Abstract
본 발명의 석회화 조직의 결손을 회복하기 위한 조성물을 필수적으로 칼슘, 규소 및 산소로 이루어진 무정형 생활성 미립물질을 포함한다. 상기 제는 a) 유기 실리케이트 공급원과 칼슘 공급원의 반응 생성물, b) 칼슘을 함유하는 테트라에틸오르토실리케이트의 가수 분해 생성물, c) 칼슘 함유 실리카 졸-겔, d) 이성분 산화칼슘 및 실리케이트 침전 물질, e) 천연 규회석형 칼슘 실리케이트의 합성 유사체 및 f) 가용성 칼슘 공급원과 실리케이트 용액의 침전 반응 생성물로 이루어진 군으로부터 선택될 수 있다. 무정형 생활성 미립물질을 제조하는 방법은 필수적으로 충분량의 가용성 칼슘 공급원의 용액을 실리케이트 또는 실리케이트 전구물질에 첨가하여 칼슘 함유 실리케이트 미립물질의 침전을 유도하는 단계로 이루어진다.
Description
생활성 미립물질과 같은 생성물은 치주 포켓, 적출부위, 상아질 과민증 치료 부위, 골조직 형성 부위 등과 같은 석회화 조직에서의 결손을 회복시키거나 치유시키는데 사용될 수 있다.
경조직의 이식된 유리질 입자로의 강한 부착에 의해 확실하게 되는 바와 같이 골조직 형성과 같은 석회화 조직에서의 결손을 회복 또는 치유하는데 있어서 생활성 미립 물질의 생체적합성은 1971년에 최초로 증명되었다. 구체적으로, 생활성 유리의 용도는 문헌(Hench et al., J. Biomed. Mater, Res. Symp., 2: 117-141(1971))에 최초로 보고되었다. 뼈는 골 표면에 인접하여 있는 생활성 유리상에 부착하므로써 성장하고, 이후 새로운 뼈가 짧은 거리에 대해 유도될 수 있다.
생활성 유리의 과립은 차후 골조직 형성 응용에 개시되었다. 미국 특허 제 4,239,113호는 뼈 시멘트(bone cement) 제조용 조성물을 개시하고 있다. 미국 특허 제 5,658,332호는 40 내지 58%의 SiO2, 10 내지 30%의 NaO, 10 내지 30%의 CaO 및 0 내지 10%의 P2O5를 함유하고 크기가 200 내지 300㎛인 생활성 유리 과립을 사용하여 부속골격 또는 감소된 대사 상태를 보이는 부위내 결손에서 골성 조직을 형성하는 방법을 개시하고 있다.
치아 조직내 결손을 회복하거나 치유하는 영역에서, 생활성 미립 물질은 90 내지 710㎛의 크기 범위 및 40 내지 55중량%의 SiO2, 10 내지 30중량%의 CaO, 10 내지 35중량%의 Na2O, 2 내지 8중량%의 CaF2및 0 내지 10중량%의 B2O3의 조성 범위를 이용하여 치주 골성 결손 회복(미국 특허 제 4,851,046호)을 위해 사용되었다. 미국 특허 제 4,851,046호에는 치주 응용에 사용하기 위한 다양한 크기 범위의 생활성 유리 입자를 함유하는 조성물이 개시되어 있다. 미국 특허 제 5,204,106호에는 280 내지 425㎛의 협소한 크기 범위내의 생활성 유리 입자가 이미 개시된 것과는 명백히 변형된 생체 반응을 유도하는 것이 개시되어 있다.
생활성 미립물질은 또한 상아질 과민증의 치료에 사용되어 왔다. 상아질은 치아 에나멜(enamel)을 기계적으로 지지하는 경질이나 탄성인 석회질 조직이다. 상아질은 치수(pulp)를 감싸 보호한다. 상아질은 또한 치수강을 치아 에나멜/시멘트질 연결부에 연결하는 다수개의 연속적인 상아질 세관을 갖는다. 이들 가는 상아질 세관은 치수로부터 방사상으로 뻗어있다. 부적당한 구강 위생 관행에 의해 또는 치주 질병 및/또는 이것의 치료 결과로서 치아로부터 에나멜과 시멘트질이 떨어져 나가는 경우에는 치아 민감성의 문제가 발생한다. 상아질 세관은 치아의 중심 코아(central core)를 외부 환경에 노출시켜 유체 충전된다. 따라서, 기류 (air current), 촉각적 또는 열적 자극으로 처리되는 경우, 치수 신경이 자극받아 통증을 인지하게 한다. 과민성 상아질의 원인을 설명하는 우세 이론인, 소위 유체역학 이론에 따르면, 상아질 세관을 통해 전달되는 자극은 이들 영역에서 통증의 원인이 되는 치수에 분포된 신경을 자극한다. 민감성 상아질을 억제하고, 통증을 개선하거나 완화시키는 것은 상아질 세관을 치유하여, 물리적 또는 화학적 수단에 의해 감각 전도체를 차단하므로써 달성될 수 있다.
상아질 세관의 부분적 폐색을 제공하는 선행 출원에 있어서, 미국 특허 제 5,735,942호("'942 특허")에는 치아 과민성의 치료를 위해 생활성 용융 유도 유리 조성물이 개시되어 있다. '942 특허에는 40 내지 60중량%의 SiO2, 10 내지 30중량%의 CaO, 10 내지 35중량%의 Na2O, 2 내지 8중량%의 P2O5, 0 내지 25중량%의 CaF2및 0 내지 10중량%의 B2O3의 중량비와 약 10㎛ 미만의 입도에 대해 일부 입자가 약 2㎛ 이하이고, 일부는 2㎛보다 큰 조성물이 개시되어 있다. '942 특허는 60%의 SiO2가 생활성 용융 유도 유리에 대해 최대 실리카 한계임을 명시하고 있다.
용융 유도 방법에서는, 다공성 수화된 실리카 겔 층의 전개를 유도하는 세가지 방법, 즉, 이온 교환, 실리카 용해 및 실록산 결합된 수화된 실리카 사슬 또는 고리를 형성하는 실란올 축합이 있다. SiO2의 함량이 증가함에 따라, 용융 유도 유리는 증가하고, 이들 반응의 속도는 감소하여 용액중 Ca+2이온의 이용율 및 표면상으로의 실리카 겔 층 전개능이 감소한다. 결과는 SiO2의 함량이 60%에 도달하게 될 때 용융 유도 유리의 생활성이 감소하고 궁극적으로는 없어지게 된다. 상기 방법은 특정 적용을 위해 물질을 변형시키고 맞추는 제형물의 능력을 방해하여, 효과적 인 조성 영역을 협소하게 한다.
용융 유도 공정과 종래 기술('942 특허)에 의해 제조된 생활성 유리는 고온, 일반적으로 약 1350℃ 정도에서 백금 도가니내에서 처리된다. 그러므로, 용융 유도 유리는 비용이 많이 들고, 제조 설비로 이동시키기가 어렵다. 고온 공정에다 다수 처리 단계는 용융 유도 공정에 의해 제조되는 생활성 유리를 고가이게 하여 구강 관리 제품을 위해 생활성 유리를 사용하는데 있어서의 실용성을 실질적으로 제한한다.
