Claims (9)
- 인간배아 줄기세포로부터 배아체가 형성되는 과정에서 혈청, 액티빈 (activin A) 및 레티노산 (retinoic acid)를 순차적으로 처리하여 췌장내배엽이 증가된 배아체를 유도하는 것을 특징으로 하는 췌장으로 분화 가능한 췌장전구세포의 분화 유도 방법.
- 제 1항에 있어서, 인간배아 줄기세포를 혈청으로 처리하여 삼배엽성 배아체를 유도하는 1단계, 상기 제 1단계의 삼배엽성 배아체를 액티빈으로 처리하여 내배엽 (endoderm)성 배아체로 유도하는 2단계, 상기 2단계의 배아체를 레티노산으로 처리하여 췌장내배엽이 증가된 배아체로 유도하는 3단계, 및 상기 3단계의 췌장내배엽이 증가된 배아체를 배양접시에 부착하여 ITS (인슐린, 트랜스페린, 셀레나이트)와 파이브로넥틴 (fibronectin)이 첨가된 무혈청 배양액에서 배양하는 4단계를 포함하는 것을 특징으로 하는 방법.
- 제 2항에 있어서, 상기 1단계는 15 ~ 25 %의 혈청이 첨가된 배양액에서 3 ~ 5일간 배양하는 것을 특징으로 하는 방법.
- 제 2항에 있어서, 상기 2단계는 액티빈 25 ~ 35 ng/ml로 처리하여 3 ~ 5일간 배양하는 것을 특징으로 하는 방법.
- 제 2항에 있어서, 상기 3단계는 5 ~ 15 μM 레티노산으로 처리하여 1 ~ 3일간 배양하는 것을 특징으로 하는 방법.
- 제 2항에 있어서, 상기 4단계는 인슐린 4 ~ 6 mg/l, 트랜스페린 40 ~ 60 mg/l, 셀레나이트 0.4 ~ 0.6 mg/l와 파이브로넥틴 3 ~ 7 μg/ml이 첨가된 무혈청 배지에서 10 ~ 15일간 배양하는 것을 특징으로 하는 방법.
- 제 2항에 있어서, 췌장전구세포를 증식과 세포이동과정을 통하여 3차원적 세포집합체를 형성시키는 단계를 더 포함하는 것을 특징으로 하는 방법.
- 제 1항 내지 7항의 어느 한 항에 따른 방법으로 분화된 췌장전구세포.
- 제 8항의 췌장전구세포를 유효성분으로 포함하는 당뇨병 치료용 세포치료제.
