Claims (10)
- 프로인슐린-양 물질을 함유하는 불순 혼합물을 pH 약 7 내지 약 10에서 역상 대공극성 아크길레이트 에스테르 공중합체 수지 지지체에 적용시킨 다음 : 이 지지체로부터 아세톤, 아세토니트릴 및 아세톤과 아세토니트릴의 혼합물중에서 선택한 유기 희석제 약 10용적% 내지 약 30용적%를 함유하는 pH 약 8 내지 약 11의 수성 용출제를 사용하여 프로인슐린-양 물질을 용출시킴을 특징으로 하여, 프로인슐린-양 물질을 함유하는 불순 혼합물로부터 불순물을 분리시키는 방법.
- 제1항에 있어서, 프로인슐린-양 물질이 인체 프로인슬린의 아미노산 서열에 상용하는 서열을 갖는 방법.
- 제2항에 있어서, 프로인슐린-양 물질이 프로인슐린에 대한 전구체인 방법.
- 제3항에 있어서, 프로인슐린-양 물질이 프로인슐린 S-설포네이트인 방법.
- 제1 내지 4항중 어느 한 항에 있어서, 대공극성 아크릴레이트 에스테르 공중합체 지지체가 XAD-7 또는 XAD-8인 방법.
- 제5항에 있어서, 프로인슐린-양 물질을 함유하는 불순 혼합물을 뱃치 조건하에서 처리하는 방법.
- 제5항에 있어서, 프로인슐린-양 물질을 함유하는 불순 혼합물을 크로마토그래피 칼럼 조건하에서 처리하는 방법.
- 제5항에 있어서, 프로인슐린-양 물질을 함유하는 불순 혼합물을 pH 약 8 내지 약 9에서 대공극성 아크킬레이트 에스테르 공중합체 지지체에 적용시키는 방법.
- 제8항에 있어서, 불순 혼합물을 지지체에 적용시킨 후, 용출시키기 전에 지지체를 pH 약 7 내지 약 8.5의 수성 완충액으로 세척하는 방법.
- 제9항에 있어서, 프로인슐린-양 물질을 pH 약 10.5의 수성 용출제를 사용하여 지지체로부터 용출시키는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
