Abstract

본 발명은 하기 구조식(I)의 세프트리악손 디소디움염 헤미헵타하이드 레이트를 제조하는 신규하고 개선된 방법에 관한 것이다.
7-아미노-3-{〔2,5-디하이드로-6-하디록시-2-메틸-5-옥소-as-트리아진-3-일)티오〕메틸}-3-세펨-4-카복실릭에시드 디소디움염을 수용액중 적당한 불활성 유기용매존재하 0℃내지 40℃의 온도에서 2-벤조-티아졸릴-티오에스텔과 같은 반응성 2-(2-아미노티아졸-4-일)-2-syn-메톡시이미노 아세틱에시드 유도체를 가하여 전환시킨 목적화합물을 매우 순수한 형태로 고수율로 분리하여 얻는 방법이다.
세프트리악손은 비경구 제제의 제3세대 세팔로스포린계 항생제로 중증 감염질환 치료에 유용한 약제이다.

Claims (4)

  1. 구조식(Ⅱ)의 7-아미노-3-{〔2,5-디하이드로-6-하이드록시-2-메틸-5-옥소-as-트리아진-3-일)티오〕메틸}-3-세펨-4-카복실릭에시드 디소디움염을 수용액중 적당한 불활성 유기용매중 0℃내지 40℃의 온도에서 구조식(Ⅲ)의 반응성 2-(2-아미노티아졸-4-일)-2-syn-메톡시이미노 아세틱에시드 유도체를 가하여 전환시키고, 얻어진 세프트리악손 디소디움염 헤미헵타 하이드 레이트를 분리함을 특징으로 하는 구조식(I)의 7-{〔2-(2-아미노티아졸-4-일)-2-syn-메톡시이미노〕아세트아미드}-3-{〔(2,5-디하이드로-6-하이드록시-2-메틸-5-옥소-as-트리아진-3-일)티오〕메틸}-3-세펨-4-카복실릭에시드디소 디움염헤미헵타하이드레이트(세프트리악손디소 시움염헤미헵타드레이트)를 제조하는 방법.
  2. 제1항에 있어서, 구조식(Ⅲ)의 반응성 아실화제로서 2-(2-아미노티아졸-4-일)-2-syn-메톡시이미노아세틱에시드-2-벤조티아졸릴-티오에스텔을 사용함을 특징으로 하는 방법.
  3. 제1항에 있어서, 적당한 불활성 유기용매로 아세톤을 사용함을 특징으로 하는 방법.
  4. 7-아미노-3-{〔2,5-디하이드로-6-하디록시-2-메틸-5-옥소-as-트리아진-3-일)티오〕메틸}-3-세펨-4-카복실릭에시드디소디움염.