높은 제조 비용 이외에, 상기 종래 기술에는 본래의 또 다른 단점이 있다. 먼저, 용융 유도 공정 자체로 인해 적합한 생활성에 요구되는 매우 높은 순도 및 관례에 따른 생활성 유리 조성물의 저함량 실리카 및 고함량 알카리를 유지시키는 것이 어렵다. 둘째, 상기 종래 기술은 그라인딩(grinding), 폴리싱(polishing), 프릿팅(fritting), 시이빙(sieving) 등을 포함하는 처리 단계를 요하며, 이들 단계 는 모두 생활성에 부정적으로 작용할 수 있는 잠재적 오염물질에 생활성 분말을 노출시킨다. 세째, 1713℃의 SiO2의 극히 높은 평형 액상선(liquidus) 온도 및 높은 SiO2함량을 갖는 실리케이트 용융물의 극히 높은 점도로 인해 통상적인 고온 공정에 의해 제조된 생활성 유리 및 유리-세라믹에 조성 제한이 가해진다.
또 다른 참고문헌으로, 미국 특허 제 5,874,101호("'101 특허")에는 개선된 방법에 의해 제조된 생활성 겔이 제안되어 있으며, 이 졸-겔 방법은 치아 과민성의 치료를 위한 건조 단계를 포함한다. 현존 생활성 유리 및 유리세라믹 및 상기 '942 특허에 기술된 바와 비교하면, '101 특허의 생활성 유리는 주로 실리카 함량으로 정해지는 Na2O-CaO-P2O5-SiO2시스템의 특이적 조성 영역으로 한정된다[참조: Ogin, M., Ohuchi, F and Hench, L.L., Compositional dependence of the foramtion of calcium phosphate films on bioglass,J. Japn.1991, 99, 1-6]. '101 특허에는 40 내지 90중량%의 SiO2, 4 내지 45중량%의 CaO, 0 내지 10중량%의 Na2O, 2 내지 16중량%의 P2O5, 0 내지 25중량%의 CaF2, 0 내지 4중량%의 B2O3, 0 내지 8중량%의 K2O 및 0 내지 5중량%의 MgO의 조성물이 개시되어 있다.
상기 선행 기술의 참고문헌 어디에도 제조가 간단하고 용이하며, 석회화 무기질 조직을 회복시키고/거나 치유하기 위한 칼슘 함유 무기물의 개시화(initiation)를 촉진하는 생활성 미립물질을 제공하지 않고 있다.
발명의 요약
본 발명은 석회화 조직의 결손을 회복하기 위해 필수적으로 CaO 및 SiO2로이루어진 무기 또는 유기/무기 복합 조성물에 관한 것이다. 유기/무기 복합 물질은 바람직하게는 가용성 칼슘 공급원을 TEOS 또는 나트륨 실리케이트와 같은 적합한 규소 공여 전구물질에 첨가하여 제조된 CaO-SiO2시스템의 무정형 이성분 산화물이다.
바람직한 구체예에서, 본 발명은 상아질 세관내에 투입되는 경우에 상아질 내측에 칼슘 함유 무기물의 형성을 개시시켜 과민성 치아와 관련된 통증을 완화시키는, 필수적으로 CaO 및 SiO2로 이루어진 무기 또는 유기/무기 복합 조성물에 관한 것이다.
바람직하게는, 복합 조성물은 입도가 약 10 마이크론 이하, 유리하게는 대개가 약 2 마이크론 이하인 크기 범위이어야 한다.
본 발명의 생활성 조성물은 다음 방법을 포함하는 여러 방법에 의해 제조되고/되거나 얻어질 수 있으나, 이들 방법으로 제한되는 것은 아니다: a) 무기 복합물을 제조하는 변형된 저온 졸-겔법; b) 처음부터 개질된 실리케이트를 제조하는 변형된 저온 졸-겔법; c) 화학약품 공급사로부터 입수한 이성분 산화물의 천연 유사체, 즉, 규회석(wollastonite)로부터의 칼슘 실리케이트를 화학적 또는 물리적으로 개질시키는 방법; 및 d) 무기물의 무정형 침전법.
본 발명은 일반적으로 필수 구성 성분으로서 칼슘, 규소 및 산소로 이루어진 무정형 생활성 미립물질의 조성물 및 이러한 조성물을 석회화 조직의 결손을 회복시키는데 사용하는 방법에 관한 것이다.
도 1은 석회화 조직의 샘플, 즉 무균 타액중에 인큐베이팅된 비처리된 상아질 디스크 표면의 전자 주사 현미경 사진 또는 "SEM"(10KV X 3200 확대)이다. 도2는 무기 입자를 생성시키는 침전 기재 방법에 의한 물질에 의해 처리(또는 회복)된 후의 처리된 상아질 디스크 표면(비처리된 디스크 표면과 동일한 방식으로 제조됨)의 SEM이다.
도 3은 비처리된 상아질 디스크 표면의 전자 주사 현미경 사진(20KV X 6000 확대)이다. 도 4는 희석 침전 방법에 의한 물질로 처리된 후의 유사하게 제조된 상아질 디스크 표면을 도시한 것이다.
도 5는 비처리된 상아질 디스크 표면의 전자 주사 현미경 사진(10KV X 1700 확대)이다. 도 6은 실시예 2 - 변형된 졸-겔 오르모실(ormosil) 방법에 의한 물질로 처리된 후의 유사하게 제조된 상아질 디스크 표면을 도시한 것이다.
도 7은 대조군의 비처리된 상아질 디스크 샘플의 전자 주사 현미경 사진(10KV X 1600 확대)이다. 도 8은 실시예 4 - 변형된 졸-겔 염기 촉매 반응으로 제조된 물질로 처리된 후의 유사하게 제조된 상아질 디스크 표면을 도시한 것이다.
본 발명자들은 놀랍게도 과민성 치아와 관련된 통증을 완화시키는 것을 포함하여, 석회화 조직의 결손을 회복하는 저비용이고 제조가 편리한 생활성 물질을 발견하였다. 본 발명자들은 또한 공정 파라미터 및 물질의 습식 화학 특성을 변화시키므로써 무기질화를 이행하는 생활성 유리를 발견하였다.
생활성 미립물질 조성물.생활성 유리를 포함하는 최근의 생활성 물질은 종종 P2O5, Na2O, B2O3, K2O, MgO, Al2O3, TiO2및 Ta2O5를 포함하는 산화물 및 CaF2를 포함하는 플루오라이드 염의 착체 리스트에 의존한다. 본 발명자들은 놀랍게도 핵생성 결정 성장이 필수적으로 오로지 Si, O 및 Ca+2을 기재로 하는 이성분 산화물 시스템을 사용하므로써 개시될 수 있음을 발견하였다. 본 발명에 따라 제조된 생활성 물질인, 천연 유사체로부터 유도된 CaO-SiO2함유 무기/유기 하이브리드 또는 무기 착체는 상기 물질의 표면이 혈액, 타액, 모조 체액과 접촉하게 놓여지면 정상적인 상아질에서 우세하게 되는 무기물 상에 대한 조성 및 결정도에 있어서 유사하게, 아파타이트(apatite) 또는 그 밖의 칼슘 함유 무기물 층을 침착시킬 수 있다.
본 발명자들은 또한 이러한 조성물으로부터 이들 종을 제거하면서 생활성을 유지시키므로써 상기 물질의 비용을 낮출수 있으며, 처리시 보다 자유롭게 하며, 감각기 및 독물학적 문제 발생의 가능성을 제거할 수 있게 한다. 본 발명의 화학 조성의 단순성은, 처리를 용이하게 하고 비용을 낮추게 도울 뿐만 아니라, 조성물을 의도하는 적용에 맞추는데 융통성을 가지게 한다. 본 발명자들은 특정 문제인 치아 과민증의 치료를 위해 핵형성 칼슘 함유 무기물로 조정되어 체내 어디에서도 골 유도 물질로서 사용되는 경우에 생물학적 활성에 필수적일 수 있는 부형제를 제거한 물질을 발견하였다.
