Description

【発明の詳細な説明】 産業上の利用分野 本発明はカプセル剤、特にゼラチンカプセル剤に関する
ものであり、又その内容物に関連を有する。
従来の技術 多（の場合において活性成分を口腔内に長時間保持する
ことは有利である。例えば殺菌剤、咽頭炎治療薬、風邪
薬及びセキ止め薬並びに歯科用組成物がこれに該当する
。消化管内腔、消化管壁を回避するため又は肝臓への最
初の通過を除くために有効薬の迅速作用を望む場合に頬
側経路又は舌下経路による薬物投与が又屡々所望される
。噛むことによって薬物を放出する軟性ゼラチンカプセ
ルに含有させた粉末又は液体としての単−投与形の製薬
組成物の提供は公知である。けれどもカプセル内容物が
嘩下の際にほとんど直ちに失われて効能を発揮しないこ
とに問題がある。
日が”′　しようとする問題点 上記の問題に対するひとつのアプローチは咀嚼に適する
不溶性成分を添加することによってゼラチンに咀噂性を
与え、かようにして咀嚼用軟ゼラチンカプセルを提供す
ることである。けれどもこのアプローチはこの問題、即
ちカプセル内容物を唖下して大きな軟ゼラチン殻が口内
に残るという問題を克服しない。
問題点を解決するだめの手段 本発明はこの問題の解決のために頬腔の流体と接触した
ときに充分な量の活性成分を与え得る単−投与形状及び
頬腔の中で有効時間の長さを保持し得る形状を提供する
。
従って本発明は下記諸成分即ち（ａ）頬腔から吸収され
るか又は頬腔作用性を有する成分を無毒性担体と共に有
する半固体組成物とら）咀嚼増強剤とを有する咀嚼用組
成物であってこの咀嚼用組成物が１００℃以下の温度で
液状である該咀嚼用組成物を含有する硬性ゼラチンカプ
セル剤を提供する。
好ましくは該硬性ゼラチンカプセルは下記諸成分即ち（
ａ）頬腔から吸収されるか又は頬腔作用性を有する成分
、わ）無毒性半固体状担体及び（Ｃ）咀嚼増強剤を有す
る咀嚼用組成物であってこの咀Ｉｌ＃用組成物が１００
℃以下の温度で液状である該咀咀用組成物を含有する硬
性ゼラチンカプセル剤である。
担体は例えば１種又は複数種のリピドであるか又は水溶
性物質例えばポリマーから成り、それらは天然物又は合
成物或は１種又は複数種の該物質の混合物であり得る。
担体の融点は３６〜６０℃の範囲内にあることが好まし
い。好適ポリマーは例えば適宜の融点と硬性とをもつポ
リエチレングリコール類（ＰＥＧ）の群から、ガテフォ
ス社（Ｇａｔｔｅｆｏｓｓｅ）から供給される付形剤の
ゲルシル群（Ｇｅｌｕｃｉｒｅ　ｒａｎｇｅ）から、又
はグイナミドノーペル社（Ｏｙｎａｍｉｔ　Ｎｏｂｅｌ
）から供給される付形剤例えばウィテプゾル（Ｗｉｔｅ
ｐｓｏｌ）　（商標名）及びソフテサン（Ｓｏｆｔｅｓ
ａｎ）　（商標名）から選択され得る。ＰＥＧは好まし
くは２００〜ｉｏ、　０００の範囲、更に好ましくは　
１０００〜２０００の範囲の分子量を有する。特に好適
なポリマーは分子１１５４０を有する。
池の物質も又担体として、単独で又は既知担体系と組合
せて使用され得る。かような物質は例えば植物性及び動
物性の任意に水素化され得る油脂；炭化水素；炭素原子
数約１２〜約１８の脂肪族アルコール及び酸；及びそれ
らのエステル並びに塩を包含する。
活性成分が液体である場合には固体担体を用いて該活性
成分を先ず半固体組成物にする。かような担体はコロイ
ド性担体、例えばコロイド性二酸化ケイ素、又はセルロ
ース誘導体例えばメチルセルロース又はエチルセルロー
スでアリｆ８　ル。他ノ適切な固体担体は当業技術者に
より容易に認識されよう。
頬腔作用剤は頬腔への適用が望まれる薬剤であって例え
ば歯科治療活性剤におけるような局所剤又は咽頭炎の治
療用又は口蓋或は歯肉に不快を起す症状の治療用の製剤
であってよい。又は核剤は頬腔粘膜を経て吸収され心臓
に影響する疾病に悩む患者に起る如き徴候を急速に救済
