Abstract
생물학적으로 분해 가능한 중합체를 바탕으로 생물학적 분해 가능한 용매를 사용해서 그 이외의 독성 용매 없이도 마이크로캡슐을 생산하는 마이크로캡슐의 제조과정에 관해 설명 하였다. 사용된 용매는 일차 에스테르이고 이 에스테르는 C1-C5-알콜과 C1-C5-모노카르본산과 에스테르의 혼합물로 이루어져 있다.
이러한 용매는 첨가물 C1-C5-알콜을 가지고 있을 수 있다. 활성 물질로써 물에 녹거나 녹지 않는 물질이 사용된다. 약제, 펩타이드, 단백질, 효소, 왁찐과 같이 물에 용해되는 활성 물질은 중합체 용액에서 물로된 용액으로써 미세분 된다. 마이크로캡슐은 용액의 분무, 현탁액, 활성 물질과 생물학적 분리 가능한 중합체로부터의 W/O-미세분을 통해 얻어진다. 이 과정은 약제, 왁찐, 효소 제조에 사용된다.

본 발명은 생물학적으로 분해 가능하고 동물학적으로 보아도 위험하지 않은 용매를 사용하여 생물학적 분해 가능한 마이크로캡슐의 제조방법에 관한 것이다. 여기서 쓰여진 개념 '마이크로캡슐'은 다음과 같은 정의에 의해 본래의 마이크로캡슐 뿐만 아니라 미세구역과 미립자를 의미한다: 마이크로캡슐은 1-1000㎛의 곡물 크기 만한 범위 내에 있는 구슬 모양의 입자를 말하며 액체나 고체 형태로 활성 물질을 함유하고 있는 내핵으로 구성되어 있고 캡슐의 벽은 중합체로 되어있다. 미세구역과 미립자의 개념은 1-1000㎛의 곡물 크기 만한 범위 내에 있는 구슬 모양 또는 구슬 모양이 아닌 입자를 말하며 활성 물질이 소위 말해 고체 용매나 현탁액으로써 깊숙이 자리를 잡고 있는 중합체 무형 기질로 되어있다. 약제, 펩타이드, 단백질, 효소, 왁찐과 같이 생물학적으로 아주 활성적이고 민감한 활성 물질들은 주사 용액으로써 많이 사용되었다. 그러나 이러한 활성 물질들은 생물학적으로 짧은 반감기를 가졌고 용액으로 투여되어서는 비활성적이다. 이것은 활성 물질의 계속적 또는 짧은 휴지기를 둔 공급이(장시간 후 해체) 효과적인 치료와 예방의 전제조건이라는 것을 의미한다. 이와 같이 활성 물질의 공급이 오랜 시간 지속되는 것은 활성물질이 생물학적으로 분해 가능한 해체체계로 되어졌기 때문이다. 이러한 해체체계는 마이크로캡슐이나 작은 막대기 모양의 이식조직의 형태 속에서 이루어 질 수 있다.
생물학적 분해 가능한 마이크로캡슐은 과거에 그 효과가 잘 입증되었다. 왜냐하면 이 입자는 통례의 주사 바늘로도 쉽게 투약되어 질 수 있기 때문이다. 잘 알려진 생물학적으로 분해 가능한 약제 해체체계는 유산과 글리콜산의 폴리에스테르의 사용으로 이루어 질 수 있다. (D.H. Lewis, controlled release of bioactive agents from lactide/glycolide polymers, in: Biodegradable polymers as drug delivery systems, M. Chasin, and R. Langer(Eds.), M. Dekker, New York, 1990, pp.1-41).