골아세포 및 그 밖의 경조직 세포를 통해 내방성장 및 조직 형성을 조장시키므로써 부분적으로 작용하는 최근에 사용되는 대부분의 생활성 물질과는 달리, 본 발명의 물질은 놀랍게도 세포 애주번트를 필요로 하지 않는다. 추가로, 본 발명에따른 조성물은 골 구조발생 단백질, 상아질 인단백질, 치골모세포 또는 이들의 숙주 세포, 또는 정의된 콜라겐 매트릭스의 보조없이 모의 체액으로부터 아파타이트 및 칼슘 함유 무기물의 침전을 유도할 수 있다. 유기 애주번트를 필요로 하지 않고 무기질화를 유도할 수 있는 능력은 본원 물질을 구강 환경에 사용하는데 적합하게 한다.
또한, 이러한 최근 사용되는 많은 물질들은 특이적 임상 용도에 적합한 다공성 및 기계적 강도와 같은 고도로 분화된 특성을 필요로 한다. 예를 들어, 리, 클락(Li, Clar) 및 헨치(Hench)는 효과적인 무기질화제가 되는데는 히드록시아파타이트 최저 형성율이 요구되며, 이러한 형성율은 조성 및 미세구조의 함수임을 보여주었다[참조: Li et al., 'An Investigation of Bioactive Glass Powders by Sol-Gel Processing, J. of Applied Biomaterial, Vol, 2, 231-239(1991)]. 본 발명자들은 또한, 놀랍게도 본 발명의 물질이 치과 응용에 적용되는 종래의 생유리의 다공성 및 기계적 강도를 필요로 하지 않아. 세관내에 투입되는 경우에 상아질 세관내측에 칼슘 함유 무기물의 형성을 개시시킨다는 것을 발견하였다.
본 발명자들은 또한 놀랍게도 생활성이 되는 본 발명의 생활성 물질이 덜 다공성인 미세구조를 갖는 종래 기술의 용융 유도 유리에 대해 조성적 한계를 초월한다는 것을 발견하였다.
필수적인 것은 아니지만, 종래 기술의 조성물에서 통상적으로 요구되는, P2O5, Na2O, B2O3, K2O, MgO, Al2O3, TiO2, Ta2O5를 포함하는 그 밖의 산화물 및 CaF2를포함하는 플루오라이드 염이 본 발명의 이성분 산화물 시스템에 함게 사용될 수 있다.
본 발명의 생활성 물질에 대해 바람직한 예시적인 응용은 상아질 과민증의 치료이다. 노출된 과민성 상아질 표면의 현미경 조사에서, 상아질의 과민성 영역과 비과민성 영역 사이에 뚜렷한 차이가 존재함을 보여준다. 과민성 상아질은 개방 세관에 의해 퍼지게 된다. 대조적으로, 비과민성 치아 표면은 천연 무기질 침착물 또는 피복물에 의해 외부 환경으로부터 떨어져 실링되는 세관에 의해 특징된다. 본 발명자들은 상아질 표면에 결합되거나 세관내 투입되면, 본 발명의 무정형 물질이 타액 분위기에 의해 자극되는 핵형성 및 무기질화를 개시하여 조성과 결정도에서 치아 무기질과 유사한, 아파타이트 또는 그 밖의 칼슘 함유 무기물 층을 침착시키므로써 외부로부터 세관을 실링한다.
상아질 과민증의 치료에 사용하기 위한 조성물과 함께 사용하기에 적합한 담체는 바람직하게는 물, 폴리올 및 이들의 혼합물과 같은 히드록실계 물질이다. 때때로 습윤제로서 언급되는 폴리올에는 글리세롤, 소르비톨, 프로필렌 글리콜, 크실리톨, 폴리프로필렌 글리콜, 폴리에틸렌 글리콜, 수소화 옥수수 시럽 및 이들의 혼합물이 포함된다. 담체로서 특히 바람직한 것은 3 내지 30%의 물, 0 내지 90%의 글리세롤 및 0 내지 80%의 소르비톨의 액체 혼합물이다. 일반적으로, 담체의 양은 약 25 내지 99.9중량%, 바람직하게는 약 70 내지 95중량%이다.
본 발명의 조성물이 상아질 과민증의 치료를 위해 치약 또는 겔의 형태로 되는 경우, 조성물에는 일반적으로 약 0.1 내지 10%, 바람직하게는 약 0.5 내지 5중량%의 천연 또는 인공 농후제가 포함될 것이다. 농후제에는 히드록시프로필 메틸셀룰로오스, 히드록시에틸 셀룰로오스, 나트륨 카르복시메틸셀룰로오스, 크산탄 검(xanthan gum), 트라가칸트 검(tragacanth gum), 카라야 검(karaya gum), 검 아라빅(gum arabic), 진두발(Irish moss), 전분, 알기네이트(alginate) 및 카라게난(carrageenan)이 포함될 수 있다.
계면활성제가 상아질 과민증의 치료를 위한 조성물에 포함될 수 있다. 이들 계면활성제는, 음이온계, 비이온계, 양이온계 또는 양쪽성 타입일 수 있다. 매우 바람직한 것은, 나트륨 라우릴 술페이트, 나트륨 도데실벤젠 술포네이트 및 나트륨 라우실사르코시네이트이다. 계면활성제는 일반적으로 약 0.5 내지 80중량%의 양으로 존재한다. 안티 카리에스(anti-caries) 보호를 위해, 플루오라이드 이온의 공급원이 일반적으로 구강용 조성물에 존재할 것이다. 플루오라이드 공급원에는 불화나트륨, 불화칼륨, 주석 모노플루오로포스페이트, 불화칼슘, 불화주석 및 나트륨 모노플루오로포스페이트가 포함된다. 이들 공급원은 약 25 내지 3500ppm 플루오라이드 이온을 방출해야 한다. 안티 카리에스제는 약 0.05 내지 3.0중량%, 바람직하게는 약 0.5 내지 1.0%의 양으로 존재할 것이다.
구강 관리 조성물에 일반적으로 존재하는 향미제는 스피어민트 및 페퍼민트오일에 기재한 것들이 있다. 다른 착향제의 예로는 멘톨(menthol), 정향(clove), 윈터그린(wintergreen), 유칼립터스(eucalyptus) 및 아니스(aniseed)가 포함된다. 향미제는 약 0.1 내지 5.0% 범위의 농도일 수 있다. 사카린, 나트륨 시클라메이트, 아스파르탐, 수크로스 등과 같은 감미제는 약 0.1 내지 5.0중량%의 수준으로포함될 수 있다. 방부제, 실리콘, 그 밖의 합성 또는 천연 중합체, 항치은염 활성제, 안티 타르타르제, 화이트닝제, 및 베이킹 소다(baking soda) 및 퍼옥사이드와 같은 통상적인 치약에 존재하는 그 밖의 바람직한 생성물을 포함하는 그 밖의 첨가제가 혼입될 수 있다. 또한, 이들 물질은 미국 특허 제 5,589,159호에 기재된 것과 같은 신규 탈감작화제 뿐만 아니라 질산칼륨, 염화칼슘 및 중탄산칼륨과 같은 종래의 탈감작화제와 함께 사용할 수 있다는 이점을 갖는다.
