シリンジ(Hamiltonシリンジ)を接合してなる気管内チューブ(スプレー点滴注射器、Penn Century of Philadelphia, PA製)を、光ファイバー喉頭鏡を用いて挿入した。点滴注射器の前記シリンジ(Hamiltonシリンジ)を利用して、インスリン(0.03mg/kg)及び担体化合物(16mg/kg)を、気道の下方部に滴下した。投与に次いで、気管内チューブを取り外し、これ以降の研究の間、呼吸速度をモニターした。
血液サンプルを、0、5、15、30、60、及び120分の時点で、尾の動脈から採り、DSL Inslin Kit #10-1600を用い、該キットに表示された操作に従って検定した。担体を用いたCmax値は、44.6±10.0であった。担体なしのCmax値は、19.4±4.3であった。

(実施例10:組換えヒト成長ホルモン(rhGH)の経口デリバリー)
リン酸緩衝液100%中に、担体化合物2-(4-(N-サリチロイル)アミノフェニル)プロピオン酸200mg/kg及びrHGH3mg/kgを含有する経口栄養(PO)投与溶液を、混合によって調製した。該溶液のpHは、約7.67から約8.09までの範囲であった。
体重200−250gのオスのSpague-Dawleyラットを、24時間断食させ、投薬の15分前に、ケタミン(44mg/kg)及びクロルプロマジン(1.5mg/kg)を投与した。該ラットに、投薬溶液1ml/kgを、POにより投与した。血液サンプルを、尾の動脈から非経口採取し、血清中rHGH濃度の測定に用いた。血清中rHGH濃度を、rHGH免疫アッセイテストキット(Genzyme Corporation Inc., Cambridge, MA製、Kit#K1F4015)によって計量した。rhGHの平均最高血清中濃度は、93±74ng/mlであった。

(実施例11:エリトロポエチン(EPO)の経口デリバリー)
水100%中に、担体化合物2-(4-(N-サリチロイル)アミノフェニル)プロピオン酸200mg/kg及びヒトエリトロポエチンクアンチキン(quantikine)IUD1mg/kgを含有する経口栄養(PO)投与溶液を、混合によって調製した。該溶液のpHは、約6.9から約7.9までの範囲であった。
体重200−250gのオスのSpague-Dawleyラットを、24時間断食させ、投薬の15分前に、ケタミン(44mg/kg)及びクロルプロマジン(1.5mg/kg)を投与した。該ラットに、投薬溶液1ml/kgを、POにより投与した。血液サンプルを、尾の動脈から非経口採取し、血清中EPO濃度の測定に用いた。血清中EPO濃度を、Elisaアッセイテストキット(R&D Systems, Minneapolis, MN製、Kit#DEPOO)によって計量した。EPOの平均最高血清中濃度は、163±136ng/mlであった。
この操作を、担体化合物を使用せずに繰り返した。平均最高血清中濃度は、0ng/mlであった。

Claims (11)
- 前記生物学的に活性なペプチドが、インスリンを含み、前記インスリンがブタインスリンまたはヒトインスリンである、請求項1に記載の組成物。
- 前記生物学的に活性なペプチドが、パラチロイドホルモンを含む、請求項1に記載の組成物。
- 前記生物学的に活性なペプチドが、エリトロポエチンを含む、請求項1に記載の組成物。
- (I)請求項1に記載の組成物;及び
(II)(a)賦形剤、
(b)希釈剤、
(c)錠剤分解物質、
(d)潤滑剤、
(e)可塑剤、
(f)着色剤、
(g)投薬媒体、または
(h)これらのあらゆる組み合わせ;
を含み、前記活性剤が、生物学的に活性なペプチドである、投薬製剤。 - 前記生物学的に活性なペプチドが、インスリンを含み、前記インスリンがブタインスリンまたはヒトインスリンである、請求項5に記載の投薬製剤。