폴리락타이드와 폴리락타이드 글리콜리드와 같은 폴리에스테르로부터 마이크로캡슐의 제조를 위한 세 과정이 잘 알려져 있다: 건조된 교질의 분리 (형상 분리). 이러한 분리는 예를 들어 용해되지 않는 물질을 첨가하거나 용해 물질을 증발시키거나(solvent evaporation), W/O 미세분으로 용제 추출을 하거나 분무 건조를 할 수 있다.(R. Jalil et al., J. of Microencapsulation 7, 297-325, 1990).이러한 모든 방법은 생물학적 분해 가능한 중합체의 용해를 위해 유기적 용매가 삽입 되어져야만 한다는 공통점을 가지고 있다. 이러한 유기적 용매는 많은 부분들이 이러한 방법을 통해서 분리 되어진다. 중합체로부터 용매의 완전한 분리는 불가능하기 때문에 마이크로캡슐에는 항상 용제의 찌꺼기가 0.01-10 % 정도 잔재한다.(G. Spenlehauer et al., Biomaterials 10, 557-563, 1989); D.H. Lewis et al., PCT WO 89/03678). 특히 지금까지 설명한 세 과정에 사용된 용매는 생물학적으로 분해 되지 않고 부분적으로는 매우 독성이 강하고 환경을 파괴하는 물질들이다(메틸렌클로이드, 클로로포름, 벤졸, 테트라하이드로퍼란, 아세토니트릴, 불소염화탄산수 등등). 이 물질들은 생물학적으로 분해 가능한 해체체계를 하는데 도움이 되지 못 한다. 예를 들어 PS-US 5'066'436에 의해 교질 분리를 통해 마이크로캡슐이 제조 되어진다. 이때 교질 분리에 유기적 용매는 방해 요소가 된다. 중합체를 위한 용매 이외에 본질적인 형상 분리와 마이크로캡슐의 세척과 정화를 위해 또 다른 유기적 용제가 사용된다. 그렇게 제조된 마이크로캡슐은 상대적으로 크고 제조 과정동안 쉽게 응고되고 그것으로 인해 나머지 용매의 분리를 더욱 어렵게 만든다. 용매 증발 또는 용매 추출상의 단점은 이미 언급된 마이크로캡슐의 독물학적으로 위험한 용매의 찌꺼기와 물에 용해되는 약제, 펩타이드, 단백질, 효소, 왁찐과 같은 활성 물질이 물로 미세분 시키는 단계에서 용해되고 그것으로 인해 부분적 또는 전부 포화되는 난점이 있다.
분무 건조는 생물학적 분해 가능한 마이크로캡슐의 제조 방법으로 잘 알려졌고 이 과정은 간단하고 빠르다. 그러나 이 단계에서는 항상 생물학적으로 분해 가능한 유산과 글리콜산의 중합체로 동그랗고 침투성이 없는 입자를 생산하기는 어렵다. 메틸렌콜로이드 속에서 용해된 폴리(d.1-유산)로는 매우 불규칙한 형태, 표면과 열이 비슷한 물질을 가진 마이크로캡슐이 제조된다. 유산과 글리콜산(PLGA)의 공중합체는 마이크로캡슐 제조에 분부 건조를 하지 않는다는 것이 PS - EP A1 315'875에 의해 알려져 있다. 이러한 공 중합체는 포낭 물질로써 중요하다. 왜냐하면 공중합체는 유산과 글리콜산의 동질 중합체로써 유기체 내에서 보다 더 빨리 분해되어진다. 예를 들어 호르몬 치료와 효소 치료를 위해 복용 간격을 둘 수 있는 것은 PLGA-중합체, 특히 각각 50%의 유산과 글리콜산[PLGA50: 50]을 가진 PLGA로 인해 활성 물질을 3-4주 넘게 방치해 둘 수 있다.
분무 건조로 인한 마이크로 포낭 형성에서의 또 다른 난점은 물질들이 포낭 되려는 특성을 가지고 있다는 데에 있다. 중합체 용액(대부분 메틸렌클로이드)에서 용해되는 물질들은 일반적으로 높은 효과와 동질의 활성 물질 분배로 마이크로캡슐이 된다. 그와 반대로 특정한 약제, 펩타이드, 단백질, 효소, 왁찐과 같이 중합체 용액 속에서 용해 될 수 없는 물질들의 마이크로 포낭 형성은 문제시 되고 있다. 이러한 활성 물질이 유기적인 중합체 용액에서 미세화된 형태로 현탁 되어지면 활성 물질의 미세함에 따라 만족스럽지 못한 제조 효과를 보이기도 한다. 물에 용해되는 활성 물질은 종종 먼저 물에 용해되고 이 활성 물질 용액은 그 다음에 중합체 용액 속에서 미세분 된다.(W/O 미세분). 그러나 잘 알려진 용매로는 W/O-미세분이 충분하게 이루어 질 수 없어서 충분한 미세분과 물리학적 안정성이 없다. 그것으로 인해 미세분된 형상들은(유착) 빠른 융합이 이루어져서 좋지 않은 포낭상태가 된다. 그렇기 때문에 마이크로 포낭 단백질이 하루에서 사흘 사이내로 해체되는 것을 볼 수 있다.
