Abstract
PURPOSE: The condensation reaction between 3,5-disubstituted-2-deoxyribofuranosyl halide and a pyrimidine derivative is effected in the presence of a compound having active hydrogens of lower than 19 pKa to prepare title compound in high yield.
CONSTITUTION: The condensation reaction between 3,5-disubstituted-2-deoxy- ribofuranosyl halide of formula I (R1 is protected hydroxyl; X is halogen) and a pyrimidine derivative of formula II (R2 is organic silyl, trifluoromethyl) is effected in the presence of a compound having active hydrogens of less than 19 pKa to prepare 3',5'-disubstituted-2'-deoxy-β-uridine of formula III. Further, the protecting group for hydroxyl group in the compound of formula III is eliminated to give 2'-deoxy-β-uridine of formla IV.
USE: 2'-Deoxy-β-uridine of formula IV is useful as carcistatic agent, antiviral agent, radiation sensitizer, etc.
