Abstract

Description
전통적인 활성제 전달 수단은 종종 생물학적, 화학적 및 물리적 장벽에 의해 심하게 제한된다. 통상적으로, 이러한 장벽들은 전달이 일어나는 동안 거치는 환경, 전달하려는 표적의 환경 또는 표적 그 자체에 의해 부과된다. 생물학적 또는 화학적 활성제는 그러한 장벽에 대해 취약하다.

활성제
본 발명에 사용하기에 적합한 활성제는 생물학적 또는 화학적 활성제이다.

아실화된 아미노산은 하나의 아미노산, 두 개 이상의 아미노산 또는 아미노산 에스테르가 아미노산에 존재하는 유리 아미노기와 반응하는 아민 변형제와 반응하여 아미드를 생성하는 것에 의해 제조될 수 있다.
적합한, 그러나 비제한적인, 아실화된 아미노산 제조에 유용한 아실화제의 예는 다음과 같은 화학식을 갖는 산 염화물(acid chloride) 아실화제를 포함한다.

변형 아미노산 생성 반응은 통상 진한 염산과 같은 적당한 산으로 pH가 약 2 내지 3에 도달할 때까지 혼합물의 pH를 조절함으로써 중단(quench)된다. 상기 혼합물을 실온에서 놓아두면 분리되어 투명한 상층 및 흰색 또는 회색이 도는 흰색 침전을 생성한다. 상층을 버리고, 변형된 아미노산을 여과 또는 기우려 따르기( decantation)하여 모은다. 상기 미정제 변형 아미노산을 물과 혼합한다. 용해되지 않는 물질을 여과하여 제거한 후 여과액을 진공 건조한다. 통상적으로 변형 아미노산의 수득률은 약 30 내지 60% 범위이며, 대개 약 45%이다. 본 발명은 또한 디아실화, 트리아실화 등과 같은 다중(multiple) 아실화에 의해 변형된 아미노산도 예상한다.
아미노산 에스테르 또는 아미드가 출발 물질이라면, 그들을 디메틸 포름아미드 또는 피리딘 같은 적당한 유기 용매에 녹이고, 5℃ 내지 70℃ 온도 범위에서, 바람직하게는 약 250℃에서, 7 내지 24 시간 동안 아미노 변형제와 반응시킨다. 아미노산 에스테르의 양에 대한 사용된 아미노 변형제의 양은 상기 아미노산의 경우와 같다.

산 침전, 재결정 또는 고체 컬럼 담체위에서 분별(fractionation)하여 변형된 아미노산을 정제할 수 있다. 분별은 아세트산/부탄올/물 같은 혼합 용매를 이동상으로 사용하여 실리카겔 또는 알루미나와 같은 고체 컬럼 담체 위에서, 트리플로로아세트산/아세토니트릴 혼합물을 이동상으로 사용하여 역상 컬럼 담체 위에서, 및 물을 이동상으로 사용하여 이온 교환 크로마토그래피로 할 수 있다. 또한 메탄올, 부탄올 또는 이소프로판올과 같이 분자량이 작은 알코올로 추출하여 무기염과 같은 불순물을 제거함으로써 변형된 아미노산을 정제할 수 있다.
변형된 아미노산은 통상 염기성 수용액(pH≥9.0)에 용해되고, 에탄올, n-부탄올 및 톨루엔/에탄올 1:1(v/v) 용액에는 부분적으로 용해되며, 중성 물에는 용해되지 않는다. 변형된 아미노산의 소디움 염과 같은 알칼리 금속염은 통상 약 pH 6-8인 물에 용해된다.
폴리아미노산 또는 펩티드에서 하나 이상의 아미노산이 아실화 및/또는 술폰화될 수 있다. 폴리아미노산 및 펩티드는 아실화된 아미노산을 하나 이상 포함할 수 있다. 선형으로 변형된 폴리아미노산 및 펩티드는 통상 아실화 아미노산을 하나만 포함하지만, 다른 폴리아미노산 및 펩티드 배열(configuration)은 아실화된 아미노산을 하나 이상 포함할 수 있다. 폴리아미노산 및 펩티드는 아실화된 아미노산과 중합되거나, 또는 중합 후에 아실화될 수 있다.

본 발명에 유용한 조성물은 하나 이상의 활성제를 포함할 수 있다.