이것은 일반적으로 불충분하게 포낭 되어진 활성 물질과 마이크로캡슐의 높은 침투성 때문이다. (H.T. Wang et al., Biomaterials, 11, 679-685 1990). 그 이외에도 물과 섞여지는 아세톤, 테트라하이드로퍼란, 디옥산, 아세토니트릴과 같은 용매는 사용되어 질 수 없다. 왜냐하면 잘 섞여지기 때문에 활성 물질이나 중합체가 침전되거나 응집되어 질 수 있기 때문이다.
본 발명의 목적은 생물학적으로 분해 가능한 중합체가 생물학적으로 분해 가능하고 독물학적으로 위험이 없는 용매 속에서 용해되는 것을 증명해 보이는 것이다. 또한 약제, 펩타이드, 단백질, 효소, 왁찐과 같은 활성 물질, 즉 생물학적으로 활발한 물질들은 분무 건조를 통해 마이크로캡슐을 제조 할 수 있다는 것을 보여주는 것이다.
특허청구 제1항 - 제10항에 의해 제조 방법상의 과제들이 보여진다. 본 발명의 제조 방법은 방법 도표에서 보다 자세히 볼 수 있고 이러한 도표는 각 단계들을 설명하고 있다. 여러 가지 과정 단계를 설명하기 위해 각각 참조 숫자를 사용하였다. 이 숫자는 각 단계를 말한다. 실시예 1-5에는 특허출원용 구조 견본들을 덧붙혀 설명하고 있다.
도표 1은 1단계에서 8단계를 차례차례 설명한 마이크로캡슐 제조 방법을 보여주는 방법 도표이다.
[용해 (Ⅰ)]
이 절차의 출발점은 δ-발레롤락톤, g-카프롤락톤, β-옥시낙산, 유산, 글리콜산의 모노 폴리에스터나 코폴리에스터와 같은 용매들 속에서 용해되는 모든 생물학적 분해 가능한 중합체들이다. 폴리유산이나 폴리글리콜산은 폴리락타이드와 폴리글리콜리드로써 표기된다.
생물학적 분해 가능한 중합체나 이러한 중합체들의 혼합물을 용해하는데는 발명에 따른 용매가 사용되고 이때 첫 용해인 중합체 용해가 일어난다. 이러한 용매에는 일차 에스테르가 있고 이 에스테르는 C1-C5-알콜과 C1-C5모노카르본산과 일차 에스테르의 C1-C5알콜과의 혼합물과 같은 것으로 구성되어 있다. 생물학적으로 분해 가능한 용매에는 예를 들어 개미산에틸에스테르, 개미산프로필에스테르, 개미산부틸에스테르, 초산에틸에스테르, 초산프로필에스테르, 초산이소프로필에스테르, 초산부틸에스테르, 낙산에틸에스테르, 길초산에틸에스테르, 그리고 에탄올, 프로판올, 이소프로판올과 혼합된 에스테르 등이 있다. 이러한 용매 및 용매 혼합물은 글리콜산, 유산, β-하이드록시낙산, δ-벨러릭락톤 과 ε-캐프로익락톤으로된 생물학적 분해 가능한 폴리에스테르 결합을 위해 놀랄만한 높은 용해력을 가지고 있다. 이러한 용매는 단량체 뿐만 아니라 하이드록시카르본산의 공중합체를 50:50, 65:35, 75:25, 85:15 등 여러 가지 상이한 비율로 용해시킬 수 있고 넓은 분자량 범위 속에서 (2000-150000)도 용해시킬 수 있다. 이때 중합체는 따로따로 또는 혼합물로써 용해되어 질 수 있고 용매는 각각 또는 용매 혼합물로써-여러 가지 다른 에스테르로된 혼합물 또는 에스테르와 알콜로된 혼합물로써 사용되어질 수 있다.
