본 발명은 접합체가 약물에 화학적으로 결합된 세포바인딩제를 포함하는, 실질적으로 고순도 및 안정성을 갖는 접합체를 제조하는 방법에 관한 것이다.
암치료는 보다 효과적으로 표적하고 암세포를 사멸하는 약학의 발달로 현저히 진보되었다. 이 때문에, 연구자들은 종양-특이 또는 종양-관련 항원에 결합하는 항체에 기초한 약물을 개발하기 위해 암세포에 의해 선택적으로 발현되는 세포-표면 리셉터 및 항원의 이점을 취하였다. 이와 관련하여, 박테리아 및 식물 독소와 같은 세포독성 분자, 방사성 핵종, 및 특정 화학치료 약물은 종양-특이 또는 종양-관련 세포 표면 항원에 결합하는 모노클로날 항체에 화학적으로 연결된다(참조 예, 국제특허출원 WO 00/02587, WO 02/060955, 및 WO 02/092127, 미국 특허 5,475,092, 6,340,701, 및 6,171,586, 특허출원공개 제 2003/0004210A1, 및 Ghetie et al., J. Immunol. 마이탄시노이드와 같은 설피드릴-함유 세포독성제에 대해 항체를 접합시키는 방법은 이전에 기술되어 있다(참조 예, 미국 특허 5,208,020, 5,416,064 및 6,441,163). 예를 들어, 미국 특허 5,208,020 및 5,416,064에는 항체가 미국 특허 4,149,003, 4,563,304 및 미국 특허 출원 공개 제 2004/0241174 A1에 기재된 바와 같은 헤테로이작용성 제제로 일차 변형된 항체-마이탄시노이드 접합체를 제조하는 방법이 기재되어 있다. 미국 특허 5,208,020 및 5,416,064에는 또한 변형된 항체를 pH 7에서 과량의 설피드릴-함유 세포독성제와 접합시키고, SephadexTM G25 크로마토그래피 컬럼상에서 정제하는 것이 기재되어 있다. 크기 배제 크로마토그래피(SEC)에 의한 항체-약물 접합체의 정제가 또한 기재된바 있다(참조 예, Liu et al., Proc. 상기 항체-약물 접합체의 제조에 대해 앞서 기재된 방법들은 복잡하다. 왜냐하면, 이들은 최적으로 원하여지는 것보다 덜 순수하거나 덜 안정적인 면역접합체를 성취하거나 생성하는데 성가신 단계들로 방해받기 때문이다. 예를 들어, pH 6.0 내지 6.5에서의 접합은 순수하고 안정한 접합체를 생성하는데 최적은 아니다. 또한, 이러한 조건하에서의 접합 반응은 일반적으로 느리며 비효율적이며, 이는 과도 한 시간 및 재료 사용을 요구한다.
순도 및/또는 안정성과 같은 생성물 질을 손상시키지 않고 하나 이상의 제조 단계를 변형 또는 없애는 것이 원하여진다. 일부 옵션이 다른 것들보다 세포바인딩제, 링커, 및 약물의 특정 조합으로 보다 효율적이게 될 정도로 지금까지 기재된 것들보다 추가의 정제 옵션을 갖는 것이 또한 원하여진다.
앞서 살펴본 바와 같이, 실질적으로 고순도이면서 동시에 보다 우수한 안정성을 갖는 세포바인딩제-약물 접합체 조성물을 제조하는 향상된 방법을 개발하는 것이 당 기술분야에 요구된다. 본 발명은 이러한 방법을 제공한다. 본 발명의 이러한 잇점 및 다른 잇점 뿐만 아니라 추가적인 발명적 특징들이 본 명세서에 제공된 본 발명의 상세한 설명으로부터 명확해질 것이다.
본 발명은 약물에 화학적으로 결합된 세포바인딩제를 포함하는 실질적으로 고순도 및 안정성을 갖는 접합체를 제조하는 방법을 제공한다. 상기 방법은 (a) 상기 세포바인딩제에 링커를 공유결합시키기 위해 세포바인딩제와 이작용성 가교제를 접촉시키고 이에 따라 이에 결합된 링커를 갖는 세포바인딩제를 포함하는 제 1 혼합물을 제조하는 단계, (b) 상기 제 1 혼합물을 접선 흐름 여과, 흡착 크로마토그래피, 흡착 여과, 선택적 침전 또는 이의 조합으로 적용하여 이에 결합된 링커를 갖는 정제된 제 1 세포바인딩제 혼합물을 제조하는 단계, (c) 약 pH 4-9를 갖는 용액에서 이에 결합된 링커를 갖는 세포바인딩제를 약물과 반응시켜 상기 정제된 제 1 혼합물에서 이에 결합된 링커를 갖는 세포바인딩제에 약물을 접합시켜 (i) 상기 링커를 통해 상기 약물에 화학적으로 결합된 세포바인딩제, (ii) 유리 약물(free drug), 및 (iii) 반응 부산물을 포함하는 제 2 혼합물을 제조하는 단계, 및 (d) 상기 제 2 혼합물을 접선 흐름 여과, 흡착 크로마토그래피, 흡착 여과, 선택적 침전 또는 이의 조합으로 적용하여 상기 링커를 통해 상기 제 2 혼합물의 다른 성분으로부터 상기 약물에 화학적으로 결합된 세포바인딩제를 정제하고, 이에 따라 정제된 제 2 혼합물을 제조하는 단계를 포함한다.
본 발명은 실질적으로 고순도 및 안정성을 갖는 세포바인딩제-약물 접합체를 제조하는 방법을 제공한다. 이러한 조성물은 상기 접합체의 고순도 및 안정성 때문에 질병 치료에 사용될 수 있다. 마이탄시노이드와 같은 약물에 화학적으로 결합되는 항체와 같은 세포바인딩제를 포함하는 조성물은 예를 들어, 미국 특허 출원 공개 제 2004/0241174 A1에 기재되어 있다. 이러한 정황에서, 실질적으로 고순도는 (a) 접합체 종의 90%이상, 바람직하게 95%이상이 모노머성이며, 그리고/또는 (b) 접합체 제조물내 유리 약물 수준이 (총 약물에 대하여) 2%미만인 것으로 간주된다.