한가지 구체례에서, 투여 전에 하나 이상의 화합물 또는 염, 폴리아미노산 또는 펩티드를 활성제와 간단히 혼합함으로써, 하나 이상의 상기 화합물 또는 염을 포함한 상기 화합물 또는 상기 화합물의 염 또는 폴리아미노산 또는 펩티드가 직접 전달 담체로 사용될 수 있다.
투여 혼합물은 투여 직전에 담체의 수용액을 활성 성분의 수용액과 혼합함으로써 제조된다. 대체 방안으로, 담체 및 생물학적 또는 화학적 활성 성분은 생산과정 중에 혼합될 수 있다. 용액은 임의로 포스페이트 완충염, 구연산, 아세트산, 젤라틴 및 아라비아고무와 같은 첨가제를 함유할 수 있다.

실시예 1 - 담체의 제조
2-(4-(N-살리실로일)아미노페닐)프로피온산(담체 B)의 제조
2-(4-아미노페닐)프로피온산 58.6g(0.355mol)과 염화 메틸렌 500ml의 슬러리를 클로로트리메틸실란 90.11ml(77.13g, 0.710mol)로 처리하고, 120분 동안 가열 환류하였다. 상기 반응 혼합물을 0℃로 냉각시키고, 트리에틸아민 184.44ml(107.77g, 1.065mol)으로 처리하였다. 5분 동안 교반한 후에, 상기 혼합물을 150ml의 염화 메틸렌에 용해된 70.45g(0.355mol)의 O-아세틸살리실로일 클로라이드 용액으로 처리하였다. 상기 반응 혼합물을 25℃까지 올린 후, 64시간 동안 교반하였다. 휘발물은 진공 제거하였다. 잔류물을 2N 수산화나트륨 수용액 내에서 1시간 동안 교반하고, 2M 황산 수용액으로 산성화하였다. 상기 고체를 에탄올/물로 2회 재결정하여 황갈색 고체를 얻었다. 여과에 의한 분리로 2-(4-(N-살리실로일)아미노페닐)프로피온산 53.05g(수득률 52%)을 얻었다.

소디움 2-(4-(N-살리실로일)아미노페닐)프로피오네이트(담체 B의 소디움 염)의 제조
2-(4-(N-살리실로일)아미노페닐)프로피온산 53.05g(0.186mol)과 에탄올300ml의 용액을 물 22ml에 용해된 NaOH 7.59g으로 처리하였다. 반응 혼합물을 25℃에서 30분 동안 및 0℃에서 30분 동안 교반하였다. 얻어진 엷은 노란색 고체를 여과에 의해 분리하여 소디움 2-(4-(N-살리실로일)아미노페닐)프로피오네이트 52.61g을 얻었다.
성질. 용해도: 200mg/ml 투명한 용액, pH=6.85. 분석. 계산값 - C:60.45, H:5.45, N:3.92, Na:6.43 측정값 - C:60.84, H:5.87, N:3.85, Na: 6.43. 녹는점 236-238℃.
담체 C의 소디움 염의 제조
마그네틱 교반기와 환류 응축기(reflux condenser)를 장치한 2L 둥근 바닥 플라스크에 염화메틸렌(250ml)에 대한 3-(4-아미노페닐)프로피온산(15.0g, 0.084mol, 1.0당량) 현탁액을 넣었다. 클로로트리메틸실란(18.19g, 0.856mol, 2.0당량)을 한번에 첨가하고, 혼합물을 아르곤 분위기 하에서 1.5시간 동안 가열 환류하였다.

무수 아세트산(acetic anhydride)(32ml, 34.5g, 0.338mol, 1.03당량), 4-메틸살리실산(50g, 0.329mmol. 1.00당량) 및 크실렌(100ml)를 마그네틱 교반기, 온도계 및 응축기가 장치된 1L 4구 플라스크에 넣었다. 상기 플라스크를 모래 중탕(sand bath)에 넣고, 탁한 흰색 혼합물을 가열하기 시작했다. 상기 반응 혼합물은 90℃ 근처에서 노란 용액 상태로 투명해졌다. 휘발성 유기물(크실렌 및 아세트산)의 대부분을 딘-스탁(Dean-Stark) 트랩으로 3시간(135-146℃) 동안 증류하였다. 추가적으로 1시간 동안 중류를 계속하였고(증류액의 총량은 110ml), 그 동안 반응용기(pot)의 온도가 204℃까지 천천히 올라갔고, 증류액이 떨어지는 속도가 느려졌다. 잔류물을 뜨거운 상태에서 알루미늄 쟁반에 부었다. 식으면서 부서지기 쉬운 노란색 유리가 생성되었다. 상기 고체를 미세 분말로 갈았다. 얻어진 올리고(4-메틸살리실레이트)를 추가적인 정제과정 없이 사용하였다.