[활성 물질의 감입]
포낭 되어질 활성 물질의 생물학적으로 분해 가능한 중합체로의 감입은 용해도에 따라 3가지로 즉 3단계, 5단계, 6단계로 이루어진다. 활성 물질로써 살아있는 유기체 속에서 생물학적 효과-약리학적 효과, 면역학적 효과, 효소적 효과-를 나타내는 물질들이 정의되어진다. 포낭될 활성 물질은 일반적으로 가루 형태이지만 액체 형태로 사용되어 질 수 있다.
[용해 (Ⅱ)]
발명에서 제시된 생물학적 분해 가능하고 에스테르나 C1-C5-알콜, 임의의 에스테르와 알콜의 혼합물로된 용매 속에서 용해되는 활성 물질들은 유기적 활성 물질 용해(두 번째 용해)가 일어나는 데에서 직접 용해되어진다. 이러한 활성 물질 용해는 선호되는 과정 단계이다. 왜냐하면 이런 활성 물질 용해는 손쉽게 이루어질 수 있고 열역학적으로도 안전하기 때문이다.
[혼합]
중합체 용해(첫 용해)와 유기적 활성 물질 용해(두번째 용해)가 함께 이루어져 혼합되어지고 거기에서 기질 '용액'을 얻는다. 여기에서 사용된 용매는 약제나 펩타이드 같은 적은 분자의 수많은 활성 물질을 위한 높은 용해력을 가지고 있다. 이러한 용매의 좋은 특성은 활성 물질과 생불학적으로 분해 가능한 중합체를 동시에 용해시킬 수 있다는 것이다. 물론 이때에 같은 용매나 생물학적으로 분해 가능한 용매의 결합이 사용되어져야 한다. 이 절차는 8단계에서 계속 되어진다.
[현탁]
물 속에서도 발명에서 거론된 용매 속에서도 충분히 용해되지 않는 활성 물질들은 생물학적 분해 가능한 용매와 함께 생물학적 분해 가능한 중합체가 가지고 있는 중합체 용액 속에서 초음파를 통해 현탁 되어지고 거기에서 기질 '현탁액'을 얻는다.
높은 효과를 내기 위해서 활성 물질은 미세화 되어진다. 입자는 일반적으로 활성 물질을 위해 10μm보다 더 작아야 한다. 절차는 8단계에서 계속 되어진다.
[용해 (Ⅲ)]
물에는 잘 녹지만 발명에서 거론된 용매에는 잘 녹을 수 없는 약제, 펩타이드, 효소, 단백질, 왁찐같은 활성 물질들은 물로된 매개물에서는 가능한 한 작은 부피 속에서 용해되어지는 장점을 가지고 있다. 물로된 매개물은 물 이외에 완충소금, 텐사이드, 고정물질, 방부제와 같은 첨가물을 함유하고 있다.
[미세분]
6단계에서 얻어진 물에 의한 활성 물질 용액은 발명에서 거론된 용매 속에서 생물학적으로 분해 가능한 중합체가 지니는 중합체 용액에서 가장 미세하게 미세분되어지고 그것을 통해 기질 'W/O-미세분'을 얻는다. 물에 의한 활성 물질 용액은 50%의 부피와 작은 열소비로 중합체 용액 속에 들어간다. 미세분은 단순하게 흔들어서도 이루어지고 미세분기나 초음파를 이용하면 더 좋은 효과를 볼 수 있다.
선호되는 미세분 방법은 10-300 초 동안 필요한 경우 더 길게 50-400W의 초음파를 사용하는 것이다. 이러한 중합체 용액에서 물에 의한 활성 물질 용액의 미세분은 놀랄만하게도 텐사이드나 표면 수축 완화제의 첨가 없이도 많은 시간동안 변화 없이 지속되고(유착은 아니다) 분무 과정 동안에는 계속적으로 미세분되어지지 않는다. 발명에서 거론된 용매는 물에 의한 활성물질 용액으로 W/O-미세분이 이루어지지만 이 미세분 과정에 용매로 일반적인 메틸렌클로이드를 사용하면 미세함과 견고함이 없다. 절차는 8단계에서 계속 되어진다.