するように企図され得る。
就中、本発明は心臓作用性薬物例えばニフェジピ：／　
（ｎｉｆｅｄｉｐｉｎｅ）、ニトログリセリン（ｎｉｔ
ｒｏ−ｇｌｙｃｅｒｉｎｅ）、イソソルバイト　シナイ
トレート（ｉｓｏｓｏｒｂｉｄｅ　ｄｉｎｉｔｒａｔｅ
）及びプロパノロール（ｐｒｏｐａｎｏｌｏｌ）の投与
の際に特に有用である。本発明は又鎮痛用薬物例えばピ
コプレノルフィン（ｂｕｐｒｅｎｏｒｐｈｉｎｅ）　　
、ペンクシシン（ｐｅｎｔａｚｏｃｉｎｅ）及びフエン
ゾシン（ｐｈｅｎｚｏｃｉｎｅ）　；ホルモン例えばオ
キシトシン、エステローケン類及びメチルテストステロ
ン；鎮静薬例えばトリアゾラム（ｔｒｉｚｏ−１ａｍ）
及び７　／ｌ／ラゼパム（ｆ　ｌｕｒａｚｅｐａｍ）の
投与時にも有用である。
本発明の使用により硬性ゼラチンカプセル殻の中へ充填
するための組成物の中へ９５重量％までの活性成分を導
入することが可能である。固体活住物質の場合に活性物
質の好適範囲は２０重量％以下である。
カプセル中の組成物は１０００ｍｇ以下の活性成分を含
み得るし、典型適には１μｇ〜２５０ｍｇの活性成分含
む。
本発明で使用される組成物は可成りの長時間にわたり口
腔内に保持されるので活性成分は最高効果を奏し得られ
る。本発明のカプセル剤は口腔内へ導入されると噛まれ
るか又は頬腔の分泌唾液中に溶ける。いずれの場合にも
カプセル殻は直ぐに分散して咀嚼用塊状物を残すがこれ
は容易に口腔内に保持される。それは噛まれることによ
って急速に分散するか又はより長い間吸収されることが
必要である場合に舌下にとどまり得る。この組成物が口
腔内に保持されるためには該組成物中に咀■６増強剤即
ち１種又は複数種の各種の安定化剤又はｇ２剤てあり得
る核剤の添加が必要である。該咀嚼増強剤は例えばガム
即ち天然、合成及び半合成ガム、例えばキサンタンガム
、トラガントガム、アカシアガム、グアルガム、又はロ
ーカストビンガム；セルロース誘導体例えばナトリウム
カルボキンメチルセルロース又はヒドロキシプロピルメ
チルセルロース（）ＩＰＭｃ）；その他の咀嚼増強剤例
えばアルギネート、ゼラチン、ポビドン（ｐＯ−ｖｉｄ
ｏｒ＋ｅ）　、ペクチン、ビーズワックスを包含する。
好ましいガムはキサンタンガムであり、マルチンデール
（Ｍａｒｔｉｎｄａｌｅ）におけるものが参照される。
この組成物のその他の成分にはゲル化剤、香味料及び吸
収増強薬が包含される。
本発明による組成物の調製に当り好適な担体はポリエチ
レングリコールから選択される。多くの製薬学的化合物
はＰＥＧに可溶であって唾液と接触すると急速に放出さ
れる。
活性成分を咀嚼増強剤及びその他の所望成分例えばゲル
化剤又は香味料と共に担体と混合し、この混合物を攪拌
し均質混合物を得るために必要な。
場合には加熱する。この混合物の適当量を標準的手段に
よりカプセル殻の中へ導入する。
咀咀増強剤は本発明による組成物を咀嚼可能とならしめ
て活性成分が奏効し又は吸収されるために充分な時間だ
け頬腔内に保持されることを助ける。
満足され得る咀嚼性と保持性とを与えるために要する安
定化剤又は懸濁剤の量は臨界的であることが見出された
。２５％までの量を使用し得るけれども本発明の好適態
様において本組成物は２〜１５重量％、最も好ましくは
約５重量％のキサンタンガムを含有する。
本発明のカプセル剤について、咀嚼を−そう満足ならし
めるために活性物質又は担体系の味を隠してしまう香味
料の添加が望ましい。
最も好ましい香味料は０．２％の量のペパーミントであ
るがこれは試験時に使用された担体系の味を隠すのに最
良であったからである。該香味料は又カプセル剤の中へ
拡散する傾向をもち、ゼラチン臭を隠蔽することを助け
る。
他゛の適切な香味料はバニラ及びオレンジを包含する。
柑橘果実香味料を使用する場合には香味増強のためにク
エン酸添加が好ましい。
