재조합 사람 레닌의 제조방법

제1도는 전프로레닌 암호화 DNA의 완전한 누클레오티드 서열 및 추론되는 아미노산 서열이고,
제2도는 전프로레닌을 암호화하고, 포유류 세포 배양물로부터 프로레닌의 분비를 초래하는 발현 벡터의 구성 이며,
제3a도 및 제3b도는 레닌-관련 단백질용 세균성 발현 벡터의 구성이고,
제6도는 AtT20세포의 배지로부터 항-레닌으로 면역 침전된35S-메티오닌 표지 단백질의 SDS-폴리아크릴아마이드 겔 이다.

Claims (7)

  1. 재조합 숙주 세포에 부합하고, 전기 재조합 숙주세포에서 후술하는 DNA서열의 발현을 수행할 수 있는 조정서열(Contr01 Sequence)에 실시 가능하게 연결된 사람레닌, 프로레닌, 또는 전프로레닌(prepro renin)을 암호화하는 DNA서열로 이루어진 발현 시스템.
  2. 제1항에 있어서, 조정 서열이 포유류 숙주와 부합하는 시스템.
  3. 제1항에 있어서, 조정 서열이 세균성 숙주와 부합하는 시스템.
  4. 레닌, 프로레닌, 또는 전프로레닌을 암호화하는 재조합 DNA서열로 형질전환된 재조합 숙주 세포를 배양하고, 배양물로부터 레닌 또는 프로레닌을 회수함을 특징으로 하여, 사람 레닌 또는 프로레닌을 제조하는 방법.
  5. 본래 조절 분비 회로를 사용하는 포유류 세포를 처리하여, 전프로레닌용 발현 시스템을 함유하도록 변형시키고, 분비 촉진제로 전프로레닌 유전자를 발현하도록 유도시킴을 특징으로 하여, 분비, 배양된 완전재조합 레닌을 제조하는 방법.
  6. 소수성 상호작용 크로마토그라피(HIC), 펩스타틴(pepstatin) 크로마토그라피, 및 음이온 교환으로 이루어진, 세포 배양 상등액으로부터 레닌을 정제하는 방법.
  7. 펩스타틴 크로마토그라피, 크기별 제외(size exclusion), 및 음이온 교환으로 이루어진, 세포 배양 상등액으로부터 프로레닌을 정제하는 방법.