[분무 건조]
4,5,7, 단계에서 얻어진 기질 '용액' '현탁액' 'W/O-미세분'은 분무 건조를 통해 마이크로캡슐로 제조될 수 있고 이때 미니 스프레이 건조기 190로도 충분하다. 분무 건조를 통해 제조된 마이크로캡슐은 일반적으로 직경이 1-50 ㎛이고 최고 100㎛이며 쉽게 주사바늘을 통과할 수 있다. 마이크로캡슐의 적재 정도는 0-50%이며 적을 때는 0-20% 사이이다.
이러한 방법으로 제조된 마이크로캡슐은 또 다른 과정을 통해 - 세척 과정, 여러 가지 상이한 곡물 크기로의 분류, 진공 상태에서의 마지막 건조, γ방사선을 통한 살균 - 처리된다.
여기서 서술된 8단계 과정은 발명에 거론된 용매의 높은 용해력과 물에 의한 활성 물질 용액의 쉬운 미세분력을 근거로 제조 효과가 크고 상이한 용해성을 가진 활성 물질들을 같은 종류의 활성 물질로 분류한다. 사용된 중합체, 적재 정도와 실질적 첨가물(해체 조정제)에 따라서 활성 물질은 하루에 해체될 수도 있고 일년이 걸릴 수도 있다.
일반적으로 할로겐화 되었거나 그렇지 않은 유기적 용매와는 반대로 발명에서 거론된 생물학적 분해 가능한 용매는 폴리(d.1.-유산) 또는 폴리유산글리콜산으로 분무 건조를 통해 마이크로캡슐을 제조할 수 있게 한다. 획득한 생산물자들은 규칙적이고 구슬 모양의 입자 형태를 띠고 반질 하고 다공질이 아닌 표면을 가지고 있다. 입자들의 유착, 응집, 변형은 거의 보이지 않는다. 발명에 거론된 용매는 동물학적으로 고려하지 않아도 되는 장점을 가지고 있다. 마이크로캡슐에 바탕이 되는 중합체와 마찬가지로 발명에서 거론된 용매는 유기체 속에서 아주 작고 무독성이고 일부는 생리학적인 구성성분으로 분해되다. 이러한 분해는 일반적으로 가수 분해를 통해 일어나며 효소적인 에스테르젠이나 수소 제거를 통해 일어날 수 도 있다. 발명에 거론된 용매와는 달리 일반적으로 사용되는 용매는 유기체 내에서 독성의 변형 물질로 형태가 변한다. 암을 유발시킬 가능성과 상이한 독성의 효과에 대한 의구심과 함께 유기체내에서 호흡독인 탄산일산화물이 메틸렌클로이드로부터 생성된다.
실시예 1은 단백질을 포함한 마이크로캡슐의 제조를 서술한 것이다: 0.09g의 소혈청알부민(Fluka, CH-Buchs)은 2.16g의 물에 용해되었다. 물에 의한 용해는 초음파발진기의 도움으로 3.0g의 폴리(d.1.-유산) (Resomer R202. 베링어잉엘하임)용액 속에서 57.0g의 초산이 소프로필에스테르로 미세분 되었다. 미세분은 초음파기 속에서(Vibracell, vc375, Sonics and Materials, Danbury, CT) 30초 동안 두 번 260W의 에너지로 냉각 하에 이루어진다.