페이스트, 겔, 린스, 분말, 검, 치실, 슬러리 및 용액을 포함하나, 이로 제한되는 것은 아닌, 상아질 과민증의 치료에 활성제의 구강 전달을 위한 다수의 비히클이 존재하나, 각각이 기술되진 않지만, 본 발명의 사상내에 있는 것이다.
골 이식체로서 사용하거나 이식가능한 표면을 형성시키기 위한 조성물에 있어서, 응용에 따라 항생물질, 구조용 섬유 및/또는 중합체 수지를 포함하는 그 밖의 물질이 본 발명의 생활성 물질에 첨가될 수 있다.
제조.본 발명의 이점은 상기 물질이 종래 기술의 용융 유도 생활성 물질 및 졸-겔 유도 생활성 물질에 포함되는 고비용이고, 시간 소모적인 처리 절차를 없애므로써 제조를 용이하게는 하는 이점이 있다.
본 발명의 생활성 물질은 개선된 처리로 인해 특정 적용에 대해 조성물을 특이적으로 맞추어 설계된 수개의 공정 및 수개의 화학적 변형에 의해, 그리고 물질의 생활성을 증진시키도록 습식 화학 특성을 조정하므로써 제조될 수 있다. 본 발명의 공정들은 Si, O 및 Ca2+만을 기재로 하는 본 발명의 생활성 물질을 제조하는데사용될 수 있다.
1. 무기 유리를 생성하는 변형된 졸-겔 방법.
유리의 통상적인 졸-겔 합성은 금속 알콕사이드 전구물질을 물 및 촉매와 혼합하고 이어서 메탈 알콕사이드 종을 "중합"하므로써 달성되며, 겔의 제조는 1) 흔히 600 내지 900℃의 온도에서 가열 소결시키는 단계, 2) 소성 열처리하는 단계, 및 흔히 3) 나노구조 물질의 완전성을 유지시키기 위해 초임계 건조시키는 단계에 의해 수행한다.
용융 유도 방법에 의해 제조된 유리와는 달리, 졸-겔 방법에 의해 제조된 유리("졸-겔 유도 유리")는 순수한 실리카 겔까지 조성물에 대한 생활성을 유지시킨다. 졸-겔 처리의 특이적인 특징은 미세다공성 물질의 제조에 있다. 이에 따른 두가지 유리한 결과는 다음과 같다: 1) 다공성 칼슘 함유 실리카 겔과 관련된 높은 표면적이 주변의 용액 중 Ca2+이온의 농도를 신속하게 증가시키도록 하며, 2) 졸-겔 방법에 의해 제조된 텍스쳐(texture)가 SiO2의 함량이 증가함에 따라 감소된 이온 교환 및 해리 속도에도 다공성 겔 층을 형성한다. 이러한 두가지 인자는 졸-겔 유도 유리에서 빠른 속도의 무기질 형성 및 조성물의 생활성 범위 연장에 대해 원인이 되는 것으로 여겨진다. 연구 결과, 용융 유도 유리와는 달리 졸-겔 제조된 실리카가 준안정 모의 체액으로부터 아파타이트 핵형성을 용이하게 하고 개시하는 고도로 가수분해된 실리카 표면을 갖는 것으로 나타났다.
본 발명자들은 놀랍게도 추가의 처리 단계인 가열 소결, 소성 열 처리 및 나노구조 물질의 완전성을 유지시키기 위한 초임계 건조 단계들(이들 단계는 제조에 용이하게 적용될 수 없으나 올바르게 형성된 졸-겔 유리"를 달성하는데는 필요하다)을 필요로 하지 않는, "변형된 졸-겔 방법"에서 졸-겔 유도 물질과 관련된 유리한 다공성과 증가된 표면적을 혼입시킬 수 있었다.
무기 물질을 생성시키기 위한 본 발명의 상기 변형된 졸-겔 방법에서, 칼슘 알콕사이드 전구물질, 즉 테트라에톡시실란(TEOS) 및 임의로 트리에틸포스페이트(TEP)는 산성 조건 및 저온 하에서 가수분해되어 겔을 형성한다. 또한, 그 밖의 적합한 성분은 당업자들에게는 자명할 것이다. 상기 방법은 또한 가수분해 반응을 위한 알칼리 조건을 사용하여 수행될 수 있다. 이는 생성되는 분말의 형태 및 크기 둘 모두를 조절하는 것으로 밝혀졌다.
2.유기적으로 변형된 실리케이트("오르모실(Ormosil)")를 생성하는 변형된 졸-겔 방법.
이러한 오르모실 방법에서는, 상기 변형된 졸-겔 방법에서 기술된 바와 같이 유기/무기 하이브리드 물질을 제조하는데 가수분해 반응이 먼저 사용된 후, 무기 망상구조의 형성이 이어진다. 이러한 망상구조의 형성 동안에, 적합하게 작용성화된 유기 잔기에 응축반응이 진행되어, 망상구조 형성에 혼입된다. 이러한 두번째 명시에서, 유기/무기 하이브리드는 문헌(Hu and Mackenzie, Journal of Material Science, 27:4416-4420[1992])에 의해 기술된 바와 같이 실란올 말단 폴리(디메틸실록산)[PDMS] 및 테트라에톡시실란[TEOS] 전구물질(규소의 공급원)로부터 제조된다. 최종 입도, 표면 변형 및 반응성 조절이 처리 파라미터를 변형하므로써, 특히촉매의 선택 및 시스템의 pH에 의해 조절될 수 있다. 물질의 가요성은 유기 및 무기 성분의 혼합비를 변화시키므로써 조절될 수 있다. Ca(II) 및 그 밖의 생활성에 중요한 화학종이 유기/무기 하이브리드 물질에 도입되는 경우, 이들 복합물은 가요성 및 생활성 모두를 나타낼 것이다. 본 발명의 물질의 입도, 표면 형태, 가요성 및 생활성을 조절하는 능력은 이들 물질을 의도하는 적용에 독특하게 맞출 수 있게 한다.
3.천연 무기물(구입가능한 실리케이트) 사용/개질
본 발명의 생활성제는 무기물에 의해 생성된 물질의 천연 유사체, 즉, 규회석으로부터 얻어진 칼슘 실리케이트를 사용하거나 변형시키므로써 제조될 수 있다. 상업용 가용성 실리케이트는 하기 화학식을 갖는다:

상기 식에서, M은 알칼리 금속이고, m 및 n은 각각 M2O의 몰당 SiO2및 H2O의 몰수이다.
상업용 실리케이트의 조성은 일반적으로 SiO2대 M2O의 중량비로 기술된다. 반응성인 주 실리카 종의 공급원으로서 역할하는 능력으로부터 나오는 가장 광범위하고, 빠르게 성장하는, 이러한 가용성 실리케이트는 많은 용도를 갖는다. 반응성 실리카 종을 용이하게 형성하는 이러한 무기 물질의 능력은 칼슘 함유 무기물을 핵형성시키는 능력에 대해 중요하다. 가용성 실리케이트의 해리 속도는 유리 비, 고형물 농도, 생성 온도, 압력, 입도 및 전체 표면적에 의존한다. 해리는 일반적으로 이온 교환 및 망상구조 붕괴와 관련된 2 단계 메카니즘으로 일어난다.