(b) N-(4-메틸살리실로일)-8-아미노카프릴산의 제조
7M 탄산칼륨 용액(45ml, 43.2g, 0.313mol, 0.95당량), 8-아미노카프릴산(41.8g, 262mol, 798당량) 및 물(20ml)를 마그네틱 교반기, 응축기 및 첨가 깔때기가 장치된 1L 둥근 바닥 플라스크에 가하였다. 디옥산(250ml)에 용해된 올리고(4-메틸살리실레이트)(44.7g, 0.329mmol, 1.0당량)용액을 흰색의 탁한 혼합물에 30분 동안 가하여 반응시켰다. 상기 반응 혼합물을 90℃에서 3시간(반응이 HPLC에 의해 완결되었다고 결정된 시간)동안 가열하였다. 상기의 투명한 오렌지색 반응 혼합물을 30℃까지 냉각시키고, 50% 황산 수용액(64g)으로 pH=2가 되도록 산성화하였다. 얻어진 고체를 여과하여 분리하였다. 흰색 고체를 50% 에탄올-물 1170ml로 재결정하였다. 상기 고체를 여과하여 회수하고, 50℃의 진공 오븐에서 18시간 동안 건조하였다. N-(4-메틸살리실로일)-8-아미노카프릴산은 흰색 고체로 분리되었다.(30.88g, 52%); 녹는점=113-114℃;1H NMR(DMSO-d6) δ12.80(s, 1H), 12.00(s, 1H), 8.73(bt, 1H), 7.72(d, 1H), 6.70(s, 1H), 6.69(d, 1H), 3.26(q, 2H), 2.26(s, 3H), 2.19(t, 2H), 1.49(m, 4H), 1.29(m, 6H). 분석. 계산값 C16H23NO4: C65.51; H7.90; N4.77. 측정값: C65.48; H7.84; N4.69.

절차
1. 용액
·크실라진(100mg/ml) : 케타민(100mg/ml) : 아세프로마진(10mg/ml)을 1:3:1의 비율로 함유하는 마취제 용액의 칵테일
·헤파린 20 units/ml를 함유하는 0.9% 염화나트륨 주사제 USP
·인슐린을 0.01N HCl로 적정된 pH=3인 증류수에 용해시켰다. 용액이 투명해질 때 pH 7인 증류수를 첨가하였다. 그 후, 0.01 NaOH로 pH를 7.5로 조절하였다.
·담체 B를 pH 7.4인 증류수에 용해시키고, 2분 동안 초음파 처리한 다음 0.1N NaOH를 사용하여 pH를 7.4로 조절하였다.
2. 동물
각 쥐(rat)의 무게를 달고, 지워지지 않는 식별 표지를 하고, 80mg/kg,10mg/kg 및 2.0mg/kg 들어있는 케타민:크실라진:아세프로마진 칵테일을 복강내에 주사하여 마취하고, 우리에 넣었다. 상기 쥐를 수술대로 옮기고 직장 탐침을 통해 항온 조절되는 가열 담요 위에 놓아 깊게 마취되도록 하였다.
각 쥐의 우측 경부 정맥으로 실리콘 튜빙을 사용하여 카테터를 꽂고, 200μl의 소금기를 함유한 헤파린을 캐뉼라에 흘려준 후에, 100μl의 혈액을 채취하여 상기 카테터의 개방을 확인하였다. 그후 헤파린이 첨가된 식염수 200μl로 상기 캐뉼라를 세척하였다.
광섬유 후두경을 사용하여 기관내(endotracheal) 튜브를 삽입하였다. 점적 주사기(Hamilton syringe)를 기관내 튜브에 부착하고, 공기 통로를 통하여 용액(100μl)을 점적하기 위해 사용하였다. 투여 후 기관내 튜브를 제거하고, 나머지 연구기간 동안 호흡속도를 모니터하였다.