미세분은 실험실 분무 건조기 내에(Mni Spray Dryer,190 Laboratorien, CH-Flawil) 다음과 같은 조건하에서 분무되어진다: 처음 온도: 55도, 분무량: 400 단계, 흡출기: 15 단계, 분무 속도: 1분에 2.4ml. 마이크로캡슐은 하얗고 자유로이 떠다니는 가루이다. 수확물; 1.8g(이론의 58%) 활성 물질 함량: 2.9%(이론의 100%)
실시예 2는 단백질을 함유한 마이크로캡슐의 제조를 설명한다: 0.09g의 소혈청알부민은 2.16g의 물에서 용해되어졌다. 물에 의한 용해는 초음파 발진기의 도움으로 (30초 동안 260W) 3.0g의 폴리(d.1-유산) (Resomer R202, 베링어잉엘하임)의 용액 속에서 57.0g의 초산에틸에스테르로 미세분 되어졌다. 미세분과 분무 건조는 보기 1과 동일한 조건하에서 이루어진다. 수확물: 1.6g(이론의 52%), 활성 물질함량: 2.9%(이론의 100%)
실시예 3은 왁찐을 내포하고 있는 마이크로캡슐의 제조를 설명한다: 0.100 g의 파상풍 톡소이드는 2.0ml의 물에 용해되었다. 이 용액은 초음파발진기의 도움으로 5.0g의 폴리(d.1-유산글리콜산) (Resomer R502,베링어잉엘하임)의 용액 속에서 100.0g의 개미산에틸에스테르로 미세분 된다. 이렇게 미세분된 것은 30초 동안 두 번 210W의 초음파에너지로 냉각 하에서 이루어진다. 미세분은 분무건조기로 다음과 같은 조건하에서 분무되어진다: 처음 온도: 55도, 분무량: 500 단계, 흡출기: 17.5 단계, 분무 속도: 1분에 2.4ml. 마이크로캡슐은 자유로이 날아다니는 하얀 가루로 쌓였다. 수확물: 0.7g(이론의 40%), 단백질량:1.82%(이론의 93%)
실시예 4는 효소를 내포하고 있는 마이크로캡슐의 제조를 설명한다: 75.8U/mg(Fluka, CH-Buchs)의 활성을 가진 메어레티히의 15mg의 괴산화물은 0.6ml 물에서 용해되어진다. 이 용액은 30초 동안 375W의 초음파를 이용해서 냉각하에 1.5g의 폴리(d.1-유산)용액에서 30.0g의 개미산에틸에스테르로 미세분 된다.
이러한 미세분은 실험실 분무 건조기로 다음과 같은 조건하에서 분무되어진다: 처음 온도: 47도, 분무량: 450 단계, 흡출기: 17단계, 분무 속도: 1분에 1.6ml. 마이크로캡슐은 자유로이 떠다니는 하얀 가루로 쌓인다. 수확물: 0.92g(이론의 61%), 효소활성: 0.177 U/mg 마이크로캡슐(이론의 23.4%)
실시예 5는 약제를 내포하고 있는 마이크로캡슐의 제조를 설명한다: 30mg의 프로스타글라딘 E2(Fluka, CH-Buchs)와 6.0g의 폴리(d.1-유산글리콜산)는 따로따로 60g의 개미산에틸에스테르에서 용해되어졌다.
이 용액은 다음과 같은 조건하에서 분무되었다: 처음 온도: 45도, 분무량: 420단계, 흡출기: 15 단계, 분무 속도: 1분에 2.4 ml. 마이크로캡슐은 자유로이 날아다니는 하얀 가루로 쌓였다. 수화물: 4.2g(이론의 70%), 적재도: 4.99㎍/㎎ 마이크로캡슐(이론의 100%)
실시예 6은 호르몬을 내포하고 있는 마이크로캡슐의 제조를 설명한다: 17-β-에스트라디올 25g은 물이 없는 125ml의 에탄올과 초산에틸에스테르 312.5ml의 혼합물 속에서 용해되어진다. 100g Resomer RG503은 812.5ml의 개미산에틸에스테르에서 용해되어진다. 두 용액을 분무 전에 저어주고 두 용액은 고정된 현탁액이 형성되면서 합쳐진다.
현탁액은 다음과 같은 조건하에서 미니 분무건조기(Fa.NIRO As, 코펜하겐)로 분무되어진다: 처음 온도: 67도, 분무속도: 1시간에 0.91. 마이크로캡슐은 하얀 가루로 쌓였고 따로따로 떨어진 둥근 입자이다. 수확물: 80g(이론의 80%), 적재량: 70.2㎍/㎎ 마이크로캡슐(이론의 85.1%)
실시예 7은 호르몬을 함유하고 있는 마이크로캡슐의 제조를 설명: 17-β-에스트라디올 10g은 물이 없는 100ml의 에탄올과 250ml의 초산에틸에스테르의 혼합물 속에서 용해된다. 90g의 Resomer RG752(베링어잉엘하임)는 650ml의 개미산에틸에스테르에서 용해된다.
