Abstract

NEW MATERIAL:An N1-(1-Alkoxy-4-hydroxybutyl)-5-fluorouracil of formula I: (R1' is alkyl group; R2 is vinyl or alkoxycarbonylmethyl group).
USE: An oncostatic drug and an intermediate for synthesis of N1-(2-tetrahydrofuryl)-5-fluorouracil, which is an anti-malignant solid cancer drug.
PROCESS: A 2,4-bistrimethylsilyl derivative of 5-fluorouracil (FU) is reacted with a compound of formula II in the presence of a Lewis acid, e.g. tin tetrachloride, etc. at -50W10°C to form a compound of formula III, which is hydroborated with a diborane when R2 is a vinyl group. The borane compound is then reacted with H2O2 to give the compound of formula I. When R2 is analkoxycarbonyl methyl group, a compound of formula III is reduced in the presence of an alkalimetal borohydride, e.g. potassium borohydride, etc. to form the compound of formula I.
