제1도는 진정세균의 16S 리보소옴 RNA의 2차 구조에 대한 도면이다. 제2도는 본 발명에 따라 살모넬라 진정세균류(Salmonela enteritidis에 대한 프로브 및 두 개의 헬퍼 올리고뉴클레오티드의 결합위치를 나타내는 진정세균의 16S 리보소움 RNA의 2차 구조에 대한 도면이다. 제3도는 본 발명에 따라 임균(Neisseria gonorrhea)에 대한 프로브 및 올리고뉴클레오티드의 결합 위치를 나타내는 진정세균의 16S 리보소옴 RNA의 2차 구조에 관한 도면이다.

Claims (27)

  1. 단일 스트랜드의 표적 핵산내에서 뉴클레오티드 프로브와 상보적 뉴클레오티드 서열간의 결합을 증가시키는 방법에 있어서, 프로브와는 다른 지역에서 표적 핵산과 혼성화하는 헬퍼 올리고뉴클레오티드를 표적 핵산에 가하고, 표적 핵산에 대한 프로브의 결합을 강화시키는 데 효과적인 양으로 헬퍼 올리고뉴클레오티드를 가하는 것으로 이루어지는 것을 특징으로 하는 방법.
  2. 제1항에 있어서, 표적 핵산이 DNA, mRNA, rRNA, tRNA 및 다른 작은 핵산으로부터 선택되는 것을 특징으로 하는 방법.
  3. 제2항에 있어서, 표적 핵산이 그것을 생성시키거나 또는 헬퍼 올리고뉴클레오티드와 뉴클레오티드 프로브의 첨가에 앞서 그것이 발견되는 유기체로부터 분리되는 것을 특징으로 하는 방법.
  4. 제2항에 있어서, 헬퍼 올리고뉴클레오드와 뉴클레오티드 프로브가 표적 핵산이 위치하고 표적 핵산과 헬퍼 올리고뉴클레오티드와 뉴클레오티드 프로브간의 혼성체 형성이 세포내에서 일어나는 세포에 주입되는 것을 특징으로 하는 방법.
  5. 제1항 내지 제4항중의 어느 하나에 있어서, 프로브가 DNA 올리고뉴클레오티드이고 헬퍼 올리고뉴클레오티드가 DNA 올리고뉴클레오티드인 것을 특징으로 하는 방법.
  6. 제5항에 있어서, 프로브는 길이가 약 10 내지 약 50개의 뉴클레오티드로 이루어지고 헬퍼 올리고뉴클레오티드는 길이가 약 10 내지 약 100개의 뉴클레오티드로 이루어지는 것을 특징으로 하는 방법.
  7. 제6항에 있어서, 프로브와 헬퍼 올리고뉴클레오티드가 디포스페이트 에스테르 골격, 알킬 또는 아릴포스포네이트 골격 또는 포스포로티오에이트 골격으로부터 선택되는 것을 특징으로 하는 방법.
  8. 제7항에 있어서, 프로브가 약 15 내지 약 40개의 뉴클레오티드로 이루어지고 헬퍼 올리고뉴클레오티드가 약 20 내지 약 50개의 뉴클레오티드로 이루어지는 것을 특징으로 하는 방법.
  9. 제6항에 있어서, 헬퍼 올리고뉴클레오티드가 프로브 바로 근처의 표적 핵산과 결합하는 것을 특징으로 하는 방법.
  10. 제7항에 있어서, 헬퍼 올리고뉴클레오티드가 프로브 바로 근처의 표적 핵산과 결합하는 것을 특징으로 하는 방법.
  11. 제6항에 있어서, 헬퍼 올리고뉴클레오티드가 프로브로부터 떨어진 지역에서 표적 핵산과 결합하는 것을 특징으로 하는 방법.
  12. 제7항에 있어서, 헬퍼 올리고뉴클레오티드가 프로브포부터 떨어진 지역에서 표적 핵산과 결합하는 것을 특징으로 하는 방법.

  18. 제6항에 있어서, 헬퍼 올리고뉴클레오티드의 혼합물이 사용되는 것을 특징으로 하는 방법.
  19. 제14항에 있어서, 헬퍼 올리고뉴클레오티드의 혼합물이 사용되는 것을 특징으로 하는 방법.

  23. 표적 핵산 검출용 키트에 있어서, 표적 핵산에서의 뉴클레오티드 서열과 상보적인 표지된 뉴클레오티드 프로브와 헬퍼 올리고뉴클레오티드로 이루어지는 것을 특징으로 하는 키트.
  24. 제23항에 있어서, 2개 이상의 헬퍼 올리고뉴클레오티드로 이루어지는 것을 특징으로 하는 키트.
  25. 제24항에 있어서, 키트가 두 개 이상의 프로브와 두 개 이상의 헬퍼 올리고뉴클레오티드를 함유하는 것을 특징으로 하는 키트.
  26. 제25항에 있어서, 각 프로브가 헬퍼 올리고뉴클레오티드를 가지고 있는 것을 특징으로 하는 키트.
  27. 제26항에 있어서, 한 개 이상의 프로브가 두 개 이상의 헬퍼 올리고뉴클레오티드를 가지고 있는 것을 특징으로 하는 키트.