이와 관련하여, 본 발명의 방법은 (a) 상기 세포바인딩제에 링커를 공유결합시키기 위해 세포바인딩제와 이작용성 가교제를 접촉시키고 이에 따라 이에 결합된 링커를 갖는 세포바인딩제를 포함하는 제 1 혼합물을 제조하는 단계, (b) 상기 제 1 혼합물을 접선 흐름 여과, 흡착 크로마토그래피, 흡착 여과, 선택적 침전 또는 이의 조합으로 적용하여 상기 제 1 혼합물의 다른 성분으로부터 이에 결합된 링커를 갖는 세포바인딩제를 정제하고, 이에 따라 이에 결합된 링커를 갖는 정제된 제 1 세포바인딩제 혼합물을 제조하는 단계, (c) 약 pH 4-9를 갖는 용액에서 이에 결합된 링커를 갖는 세포바인딩제를 약물과 반응시켜 상기 정제된 제 1 혼합물에서 이에 결합된 링커를 갖는 세포바인딩제에 약물을 접합시켜 (i) 상기 링커를 통해 상기 약물에 화학적으로 결합된 세포바인딩제, (ii) 유리 약물(free drug), 및 (iii) 반응 부산물을 포함하는 제 2 혼합물을 제조하는 단계, 및 (d) 상기 제 2 혼합물을 접선 흐름 여과, 흡착 크로마토그래피, 흡착 여과, 선택적 침전 또는 이의 조합으로 적용하여 비-접합 약물, 반응물 및 부산물을 제거할 뿐만 아니라 실질적으로 정제된 세포바인딩제-약물 접합체를 획득하는 단계를 포함한다.
바람직하게, 접선 흐름 여과(TFF, 교차 흐름 여과, 한외여과 및 정용여과로도 알려짐) 및/또는 흡착 크로마토그래피 수지가 정제 단계에 이용된다. 그러나, TFF가 제 1 정제 단계(단계 b)에 사용되는 경우, (단계 c)에서 pH 6.0-6.5에서의 접합이 이용되며, 흡착 크로마토그래피 수지가 제 2 정제 단계(단계 d)에 이용되며, 상기 흡착 크로마토그래피 수지는 비이온 교환 수지인 것이 바람직하다. 다른 바람직한 구현으로, TFF는 두 정제 단계 모두에 이용되거나, 흡착 크로마토그래피 수지는 두 정제 단계 모두에 이용된다. 택일적으로, 흡착 크로마토그래피 수지가 제 1 정제 단계에 사용되며, TFF는 제 2 정제 단계에 이용된다. TFF와 흡착 크로마토그래피 수지의 조합이 또한 제 1 및/또는 제 2 정제 단계에 이용될 수 있다.
Pellicon 타입 시스템(Millipore, Billerica, MA), Sartocon Cassette 시스템(Sartorious AG, Edgewood, NY) 및 Centrasette 타입 시스템(Pall Corp., East Hills, NY)을 포함하는 어느 적절한 TFF 시스템이 이용될 수 있다.
어느 적절한 흡착 크로마토그래피 수지가 이용될 수 있다. 바람직한 흡착 크로마토그래피 수지는 히드록시아파타이트 크로마토그래피, 소수성 전하 유도 크로마토그래피(HCIC), 소수성 상호작용 크로마토그래피(HIC), 이온 교환 크로마토그래피, 혼합 방식 이온 교환 크로마토그래피, 고정화 금속 친화성 크로마토그래피(IMAC), 염료 리간드 크로마토그래피, 친화성 크로마토그래피, 역상 크로마토그래피, 및 이의 조합에 대한 수지들을 포함한다. 적절한 히드록시아파타이트 수지의 예는 세라믹 히드록시아파타이트(CHT Type I 및 Type II, Bio-Rad Laboratories, Hercules, CA), HA Ultrogel 히드록시아파타이트(Pall Corp., East Hills, NY), 및 세라믹 불화인회석(CFT Type I 및 Type II, Bio-Rad Laboratories, Hercules, CA)를 포함한다. 적절한 HCIC 수지의 예는 MEP Hypercel 수지(Pall Corp., East Hills, NY)이다. 적절한 HIC 수지의 예는 Butyl-Sepharose, Hexyl-Sepaharose, Phenyl-Sepharose, 및 Octyl Sepharose 수지(모두 GE Healthcare, Piscataway, NJ), 뿐만 아니라 Macro-prep Methyl 및 Macro-Prep t-Butyl 수지(Biorad Laboratories, Hercules, CA)를 포함한다. 적절한 이온 교환 수지의 예는 SP-Sepharose, CM-Sepharose, 및 Q-Sepharose 수지(모두 GE Healthcare, Piscataway, NJ), 및 Unosphere S 수지(Biorad Laboratories, Hercules, CA)를 포함한다. 적절한 혼합 방식 이온 교환기의 예는 Bakerbond ABx 수지(JT Baker, Phillipsburg NJ)를 포함한다. 적절한 IMAC 수지의 예는 Chelating Sepharose 수지(GE Healthcare, Piscataway, NJ) 및 Profinity IMAC 수지(Bio-Rad Laboratories, Hercules, CA)를 포함한다. 적절한 염료 리간드 수지의 예는 Blue Sepharose 수지(GE Healthcare, Piscataway, NJ) 및 Affi-gel Blue 수지(Bio-Rad Laboratories, Hercules, CA)를 포함한다. 적절한 친화성 수지의 예는 세포바인딩제가 항체인 Protein A Sepharose 수지(예, MabSelect, GE Healthcare, Piscataway, NJ), 및 예를 들어, Lentil Lectin Sepharose 수지(GE Healthcare, Piscataway, NJ)와 같이 세포 바인딩제가 적절한 렉틴 결합부를 갖는 렉틴 친화성 수지를 포함한다. 택일적으로 상기 세포바인딩제에 특이적인 항체가 사용될 수 있다. 이러한 항체는 예를 들어, Sepharose 4 Fast Flow 수지(GE Healthcare, Piscataway, NJ)에 고정될 수 있다. 적절한 역상 수지의 예는 C4, C8 및 C18 수지(Grace Vydac, Hesperia, CA)를 포함한다.
