Abstract

본 발명은 에스트라디올 또는 그의 제약적으로 허용가능한 유도체를 단독으로 또는 게스타겐과 함께 인간 또는 동물피부에 적용시 방출을 제어하기 위한 유효물질 함유 패치에 관한 것으로서, 지지층과 : 상기 지지층에 결합되며, 조합된 유효물질 방출에 적당하며, 감압성 접착제를 사용하여 제조되는 유효물질 함유 저장부와 : 분리가능한 보호층으로 구성되며, 상기 감압성 접착제가 울 왁스 및 산화아연을 함유하는 것을 특징으로 한다.

Description

호르몬의 경피 적용용 에스트라디올 함유 패치{Estradiol-containing patch for transdermal application of hormones}
본 발명은 에스트라디올 또는 그의 제약적으로 허용 가능한 유도체를 단독으로 또는 게스타겐과 함께 인간의 피부 또는 동물의 피부에 적용시 방출을 제어하기 위한 유효물질 함유 패치(patch)에 관한 것이다.
에스트로겐 - 및/또는 게스타겐 함유 패치는 이미 공지되어 있다. 그러나 이러한 패치는 에탄올을 함유하고 있거나 또는 시간이 경과함에 따라 유효물질이 재결정되는 잠재적인 위험이 있다는 단점을 갖고 있다.
DE-OS 3205258호 및 EP 0285563호에는 패치에 에스트라디올 및 에탄올을 동시에 투여하는 것이 공지되어 있다. 그러나, 이러한 패치의 제조는 매우 복잡함은 물론 이러한 패치는 유연성이 부족하기 때문에 부착된 후에는 착용의 편안함이 전혀 없었다.
EP 0285563호에는 에스트로겐 및 게스타겐의 혼용을 위한 경피적 치료 시스템이 기술되어 있다. 저장부는 유효물질 제제 및 선택적으로 막 및 경피흡수 증강제로서 에탄올을 함유한다. 유효물질의 방출은 막에 의하여 주로 제어되기 때문에, 경피적 치료 시스템은 본 발명에 따른 유효물질 패치와는 근복적으로 상이하다. 상기 EP 0285563호에는 기술된 패치에 있어서, 접착제는 피부에 패치를 고정시키는 기능만을 갖고 있다. 이것이 유효물질 방출의 제어에 공헌할 수 있다는 사실은 그의 주요 임부를 구성하는 것이 아니고 부차적인 효과에 지나지 않는다. 이 패치는 유효물질 제제가 불투과성 지지층(backing layer), 및 접착층을 갖는 막으로 구성되는 파우치(pouch) 내에 존재하기 때문에 소위 말하는 "파우치 패치(pouch patch)"이다. 복잡한 구조 때문에 이 패치의 제작에는 개개의 부품을 따로따로 준비한 다음에 추가적인 처리단계에서 결합하여 패치를 형성하여야만 하기 때문에 많은 비용이 소요된다.
EP 0275716호에는 중합체 층에 용해 또는 미세분산되어 있는 하나 이상의 에스트로겐의 동시투여를 위하여 본 발명의 단일층 시스템과는 대조적으로 경피적 이층시스템이 기술되어 있다. 이 경우 감압성 층이 유효 화합물은 별도로 하고 경피흡수를 증진시키는 물질을 함유하고 있다. 중합체 및 감압성 층은 폴리아크릴레이트, 실리콘 또는 폴리이소부틸렌으로 구성될 수도 있다.
EP 0328806호에는 막이 없는 경피적 치료시스템이 기술되어 있는데, 이 경우 상기 시스템의 매트릭스는 폴리아크릴레이트 접착제, 용매, 침투증강제 및 에스트로겐, 그 유도체 및 이들의 혼합물로 구성되어 있다.
WO 87/07138호에는 지지층, 유효물질을 함유하는 매트릭스, 및 분리가능한 보호층에 의하여 커버되는 감압성 접착제로 구성되는 에스트라디올 패치가 기술되어 있다. 매트릭스 및 감압성 접착제의 제조는 균질화, 가스제거, 코팅, 건조 및 개개의 조각으로 분할하는 적용기술에 의하여 많은 비용이 요구되는 공정단계로 수행된다. 일예로서, 지지층은 감압성 접착제로 더욱 코팅되어야 하는데, 이는 추가적인 공정 단계를 의미한다. 개개의 조각(부분)의 결합은 개별적인 공정 단계에서 이루어진다. 따라서, 패치의 전체적인 제조에는 비용이 많이 소요되고 제조하기가 복잡하다.
미국특허 제4624665호에는 저장부(reservoir)에 유효물질을 미소캡슐형태로 함유하는 시스템이 기술되어 있다. 저장부는 지지층과 막사이에 매입되어 있다. 시스템의 외측 가장자리에는 감압성 접착제가 제공되어 있으며, 이 시스템의 구조 및 제조는 유효물질이 미소캡슐화되고 균질상으로 분포되어야 하고 그 다음 추가적인 공정단계에서 지지층과 막 사이에 매입되어야 하기 때문에 매우 복잡하다. 이외에, 그 다음에 이 시스템에는 접착성 있는 가장자리가 제공되고 보호층으로 커버되어야만 한다.
또한, EP 0186019호에는 고무/접착제 수지 덩어리에 수팽창성 중합체를 첨가한 유효물질 패치가 기술되어 있는데, 이 패치로 부터 에스트라디올이 방출될 수 있다. 그러나, 상기 유효물질 패치로부터 에스트라디올의 방출은 너무나도 낮아 치료요건을 충족시키지 못한다.