0, 10, 30, 60, 90, 120 및 180분에 혈액 시료(500μl)를 채취하고, 약 200μl의 헤파린 첨가 식염수를 캐뉼라에 흘려주었다. 혈액을 1ml 투베르쿨린 주사기(20μ/ml헤파린 100μl를 포함하는)에 모아서 직접 마이크로 원심분리(microcentrifuge) 튜브에 넣었다.
혈액 시료를 즉시 10,000rpm에서 4분 동안 원심분리하였다. 약 30μl의 분취량(aliquot)을 꼬리표를 단 1ml의 실리콘 처리된 에펜도르프 튜브로 옮기고, 이어지는 포도당 측정을 위한 연구가 완결될 때까지 냉동하였다. 남은 시료들은 인슐린 측정을 위하여 -70℃로 냉동시켰다(Ultra-sensitive RIA).
분석
1. 포도당 측정
엑타켐 DT 슬라이드(Ektachem DT Slide)(GLU) 방법을 사용하여 단식 혈당 수준(plasma glucose level)을 측정하였다. 상기 분석은 산소 분자와 포도당의 효소 촉매 반응과, 이어지는 고도로 착색된 적색 염료를 생성하는 두 번째 반응에 기초한다. 색깔의 강도는 시료내의 포도당 양에 비례한다.
2. 약역학적 분석
시간에 대한 평균 혈장 인슐린 농도에 대해 표준 구획 및 비구획 약역학 분석을 하였다. 통상 구획 분석과 비구획 분석이 구획 모델의 정당성을 증명하기 위하여 동시에 행해진다.
a. 구획분석(Compartmental analysis)
돼지 인슐린의 기관내 분사(spray-intracheal(IT)) 투여에서 있어서, 최소자승 비선형 회귀분석법(least-squares nonlinear regression method)(PKanalyst, Micromath, Salt Lake City, UT)을 사용하여 평균 인슐린 농도(CINS) 대 시간 양상을 1구획 모델, 1차 흡수에 맞추었다. 다음의 식에 따라 평균 인슐린농도를 맞추었다.
여기서 A=Dka/Vd(ka-ke)는 시간 0에서 체내의 약물 농도이고, ka는 1차 흡수 속도 상수이고, ke는 1차 제거 속도 상수이다.
사다리꼴 법칙에 의해 폐 농도-시간 곡선의 아래부분 면적(area under thelung concentration-time curve=(AUC0→t))을 계산하였다. 측정된 최종 Cins를 말기의 1차 속도 상수로 나누어 무한대 보간법을 수행하였다. 상기 두 화합물의 합으로 곡선 아래부분의 전체면적을 계산하였다.
제거 반감기는 0.693/ke로부터 계산되는데, ke는 농도-시간 양상의 로그(log)의 말기의 기울기이다.
b.비구획 분석(Noncompartmental analysis)
표준 기술(standard technique)을 사용하여 시간에 대한 인슐린 플라즈마 농도에 대한 비구획 분석을 하였다. 치료를 위한 비조절 평균 혈장농도(CPins)로부터 피크 CPins에 대한 시간(Tmax) 및 피크 CPins(Cmax)를 계산하였다. AUC는 사다리꼴 법칙에 의해 계산되었다. 약역학 변수를 계산하기 위한 다음의 정맥투여에 따른 자료가 없기 때문에 MRT를 계산하지 않았다.
3.약역학 분석
처리를 위한 평균 혈당 수준 대 시간 양상으로부터 최소 혈당 농도%(percentage minimum plasma glucose concentraion=%MPGC) 및 각 %MPGC를 얻기 위한 시간 T(T%MPGC)를 결정하였다.
곡선 윗부분의 면적 효과(area above the curve effect =AACE)는 다음과 같이 계산되었다.
여기서 AUCE는 사다리꼴 법칙을 사용하여 계산된 곡선 아래 면적 효과를 나타낸다.
0→t에서 혈당의 전체 감소 %(percentage total reduction in plasma glucose=%TRPG0→t)는 다음의 식을 사용하여 결정되었다.
4.통계 분석
인슐린 투여량간의 AUC차이는 α<0.05로 가정한 Sheffe's 다중 비교 검사(multiple comparison test)를 사용하여 그 유의성을 검사하였다. 인슐린 단독인 것과 담체 B와 결합된 것 사이의 AACE0→3(혈당수준) 차이는 터키(Turkey's) 테스트 및 α<0.05로 가정한 분산분석을 사용하여 그 유의성을 검사하였다.