実施例 以下の非限定的諸例によって本発明を更に充分に例示す
る。カプセルの中へ充填される咀嚼用組成物は下記諸方
法のうちのいかなる方法によっても調製される。基礎的
調製法は担体系の液化、次に他の付形剤との混合を包含
する。次にこの混合物を溶融下にカプセル中へ充填する
。
下記の系を使用し得る： （１）水浴上のビーカー内で該混合物を調製してから注
射器によりカプセルの中へ手作業で充填する。
〔２）熱板（ホットプレート）上のビーカー内で該混合
物を調製してから自動式ハイパーポンプ（Ｈｉｂａｒ　
ｐｕｍｐ）によりカプセルへ充填する。
（３）エラノコ８型（ｌｌｉｌａｎｃｏ　Ｍｏｄｅｌ　
３）カプセル充填機との共用でエランコ　ロトプ８型（
Ｅｌａｎｃ。
ＲＯｔＯｐ　８　）カプセル充填機を使用する。
〔４）不銹鋼製ジャケット付容器の中で該混合物を調製
してからハロヘフリゲル式（Ｈａｒｒｏ　Ｈｏｅｆｌｉ
−ｇｅｔ）　Ｋ　Ｆ　Ｍ充填機を使用する。
（５）該混合物調製のためにギスチ式（Ｇｕｉｓｔｉ）
混合器を用い、この混合物をヘフリゲル及びカルブ式（
Ｈｏｅｆｌｉｇｅｒ　ａｎｄ　Ｋａｒｇ）　Ｇ　Ｋ　Ｆ
　１５０　ＯＬ充填機の使用でカプセル中へ充填する。
上記諸方法は各バッチについて各種の寸法の咀嚼用カプ
セル剤製造のために使用され得る。

Claims (14)

    【特許請求の範囲】
  1. （１）下記諸成分即ち（ａ）頬腔から吸収されるか又は
    頬腔作用性を有する成分を無毒性担体と共に有する半固
    体組成物と（ｂ）咀嚼増強剤とを有する咀嚼用組成物で
    あってこの咀嚼用組成物が１００℃以下の温度で液状で
    ある該咀嚼用組成物を含有する硬性ゼラチンカプセル剤
    。
  2. （２）下記諸成分即ち（ａ）頬腔から吸収されるか又は
    頬腔作用性を有する成分、（ｂ）無毒性半固体状担体及
    び（ｃ）咀嚼増強剤を有する咀嚼用組成物であってこの
    咀嚼用組成物が１００℃以下の温度で液状である該咀嚼
    用組成物を含有する硬性ゼラチンカプセル剤。
  3. （３）咀嚼増強剤が安定化剤又は懸濁化剤である特許請
    求の範囲第１又は２項記載のカプセル剤。
  4. （４）咀嚼増強剤がガムである特許請求の範囲第３項記
    載のカプセル剤。
  5. （５）ガムがキサンタンガムである特許請求の範囲第４
    項記載のカプセル剤。
  6. （６）担体が分子量範囲２００〜１０，０００を有する
    ポリエチレングリコールである特許請求の範囲第１〜５
    項のいずれか１項記載のカプセル剤。
  7. （７）担体が分子量範囲４００〜４，０００を有するポ
    リエチレングリコールである特許請求の範囲第１〜５項
    のいずれか１項記載のカプセル剤。
  8. （８）ポリエチレングリコールが分子量範囲１０００〜
    ２０００を有する特許請求の範囲第７項記載のカプセル
    剤。
  9. （９）咀嚼用組成物が９５重量％以下の活性成分を含有
    する特許請求の範囲第１〜８項のいずれか１項記載のカ
    プセル剤。
  10. （１０）咀嚼用組成物が２０％以下の固体活性成分を有
    する特許請求の範囲第９項記載のカプセル剤。
  11. （１１）活性成分が心臓作用性薬物又は鎮痛薬である特
    許請求の範囲第１〜１０項のいずれか１項記載のカプセ
    ル剤。
  12. （１２）頬腔から吸収されるか又は頬腔作用性をもつ成
    分が液体であり担体が固体である特許請求の範囲第１項
    記載のカプセル剤。
  13. （１３）担体がコロイド状二酸化ケイ素又はセルロース
    誘導体である特許請求の範囲第１２項記載のカプセル剤
    。
  14. （１４）香味料を更に含有する特許請求の範囲第１〜１
    ３項のいずれか１項記載のカプセル剤。