DE-OS 2006696호에는 침투작용을 갖는 패치 또는 접착붕대가 기재되어 있는데, 이 경우 피임물질이 접착성분 또는 접착필름에 혼합되어 있다. 접착필름은 아크릴레이트일 수도 있다.
따라서, 본 발명은 상술한 종래 기술이 갖고 있는 문제점을 해결하기 위하여 안출된 것으로서, 본 발명의 목적은 충분한 유효물질 방출을 가지며 안정한(즉 재결정이 없는)에스트로겐 - 및/또는 게스타겐 함유 패치를 제공하는데 있는바, 이 경우 상기 유효물질 방출은 보관에 의하여 변화를 받지 않는다.
상기 본 발명의 목적은 울 왁스(wool wax : 양모에서 추출되는 왁스) 및 산화아연을 함유하는 에스트로겐 - 및/또는 게스타겐 함유 감압성 접착제에 의하여 달성됨을 알았다.
따라서, 본 발명의 상기 목적은 독립 청구항에 따른 유효물질 함유 패치에 의하여 달성되며, 종속 청구항은 특히 본 발명의 주제의 바람직한 실시예에 관한 것이다.
울 왁스는 제약 원료로서 공지되어 있으며 약전에 기술되어 있다. 울 왁스는 유효물질에 대한 그의 유리능력 때문에 사용되며, 피부에 의한 충분한 내성 및 물에 대한 흡수능력이 우수하다.
또한 산화아연은 약전에 기술되어 있다. 산화아연은 온화한 소독제로서 작용하며, 소염제이며, 더욱이 약 알칼리성 반응을 나타낸다.
본 발명에 따른 유효물질 패치는 인간 및 수의학에서 치료목적으로는 물론 화장품용으로 사용될 수 있다.
충분한 유효물질 방출을 가지며 재결정이 없는 에스트로겐 - 및/또는 게스타겐 함유 패치는 그의 저장부에 에스트라디올 및 그의 제약적으로 허용 가능한 유도체를 단독으로 또는 게스타겐과 함께, 저장부의 구성 성분 전체에 대하여 1-20중량%의 농도로, 실제로는 1:1 ~ 1:10 의 몰비로 함유한다.
에스트라디올 함유 저장부는 노화방지제, 가소제, 산화방지제 및 흡수증강제로 구성되는 그룹에 속하는 성분을 최소한 하나 함유 할 수도 있다. 적당한 가소제는 당업자에게 공지되어 있으며, 예를들면 DE 3743946호에 기술되어 있다. 일반적으로, 에스트라디올 함유 저장부는 가소제를 5중량%까지의 비율로 함유한다.
더욱이, 저장부는 1중량%의 농도로 노화방지제를 함유하며, 이는 당업자에게 공지된 기술로서 예를 들면 DE 3743946호에 기재되어 있다.
또한, 불투과성 지지층 및 분리가능한 보호층에 대한 재질은 당업자에게 공지되어 있다.
에스트라디올 함유 저장부는 용액, 분산액 및 핫 멜트(hot melt)로부터 제조될 수도 있다.
저장부가 피부에 대한 충분한 고유의 접착성을 갖지 않는 경우, 추가적인 유효물질 없는 감압성 접착제 또는 주위에 감압성 접착성 있는 가장자리가 제공될 수 있다. 이와 같은 방식으로 경피 패치는 전체 적용기간에 걸쳐 피부에 부착되는 것이 보증된다.
경피 에스트라디올 함유 패치의 특히 바람직한 구조는 매트릭스 시스템이며, 이 경우 잘알려진 바와 같이 매트릭스는 유효물질 방출의 제어를 담당하여 하이구치(Higwchi)에 의한 √t-법에 따르게 된다. 그러나, 이는 막 시스템이 특별한 경우에 유리하지 않는다는 것을 의미하는 것은 아니다. 막 시스템에 있어서, 유효물질 방출제어 막은 저장부와 감압성 접착층 사이에 위치된다.
경피 패치의 두께는 치료 요건에 따르게 되며, 치료요건에 따라 조절될 수 있다. 통상적으로, 그 두께는 0.03-0.6mm 이다.
본 발명을 실시예에 의하여 상세히 설명하면 다음과 같다.
실시예 1 :
97.86g의 두로탁(Durotak) 387-2287 용액(50.2g의 고형물질), 4.0g의 울 왁스, 21.56g의 에탄올, 및 10.78g의 초산 에틸 에스테르를 실온에서 2-3시간동안 교반하여 균질화시킨 다음, 2.0g의 에스트라디올 헤미하이드레이트 및 7.0g의 노레틴드론 아세테이트를 가하여 약 1시간동안 교반하였다. 그 다음 16.8g의 산화아연을 가하여 추가적으로 30분동안 교반하였다. 얻어진 유효물질 함유 접착성 덩어리를 분리가능한 보호층(실리콘으로 일측에 코팅된 호스타판 RN 100-훽스트 디아포일)에 코팅하여 대략 80g/㎡의 단위면적당 중량을 갖는 유효물질 함유 저장부를 얻었다. 이 저장부에 불투과성 지지층(폴리에스테르 필름, 두께 19㎛)을 적층시켰다. 그 다음에 16㎠의 유효물질 패치를 천공하였다.
실시예 2 및 3
표1에 명기된 바와같은 원료량으로 실시예 1과 같이 행하여 제조하였다.
인간피부침투를 측정하기 위하여, 피부를 프란츠 세포에 클램프(고정)시켰다. 1.539㎠의 표면을 갖는 에스트로겐 - 및/또는 게스타겐 함유 패치를 상기피부에 접착시키고 유효물질 방출을 37℃에서 측정하였다.(수용체 매질 : 0.9% 염화나트륨 용액 + 0.1% NaN3).
재결정 현상에 대한 시험을 역광선(counterlight)에서 수행하였다.
그 결과는 표2에 나타낸 바와같다.