실시예 2a
(i) 담체 B 5mg/kg과 혼합된 인슐린 0.05mg/kg을 폐기관내 분사 점적한 경우 혈장인슐린 및 혈당 - 시간 양상은 도 1 및 2에서 각각 나타낸다.
(ii) 및 (iii) 도 3과 4 및 아래 표 1은 인슐린 0.05 및 0.01 mg/kg과 각각 혼합된 담체 B 16mg/kg의 폐 기관내 분사 점적한 경우 혈당-시간 양상 및 약물 동력학 변수들을 보여준다. 담체의 존재 및 비존재 하에서 인슐린을 투여하는 경우에, AACE0→3의 유의성 차이(p<0.05)를 관찰하였다. 인슐린(담체 B 16mg/kg과 결합된) 투여량 0.01 및 0.05에 대한 %TRGP0→3는 각각 10.5±1.5 내지 36±9% 및 47±10 내지 65.7±5%까지 상승하였다. 인슐린 0.05mg/kg을 단독으로 사용한 경우와 담체 B 5mg/kg와 함께 투여한 것 사이에 %TRGP0→3값의 통계적으로 유의한 차이가 없었다. 담체 B에 대해 발견된 이러한 투여량-효과 관계는 인슐린의 약물동력학에 미치는 담체의 영향(증가된 생체 이용률) 때문인 것 같다.
상기의 데이터는 인슐린의 생체 이용률을 상당히 증가시키는 담체 B의 잠재능, 및 포도당 수준에 미치는 담체 B의 영향을 시사한다.
(iv) 인슐린 투여량의 함수로 나타낸 기관내 분사 점적에 따른 평균 인슐린 혈장 농도를 도 5에서 보여준다. 구획 분석 및 비구획 분석에서 얻어진 약물동력학 변수들은 아래의 표 2에 정리하였다. 평균 혈장 수준의 비선형 커브 맞추기는 플라즈마 농도가 낮아지는 것은 모든 경우에 1차 흡수에 대해 1차 지수함수적으로 감소하는 것으로 가장 잘 기술되었다. 상대적인 모델 선택 표준(MSC)과 비교함으로써 모형화 우수성에 대한 평가를 하였다. 상기 변형된 아카이케 정보(Akaike information) 표준은 모델의 적합성의 비교를 가능하게 한다. MSC 값은 모델의 적합성을 나타낼 때 1.3 내지 3.8의 범위이다.
이 연구에서 얻어진 평균 피크 혈장 농도는 돼지 인슐린 0.001mg/kg (0.026U/kg), 0.005mg/kg (0.13U/kg), 0.01mg/kg (0.26U/kg), 0.05mg/kg (1.3U/kg), 0.1mg/kg (2.6U/kg), 0.5mg/kg 및 1mg/kg에 대하여 각각 57.5μU/ml (Tmax=10분), 18.8μU/ml (Tmax=20분), 71.3μU/ml (Tmax=10분), 48.95μU/ml (Tmax=10분), 213.1μU/ml (Tmax=10분), 354μU/ml (Tmax=20분) 및 902μU/ml (Tmax=20분)이었고, 그것은 곡선모형화 후에 얻어진 값과 가까운 일치를 보여준다.
인슐린 투여량에 대한 함수로서의 로그 혈장 농도-시간 도표(도 5)는, 토끼에 0.1u/kg의 소 인슐린을 정맥투여한 후에 얻은 이전의 보고보다(t1/2=30분), 특정된 반감기가 상당히 길어질수록(7.8 내지 142.4분. 표 2 참조.) 혈장농도가 감소함을 나타낸다. 상기 관찰결과는 "flip-flop" 케이스의 존재를 시사하는 것으로, 즉 제거보다는 흡수가 혈장 농도 감소를 지배하는 것을 나타낸다. 그러나 이러한 흡수 반감기의 증가는 흡수가 속도 결정 단계라는 것을 암시하는 것은 아니다. 오히려, 이 사건은 점적부피당 인슐린 농도가 증가함에 따라 인슐린이 헥사머(hexamer)에서 모노머(monomer) 형으로 전환되거나, 느리게 "용해" 되는 결과일 수 있다.