본 발명의 방법에 따라, 이에 결합된 링커를 갖는 세포바인딩제 뿐만 아니라 반응물 및 다른 부산물을 포함하는 제 1 혼합물이 제조된다. 반응물 및 부산물로부터 변형 세포바인딩제의 정제는 상기 제 1 혼합물을 정제 공정에 적용하여 수행된다. 이와 관련하여, 상기 제 1 혼합물은 예를 들어, 멤브레인-기초 접선 흐름 여과 공정과 같은 접선 흐름 여과(TFF), 흡착 크로마토그래피, 흡착 여과, 또는 선택적 침전, 또는 어느 다른 적절한 정제 공정 뿐만 아니라 이의 조합을 이용하여 정제될 수 있다. 이러한 제 1 정제 단계는 정제된 제 1 혼합물을 제공한다. 즉, 본 발명에 따른 정제 전의 상기 제 1 혼합물에 비하여 증가된 농도의 결합된 링커를 갖는 세포바인딩제 및 감소된 양의 결합되지 않은 이작용성 가교제를 제공한다.
결합된 링커를 갖는 세포바인딩제의 정제된 제 1 혼합물을 얻기 위해 상기 제 1 혼합물의 정제 후, 약 pH 4-9를 갖는 용액에서 상기 결합된 링커를 갖는 세포바인딩제와 약물을 반응시킴으로써 상기 제 1 정제 혼합물에서 결합된 링커를 갖는 세포바인딩제에 약물이 접합되며, 그 결과, (i) 상기 링커를 통해 상기 약물에 화학적으로 결합된 세포바인딩제, (ii) 유리 약물, 및 (iii) 부산물을 포함하는 제 2 혼합물이 생성된다. 접합 반응은 약 pH 4-9에서 수행되며, 상기 반응은 바람직하게 약 pH 6 또는 그 이하에서, 또는 약 pH 6.5 또는 그 이상에서 수행되며, 가장 바람직하게는 약 pH 4-6에서, 또는 약 pH 6.5-9에서, 특히 4 내지 6 미만의 pH 또는 6.5이상 내지 9의 pH에서 수행된다. 접합 단계가 약 6.5 또는 그 이상의 pH에서 수행되는 경우에 일부 설피드릴-함유 약물은 디설피드-결합 형성에 의해 다이머화되는 경향이 있다. 반응 혼합물로부터 미량 금속 및/또는 산소의 제거 뿐만 아니라 산화방지제의 임의의 첨가 또는 보다 반응적인 이탈기를 갖는 링커의 사용, 또는 1 분취액이상의 약물 첨가가 이러한 상황에서 효율적인 반응을 위해 필요할 수 있다.
임의로, 변형된 세포바인딩제의 정제는 생략될 수 있다. 이러한 경우에, 상기 약물은 가교제와 함께 동시에 첨가되거나, 또는 상기 세포바인딩제에 가교제를 첨가한 후 1, 2, 3 또는 그 이상의 시간 후와 같이 다소 늦은 시점에 첨가될 수 있다.
본 발명의 방법은 임의로 상기 세포바인딩제-약물 접합체의 용해도 및 회수를 증가시키기 위해 본 발명의 방법에 사용된 접합 단계에 수크로즈의 첨가를 포함한다. 바람직하게, 수크로즈는 약 0.1-20%(w/v)(예, 약 0.1%(w/v), 1%(w/v), 5%(w/v), 10%(w/v), 15%(w/v), 또는 20%(w/v))의 농도로 첨가된다. 바람직하게, 수크로즈는 약 1-10%(w/v)(예, 약 2%(w/v), 약 4%(w/v), 약 6%(w/v), 또는 약 8%(w/v))의 농도로 첨가된다. 또한, 접합 반응은 완충제의 첨가를 포함할 수 있다. 당해 기술분야에 알려진 어느 적절한 완충제가 사용될 수 있다. 적절한 완충제는 예를 들어, 시트레이트 버퍼, 아세테이트 버퍼, 숙시네이트 버퍼, 및 포스페이트 버퍼를 포함한다.
접합 단계 이후에, 상기 제 2 혼합물은 정제 단계가 아루어진다. 이와 관련하여, 상기 제 2 혼합물은 상기 언급한 바와 같이, 예를 들어, 멤브레인-기초 접선 흐름 여과 공정과 같은 접선 흐름 여과(TFF), 흡착 크로마토그래피, 흡착 여과, 또는 선택적 침전, 또는 어느 다른 적절한 정제 공정 뿐만 아니라 이의 조합을 이용하여 정제될 수 있다. 이러한 제 2 정제 단계는 정제된 제 2 혼합물을 제공한다. 즉, 본 발명에 따른 정제 전의 상기 제 2 혼합물에 비하여 상기 링커를 통해 화학적으로 결합된 세포바인딩제의 증가된 농도 및 상기 제 2 혼합물의 하나 이상의 다른 성분의 감소된 양을 제공한다.
상기 세포바인딩제는 세포, 전형적으로 그리고 바람직하게 동물 세포(예, 사람 세포)에 바인딩하는 어느 적절한 제제일 수 있다. 상기 세포바인딩제는 바람직하게 펩타이드 또는 폴리펩타이드이다. 적절한 세포바인딩제는 예를 들어, 항체(예, 모노클로날 항체 및 이의 프레그먼트), 림포카인, 호르몬, 성장인자, 영양분-수송 분자(예, 트랜스페린) 및 세포 표면상의 표적 분자를 특이적으로 바인딩하는 어느 다른 제제 또는 분자를 포함한다.
