Claims (11)
- 다음 반응식에 따라a) 5-히드록시-1,3-벤젠디카르복실산을 n-부탄올 및 산성 촉매작용에 의해 에스테르화시켜 화학식 VIII의 5-히드록시-1,3-벤젠디카르복실산 n-부틸 디에스테르를 수득하는 단계;b) 화학식 VIII의 화합물을 이소세리놀 과량으로 아미드화시켜 N,N'-비스-(2,3-디히드록시프로필)-5-히드록시-1,3-벤젠디카르복스아미드의 수용액을 수득하고, 이를 임의로 화학식 III의 나트륨 염으로 변형시키는 단계;c) 화학식 III의 화합물을 화학양론적 양 또는 1% 과량의 ICl로 요오드화하여 화학식 IV의 N,N'-비스-(2,3-디히드록시프로필)-5-히드록시-2,4,6-트리요오도-1,3-벤젠디카르복스아미드를 수득하는 단계;d) 화학식 IV의 화합물을 수용액 상태의 화학식 IX의 화합물인 2-클로로-N-메틸-아세트아미드로 알킬화하여 습 생성물로서 사용되는, 화학식 VII의 N,N'-비스-(2,3-디히드록시프로필)-2,4,6-트리요오도-5-[2-(메틸아미노)-2-옥소에톡시]-1,3-벤젠디카르복스아미드를 수득하는 단계; 및e) 염기성 조건에서 화학식 VII의 화합물을 스마일 (Smiles') 재배열하고 이어서 정제하여 화학식 I의 이오메프롤을 수득하는 단계를 포함하는 N,N'-비스[2,3-디히드록시프로필]-5-[(히드록시아세틸)메틸아미노]-2,4,6-트리요오도-1,3-벤젠디카르복스아미드의 제조 방법.
- 제1항에 있어서, 6 몰%와 등가인 촉매량의 H2SO4 또는 p-톨루엔술폰산을 사용하는 방법.
- 제1항 또는 2항에 있어서, 냉각시켜 미리 농축시킨 최종 반응 혼합물로부터 직접 결정화시킴으로써 화학식 VIII의 화합물을 회수하는 방법.
- 제1항 또는 2항에 있어서, 화학식 VIII의 화합물을 유기 용매를 미리 제거한 알칼리 수용액으로부터 용융된 잔사로 침전시킴으로써 회수하고, 이어서 이를 무기 염기 또는 암모니아 수용액으로 처리한 다음 조절 냉각시켜 부분적으로 결정성인 고상물로서 화학식 VIII의 화합물을 수득하는 방법.
- 제4항에 있어서, 고상물로서 회수된 화학식 VIII의 화합물을 n-부탄올 중에 재용해시켜 후속 단계에서 그대로 사용되는 용액을 수득하는 방법.
- 제1항 또는 2항에 있어서, 단계 a)에서 수득한 화학식 VIII의 n-부틸 디에스테르를 회수하지 않고, 화학식 VIII의 화합물과 이소세리놀의 아미드화 반응 말기에 반응 혼합물을 물로 희석한 다음 강산성 양이온 교환 수지를 함유하는 제1 컬럼을 통하여 과량의 이소세리놀로부터 또한 제1 컬럼과 연결된, 제2 약염기성 음이온 교환 수지를 통하여 이온성 불순물로부터 정제하는 방법.
- 제1항 또는 제2항에 있어서, 단계 b)를 용융 상태에서 120% 이소세리놀 과량 중 90 내지 95 ℃의 온도에서 12시간 동안 수행하고, 상기 반응으로부터 형성된 n-부탄올을 진공 하에서 증류 제거하고, 반응 완료 후 그 물질을 물로 처리하여 수용액을 수득하고, 이를 양이온 교환 수지를 통하여 정제한 다음 NaOH로 pH 9 내지 10으로 조정하여 화학식 III의 화합물에 대응하는 나트륨 염의 수용액을 수득하는 방법.
- 제5항에 있어서, 반응 단계 b) 전에 용매를 제거하여 용융 잔사로서 화학식 VIII의 화합물을 수득하고, 이를 최종적으로 청구항 7의 조건 하에서 처리하는 방법.
- 제7항에 있어서, 과량의 이소세리놀을 회수하고 수지로부터 희석 암모니아 용액으로 용출시킴으로써 공정으로 재순환시키고, 상기 용액을 잔사로 농축시킨 다음 에탄올 용액 중에서 이소세리놀 옥살레이트를 형성시킴으로써 정제하고, 이어서 강산성 폴리스티렌 매트릭스 이온 교환 양이온 수지를 통하여 정제하고, 여기서 이소세리놀은 희석된 암모니아 용액으로 용출시킴으로써 회수하는 방법.
- 제1항 또는 제2항에 있어서, 요오드화를 요오드화제로서 중성 수용액 중의 ICl을 사용하여 pH 6 내지 7의 실온에서 요오드화제의 첨가량이 화학양론적 양 또는 약 과량 (약 1%)과 등가이도록 전위차계를 사용하여 조절하면서 수행하여 용액을 수득하고, 이를 이어서 직접 알킬화시키는 방법.
- 제1항 또는 제2항에 있어서, 화학식 IV의 화합물을 화학식 IX의 화합물로 알킬화시키는 단계를 중성 pH의 95 ℃의 온도에서 수행하고, 여기서 알킬화제를 기질 몰 당 1.8 내지 2.2 몰의 양으로 첨가하고, 상기 반응 말기에 형성된 현탁액을 냉각시켜 화학식 VII의 화합물의 회수 단계에 공급하는 방법.
