Description
Claims (13)
- 아르기닌을 탈카복실화함으로써 아그마틴을 제조하는 방법에서, 아르기닌 데카복실라제 유전자를 증폭 발현시키는 재조합체 또는 상기한 재조합체로부터 수득되는 아르기닌 데카복실라제 또는 아르기닌 데카복실라제 함유물을 사용함을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 아그마틴의 제조방법에서 아르기닌 데카복실라제 유전자를 증폭 발현시키는 재조합체를 사용함을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 재조합체에서 아르기닌 데카복실라제의 발현량이 균체내 단백질의 5% 초과임을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 아르기닌 데카복실라제 유전자가 에스케리키아 콜리 기원 adi 유전자임을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 재조합체가 pUC계 플라스미드, pBR322계 플라스미드 또는 이의 유도체 기원의 벡터 DNA와 아르기닌 데카복실라제 유전자를 연결시켜 수득된 재조합 DNA를 사용한 형질전환체임을 특징으로 하는 아그마틴의 제조방법.
- 제5항에 있어서, 재조합 DNA가 trc 프로모터를 함유함을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 재조합체가 에스케리키아 콜리를 숙주로 함을 특징으로 하는 아그마틴의 제조방법.
- 제7항에 있어서, 재조합체가 FERM-P18285임을 특징으로 하는 아그마틴의 제조방법.
- 제7항에 있어서, 재조합체가 아그마틴에서 푸트레신으로의 전환을 촉매하는 아그마티나제 결핍 에스케리키아 콜리임을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 아르기닌에서 아그마틴으로의 전환반응을 위한 반응계의 pH가 pH 6 미만으로 조절됨을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서, 아르기닌에서 아그마틴으로의 전환반응을 위한 반응계에 피리독살 포스페이트를 첨가함을 특징으로 하는 아그마틴의 제조방법.
- 제1항에 있어서,아르기닌 데카복실라제 유전자를 증폭 발현시키는 재조합체를 적어도 30℃미만에서 배양함으로써 균체내에 아르기닌 데카복실라제를 축적시키는 공정 및아르기닌 데카복실라제 유전자를 증폭 발현시키는 재조합체 또는 재조합체로부터 수득되는 아르기닌 데카복실라제 또는 아르기닌 데카복실라제 함유물을 사용하여 아르기닌을 탈카복실화함으로써 아그마틴을 제조하는 공정을 포함함을 특징으로 하는 아그마틴의 제조방법.
- 아르기닌 데카복실라제 유전자를 증폭 발현시키는 재조합체를 배지중에 배양하고 배지중 및/또는 세포중에 아르기닌 데카복실라제를 축적시킴을 특징으로 하는 아르기닌 데카복실라제의 제조방법.
