 피부에 점착시키기에 충분한 접착력을 갖고, 매트릭스(a)와 릴리이스 라이너(d) 사이에 위치한 (b) 부분은 휘발성 약제 또는 이것의 염중 하나, 또는 둘 모두에 대해 투과성이 됨을 특징으로 하는, 화학적으로 염기성인 휘발성의 약제학적 활성 성분의 경피투여를 위해 사용되는 패치.
  2. 제1항에 있어서, (a) 또는 (b)또는 둘 모두가 활성 성분을 유리 염기의 형태로 더 함유함을 특징으로 하는 패치.
  3. 제1항에 또는 제2항에 있어서, 염이 염산염 또는 브롬화 수소산염 또는 둘 모두를 포함함을 특징으로 하는 패치.
  4. 제1항 내지 제3항 중 어느 한 항에 있어서, 매트릭스(a)가 약리학적으로 허용될 수 있는 염에 대한 용해제 또는 투과 증강제 또는 둘 모두를 함유함을 특징으로 하는 패치.
  5. 제1항 내지 제4항 중 어느 한 항에 있어서, 활성성분이 바람직하게는 L-형태인 데프레닐이며, 데프레닐성분은 적합하게는 데프레닐 염기로서 계산하여 약 5내지 약 75mg의 양으로 존재함을 특징으로 하는 패치.
  6. 제1항 내지 5항 중 어느 한 항에 있어서, 요소(b)가 다수의 층, 바람직하게는 2내지 3개의 층으로 이루어지며, 이중 적어도 하나는 염기성기를 함유하고, 매트릭스(b)는 적합하게는 요소 (b)의 2개의 층 사이에 적층되며, 이중 한층 B1은 릴리이스 라이너(d) 상에 위치하고, 한 B2는 뒷층(c) 상에 위치하며, 층 B1은 층 B2보다 더 큰 점착 활성을 갖고, 릴리이스 라이너(d)에 더 근접한 층(b)는 바람직하게는 릴리이스 라이너(d)로 부터 멀리 잇는 층(b) 보다 더 낮은 함량의 염기성를 가짐을 특징으로 하는 패치.
  7. 제1항 내지 6항중 어느 한 항에 있어서, 매트릭스(a)가 압력-민감성 접착제로 이루어지며, 바람직하게는 요소(a)가 모든, 그러나 한 측면에 요소(b)에 의해 둘러싸이고, 나머지 측면은 요소(a)의 점착활성의 도움으로 피부에 접착하도록 예정되거나, 또는 매트릭스(a)가 뒷층(c)와 직접 접촉할 정도로 요소(b)에 의해 둘러싸임을 특징으로 하는 패치.
  8. 제1항 내지 7항 중 어느 한 항에 있어서, 요소(b)가 염으로 부터 유리염기를 유리시키기 위해 염기성기를 구성하는 아미노기를 함유하는 접착제를 포함하고, 요소(b)가 적합하게는 릴리이스 라이너(d)와 직접 접촉하고, 적어도 하나의 뒷층(c) 및 릴리이스 라이너(d)는 유리하게는 합성수지의 박, 특히 알루미늄 중착박으로 이루어지며, 바람직하게는 합성수지의 폴리에스테르임을 특징으로 하는 패치.
  9. 제1항 내지 8항 중 어느 한 항에 있어서, 피부에 접촉하도록 예정된 경우, 패치의 크기가 약 10내지 약 25cm2임을 특징으로 하는 패치.
  10. (ⅰ)휘발성의 약제학적 활성성분의 약리학적으로 허용될 수 있는 염과, 압력-민감성 접착제 재료와 용매 또는 희석제의 유체 조성물의 혼합물을 형성시키고, 이 혼합물로부터 용매 또는 희석재를 증발시켜서, 층 또는 입자 형태로 균일한 기재의 매트릭스(a)를 생성시키는 단계, (ⅱ)염으로 부터의 유리 염기의 생성에 적합한 염기를 함유할 수 있는 상기 압력-민감성 접착제 조성물 및 용매 형태의 구멍 단면을 가질 수 있는 적어도 하나의 층에 접착제 조성물을 성층시키고, 이러한 층으로 부터 용매 또는 희석재를 증발시켜서 층(b)를 생성시키는 단계, (ⅲ)매트릭스(a)를 층(b)로 적층시키는 단계, 및 (ⅳ)유리염기에 대해 투과성인 매트릭스(a) 또는 하나의 층(b)가 릴리이스 라이너(d)와 직접 접촉할 정도로 매트릭스(a) 및 층(b)에 상기 뒷층(c) 및 상기 릴리이스(d)를 도포시키는 단계로 이루어지며, 활성 성분으로 중강된 매트릭스(a), 및 적어도 하나의 층(b)개 패치를 피부에 점착시키기에 충분한 점착력을 갖고, 매트릭스(a)가 조성물(b)로 처리될 때가지, 상기 염이 분해되지 않을 정도의 조건하에 유지되고, 단계(ⅰ) 및 (ⅱ)가 어떠한 바람직한 순서로도 수행됨을 특징으로 하는, 제1항 내지 9항중 어느 한 항에 따르는 패치의 제조방법.
