Abstract
혈액등의 액체시료중에 포함되는 유약한 세포를 분류계수하기 위한 시약을 제공하는 것으로서 하기하는 (1)로부터 (4)를 함유하는 유약백혈구를 함유하는 피검사시료와 혼합시키므로서 유약백혈구의 각군을 다른 세포군으로부터 판별가능한 정도로까지 차이가 생기게 하는 유약세포측정용시약이다.
(1) 혈구의 세포질 및 세포막을 고정화시키기 위한 폴리옥시에틸렌계 비이온계면활성제
(2) 혈구의 세포막에 손상을 주어 축소화하기위한 가용화제
(3) 혈구의 세포질 및 세포막을 고정화하기 위한 아미노산 및
(4) 액의 PH를 5.0-9.0침투압력을 150-600mOsm/kg 전기전도도를 6.0-9.0ms/cm로하는 완충액.

제 1 도는 혈액시료를 본 발명의 시약을 사용해서 측정한 때의 DC신호강도와 RF신호강도를 축으로하는 2차원 산포도의 모식도의 1예, 제 2 도는 만성골수성 백혈병환자로부터 채취된 말초혈액을 본 발명의 시약을 사용해서 측정한 때의 DC신호강도와 RF신호강도를 축으로하는 2차원산포도,
제 3 도는 임파선계의 유약구가 출현한 환자로부터 채취된 말초혈액을 본 발명의 시약을 샤용해서 측정한 때의 DC신호강도와 RF신호강도를 축으로하는 2차원 산포도.
Claims (6)
- 하기하는 (1)로부터 (4)를 함유하는 유약백혈구를 함유하는 피검사시료와 혼합하므로서 유약백혈구의 각군을 다른 세포군으로부터 판별가능한 정도까지 차이가 생기게하는 유약세포측정용시약(1) 혈구의 세포질 및 세포막을 고정화 시키기위한 다음식으로 표시되는 폴리옥시에틸렌계의 비이온계면 활성제 :R1-R2-(CH2CH2O)n-H[식중 R1은 탄소수 10-25의 알킬, 알케닐 또는 알키닐기 :R2는또는 COO : n은 10-40의 정수를 나타낸다.](2) 혈구의 세포막에 손상을 주어 축소화하기 위한 가용화제로서 다음의 2군으로부터 선택되는 적어도 1종의 가용화제 :(a) 제 1 의 군의 가용화제 :· N-라우로일사르코신산나트륨· 라우로일메틸 β-알라닌나트륨· 라우로일사르코닌(b) 제 2 군의 가용화제 :· n-옥틸 β-글루코시드· CHAPS(3-[(3-클로르아미드프로필)디메틸 암모니오]-1-프로술포네이트· CHAPSO(3[(3-클로르아미드프로필)디메틸암모니오]-2-히드락시-1-프로판술포네이트· MEGA 8(옥타노일-N-메틸글루카미드)· MEGA 9(노나노일-N-메틸글루카미드)· MEGA 10(데카노일-N-메틸글루카미드)· 슈크로오스모노카프레이트· N-포르밀메틸로이시루알라닌(3) 혈구의 세포질 및 세포막을 고정화하기 위한 아미노산 및(4) 액의 PH를 5.0-9.0침투압력을 150-600m 0 sm/kg전기전도도를 6.0-9.0ms/cm로하는 완충액