본 명세서에 사용된 용어 "항체"는 Fab, F(ab')2, dsFv, sFv, 다이어바디(diabodies)와 같은 어느 면역글로블린, 어느 면역글로블린 프레그먼트, 또는 세포 표면상의 (예를 들어, 상보성결정지역(complementarity determining region, CDR)을 함유하는) 항원에 결합할 수 있는 면역글로블린 키메라를 칭한다. 어느 적절한 항체가 세포바인딩제로 사용될 수 있다. 당업자는 적절한 항체의 선택이 표적하고자 하는 세포집단에 따라 달라질 것임을 인식할 것이다. 이와 관련하여, 특정 세포 집단(전형적으로 그리고 바람직하게 병에 걸린 세포 집단)에서 선택적으로 발현되는 세포 표면 분자(즉, 항원)의 타입 및 수는 본 발명의 조성물에 사용되는 적절한 항체의 선택을 결정할 것이다. 세포 표면 발현 프로필은 종양 세포 타입을 포함하는 광범위하게 다양한 세포 타입에 대해 알려져 있으며, 또는 알려지지 않은 경우에 통상적인 분자생물학 및 조직 화학 기술을 이용하여 검출될 수 있다.
상기 항체는 폴리클로날이거나 모노클로날일 수 있으나, 가장 바람직하게 모노클로날 항체이다. 본 명세서에 사용된 "폴리클로날" 항체는 전형적으로 면역화 동물의 혈청에 함유된 항체 분자의 이종성 집단을 칭한다. "모노클로날" 항체는 특정 항원에 특이적인 항체 분자의 동종성 집단을 칭한다. 모노클로날 항체는 전형적으로 B 림프구("B 세포")의 단일 클론에 의해 생성된다. 모노클로날 항체는 표준 하이브리도만 기술을 포함하는 당업자에게 알려진 다양한 기술을 이용하여 획득될 수 있다(참조 예, Kohler 및 Milstein, Eur. J. 상기 모노클로날 항체는 어느 적절한 동물로부터 분리되거나 생성될 수 있으나, 바람직하게 포유류에서, 보다 바람직하게 마우스 또는 사람에서, 가장 바람직하게는 사람에서 생성된다. 마우스에서 항체를 생성하는 방법은 당업자에게 잘 알려져 있으며 본 명세서에 설명된다. 사람 항체와 관련하여, 당업자는 폴리클로날 항체가 적절한 항원으로 백신접종되거나 면역화된 사람 대상자의 혈청으로부터 분리될 수 있다는 것을 인식할 것이다. 택일적으로, 사람 항체는 마우스와 같은 사람이 아닌 동물에서 사람 항체를 생성하는 공지 기술을 적용하여 생성될 수 있다(참조 예, 미국 특허 5,545,806, 5,569,825, 및 5,714,352, 및 미국 특허 출원 공개 제 2002/0197266 A1).
사람에서 치료학적 적용을 위한 이상적인 선택이지만 사람 항체, 특히 사람 모노클로날 항체는 전형적으로 마우스 모노클로날 항체보다 생성하기 어렵다. 그러나, 마우스 모노클로날 항체는 사람에 투여되는 경우에 신속한 숙주 항체 반응을 유도하며, 이는 항체-약물 접합체의 치료학적 또는 진단학적 잠재력을 감소시킬 수 있다. 이러한 문제를 회피하기 위해, 모노클로날 항체는 바람직하게 사람 면역 시스템에 의해 "외래"로 인지되지 않는다.

마이탄시노이드에 부가적으로, 상기 접합체에 사용되는 세포독성제는 탁산 또는 이의 유도체일 수 있다. 탁산은 파클리탁셀(Taxol®), 세포독성 천연 산물, 및 도세탁셀(Taxotere®), 반-합성 유도체를 포함하는 화합물류이며, 이들은 모두 암치료에 널리 사용된다. 탁산은 튜블린의 디폴리머화를 저해하여 세포 사멸을 일으키는 유사분열 스핀들 활성 억제제이다. 도세탁셀 및 파클리탁셀은 암 치료에 유용한 제제이나, 이들의 항종양 활성은 정상 세포에 대한 이들의 비-특이적 독성 때문에 제한된다. 또한, 파클리탁셀 및 도세탁셀과 같은 화합물 자체는 세포바인딩제의 접합체에 사용되기에 충분히 강력하지 못하다.

항체와 같은 세포바인딩제에 탁산을 접합시키는 방법과 함께 본 발명의 정황에 사용될 수 있는 탁산 합성 방법은 미국 특허 5,416,064, 5,475,092, 6,340,701, 6,372,738, 6,436,931, 6,596,757, 6,706,708, 및 6,716,821, 및 미국 특허 출원 공개 제 2004/0024049 A1에 상세히 기재되어 있다.
세포독성제는 또한 CC-1065 또는 이의 유도체일 수 있다. CC-1065는 스트렙토마이세스 젤렌시스(Streptomyces zelensis)의 배양액으로부터 분리된 강력한 항종양 항생물질이다. 메소트렉세이트, 다우노루비신, 독소루비신, 빈크리스틴, 빈블라스틴, 멜팔란, 미토마이신 C, 클로람부실, 칼리키아미신, 튜블리신 및 튜블리신 유사체, 듀오카마이신 및 듀오카마이신 유사체, 돌래스타틴 및 돌라스타틴 유사체와 같은 약물이 또한 본 발명의 정황에 사용될 수 있다. 독사루비신 및 다우노루비신 화합물(참조 예, 미국 특허 6,630,579)이 또한 약물로 사용될 수 있다.