공급되는 바와 같은, 칼슘 실리케이트는 본 명세서에 부합하지 않거나 요망되는 작용성을 갖지 않을 수 있어, 특정 변형, 즉, 표면 변형, 입도 조절 및/또는 그 밖의 필요한 처리가 이행될 수 있다. 처음 접수되면, 알파 아에사르(Alfa Aesar)사로부터의 상용 물질은 본 발명자들의 평가로는 적합한 활성이었다. 그러나, 물질을 5㎛ 입도로 볼-밀링(ball-milling)한 후, 물질이 개선된 생활성을 제공하였다. 규회석으로부터 얻은 천연 칼슘 실리케이트는 본 발명자들의 시험관내 평가로 생활성을 나타냈다.
칼슘 실리케이트, (메타(CaSiO3), 오르토(Ca2SiO4) 등을 포함하는 이들 물질은 알파 아에사르 및 알드리치(Aldrich)사와 같은 중 화학약품 공급 회사를 통해 상업적으로 입수할 수 있다.
4.무기 입자를 생성하는 침전 기재 방법
본 발명의 네번째 명시는 본 발명의 가장 바람직한 방법이다. 본 침전 기재 방법에서, 졸-겔 절차는, '졸-겔' 단계를 거치지 않고 무정형 산화물을 단순히 침전시키므로써 졸-겔(또는 "변형된 졸-겔" 방법) 고유의 어려운 처리 파라미터를 제거하는 침전 방법으로 변형된 것이다. 본 방법에서, 무기 입자는 TEOS의 조절된 가수분해와 이로써 형성된 칼슘 공급원, 즉 칼슘 니트레이트와 같은 칼슘 염 또는 칼슘 메톡사이드와 같은 알콕사이드 유도체로부터 공여된 혼입되는 반응 이온을 침전 방법으로 축합하므로써, 졸-겔 방법에서 이미 기재된 바와 같이 생성된다. 무기 입자는 본 발명의 물질에 생활성을 부여하는 특정 제(agent), 이온, 중합체 또는 콜로이드 입자를 함유한다.
전통적인 졸-겔(또는 본 발명자들의 "변형된" 졸-겔) 절차에서, 실리케이트 겔은 종종 촉매로서 무기산(예를 들어, HCl) 또는 염기(예를 들어, NH3)를 사용하여 단량체 4작용성 알콕사이드 전구물질을 가수분해시키므로써 합성된다. 작용기 수준에서, 일반적으로 다음 세 반응이 졸-겔 방법을 기술하는데 사용된다:
상기 식에서, R은 알킬기 C x H 2x+1 이다. 가수 분해 반응은 알콕사이드 기(OR)를 히드록실기(OH)로 대체한다. 이후, 실란올기를 포함하는 축합 반응이 실록산결합(Si-O-Si) 플러스 부산물인 알코올(ROH) 또는 물을 생성시킨다. 대부분의 조건 하에서, 축합은 가수분해가 완료되기 전에 수행된다.
물 및 알콕시실란은 비혼화성이므로, 일반적으로 알코올과 같은 공통 용매가 졸-겔 방법에서 균질화제로서 사용된다. 그러나, 가수분해 반응의 부산물이 초기에 상 분리된 시스템을 균질화시키기에 충분하므로, 겔이 추가의 용매 없이 규소 알콕사이드-물 혼합물로부터 제조될 수 있다. 본 발명자들은 가수분해 단계 후, 반응성 종, 즉, 칼슘 니트레이트가 혼합 용기에 첨가되는 경우, 실리카는 졸내 표면 전하 탈안정화로 인해 침전할 것이다. 이러한 침전된 물질은 본 발명의 모델에서 시험되었을 때, 놀랍게도 상아질 세관 폐색제로서의 우수한 결과와, 전자 주사 현미경 사진("SEM")으로 판단하여, 두개의 석회화 조직 샘플 또는 두개의 상아질 디스크 표면, 하나의 비처리된 샘플(도 1) 및 상기 바람직한 방법에 의해 제조된 물질로 처리된 샘플(도 2)를 비교하면 도면에서 도시된 바와 같은 우수한 상아질 무기질화제를 달성한다.
침전 공정의 매우 바람직한 방법, "희석 침전 공정"에서, 무정형 CaO-SiO2무기 물질은 희석된 실리케이트 공급원, 즉 칼륨 또는 나트륨 실리케이트 수용액과 이미 기재된 바와 같은 가용성 칼슘 공급원의 반응에 의해 생성된다. 이러한 절차는 최종 크기 또는 농도와 같은 물리적 특성 뿐만 아니라 최종 침전물의 조성 둘 모두를 조작할 수 있게 한다. 이러한 희석 공정에 의해 제조된 물질은 도 3 및 4(두개는 유사하게 제조된 상아질 표면, 비처리된 샘플 및 처리된 샘플)에서 도시된바와 같이, 수압 전도 시험 및 SEM에 의해 확인되는 수성 분산액 및 프로토타입 치약 조성물 둘 모두에 매우 효력을 갖는 것으로 밝혀졌다.
이론에 결부되지 않기를 바라는 경우, 본 발명에서 생활성을 위한 Ca(II) 및 그 밖의 중요한 화학종이 성형 실리카에 도입되어 미립 하이브리드 물질을 형성할 수 있다. 일반적으로, 정전기 반발에 의한 콜로이드의 안정화는 콜로이드 화학자들에게 널리 공지된 DLVO 이론(Derjaguin, Landau, Verivery and Overbeak theory, 현탁 안정도를 설명)에 의해 성공적으로 기술되어 있다. 실리카는 DLVO 이론에 부합하지 않는데, 그 이유는 등전점에서도 응집을 방해하는 흡수된 물 층에 의해 명백히 안정화되기 때문이다. 수성 실리카 졸로의 양이온 첨가는 수화도를 감소시키고, 실리카를 탈안정화시킬 수 있다. 알렌(Allen) 및 마티제빅(Matijevic)(J. Colloid and Interface Sci, 31[3] 1969 287-196, 33[3](1970) 420-429, 35[1](1971) 66-76)은 졸로의 염 첨가가 하기 방식으로 이온 교환을 유발할 것임을 보여주었다:

상기 식에서, Mz+은 전하 Z의 가수분해되지 않은 양이온이다.
실란올기는 물에 대한 흡착 부위이기 때문에, 이온 교환에 의한 SiOH의 제거는 수화의 양을 감소시키고, 콜로이드 안정성을 낮춘다. 이 반응은 여과에 의해 용액으로부터 용이하게 제거될 수 있고, 치과 물질로서 적용하기에 적합한 비히클 중에 재현탁될 수 있는 Ca(II) 함유 실리케이트를 침전시킨다. 잔류하는 용액은재사용되어 물질의 비용 및 제조를 용이하게 할 수 있다.
본 발명의 첨전 기재 방법에서, 나트륨 실리케이트[CAS# 1344-09-8]는 규소 공급원으로 대체될 수 있다. 적합한 양의 가용성 칼슘 공급원, 즉, Ca(NO3)2의 나트륨 실리케이트 용액으로의 첨가는 조성이 이미 기술된 물질과 유사한 칼슘 함유 실리케이트를 침전시킨다. 실리케이트 용액에 첨가되는 Ca2+의 양은 화학량론적 계산 및 실험적 설계 둘 모두에 의해 결정되었다.
