Claims (12)
- 배지에 인터로이킨-2를 생성할 수 있는 에스케리키아콜리의 형질 전환체를 배양하여 인터로이킨-2를 제조하는 방법에 있어서, 에스케리키아콜리를 pH4.8∼6.0의 배지에 접종하고, 이 pH범위를 유지하면서 성장시킴을 특징으로 하는 개량방법.
- 제1항에 있어서, 형질 전환체가 제1도의 아미노산 서열번호 1∼133을 함유한 인터로이킨-2 또는 인터로이킨-2와 유사한 생리적 활성을 갖는 폴리펩티드를 코오딩하는 DNA를 함유하는 방법.
- 제2항에 있어서, DNA가 인터로이킨-2를 코오딩하는 방법.
- 제2항에 있어 형질 전환체가 DNA로부터 상류에 프로모터를 함유하는 방법.
- 제4항에 있어서, 프로모터가 트립토판 프로모터 또는 트립토판 프로모터들인 방법.
- 제1항에 있어서, 에스케리카콜리가 에스케리키아콜라 K-12로부터 유래된 방법.
- 제1항에 있어서, pH가 5.0∼5.8인 방법.
- 제1항에 있어서, 배지의 pH를 염기성 물질인 무기염기에 의해 조절하는 방법.
- 제8항에 있어서, 무기염기가 암모니아수인 방법.
- 제1항에 있어서, 배지가 글루코오즈 및 카스아미노산을 가한 M-9 배지(변형된M-9배지)인 방법.
- 제1항에 있어서, 교반하면서 통기하에 성장시키는 방법.
