제2도는 ConA로 자극받은 Cl. Ly1+2-/9세포로부터 분리된 mRNA의 수크로즈 구배 침전 부분의 MCGF 활성양을 나타낸다. 18s 및 28s의 위치는 리보좀성 피크를 나타내고,
제3도는 비만세포 증식 인자 활성 및 다계세포 증식인자 활성을 나타내는 cDNA 클론을 운반하는 플라스머드 pcD-MCGF를 나타내며,
제4도는 제3도의 삽입 cDNA의 제한 엔도뉴클레아제절단 지도를 나타낸다.
Claims (13)
- 포유동물의 다계 세포증식인자 활성(mammalian multi-lineage cellular growth factor activity) 및 또는 포유동물의 비만세포 증식인자 활성(mast cell growth factor activity)를 나타내는 폴리펩타이드를 제조하는 방법에 있어서, a) 상기 폴리펩타이드를 코드화하는 뉴클리오티드 서열을 함유하는 벡터(vactor)를 형성하고(여기에서 뉴클리오티드 서열은 벡터를 함유하는 숙주에 의해서 발현될 수 있다.); b) 그 벡터를 숙주내로 넣고; c) 뉴클리오티드서열을 상기의 폴리펩타이트로 전사시키기에 적합한 조건하에서 벡터를 함유한 숙주를 유지시킴을 특징으로 하는 방법.
- 제1항에 있어서, 뉴클레오티드 서열이 상기의 폴리펩타이드를 코드화하는 mRNA 서열로부터 유도된 cDNA 서열인 방법.
- 제1 또는 2항에 있어서,숙주가 벡터로 형질변환(transform)되었거나, 또는 감염(transfect)된 포유동물세포인 방법.
- 제3항에 있어서, 폴리펩타이드가 글리코실화(glycosylate)된 방법.
- 제1내지 4항중의 어느 하나에 있어서, 벡터가 제2 뉴클리오티드 서열에 연결된 폴리펩타이드를 코드화하는 뉴클리오티트를 함유하고, 이 제2 뉴클리오티드 서열은 폴리 펩타이트를 코드화하는 뉴클리오티트서열의 발현을 촉진시키는 프로모터 서열(promotersequence)을 함유하는 방법.
- 제5항에 있어서, 제 2뉴클레어티드 서열이 SV40 바이러스 초기부분 프로모터V40 바이러스 후기 부분 폴리아데닐화 서열을 함유하는 방법.
- 제1 내지 6항의 어느 하나에 있어서, 뉴클레오티드 서열이 조혈 세포증식 활성을 갖는 폴리펩타이드를 코드화하는 방법.
- 제1 내지 7항중의 어느 하나에 있어서, 폴리펩타이드를 코드화하는 뉴클레오티드 서열이 실제로 제1도에 나타낸 서열인 방법.
- 제1 내지 8항중의 어느 하나에 있어서, 폴리펩타이드를 코드화하는 뉴클레오티드서열이 제1도에 나타난 뉴클레오티드 서열과는 다르지만, 제1도에 나타낸 클레오티드서열과 하이 브리드화될 수 있는 방법.
- 제1 내지 9항중의 어느 하나에 있어서, 뉴클레오티드 서열에 폴리펩타이드 선두서열의 적어도 한부분을 코드화하는 부분이 포함되는 방법.
- 세포를 제1도의 하미노산 서열의 거의 전부분을 갖는 폴리펩타이드와 접촉시킴을 특징으로 하여 세포증식을 증진시키는 방법.
- 제11항에 있어서, 세포증식을 시험관내에서 증진시키는 방법.
- 제1도에 나타낸 DNA 서열의 거의 전부분을 함유하는 벡터로 감염 또는 형질전환된 원핵생물(prokarvotic microganism)또는 진핵 생물(eukaryotic cell)을 수성 영양 배지에서 배양시킴을 특징으로 하여, 포유동물 다계 세포증식인자 활성을 나타내는 폴리펩타이드를 제조하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