상기 약물 접합체는 시험관내 방법에 의해 제조될 수 있다. 약물 또는 프로드럭을 항체에 연결하기 위해, 연결기가 사용된다. 적절한 연결기가 당해 기술분야에 잘 알려져 있으며, 디설피드기, 산 분해성 기, 광 분해성 기, 펩티다아제 분해성 기, 및 에스테라아제 분해성 기를 포함한다. 바람직한 연결기는 디설피드기이다. 예를 들어, 접합체는 항체와 약물 또는 프로드럭 사이의 디설피드 교환 반응을 이용하여 제작될 수 있다. 약물 분자는 또한 혈청 알부민과 같은 매개 캐리어 분자를 통해 세포바인딩제에 연결될 수 있다.

분할성 링커가 본 발명의 방법에 사용되나, 비-분할성 링커가 또한 상기한 접합체를 생성하는데 사용될 수 있다. 비-분할성 링커는 마이탄시노이드, 탁산, 또는 CC-1065 유사체와 같은 약물을 안정한 공유 방식으로 세포바인딩제에 연결할 수 있는 어느 화학 부이다. 따라서, 비-분할성 링커는 상기 약물 또는 세포바인딩제가 활성을 보유하는 조건에서 산-유도 분할, 광-유도 분할, 펩티다아제-유도 분할, 에스테라아제-유도 분할, 및 디설피드 결합 분할에 실질적으로 저항적이다.

하기 실시예는 본 발명을 보다 설명하나 이의 범위를 어느 방식으로 한정하려는 것은 아니다.
실시예 1
본 실시예는 TFF를 이용한 헤테로이작용성 변형제로 변형된 항체의 정제를 나타낸다.
huN901 모노클로날 항체(최종 농도 8mg/ml)를 N-숙신이미딜 4-(2-피리딜디티오)펜타노에이트(SPP, 5.6-배 몰 초과)로 약 189분간 20℃에서 50mM NaCl, 2mM EDTA, 및 5% 에탄올을 함유하는 50mM 포타슘 포스페이트 버퍼(pH 7.5)에서 배양하였다. 제 1 그룹으로, 반응 혼합물은 평형된 SephadexTM G25F 수지 컬럼을 이용하여 정제되고, 50mM NaCl 및 2mM EDTA를 함유하는 50mM 포타슘 포스페이트 버퍼(pH 6.5)에서 용출되었다. 제 2 그룹으로, 반응 혼합물은 Pellicon XL TFF 시스템(Millipore, Billerica, MA)를 이용하여 정제되고, 항체는 10,000 분자량 컷오프 멤브레인(UltracelTM 재생 셀룰로오즈 멤브레인, Millipore, Billerica, MA)을 이용하여 50mM 포타슘 포스페이트, 50mM NaCl(pH 6.5) 및 2mM EDTA내로 초여과되었다(5 볼륨). 두 시료는 모두 50mM NaCl 및 최종 농도 3% DMA를 함유하는 포타슘 포스페이트 버퍼에서 pH 6.5에서 18시간동안 DM1(바인딩되지 않은 링커보다 1.7배 몰 초과)과 접합되었다.
두 그룹 모두에서, 수율은 조합된 변형 및 정제 단계에 있어서 분광광도계로(280nm 파장) 측정되었다. 343nM에서 8,080M-1cm-1의 흡광계수를 갖는 피리딘-2-티온을 방출시키는 디티오트레이톨 처리로 링커/항체 비율이 또한 측정되었다. 약물/항체 비율은 접합 단계에 있어서 분광광도계로(280nm 및 252nm 파장) 측정되었다. 또한, SPP-관련 소분자종의 제거는 Hisep HPLC에 의해 측정되었다.

실시예 2
본 실시예는 흡착 크로마토그래피를 이용한 헤테로이작용성 변형제로 변형된 항체의 정제를 나타낸다.
huB4 항체는 50mM NaCl, 2mM EDTA, 및 5% 에탄올을 함유하는 50mM 포타슘 포스페이트 버퍼(pH 6.5)에서 실온에서 120분간 N-숙신이미딜 4-(2-피리딜디티오)부타노에이트(SPDB, 5.4배 몰 초과)로 변형되었다. 제 1 그룹으로, 반응 혼합물은 실시예 1에 기재된 SephadexTM G25F 수지를 이용하여 정제되었다. 제 2 그룹으로, 반응 혼합물은 세라믹 히드록시아파타이트 컬럼(CHT, Bio-Rad Laboratories, Hercules, CA)에 로딩되었으며, 이는 12.5mM 포타슘 포스페이트 버퍼(pH 6.5)로 평형되고 80mM 포타슘 포스페이트 버퍼(pH 6.5)로 용출되었다.
두 그룹 모두에서 수율 및 링커/항체 비율은 실시예 1에 기재된 바와 같이 측정되었다. 제 1 그룹은 91% 수율 및 4.2 링커/항체 비율을 가졌다. 제 2 그룹은 89% 수율 및 4.2 링커/항체 비율을 가졌다.
CNTO95 항체(최종 농도 10mg/ml)는 2.7% 수크로즈 및 5% 에탄올을 함유하는 10mM 소디움 포스페이트 버퍼(pH 7.5)에서 20℃에서 120분간 N-숙신이미딜 4-(2-피리딜디티오)부타노에이트(SPDB, 4.5배 몰 초과)로 변형되었다. 제 1 그룹으로, 반응 혼합물은 12.5mM NaCl 및 0.5mM EDTA를 함유하는 12.5mM 포타슘 포스페이트 버퍼(pH 6.6)에서 SephadexTM G25F 수지를 이용하여 정제되었다. 제 2 그룹으로, 반응 혼합물은 SP Sepharose Fast Flow 컬럼(GE Healthcare, Piscataway, NJ)에 로딩되었으며, 이는 10mM 소디움 포스페이트 버퍼(pH 7.5)로 평형되고 50mM NaCl을 함유하는 50mM 포타슘 포스페이트 버퍼(pH 7.5)로 용출되었다.
두 그룹 모두에서, 수율 및 링커/항체 비율은 실시예 1에 기재된 바와 같이 검출되었다. 제 1 그룹은 96% 수율 및 4.0 링커/항체 비율을 가졌다. 제 2 그룹은 97% 수율 및 4.1 링커/항체 비율을 가졌다.