사다리꼴 법칙을 사용하여 얻은, 무한대까지의 커브 아래 면적(AUC0→∞)은 돼지 인슐린 0.001mg/kg(0.026U/kg), 0.005mg/kg(0.13U/kg), 0.01mg/kg(0.26U/kg), 0.05mg/kg(1.3U/kg), 0.1mg/kg(2.6U/kg), 0.5mg/kg 및 1mg/kg에 대하여 각각 2930, 1334, 5050, 5260, 15374, 67676 및 200230μU min/ml 이었다.
Sheffe's 테스트를 사용한 투여량 사이의 다중(multiple) 비교는 AUC 값에 있어서 유의성 차이(p>0.05)를 보여주지 않았다. 이것은 고도의 대상간 가변성 때문일 것이다(표 2에서 AUC에 대한 표준편차 참조.). 폐 투여에 따른 돼지 인슐린의 흡수정도 및 피하 투여에 따른 인간 인슐린의 흡수 정도가 동등하다고 가정함으로써, 사용된 인슐린의 상대적인 생체 이용률을 계산할 수 있다. 이러한 가정에 근거할 때, 돼지 인슐린의 상대적인 생체 이용률은 12.46%이다.
점적된 인슐린의 다양한 투여량에 대한 혈당-시간 양상 및 약물동력학 변수(PD) 자료는 도 6 및 아래의 표 3에서 보여준다. 그 결과는 투여량의 증가(0.01에서 1mg/kg로)는, 최소값에 도달하는 시간 T (T%MPGC)의 상당한 변화 없이, 최소 혈당 % (%MPGC) 값을 70.2에서 13.1로 상당히 감소시킨다는 것을 보여준다. 0에서 3시간까지 혈당의 전체 감소량(%TRPG0→3hr)(표 3 참조.)은 10.5에서 73.7%로 상당히 상승하는데, 이것은 인슐린 투여량이 증가함에 따라 흡수 정도가 증가함으로써 고도의 생체 이용률이 달성되었다는 것을 의미한다.
포도당 반응에 있어서 대상간 가변성 (CV<30%)은 인슐린 반응보다 작다. 따라서, 순환계에서 호르몬의 출현이 그것의 생물학적 활성으로 해석되는만큼 인슐린 가변성은 완충(dampening)된다.
실시예 2b
폐 기관내 분사 점적으로 돼지 인슐린 0.05mg/kg 및 담체 B 16mg/kg을 포함하는 조성물을 쥐에게 투여하였다. 돼지 인슐린 0.05mg/kg의 단독 조성물 역시 폐 기관내 분사 점적으로 쥐에게 투여하였다. 결과는 도 7 및 표 4 및 5에서 설명된다.

비교 실시예 2c
아래의 표 6에서 보여준 것과 같이, 폐 기관내 분사 점적으로 돼지 인슐린 조성물의 투여량을 단계적으로 증가시켜 쥐에게 투여하였다. 결과는 도 8 및 표 6 및 7에서 설명된다.
실시예 2d
돼지 인슐린 0.05mg/kg 및 담체 B 5mg/kg의 조성물을 폐 기관내 분사 점적으로 쥐에게 투여하였다. 돼지 인슐린 0.05mg/kg 단독 조성물을 또한 폐 기관내 분사 점적에 의해 투여하였다. 혈장 인슐린 수준은 도 9 및 표 5 및 8에서 설명된다.
실시예 2e
돼지 인슐린 0.01mg/kg 및 담체 B 16mg/kg의 조성물을 폐 기관내 분사 점적으로 쥐에게 투여하였다. 돼지 인슐린 0.01mg/kg의 단독 조성물 역시 폐 기관내 분사 점적에 의해 쥐에 투여되었다. 혈장 인슐린 수준은 도 10 및 표 5 및 9에서 설명된다.
