Claims (12)
- 프락토실 트랜스퍼라제를 생산하여 자당을 하기 화학식 1의 프락토올리고당 및 하기 화학식 2의 네오프락토올리고당으로 분해하는 토양 유래 페니실리움 시트리눔(Penicillium citrinum) KCTC 18080P.<화학식 1>G: 글루코스, F: 프락토스<화학식 2>G: 글루코스, F: 프락토스
- 미생물을 이용한 프락토올리고당의 생산방법에 있어서, 제 1항에 따른 미생물 페니실리움 시트리눔 KCTC 18080P를 이용하는 것을 특징으로 하는 프락토올리고당 및 네오프락토올리고당의 생산방법.
- 제 2항에 있어서,(a) 제 1항에 따른 미생물 페니실리움 시트리눔 KCTC 18080P를 1차 종배지에서 26-28℃, 100-200rpm에서 2일 동안 진탕배양하여 활성화시킨 종배양단계;(b) 발효배지에서 26-28℃, 200-500rpm에서 72시간 동안 진탕배양하면서 공기를 0.5-1v/vm 속도로 주입하여 균체를 대량생산하는 단계; 및(c) 원심분리하여 균체만을 회수하고 0.85% 생리식염수로 2회 세척한 후, 자당농도가 처음 삽입한 브릭스(Brix) 60-70인 자당용액에서 35-50℃, 100-300rpm, 및 pH 4-7로 20-50시간 동안 배양하는 단계를 포함하는 것을 특징으로 하는 프락토올리고당 및 네오프락토올리고당의 생산방법.
- 제 2항에 있어서,(a) 제 1항에 따른 미생물 페니실리움 시트리눔 KCTC 18080P를 종배양하는 단계;(b) 상기 배양된 균체를 대량생산하는 단계;(c) 상기 대량생산된 균체를 담체에 혼합하여 비드를 제조한 후, 이를 컬럼에 충진하는 단계; 및(d) 상기 비드가 충진된 컬럼에 자당용액을 통과시키는 단계를 포함하는 것을 특징으로 하는 프락토올리고당 및 네오프락토올리고당의 생산방법.
- 제 4항에 있어서, 상기 종배양은 26-28℃, 100-200rpm에서 2일 동안 진탕배양하는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 대량생산은 공기를 0.5-1v/vm 속도로 주입하면서 26-28℃, 200-600rpm에서 42-72시간 동안 진탕배양하는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 담체는 알지네이트 겔(Alginate gel), 한천, 광교차결합 수지(Photo cross-linkage resin), 아크릴아미드 겔(Acrylamide gel), 카파-카라지난(κ-Carrageenan), 키토산(Chitosan) 및 젤라틴(Gelatin)으로 이루어진 군으로부터 선택되는 하나인 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 담체의 농도가 1 내지 2%인 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 비드는 균체와 담체의 혼합액을 직경 0.5 내지 1mm의 주사바늘을 통하여 1-2% 염화칼슘 수용액에 투하함으로써 제조되는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 자당용액은 브릭스 60 내지 70인 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 자당용액을 35-55℃에서 100-300㎖/ hrs의 유속으로 컬럼에 통과시키는 것을 특징으로 하는 방법.
