Abstract
Description
Claims (15)
- 세포 배지 중에서 배아 줄기세포를 배양하여 배아체를 형성하는 단계;액티빈A를 이용하여 상기 배아체를 부화하는 단계; 및RA를 이용하여 상기 배아체를 부화하여 이를 인슐린생성 전구세포로 발육시키는 단계를 상기 순서대로 포함하는 배아 줄기세포의 유도방법.
- 제1항에 있어서, 상기 배아 줄기세포가 생쥐 기원의 세포일 경우, 상기 배지가 10% 우태혈청을 함유한 DMEM이고, 상기 배아 줄기세포가 인간 기원의 세포일 경우, 상기 배지가 CDM인 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제1항에 있어서, 상기 액티빈 A의 농도가 약 50-300 ng/ml 배지이고, 상기 RA의 농도가 약 1×10-7-1×10-5 mol/L인 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제3항에 있어서, 세포 배지 중에서 배아 줄기세포를 배양하여 배아체를 형성하는 단계 전에, 매트리겔로 배지를 수용하는 배양용기를 피복하는 단계를 더 포함하는 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제4항에 있어서, 상기 매트리겔의 배지 중에서의 질량백분율이 약 1%인 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제5항에 있어서, 상기 배아체를 액티빈A와 함께 20시간 내지 4일간 부화한 후, 액티빈A를 함유하지 않는 배지 중에서 6시간 내지 8시간 동안 배양하고 나서, RA를 이용하여 20시간 내지 4일간 부화하여, 인슐린생성 전구세포로 발육시키는 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제6항에 있어서, 이미 획득한 인슐린생성 전구세포를 성숙인자를 함유한 배지 중에서 부화하여 인슐린생성 전구세포를 증가시키는 단계를 더 포함하는 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제7항에 있어서, 상기 성숙인자가 bFGF를 포함하는 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제8항에 있어서, 상기 bFGF의 농도가 8-12 ng/ml인 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제9항에 있어서, N2 보충제, B27 보충제, 라미닌 및 니코틴아미드를 함유한 배지 중에서 상기 증가된 인슐린생성 전구세포를 부화하여 인슐린생성세포로 발육 시키는 단계를 더 포함하는 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 제10항에 있어서, 상기 배지가 N2 보충제, B27 보충제, 1 ㎍/㎖의 라미닌, 10 ng/㎖의 bFGF, 인슐린-트랜스페린-셀렌-A(1:100)및 10 mmol/L의 니코틴아미드를 함유한 DMEM/F12 또는 CDM인 것을 특징으로 하는 배아 줄기세포의 유도방법.
- 액티빈A와 RA를 포함한 배아 줄기세포의 유도에 사용되는 키트.
- 제12항에 있어서, 매트리겔을 더 포함하는 것을 특징으로 하는 키트.
- 제13항에 있어서, 일종의 일차 배지를 더 포함하는 것을 특징으로 하는 키트.
- 제14항에 있어서, 혈청을 더 포함하는 것을 특징으로 하는 키트.