실시예 3 - 돼지 인슐린의 폐 전달
실시예 3a
물에 용해된 돼지 인슐린 0.1mg/kg 및 담체 B의 소디움 염 7.5mg/kg의 폐 전달 투여(pulmonary delivery dosing) 조성물을 제조하였다. 돼지 인슐린 0.1mg/kg의 단독 및 담체 B의 소디움 염 7.5mg/kg의 단독의 제어 투여(control dosing) 조성물 또한 제조하였다. pH 7.3-7.6 상태인 폐 투여 조성물 0.3ml/kg 투여량을, 다음과 같은 절차에 의해, 단식하지 않은(non-fasting) 보통의 쥐 5마리에게 투여하였다. 1과 1/2 포퍼 앤드 썬즈 가비지(Popper and Sons gavage) 바늘을 동물의 목 아래로 약 몇 cm 주입하였다. 바늘 끝을 바늘이 포켓으로 떨어질 수 있는 복부 면(ventral side)을 향하여 조작하였고, 추가적인 조작으로 기관(trachea)에 이르게 하였다. 바늘이 기관 내에 있을 때 투여 용액을 바늘을 통하여 전달하였다.
혈액 시료를 주기적으로 말초 동맥(tail artery)을 통하여 채취하였고, 혈당수준을 엑타켐 DT 슬라이드(Ektachem DT slide)(Johnson & Hohnson Clinical Diagnostics사, 로체스터, 뉴욕)을 사용하여 측정하였다.
결과는 담체 B의 소디움 염 7.5mg/kg을 갖는 인슐린 0.1mg의 경우(■), 인슐린 0.1mg/kg 단독(●) 및 담체 B의 소디움 염 7.5mg/kg 단독(▲)에 대하여 도 11에서 설명된다.
실시예 3b
투여 조성물을 돼지 인슐린 0.5mg/kg 및 담체 B의 소디움 염 7.5mg/kg으로 바꾸고, 물에서 pH가 6.6-6.9 상태인 투여 조성물에 대하여 실시예 3a의 절차를 반복하였다. 돼지 인슐린 0.5mg/kg의 단독 및 담체 B의 소디움 염 7.5mg/kg의 단독의 제어 투여(control dosing) 조성물 또한 제조하였다.
결과는 담체 B의 소디움 염 7.5mg/kg을 갖는 인슐린 0.5mg/kg의 경우(■), 인슐린 0.5mg/kg 단독(●) 및 담체 B의 소디움 염 7.5mg/kg 단독(▲)에 대하여 도 12에서 설명된다.
실시예 4 - 인슐린 폐 전달
표 10에 나타낸 담체 화합물 B, 담체 화합물 C 소디움 염 및 담체 화합물 D를 다음과 같이 검사하였다. 각 쥐를, 무게를 달고, 지워지지 않는 식별 표지를 하고, 토라진(thorazine)(3mg/kg) 및 케타민(44mg/kg)을 근육 주사하여 마취하였다.
주사기(Hamilton syringe)가 부착된 기관내 튜브(Penn Century사(필라델피아, PA)에서 구입한 분사점적기)를 광섬유 후두경을 사용하여 삽입하였다. 공기통로(airway)의 하부 부분으로 인슐린(0.03mg/kg) 및 담체 화합물(16mg/kg)이 함유된용액 0.4ml/kg을 점적하기 위해 점적기의 주사기를 사용하였다. 투여 후 기관내 튜브를 제거하고, 호흡속도를 연구의 나머지 부분을 통하여 모니터하였다.
혈액 시료(500μl)를 0, 10, 30, 60, 90 및 120분에 말초 동맥을 통하여 채취(withdrawn)하여 DSL 인슐린 킷(Insulin Kit) #10-1600으로 킷에 약술된 절차에 따라 분석하였다. 혈청(serum) 인슐린 수준은 도 13에서, Cmax는 아래의 표 10에서 설명된다.

- 활성제를 필요로 하는 동물에 생물학적 활성제를 투여하는 방법으로서, 상기 방법이 (A) 활성제 및 (B) 아실화된 아미노산, 술폰화된 아미노산, 아실화된 아미노산을 포함하는 폴리아미노산, 술폰화된 아미노산을 포함하는 폴리아미노산 또는 이들 중 어떤 것의 조합 중 어느 하나를 포함하는 담체를 포함하는 조성물을, 상기 동물에 폐 경로를 통하여 투여하는 것을 포함하는 방법.
- 제 1 항에 있어서, 상기 활성제가 생물학적 활성제, 화학적 활성제 및 이들의 조합으로 구성된 군(group)에서 선택되는 방법.
- 제 2 항에 있어서, 상기 생물학적 활성제가 하나 이상의 펩티드, 무코 다당류(mucopolysaccharide), 탄수화물 또는 지방을 포함하는 방법.