본 발명의 침전 기재 방법은 다른 방법에 비해 수개의 이점을 갖는다. 이 방법은 표준 치약에 혼입된 후의 최종 생성물을 오염시킬 수 있는 부산물, 즉, 에탄올 또는 잔류 TEOS을 전혀 생성시키지 않는다. 또한, 이 방법의 화학 반응은 입도가 반응 동안에 보다 용이하게 조절될 수 있게 하여, 추가의 크기 감소에 대한 필요성과 비용이 많이 들뿐만 아니라 최종 생성물을 오염 가능성도 증가시킬 수 있는 밀링(milling)단계를 없앨 수 있다. 추가로, 반응이 단지 두가지 성분, 즉 나트륨 실리케이트 및 적합한 칼슘 공급원만을 필요로 한다. 이들 화학종 둘 모두는 일반적인 화학약품 공급 회사로부터 용이하게 입수할 수 있으며, 다른 방법과 비교하여 비교적 저렴하다.
상기 공정에서, 침전되고 잔류 용매와 분리되면 상기 물질은 세척, 고열에서의 건조 또는 초임계 건조를 포함하는 추가의 특별한 처리를 필요로 하지 않으며, 또한 상기 물질은 소성 절차 또는 유기 용매 추출을 포함하지 않는다.
추가로, 중요한 것은 아니지만, 상기 방법에서, 침전된 물질은 화학적으로조절되어 추가의 밀링, 그라인딩 또는 크기 감소를 필요로 하지 않도록 충분히 작은 입도의 무정형 물질을 생성하는 것이 바람직하다. 따라서, 샘플 오염 및 추가 처리 비용 가능성을 피한다.
대기 조건에서 수행되는 바람직한 방법의 화학 반응은 생물학적 활성 면에서 전통적인 졸-겔 방법에 의해 제조된 생활성 유리와 유사한 고도의 다공성 칼슘 함유 실리카를 생성한다. 이러한 무기 물질은 다른 방식으로 최근 입수할 수 있는 열 소결된 생활성 유리 물질과는 유사하지 않다.
반응이 이 성분 시스템, 즉, CaO-SiO2만을 기재로 하는 칼슘 함유 산화물 물질을 생성하고, 반응이 제조 설비 요원에게 안전하고 널리 알려졌을 뿐만 아니라 헬쓰 케어 분야에 잘 알려져 있는 용매 중에서 실온에서 일어난다는 것은 매우 바람직하다.
이하 실시예에서 제조되는 생활성 물질은 다음과 같은 다양한 적용에 적합하나, 이로 제한되는 것은 아니다: a) 물질을 골성 결손의 회복을 위해 조직 배지 중에 넣었을 때 골 세포 부착 및 활성의 증진하여, 세포로 접종되는 경우, 골 조직 형성이 개시된다; b) 뼈의 강 및 치아의 관을 충전시키는 이식 조성물; c) 재무기질화 치아에 사용하기 위한 구강 관리 조성물; d) 치주 골성 결손 회복을 위한 구강 관리 조성물 및 상아질 과민증의 치료에 사용하기 위한 구강 관리 조성물. 고려되는 골성 결손은 낭종성 결손 치유, 절제시 양성 또는 악성 종양 부위, 뼈 손실, 지연 또는 비결합 부위를 포함하는 골절 치유 부위, 관절 치유 부위, 골다공증관련 결손, 및 치주 결손을 포함하나 이로 제한되는 것은 아니다.
실시예
하기 실시예는 단지 예시의 목적으로 제시되는 것이며, 실시예에 기재되지 않은 변화 또는 변경이 이루어질 수 있다.
상아질 과민증의 치료로 맞추어진 실시예에서, 실험은 미국 특허 제 5,270,031호에 기재된 상아질 과민증의 변형된 시험관내 모델을 사용하여 수행하였다. 본 방법에서, 카리에스 또는 리페어(repair)가 없는 손상되지 않은 사람이 제 3대 구치를 금속 톱을 사용하여 치아의 세로축에 수직으로 약 1mm 두께의 절단면으로 절개하였다. 상아질을 함유하고, 에나멜을 함유하지 않는 절단면을 시험을 위해 보유하였다. 이들 절단면을 이후 EDTA(에틸렌디아민 테트라아세트산) 용액으로 에칭하여 도말층(smear layer)을 제거하였다. 저널(Journal of Dental Research 57:187(1978))에 보고된 바와 같이, 디스크를 분할 챔버 장치(split chamber device)내 구비시켰다. 이러한 특수 방수 챔버를 사람 체액의 오스몰 농도와 유사하게 의도되는 조직 배양 유체를 함유하는 가압된 유체 저장기에 연결시켰다. 가압된 N2및 CO22가스의 혼합물을 사용하므로써, 유체를 생리적 pH로 유지시켰다. 상기 장치는 룰러(ruler) 상에 구비된 유리 모세관 또는 그 밖의 특정 기구를 포함하고 있다. 기포를 유리 모세관내로 주입하고, 시간이 함수로서 기포의 대체율을 측정하므로써 상아질 디스크를 통한 유체 유량을 측정할 수 있었다. 유체는 사실상 정상 치아의 내부로부터 상아질 세관 밖으로 흐르는 것으로 보고되었다.
상아질 디스크의 기준 유체 유량을 측정한 후에, 실험 혼합물인 치약, 겔 또는 구강세척액을 임상적으로 용도가 유사한 방식으로 외측 디스크 표면에 도포하였다. 정해진 도포 기간 후에, 실험 물질을 헹구어내고, 도포 후(lpacute) 수압 전도를 측정하였다. 이러한 방식으로, 다양한 실험 물질이 단독으로 또는 최종 생성물의 성분으로서 상아질 세관에서의 유체 유동을 방해하는 능력에 대해 시험할 수 있었다. 이후, 실험 물질의 도포에 의해 유도된 유동 감소율%을 계산할 수 있었다.
제시된 모든 실시예에 대해, 타액 인큐베이션이 유동 감소를 달성하는데 필요하였다. 신규 제(agent)의 무기질화 효능을 연구하기 위해서는, 시간 연장 뿐만 아니라 표준 상아질 수압 전도 측정의 변경이 필요하다. 초기 도포 후 측정이 이루어진 후, 분할 챔버를 장치로부터 제거하고, 살균 여과된 사람 타액의 저장기를 챔버에 부착하여 유체가 처리된 교합 상아질 표면을 씻어내도록 하였다. 챔버의 외부를 파라필름(parafilm)으로 감싸 공기 접촉과 이에 다른 유체 손실을 피하도록 하였다. 이후, 상기 유닛을 체온으로 유지되는 인큐베이터내에 두었다. 충분한 시간이 경과한 후, 상기 유닛을 제거하고, 타액 저장기를 폐기하고, 분할 챔버를 수압 전도 장치에 재부착시켰다. 이때, 상아질 유체 유량을 재측정하고, 실험 물질을 재도포할 수 있었다. 하기 실시예에서, 보고된 유량 감소는 3회 처리 및 타액 인큐베이션 40 시간 후에 얻은 것이다.
본 명세서 및 청구범위 전반에 걸쳐, 하기 실시예에서 모든 온도는 섭씨이며, 모든 부 및 %는 다르게 명시하지 않은 한 중량에 의한 것이다.
실시예 1(변형된 졸-겔 - 오르모실)
출발 물질은 시약 등급 칼슘 니트레이트, TEOS(테트라에톡시실란), PDMS(폴리디메틸실록산)이었다. 시약 등급 2-프로판올 및 테트라히드로푸란을 용매로서 사용하고, HCl을 촉매로서 사용하였다. TEOS(10g) 및 실란올 말단 PDMS(5.9g)을 2-프로판올(4.8ml)와 테트라히드로푸란(3.2ml)의 혼합물과 혼합하였다. 이 용액을 용액 A라 하였다. 적합한 양의 칼슘 니트레이트(0.42g)를 증류수와 HCl 용액(35%)에 용해시켰다. 2-프로판올(7.0ml)를 교반하면서 용액에 첨가하고, 이를 용액 B라 하였다. 이러한 방법은 문헌(K. Tsuru et al., Mat. Res. Soc. Symp. Proc. Vol. 435, p. 404, 1996)에 개요된 생활성 오르모실(Ormosil)의 합성에 기초한다. 용액 A와 B를 혼합하고, 이후, 30분 동안 80℃에서 교반하면서 환류시켰다.