본 실시예에서 얻어진 데이타는 흡착 크로마토그래피가 헤테로이작용성 변형제로 변형된 항체를 정제하는데 사용될 수 있음을 입증한다.
실시예 3
본 실시예는 pH 6.5 이상에서 약물과 변형 항체를 접합시키는 유익한 효과를 나타낸다.
제 1 실험으로, CNTO95 항체를 실시예 2에 기재된 바와 같이 변형하고 정제하였다. 변형된 항체는 그 다음 두 그룹으로 나뉘었다. 제 1 그룹에서 접합은 12.5mM NaCl, 0.5mM EDTA, 3% DMA, 및 링커당 1.7 배 몰 초과 약물을 함유하는 12.5mM 포타슘 포스페이트(pH 6.5)에서 20℃에서 수행되었다. 제 2 그룹에서, 접합 반응은 pH 7.5에서 수행되었다. 접합된 항체는 NAP-10 컬럼에 걸쳐 정제되었다.
약물/항체 비율은 두 그룹 모두에 대해 측정되었다. 그 결과 데이타를 표 2에 나타내었다.

제 2 실험으로, huB4 인간화 모노클로날 항체는 (a) 항체에 비해 4.9배 몰 초과의 SPDB 또는 (b) 항체에 비해 4.8배 몰 초과의 SPDB로 변형되었다. 양 상황에서, 반응은 5% 에탄올에 담긴 50mM 포타슘 포스페이트, 50mM 포타슘 클로라이드, 및 2mM EDTA(pH 6.5)에서 실온에서 총 120분간 수행되었다. 시료 (a)는 pH 6.5에서 50mM 포타슘 포스페이트, 50mM 소디움 클로라이드, 및 2mM EDTA에서 평형된 SephadexTM G25F 수지 컬럼에 걸쳐 정제되었다. 시료 (b)는 크로마토그래피 버퍼가 pH 7.5로 적정된 것을 제외하고 동등하게 정제되었다. 양 시료는 최종 농도 디메틸아세트아미드(DMA) 3%에서 실온에서 18시간동안 DM4(바인딩된 링커에 비해 1.7배 몰 초과)로 접합되었다.
따라서, 시료 (a)는 pH 6.5에서 접합되었고, 시료 (b)는 pH 7.5에서 접합되었다. 그 다음 상기 시료들은 pH 6.5에서 9.6mM 포타슘 포스페이트 및 4.2mM 소디움 클로라이드로 평형된 SephadexTM G25F 수지 컬럼으로 정제되었다. 두 시료는 모두 4℃에서 7개월까지 배양되었고, 간격을 두고 방출 유리 약물을 분석하였다. 그 결과 데이타를 표 3에 나타내었다.

표 3에 데이타로 나타낸 바와 같이, 유리 약물의 방출은 pH 6.5에서 접합된 시료 (a)에 비해 pH 7.5에서 접합된 시료 (b)에서 실질적으로 보다 느렸다. 따라서, pH 7.5에서 제조된 약물 접합체 산물은 pH 6.5에서 제조된 약물 접합체 산물에 비해 시간에 걸친 방출과 관련하여 보다 안정한 것으로 나타났다. pH 7.5에서의 접합은 또한 pH 6.5에서보다 보다 나은 약물 편입을 보여, 이에 따라 사용되는 약물이 보다 적게 요구된다.
실시예 4
본 실시예는 pH 6.0이하에서 변형 항체와 약물을 접합시키는 유익한 효과를 나타낸다.
huN901 모노클로날 항체(최종 농도 8mg/ml)를 50mM NaCl, 2mM EDTA, 및 5% 에탄올을 함유하는 50mM 포타슘 포스페이트 버퍼(pH 7.5)에서 N-숙신이미딜 4-(2-피리딜디티오)펜타노에이트(SPP, 5.6배 몰 초과)와 함께 약 180분간 20℃에서 배양하였다. 제 1 그룹으로, 반응 혼합물은 50mM NaCl 및 2mM EDTA를 함유하는 50mM 소 디움 시트레이트 버퍼(pH 5.0)에서 평형되고 용출된 SephadexTM G25F 수지 컬럼을 이용하여 정제되었다. 제 2 그룹으로, 반응 혼합물은 50mM NaCl 및 2mM EDTA를 함유하는 50mM 포타슘 포스페이트 버퍼(pH 6.5)에서 평형되고 용출된 SephadexTM G25F 수지 컬럼을 이용하여 정제되었다. 두 시료는 모두 최종 농도 디메틸아세트아미드(DMA) 3%에서 실온에서 3, 19, 25, 48, 및 120시간동안 DM4(바인딩된 링커에 비해 1.7배 몰 초과)와 접합되었다.
따라서, 제 1 그룹의 시료는 50mM NaCl 및 2mM EDTA를 함유하는 50mM 소디움 시트레이트 버퍼(pH 5.0)에서 접합되었으며, 제 2 그룹의 시료는 50mM NaCl 및 2mM EDTA를 함유하는 50mM 소디움 포스페이트 버퍼(pH 6.5)에서 접합되었다. 그 다음 시료들은 50mM NaCl을 함유하는 50mM 포타슘 포스페이트 버퍼(pH 6.5)에서 평형 및 용출된 SephadexTM G25F 수지 컬럼을 이용하여 정제되었다.
두 그룹 모두에서, 링커/항체 비율은 343nM에서 8,080M-1cm-1의 흡광계수를 갖는 피리딘-2-티온을 방출시키는 디티오트레이톨 처리에 의해 측정되었다. 약물/항체 비율은 접합 단계에 있어서 분광광도계로(280nm 및 252nm 파장) 측정되었다.
제 1 그룹은 4.3 링커/항체 비율을 가졌다. 제 2 그룹은 4.2 링커/항체 비율을 가졌다.