표에서 보는 바와 같이, 본 발명에 따른 패치는 비교생성물과 비교하여 인간피부를 통해 명백히 향상된 침투를 달성하였다. 동시에 본 발명에 따른 실시예에서는 재결정이 전혀 일어나지 않았다.
본 발명에 따른 유효물질 패치는 인간 및 수의학에서 치료 목적으로는 물론 화장품 용으로 사용될 수 있다.

Claims (15)

  1. 에스트라디올을 단독으로 또는 게스타겐과 함께 인간 또는 동물 피부에 적용시 방출을 제어하기 위한 유효물질 함유 패치에 있어서, 지지층과 ; 상기 지지층에 결합되며, 조합된 유효물질 방출에 적당하며, 감압성 접착제를 사용하여 제조되는 유효물질 함유 저장부와 ; 분리가능한 보호층으로 구성되며, 상기 감압성 접착제가 울 왁스 및 산화아연을 함유하는 것을 특징으로 하는 유효물질 함유 패치.
  2. 제1항에 있어서, 저장부는 1-30중량%의 울 왁스 및 1-30중량%의 산화아연을 함유하는 것을 특징으로 하는 유효물질 함유 패치.
  3. 제1항 있어서, 저장부는 에스트라디올을 단독으로 또는 게스타겐과 함께 1-20중량% 함유하는 것을 특징으로 하는 유효물질 함유 패치.
  4. 제1항에 있어서, 저장부는 에스트라디올을 단독으로 또는 게스타겐과 함께 1:1∼1:10의 몰비로 함유하는 것을 특징으로 하는 유효물질 함유 패치.
  5. 제1항에 있어서, 감압성 접착제는 점착수지를 1-50중량%의 농도로 함유하는 것을 특징으로 하는 유효물질 함유 패치.
  6. 제1항에 있어서, 저장부는 노화방지제, 산화방지제 및 흡수증강제의 그룹에 속하는 하나 이상의 성분을 함유하는 것을 특징으로 하는 유효물질 함유 패치.
  7. 제1항에 있어서, 감압성 접착제는 감압성 용액 접착제, 감압성 분산액 접착제 또는 감압성 핫멜트(hot melt) 접착제 임을 특징으로 하는 유효물질 함유 패치.
  8. 제1항에 있어서, 지지층은 저장부의 성분에 대하여 불투과성 임을 특징으로 하는 유효물질 함유 패치.
  9. 제1항에 있어서, 저장부는 수개의 층으로 구성됨을 특징으로 하는 유효물질 함유 패치.
  10. 제1항에 있어서, 유효물질 방출 제어막이 저장부와 감압성 접착층 사이에 형성됨을 특징으로 하는 유효물질 함유 패치.
  11. 제1항에 있어서, 저장부에는 주위에 감압성 접착성있는 가장자리가 형성됨을 특징으로 하는 유효물질 함유 패치.
  12. 제1항에 있어서, 유효물질 함유 패치의 두께는 0.03-0.6mm 임을 특징으로 하는 유효물질 함유 패치.