환류 후, 혼합물을 빙수를 사용하여 25℃로 켄칭(quenching)시키고, 용기내로 주조하여 대기 조건 하에서 겔이 되게 하였다. 겔화 후, 평평하고 불규칙하게 성형된 파편들을 크기가 약 10 마이크론 이하인 범위로 밀링하는데, 대부분은 크기가 약 2마이크론 이하의 범위였다. 밀링은 볼-밀링(ball-milling), 공기 충격 밀링(air-impact milling),MICROS수퍼파인 밀링(MICROSsuperfine milling), 로터리 커터 밀링(rotary cutter milling), 햄머 밀링(hammer milling) 및 케이지 밀링(cage milling)을 포함하나, 이로 제한되는 것은 아닌 여러 가지 방법에 의해 수행될 수 있다. 이제 입도가 알려진 상기 물질을 글리세린중에 분산시키고, 여러 농도에서 슬러리로서 본 발명의 시험관내 모델로 시험하였다.
본 발명의 본 구체예에서는, 20% 슬러리의 반복 도포시 표 1에 보여지는 바와 같이 우수한 상아질 유체 유동 감소능을 나타냈다.

반복된 도포 및 여과된 타액 인큐베이션 사이클은, 비생활성 물질이 평가되는 경우에는 현저한 유체 유동 감소를 유발하지 않았음을 유의해야 한다. 또한, 상기 사이클 방법에 노출된 비처리된 상아질 표면 및 플라세보 처리된 상아질 표면을 SEM으로 확인하였다. 미세하게 분쇄된 경석으로 처리된 상아질 표면이 이러한 방식으로 조사되는 경우에 아무런 폐색도 관찰되지 않았다. 유사하게, 상기 무기질화 모델에서와 같이 살균된 타액 처리 사이클 만으로 노출된 비처리된 상아질 표면의 형태는 SEM 평가 후, 새로 컷팅되어 제조된 상아질 시편과는 크게 다르지 않았다.
실시예 2(변형된 졸-겔 - 오르모실)
본 발명의 제 2 구체예는 실시예 1에서와 같은 전구물질로부터 제조되고, 유사한 방식으로 처리되었다. 그러나, 본 실시예에서, 본 발명자들은 물질 중 칼슘 니트레이트(1.25g), 즉 칼슘 이온 공급원의 총량을 현저히 증가시켰다. 이러한 증가가 특이적인 기계적 강도 및 원래 물질의 유연성을 감소시켰지만, 형성된 조성물은 글리세린 분산액중 여러 가지 농도에서 보다 우수한 상아질 유체 유동 감소성(표 2)을 나타냈다.

상기 물질은 이러한 변형된 졸-겔 오르모실 방법에 의해 제조된 물질로 처리된 후의 상아질 디스크 표면을 보여주고 있는 도 6에서 입증되는 바와 같이, 실시예 1에 기재된 물질보다 신속하고 완전한 폐색을 유도하였다. 도 5는 유사하게 제조된 처리되지 않는 상아질 표면을 보여준다.
실시예 3 (변형된 졸-겔 - 무기 물질)
완전 무기 물질을 대기 조건 하에서 졸-겔 방법에 의해 제조하였다. 이 물질을 실시예 2와 유사하게 전구물질로서 사용하였으나, PDMS 및 테트라히드로푸란을 생략하였다. 2-프로판올은 TEOS에 대한 용매였으나, 본 실시예에서는 사용하지 않았다. TEOS는 본 실시예에서는 단순히 규소의 공급원이다. 절차는 실시예 1 및 2와 동일하다. 그러나, 본 실시예의 생성물은 칼슘, 규소 및 산소만을 함유한다. 이러한 물질이 기계적 강도는 약하였지만, 세관 폐색제로서는 매우 효과적이었다. 또한, PDMS 및 테트라히드로푸란의 부재는 독물학적 프로파일에 대해 유리하였다.

실시예 4(변형된 졸-겔 방법 - 염기 촉매작용 반응)
염기 촉매작용 졸-겔 방법을 사용하여 형성되는 미립물질 현탁액의 형태 및 크기를 조절하였다. 출발 물질은 다음과 같다: 50ml의 TEOS, 75ml의 에탄올, 70ml의 물, 0.575g의 수산화암모늄(30%) 및 3.6g의 칼슘 니트레이트. 이들 반응을 위해, 암모니아를 구형 입자의 형성을 유도하는 촉매로서 사용하였다. 이러한 절차는 문헌(1968 Stoebber and Fink article, "Controlled Growth of Monodisperse Silica Spheres in the Micron Size Range"(Journal of colloid and Interface Science, 26, 62-69, 1968)에 기초한다. 형성된 미립물질 조성물은 매우 효과적인 무기질화제인 것으로 입증되었다(표 4). 전자 주사 현미경 사진에서는 상기 복합 물질로 처리된 샘플에서 세관 구멍의 무기질화로 인해 유도된 상아질 세관의 완전한 피복 상태를 보여준다. 비처리된 샘플은 치아의 과민성 영역을 나타내는 수많은 개방 세관에 의해 표시되었다. 또한, 미립 물질의 모양 및 형태를 화학적으로 조절하는 본 발명의 능력은 본 실시예를 가장 용이하게 제형하고 처리하도록 하였다. 또한, 본 물질의 원소 분석에서는 생활성 유도비로 다량의 Ca2+및 Si이 확인되었다. 도 7 및 6은 두개의 유사하게 제조된 상아질 표면을 비교하고 있다. 두개중 하나는 비처리되었고, 나머지 하나는 실시예 8의 조성물로 처리되었다.

실시예 5 (천연 무기물)
알파 아세사르사로부터의 광물 규회석으로부터 유도된 칼슘 오르토-실리케이트[(CaO)2-SiO2, FW:0.088-0.044mm(174.24-325 메시 US)]를 사용하였다. 공표된 연구에서는 탄산칼슘과 실리카의 화학량론적 혼합물로부터 2시간 동안 1500℃에서 합성된 준규회석은 모의 체액 분위기에서 생활성임을 지시하였다[참조: De Aza PN. et al., Bioactivity of Pseudowollastonite in Human Saliva, Journal of Dentistry, 27(1999), 107-113]. 알파 아에사르사로부터 공급된 바와 같은, 입도는 이러한 적용에 부적합하여, 이 물질을 미세다공성 기판의 무기질화에 유리한 평균 입도로 글리세롤 중에서 볼 밀링하였다. 표 5에 기재된 바와 같이, 형성된 미립 조성물은 효과적인 무기질화제인 것으로 입증되었다.

실시예 6 (무기 입자를 생성하는 침전 기재 방법)
상업적으로 입수할 수 있는 칼슘 니트레이트 및 나트륨 실리케이트 용액을 사용하여 침전물을 제조하였다. 72.8중량%의 40% 나트륨 실리케이트 용액에, 탈이온수중의 27.2중량%의 75% 칼슘 테트라하이드레이트 용액을 첨가하고, 혼합기를 나트륨 실리케이트 용액에 대해 최대 교반되도록 고속으로 작동시켰다. 즉시 침전이일어났다. 침전물(칼슘 실리케이트, 무기 침전물 또는 "CSIP")을 40시간 동안 60℃에서 건조시켰다.