이 두 용액은 저어주면 하나가 되고 다음과 같은 조건하에서 미니 분무건조기로 분무되어진다: 처음 온도: 70도, 분무 속도: 1시간에 0.9. 마이크로캡슐은 하얀 가루로 쌓이고 따로따로 떨어진 둥근 모양의 입자이다. 수확물:100g(이론의 85%), 적재도: 89.6 ㎍/㎎마이크로캡슐(이론의 89.6%)
실시예 8은 합성의 왁찐을 함유하는 마이크로캡슐의 제조를 설명: 파상풍 톡신 947-967의 펩타이드 연속으로 된 0.02g의 합성 단백질은 1.0ml의 물에 용해된다. 이 용액은 초음파 발진기의 도움으로 2.0g 폴리(d.1-유산글리콜산) (Resomer Rg752, 베링어잉엘하임)의 용액 속에서 40.0g의 개미산에틸에스테르로 미세분 되었다. 이러한 미세분은 30초 동안씩 2번 210W의 초음파 에너지로 냉각 하에서 이루어진다.
미세분은 실험실 분무 건조기로 다음과 같은 조건하에 분무된다: 처음 온도: 50도, 분무량: 450 단계, 흡출기:14 단계, 분무 속도: 1분에 2.6 ml. 마이크로캡슐은 자유로이 날아다니는 하얀 가루로 쌓인다. 수확물: 1.12g(이론의 56%), 단백질량: 0.66%(이론의 0.6%)
생물학적 분해 가능한 용매가 제시되었다는 것이 이 발명의 주요점이고 그것으로 제조된 마이크로캡슐은 무독성이다. 마이크로캡슐은 용액의 분무, 현탁액, 활성물질과 중합체의 미세분을 통해 이루어진다. 활성 물질로서는 물에 용해되는 것과 안되는 물질들이 적합하다. 펩타이드, 단백질, 왁찐과 같이 물에 용해되는 활성 물질은 중합체 용액에서 물에 의한 용액으로써 미세분 되었다. 약제, 왁찐, 효모의 조제에서 사용된 과정도 있다.
Claims (7)
- 생물학적으로 분해 가능한 중합체와 활성물질을 함유하는 마이크로캡슐의 제조방법으로서, a) (유산, 글리콜산, β-옥시낙산, δ-발레롤락톤, g-카프롤락톤으로부터 선택되는) 생물학적으로 분해 가능한 중합체를 비독성의 생물학적으로 분해되는 용제에 용해시키고, b) 하나의 활성물질을, 단계 a)에서 획득한 생물학적으로 분해 가능한 중합체의 용액에, 생물학적으로 분해 가능한 용제 중의 활성물질의 용액의 형태로나 활성물질의 미분화 분발의 형태로 또는 수성 매체중의 활성 물질의 용액의 형태로 혼합하며, c) 단계 b)에서 획득한 용액, 현탁액 또는 w/o-미세분의 분산을 분무 건조시키는 것으로 이루어지는 것을 특징으로한 마이크로캡슐의 제조방법.
- 제1항에 있어서 생물학적으로 분해 가능한 용제로서 C1-C5-알콜과 C1-C5-모노카르본산, C1-C5-알콜의 에스테르가 사용되는 것을 특징으로한 마이크로캡슐의 제조 방법.
- 제1항에 있어서 생물학적 분해 가능한 용제로서 개미산 에틸에스테르가 사용되어지는 것을 특징으로한 마이크로캡슐의 제조방법.
- 제1항에 있어서 생물학적 분해 가능한 용제로 초산에틸에스테르 또는 초산이소프로필에스테르가 사용되어 지는 것을 특징으로한 마이크로캡슐의 제조 방법.
- 제1항에 있어서 생물학적 분해 가능한 용제에 용해되지 않는 활성 물질이 수성 매체에 용해되며 획득한 용액은 상기 생물학적 분해 가능한 용매에 분산되는 것을 특징으로 한 마이크로캡슐의 제조방법.
- 제1항에 있어서 활성 물질로서 약제, 펩타이드, 단백질, 효소, 백신이 사용되는 것을 특징으로한 마이크로캡슐의 제조방법.
- 제1항에 있어서 활성 물질은 고체나 액체 형태로 사용되는 것을 특징으로한 마이크로캡슐의 제조방법.