표 4에 나타낸 데이타로부터 알 수 있는 바와 같이, pH 5.0에서 변형 항체와 약물을 접합하여 제조된 접합체는 pH 6.5에서 접합이 이루어진 접합체보다 접합 반응 코스 도중에 보다 높고 보다 안정한 결합 약물 수준을 달성한다. 증가된 안정성에 부가적으로, 상기 결과는 보다 높은 약물/항체 수준은 pH 6.5의 접합에서 동일한 양의 약물을 이용하는 경우에 비해 pH 5.0에서 접합되는 경우에 달성됨을 보여주며, 이에 따라 pH 5.0에서 약물이 보다 효율적으로 사용됨을 보여준다.
두 그룹 모두에서, 접합체 모노머 양은 시간에 걸쳐 검출되었다. 그 결과 데이타를 표 5에 나타내었다.

실시예 5
본 실시예는 pH 6미만에서 변형 항체에 약물을 접합시키는 잇점을 더욱 나타낸다.
BIWA 4 항체는 50mM 포타슘 포스페이트 버퍼(pH 6.5), 50mM NaCl, 2mM EDTA, 및 5% 에탄올에서 실온에서 120-140분간 SPP(표 6에 나타낸 바와 같이 몰 초과의 SPP)로 변형되었다. 변형 항체 분액을 다양한 pH 값(pH 4.6-6.5)을 갖는 버퍼에서 평형된 각 NAP 25 컬럼에서 정제되었다. pH 4.6-5.9 버퍼는 35mM 소디움 시트레이트, 150mM 소디움 클로라이드, 및 2mM EDTA로 구성되었다. pH 6.5 버퍼는 2mM EDTA를 함유한 PBS이었다.
각 pH에서 변형된 항체는 디메틸아세트아미드(DMA, 최종 농도 3%)에서 DM1(링커에 비해 1.7배 몰 초과)과 접합되었다. 실온에서 17-18시간동안 배양 후, 접합된 항체 시료를 PBS(pH 6.5)에서 평형된 NAP 25 컬럼에서 크로마토그래피로 정제하였다. 링커/항체 비율(표 6에서 L/A)은 343nM에서 8,080M-1cm-1의 흡광계수를 갖는 피리딘-2-티온을 방출시키는 디티오트레이톨 처리에 의해 측정되었다. 약물/항체 비율은 접합 단계에 있어서 분광광도계로(280nm 및 252nm 파장) 측정되었다. 접 합체 모노머, 고분자량종, 및 저분자량종은 0.2M 포타슘 클로라이드 및 20% 이소프로판올을 함유하는 0.2M 포타슘 포스페이트 버퍼(pH 7.0)에서 평형 및 디벨로핑된 TSKG3000SWXL 컬럼을 이용한 SEC-HPLC에 의해 검출되었다.

표 6에 나타낸 데이타는 SPP-변형 BIWA 4와 DM1의 접합이 pH 6.5에서의 접합에 비해 pH 6.0이하에서 효율적임을 나타낸다. 특정 최종 약물/항체에 이르는 링커와 약물의 양, 특히 SPP 링커와 DM1의 양은 보다 낮은 pH에서 감소하였다. 또한, 보다 낮은 pH에서 접합체 모노머, 고분자량종, 및 저분자량종의 수준이 보다 최적이었으며, 수율이 향상되었다.
실시예 6
본 실시예는 변형 항체 정제 단계가 임의로 생략될 수 있음을 나타낸다. 상기 약물은 이작용성 변형제와 동시에 첨가되거나 다소 늦은 시기에 첨가될 수 있다.
변형제 다음에 약물을 첨가하는 예로, 인간화 모노클로날 항체 CNTO95는 20℃에서 120분간 항체에 걸쳐 4.6 몰 초과의 SPDB에서 이작용성 변형제 SPDB와 20mg/mL의 농도로 변형되었다. 변형 버퍼는 5.3% 수크로즈 및 5% 에탄올을 함유하는 44mM 포스페이트 버퍼(pH 7.5)이었다. 변형 항체의 한 분액은 12.5mM NaCl을 함유하는 12.5mM 포타슘 포스페이트 버퍼(pH 7.5)에서 평형 및 용출된 SephadexTM G25F 수지(표준 4-단계 공정)에 걸쳐 정제되었으며, 후속적으로 실온에서 20시간동안 12.5mM NaCl 및 10% DMA를 함유하는 12.5mM 포타슘 포스페이트 버퍼(pH 7.5)에서 최종 변형 항체 농도 10mg/mL에서 DM4(바인딩된 링커에 비해 1.7배 몰 초과 약물)와 접합되었다. 변형 항체의 제 2 분액은 추가 정제없이 120분 변형 반응의 마지막에 즉시 접합되었다(3단계 공정).
변형 반응 혼합물의 단백질 및 버퍼 농도는 10mg/mL의 변형 단백질 농도 및 5.9mM NaCl 및 2.7% 수크로즈를 함유하는 28mM 포타슘 포스페이트(pH 7.5)의 버퍼 조성을 수득하도록 조절되었다. 그 다음 DM4를 첨가하고(출발 SPDB에 비해 1.7배 몰 초과), DMA를 최종 농도 10%로 조절하였다. 실온에서 20시간 배양 후, 접합 항체의 두 분액을 pH 5.5에서 10mM 히스티딘 및 10% 수크로즈에서 평형된 SephadexTM G25F 수지에서 정제되었다.
링커/항체 비율(L/A)은 343nM에서 8,080M-1cm-1의 흡광계수를 갖는 피리딘-2-티온을 방출시키는 디티오트레이톨 처리에 의해 측정되었다. 약물/항체(D/A) 비율 및 수율은 접합 단계에 있어서 분광광도계로(280nm 및 252nm 파장) 측정되었다. 모 노머의 퍼센트는 SEC-HPLC에 의해 검사되었다. 유리 약물의 퍼센트는 Hisep 컬럼에서 HPLC에 의해 검사되었다. 이러한 분석 결과를 표 7에 나타내었다.