실시예 6에서, CSIP 입도는 밀링(예를 들어, 공기 충격 밀, 단순한 볼 밀 등)에 의해 감소시켰다. 그러나, pH를 변경시키거나 나트륨 실리케이트 용액을 희석시키므로써 입도를 변경시키면서 침전물의 최종 입도를 낮추는 높은 칼슘 니트레이트 농도를 유지시킬 수 있다. 우수한 세관 폐색 효능은 수압 전도도 및 도 3-4에 도시된 SEM 둘 모두에 의해 판단하였다.
실시예 7
과민성 치아에 사용되는 경우 통증의 완화를 추가로 보조하는 치약 조성물을 하기 제형을 사용하여 제조하였다. 본 실시예에서 탈감작화제는 상기 언급된 오르모실, 무기 복합물, 합성 유도된 무기 물질, 또는 졸-겔 유도 물질, 또는 적합 조성으로 나트륨 실리케이트 유도된 물질중 어느 하나일 수 있다. 각 성분에 대해 설정된 중량%는 전체 제형 100%의 값을 기준으로 한다.

제 2 치약 제형을 오르모실, 무기 복합물, 합성 유도된 무기 물질 또는 졸-겔 유도 물질중 어느 하나로 이루어진 탈감작화제를 포함하여 제조하였다. 중량% 기준과 제조 방법은 실시예 7에서 언급된 것과 동일하다.

실시예 9
제 3 치약 제형을 2상 시스템으로서 제조하였다. 이 치약은, 각 상이 패키지를 분할하는 격막에 의해 따로 유지되는 튜브로 존재할 수 있고, 이것은 각각의 상을 동시에 분배하는 이중 챔버 펌프로 존재할 수 있으며, 또는 순차적으로 도포되도록 의도되는 별도의 튜브로 패키징될 수도 있다.

활성 성분은 또한 치아 과민증을 완화시키도록 설계된 여러가지 치과용 린스 제형으로 제형될 수 있다. 극히 미세한 입도는 오르모실, 무기 복합물, 합성 유도 무기 물질, 졸-겔 유도 물질 또는 광물 전구물질로부터의 천연 유도 칼슘 실리케이트 중 어느 하나의 균일한 분산액을 보장하는데 요구된다. 이는 물질의 화학적 개질 또는 기계적 밀링에 의해 달성될 수 있다.

Claims (20)
- 필수적으로 칼슘, 규소 및 산소로 이루어진 무정형 생활성 미립 물질을 포함하는, 석회화 조직에서의 결손을 회복시키기 위한 조성물.
- 제 1 항에 있어서, 무정형 물질이a) 유기 실리케이트 공급원과 칼슘의 공급원의 반응 생성물;b) 칼슘을 함유하는 테트라에틸오르토실리케이트의 가수분해 생성물;c) 칼슘 함유 실리카 졸-겔;d) 이성분 산화칼슘 및 실리케이트 침전 물질;e) 천연 규회석형 칼슘 실리케이트의 합성 유사체 및f) 가용성 칼슘 공급원과 실리케이트 용액의 침전된 반응 생성물로 이루어진 군으로부터 선택됨을 특징으로 하는 조성물.
- 제 2 항에 있어서, 무정형 물질이 가용성 칼슘 공급원과, 칼륨 수용액 또는 나트륨 실리케이트 용액으로부터 선택된 희석된 실리케이트 공급원의 침전된 반응 생성물임을 특징으로 하는 조성물.
- 제 1 항 내지 제 3항 중의 어느 한 항에 있어서, 무정형 물질의 입도가 약 10마이크론 이하의 범위내에 있음을 특징으로 하는 조성물.
- 제 4 항에 있어서, 무정형 물질 대부분이 입도가 약 2마이크론 이하인 입자를 포함함을 특징으로 하는 조성물.
- 제 1 항 내지 제 5항 중의 어느 한 항에 있어서, 과민성 치아의 상아질 세관내에 칼슘 함유 무기질의 형성을 유발시키므로써, 노출된 상아질 표면 및 개방 상아질 세관의 존재로 인한 치아 과민증을 완화시키는 치과 생성물임을 특징으로 하는 조성물.
- 제 6 항에 있어서, 담체 및 약 1 내지 25%의 무정형 물질을 포함함을 특징으로 하는 조성물.
- 제 7 항에 있어서, 담체가 0 내지 30%의 물, 0 내지 90%의 글리세롤 및 0 내지 80%의 소르비톨을 포함함을 특징으로 하는 조성물.
- 제 7 항 또는 제 8항에 있어서, 무정형 물질이 조성물의 약 5 내지 10%이고, 조성물이 치약임을 포함함을 특징으로 하는 조성물.
- 석회화 조직에서의 결손을 회복시키기 위한 생성물을 제조함에 있어서의, 필수적으로 칼슘, 규소 및 산소로 이루어진 무정형 생활성 미립 물질의 용도.
- 제 10항에 있어서, 과민성 치아의 상아질 세관내에 칼슘 함유 무기질의 형성을 유발시키므로써, 노출된 상아질 표면 및 개방 상아질 세관의 존재로 인한 치아 과민증을 완화시키는 치과 생성물을 제조함에 있어서의 용도.

- 제 13항에 있어서, 치아의 탈감작화에 사용하기 위한 무정형 생활성 미립 물질.
- 제 13 항 또는 제 14항에 있어서,a) 유기 실리케이트 공급원과 칼슘의 공급원의 반응 생성물;b) 칼슘을 함유하는 테트라에틸오르토실리케이트의 가수분해 생성물;c) 칼슘 함유 실리카 졸-겔;d) 이성분 산화칼슘 및 실리케이트 침전 물질;e) 천연 규회석형 칼슘 실리케이트의 합성 유사체 및f) 가용성 칼슘 공급원과 실리케이트 용액의 침전된 반응 생성물로 이루어진 군으로부터 선택된, 무정형 생활성 미립물질.
- 석회화 조직에서 결손을 회복시키기 위한 무정형 생활성 미립 물질을 제조하는 방법에 있어서, 필수적으로 충분량의 가용성 칼슘 공급원 용액을 실리케이트 또는 실리케이트 전구물질 용액에 첨가하여 칼슘 함유 실리케이트 미립물질을 침전시키는 단계로 이루어진 방법.
- 제 16 항에 있어서, 가용성 칼슘 공급원이 칼슘 염 및 알콕사이드 유도체 및 이들의 혼합물로 이루어진 군으로부터 선택됨을 특징으로 하는 방법.
- 제 16 항 또는 제 17 항에 있어서, 실리케이트 또는 실리케이트 전구물질 용액이 칼륨 수용액 및 나트륨 실리케이트 수용액의 군으로부터 선택된 희석된 실리케이트 공급원임을 특징으로 하는 방법.
- 과민성 치아의 치료를 위한 무정형 생활성 미립물질을 제조하는 방법에 있어서, 필수적으로, 금속 알콕사이드를 물 및 촉매와 혼합하는 단계와 이러한 무정형 생활성 미립물질을 제조하기 위한 실리케이트 에스테르를 중합하는 단계로 이루어지는 방법.
- 제 19 항에 있어서, 금속 알콕사이드가 테트라에톡시실란 및 실란올 말단 폴리(디메틸실록산)으로부터 제조됨을 특징으로 하는 방법.