실시예 7
본 실시예는 헤테로이작용성 변형제로 변형된 다음 마이탄시노이드로 접합된 항체를 정제하는 향상된 수단을 나타낸다.
실시예 1에 기재한 바와 같이, SPP(7배 몰 초과)로 변형되고 SephadexTM G25F 수지에서 정제된 huN901 항체를 마이탄시노이드 DM1(3% 최종 농도 디메틸아세트아미드(DMA)에 용해된, 링커에 비해 1.7배 몰 초과)과 접합하였다.
제 1 접합체 시료를 포스페이트 완충 염수(PBS, pH 6.5)에서 SephadexTM G25F 수지상의 표준 크로마토그래피에 의해 정제하였다.
제 2 접합체 시료는 실시예 1에 기재된 바와 같이 Pellicon XL TFF 시스템(Millipore, Billerica, MA)에 의해 정제하였다.
제 3 접합체 시료는 50mM Tris(pH 8.0)에서 평형되고, 50mM 소디움 아세테이트(pH 4.0)으로 용출된 MEP Hypercell 수지 컬럼을 이용하여 정제하였다.
제 4 접합체 시료는 50mM 소디움 포스페이트(pH 6.5)에서 평형되고, 0.2M NaCl 및 50mM 소디움 포스페이트(pH 6.5)로 용출된 UNOsphere S 수지 컬럼을 이용하여 정제하였다.
제 5 접합체 시료는 50mM 소디움 포스페이트(pH 6.5)에서 평형되고, 0.3M NaCl 및 50mM 소디움 포스페이트(pH 6.5)로 용출된 CHT 수지(Bio-Rad Laboratories, Hercules, CA) 컬럼을 이용하여 정제하였다.
제 6 접합체 시료는 35mM 소디움 시트레이트, 10mM 소디움 클로라이드(pH 5.0)에서 평형되고 0.25M NaCl, 35mM 소디움 시트레이트(pH 5.0)로 용출된 SP Sepharose 수지 컬럼을 이용하여 정제되었다.
접합체 모노머는 0.2M 포타슘 클로라이드 및 20% 이소프로판올을 함유하는 pH 7.0의 0.2M 포타슘 포스페이트 버퍼에서 평형 및 디벨로핑된 TSKG3000SWXL 수지 컬럼을 이용하여 SEC-HPCL에 의해 검출되었다. 접합 단계 수율은 접합된 항체의 수율을 (280nM 파장에서 분광 광도계로 측정된) 접합된 변형 항체의 양으로 나누어 검출되었다.
이러한 분석 결과를 표 8에 나타내었다.

표 8의 결과는 본 발명의 정제 방법 모두(그룹 2-6)는 대조군(그룹 1)으로 획득된 수율과 유사한 수율을 제공하였다. 각 본 발명의 크로마토그래프 방법은 접합체 모노머의 수준 향상을 생성하였으며 정률증가하는 경향이 있다.
CHT(세라믹 히드록시아파타이트)에 부가적으로, CFT(세라믹 플루오로아파타이트)가 유사한 크로마코그래프 조건하에서 사용될 수 있다. 택일적으로 원하는 산물(실질적으로 모노머성 접합체)은 상기 수지에 의해 보유되지 않으나, 고분자량종이 보유되어 이에 따라 원하는 산물로부터 분리되도록 CHT 및 CFT 수지 모두 비-흡착 방식으로 사용될 수 있다.
접합을 위한 표준 버퍼/용매 조성물은 (실시예 1에서 사용된 것과 같이) pH 6.5의 3% DMA, 50mM 포타슘 포스페이트, 50mM NaCl, 및 2mM EDTA를 포함하나, 다른 조성물이 본 명세서에 기술한 일부 크로마토그래프 단계에 더욱 호환될 수 있으며, 표준 방법에 비해 다른 잇점을 제공한다. 예를 들어, 접합은 pH 6.5에서 3% DMA, 12.5mM 포타슘 포스페이트, 12.5mM NaCl, 및 0.5mM EDTA에서 수행될 수 있다. 이러한 조건하에서, huB4 항체에 편입된 링커의 양에 비해 편입된 DM4의 양은 표준 조 건에 비해 약 10% 높았다. 또한, 이러한 조건은 양이온 교환 및 CHT 수지와 같이 수지상의 로딩과 호환될 수 있다.

Claims (41)
- (a) 세포바인딩제에 링커를 공유결합시키기 위해 세포바인딩제와 이작용성 가교제를 접촉시키고 이에 따라 이에 결합된 링커를 갖는 세포바인딩제를 포함하는 제 1 혼합물을 제조하는 단계,(b) 상기 제 1 혼합물을 접선 흐름 여과, 선택적 침전, 흡착 여과, 또는 흡착 크로마토그래피 수지에 적용하여 이에 결합된 링커를 갖는 정제된 제 1 세포바인딩제 혼합물을 제조하는 단계,(c) 약 pH 4-9를 갖는 용액에서 이에 결합된 링커를 갖는 세포바인딩제를 약물과 반응시켜 상기 정제된 제 1 혼합물에서 이에 결합된 링커를 갖는 세포바인딩제에 약물을 접합시켜 (i) 상기 링커를 통해 상기 약물에 화학적으로 결합된 세포바인딩제, (ii) 유리 약물(free drug), 및 (iii) 반응 부산물을 포함하는 제 2 혼합물을 제조하는 단계, 및(d) 상기 제 2 혼합물을 접선 흐름 여과, 선택적 침전, 흡착 여고, 또는 흡착 크로마토그래피 수지에 적용하여 상기 링커를 통해 상기 제 2 혼합물의 다른 성분으로부터 상기 약물에 화학적으로 결합된 세포바인딩제를 정제하고, 이에 따라 정제된 제 2 혼합물을 제조하는 단계를 포함하는 세포바인딩제-약물 접합체 제조 방법.
